A simple and sensitive method for detecting diprophylline (DPP) was developed based on the fluorescence quenching of glutathione-capped CdTe quantum dots (GSH–CdTe QDs) by using diprophylline in a KH 2 PO 4 –Na 2 HPO 4 medium. Parameters affecting the quenching efficiency, including types and pH of buffer solutions as well as temperature, reaction time, adding sequence, and interfering substances, were investigated and optimized. In optimum conditions, the calibration plot of the quenched fluorescence intensity F 0 / F with a DPP concentration range of 1.67×10 –6 mol L −1 to 1.33×10 –5 mol L −1 was linear. The detection limit (with signal to noise ratio of 3) for DPP was 2.24×10 –7 mol L −1 . The proposed method was successfully applied for detecting DPP in human serum. The recovery of the method was in the range of 87.41% to 117.94%. Finally, the possible quenching mechanism of GSH–CdTe QDs and DPP was also discussed.

The hydrolysate of β-casein generated by bromelain exhibits potent DPP-IV inhibitory activity. • β-Casein hydrolysate significantly increased glucose consumption in insulin-resistant HepG2 cells. • MPIQAF, a β-casein-derived Xaa-Pro motif peptide, inhibits DPP-IV and alleviates glucose metabolism disorders. Bioactive peptides have shown therapeutic potential for type 2 diabetes by inhibiting dipeptidyl peptidase-IV (DPP-IV). In this study, β-casein (β-CN) hydrolysates were prepared using enzymatic hydrolysis and characterized spectroscopically, followed by activity analysis. The results showed that the particle size and surface hydrophobicity of β-CN were significantly reduced after enzymatic hydrolysis ( p < 0.05). Fluorescence spectroscopy revealed a red shift in the maximum emission wavelength, and secondary structure analysis indicated a conformational transition toward random coils and β-turns. Bromelain hydrolysate inhibited DPP-Ⅳ activity and significantly increased glucose consumption and glycogen synthesis in IR-HepG2 cells ( p < 0.05). A total of 439 peptide sequences were identified from the bromelain hydrolysate, among which 34.40% had a bioactivity score above 0.5, and most showed potential DPP-IV inhibitory ability. Through in silico analysis, the Xaa-Pro-type peptide MPIQAF was screened and showed strong binding affinity with DPP-IV, suggesting its hypoglycemic potential. MPIQAF was further synthesized and experimentally validated, and its direct DPP-IV inhibitory IC 50 was determined to be 2.832 mM. The peptide could ameliorate insulin resistance-induced glucose metabolism disorders in a dose-dependent manner and significantly reduce DPP-IV levels (p < 0.05). In summary, the bromelain hydrolysate, as a multi-peptide mixture, exhibited stronger overall DPP-IV inhibitory activity and holds promise for direct application as a functional food ingredient. The purified peptide MPIQAF, although a weak direct inhibitor, provides a valuable lead compound for mechanistic studies and future structural optimization toward glucose regulation. In this work, a diphenyl substituted diketopyrrolopyrrole (DPP) and its two derivatives end-capped with fluorine and n -butyl respectively, namely PDPPP, FPDPPPF, and RPDPPPR, are designed and synthesized. The resulting molecules exhibit similar energy structures, i.e. both relatively narrow optical band gaps (1.75–1.79 eV) and deep highest occupied molecular orbital (HOMO) energy levels (−5.18 to −5.25 eV), implying that all of them are potentially good electron donors for organic solar cells (OSCs). However, three molecules show different photovoltaic performances when they are blended with [6,6]-phenyl-C 61 -butyric acid methyl ester (PC 61 BM) to fabricate OSCs: the RPDPPPR-based device gives the highest power conversion efficiency (PCE) of 1.59%, whereas the PCEs of PDPPP and FPDPPPF-based OSCs are 0.46% and 0.55%, respectively. Through atomic force microscopy (AFM), X-ray diffraction (XRD) and space charge limited current (SCLC) characterizations, the prominent role of end-groups in the photovoltaic properties of DPP derivatives is disclosed: terminal alkyl chains in RPDPPPR can promote molecular crystallization and lead to the formation of finer phase-separation domains in the blended film, which are in favor of charge generation and transportation in the photovoltaic devices. Thus, RPDPPPR provides the best photovoltaic property among three DPP molecules.

In this paper, a non-fullerene electron acceptor , SF(DPPFB) 4 , which owns a spirobifluorene core and four diketopyrrolopyrrole (DPP) arms end capped by 4-fluorobenzene, is designed and synthesized for solution-processable organic solar cells (OSCs). SF(DPPFB) 4 shows similar absorption bands as those of its non-fluorinated parent compound, SF(DPPB) 4 . However, the terminal fluorine atoms reduce the energy levels of SF(DPPFB) 4 , especially, its lowest unoccupied molecular orbital level decreases by 0.04 eV than that of SF(DPPB) 4 , which enlarges the energy offset between the electron donor and acceptor, favorable for the dissociation of excitons in OSCs. Moreover, the fluorination improves the electron mobility of SF(DPPFB) 4 . Thus, the OSCs with poly(3-hexylthiophene) as the electron donor and SF(DPPFB) 4 as the electron acceptor can provide a maximum power conversion efficiency of 4.42%, with a lifted short-circuit current ( J sc ) of 8.48 mA cm −2 .

Background: Camel milk is considered to be an important source of bioactive peptides with potential anti-diabetic effects. However, the mechanism by which these active peptides exert their anti-diabetic effects is not clear. The aim of this study was to systematically evaluate the in vivo anti-diabetic effects of Leucine-Leucine-Proline-Lysine (LLPK), a novel dipeptidyl peptidase-4 (DPP-4) inhibitory peptide identified from the in vitro gastrointestinal digestion product of camel milk. Methods: Adb/dbdiabetic mouse model was used, and LLPK was administered to mice at doses of 50 mg/kg BW and 100 mg/kg BW as a daily oral gavage for 30 days. The effects of LLPK on fasting blood glucose (FBG), oral glucose tolerance test (OGTT), insulin tolerance test (ITT), and serum lipid levels were monitored, and possible mechanisms of action were elucidated using proteomics. Results: The results demonstrated that LLPK significantly improved diabetic symptoms, including FBG, OGTT, ITT, and serum lipid levels indb/dbdiabetic mice. Furthermore, significantly increased levels of serum glucagon-like peptide 1 (GLP-1) and reduced serum DPP-4 activity were observed in the LLPK-treated group compared to the control group. Hepatic proteomics indicated that LLPK improved glucose and lipid metabolism via the PPAR signaling pathway, where the key targets were Scd1, Acox1, Acaa1b, Slc27a1, Acsl1, and Ehhadh. Conclusions: In summary, this study provided new insights into the anti-diabetic mechanisms of camel milk and supported the development of camel milk-based anti-diabetic functional foods or nutraceuticals.

Given the strong hydration propensity of fluoride ions, it is difficult to detect fluoride, especially inorganic fluoride, in aqueous samples. Resolving the issue of fluoride detection in aqueous samples is a scientific undertaking of great practical significance. Herein, we propose a new method for the sensitive and selective detection of fluoride in aqueous samples without the addition of organic solvents. The method involves surface-enhanced Raman spectroscopy using 1,4-diketo-3,6-diphenylpyrrolo[3,4- c ]pyrrole (DPP) compounds and Ag nanoparticles . The method is based on a diketopyrrolopyrrole compound linked to 1-butyl iodide (DPP1), which can sense fluoride sensitively and selectively. When DPP1 was combined with Ag NPs and reacted with tetrabutylammonium fluoride or inorganic fluoride in aqueous samples, an obvious Raman enhancement was obtained at the excitation wavelength of 633 nm. This response arises because the introduction of fluoride anions into the system changes the molecular orientation of DPP1 on the Ag NP substrate from horizontal to vertical, inducing a signal enhancement in the Raman spectrum . This system can detect inorganic fluoride at concentrations as low as 1.0 μmol L −1 (0.018 ppm), which is far below the public health service recommended levels for drinking water (0.7–1.2 ppm). Furthermore, using the proposed method, a linear response for fluoride in the concentration range of 1.0 × 10 −3 –1.0 × 10 −6 mol L −1 was obtained, which makes fluoride detection possible in practical samples, such as fluoride-containing toothpaste .

Dipeptidyl peptidase-IV (DPP-IV) is a key therapeutic target for type 2 diabetes, and food-derived inhibitory peptides are promising alternatives to synthetic drugs. In this study, bullfrog muscle protein hydrolysates were prepared using different proteases, among which the compound protease hydrolysate showed the strongest DPP-IV inhibitory activity. Ultrafiltration revealed that the <3 kDa fraction exhibited the highest potency. Three peptides (VPVG, LPVQ, and VPLG) were identified by LC–MS/MS combined with in silico screening. Among them, synthesized VPVG showed the strongest inhibitory activity (IC 50 = 128.91 μM). Enzyme kinetics indicated a mixed-type inhibition mechanism, while molecular docking and molecular dynamics simulations demonstrated stable interactions with the DPP-IV active site. The peptides showed good stability under heat treatment, varying pH conditions, and simulated gastrointestinal digestion. They exhibited no cytotoxicity in Caco-2 cells, inhibited cellular DPP-IV activity, and significantly reduced blood glucose levels in a hyperglycemic zebrafish model.

An injectable hydrogel sustained drug release system could be a promising technique for in situ treatment. Herein, an injectable hydrogel was prepared for photothermal-chemo therapy of cancer based on the thermosensitive liposomal hydrogel (Lip-Gel). The Lip-Gel system was fabricated by encapsulation of the NIR-II photothermal agent (DPP–BTz) and chemotherapy drugs (GEM) in thermosensitive liposomes and then combined with hydrogel precursor solution. The hydrogel precursor was used as an injectable flowing solution at room temperature and transferred into a cross-linked gel structure at physiological temperature. After being injected into the tumor, DPP–BTz in the Lip-Gel system can generate heat under irradiation of 1064 nm laser, breaking the thermosensitive liposomes and releasing GEM to kill tumor cells. From the treatment results, the Lip-Gel system showed a significant antitumor effect through chemo-/photothermal therapy combination therapy triggered by the NIR-II laser. This work provides a useful scheme for the development of drug delivery and drug treatment directions for local cancer therapy.

Goat milk, distinguished by its high β-casein content and superior digestibility compared with bovine milk, represents a promising source of bioactive peptides. Dipeptidyl peptidase-IV (DPP-4) inhibitors are vital for managing type 2 diabetes mellitus by enhancing incretin-mediated glycemic control. Naturally derived DPP-4 inhibitory peptides are of interest as food-compatible candidates for functional ingredient development. This study aimed to identify and characterize DPP-4 inhibitory peptides from goat milk proteins through an integrated computational and experimental approach. Eleven major goat milk proteins were subjected to in silico hydrolysis using Alcalase, Pepsin, and Trypsin, generating 319 peptides (3–15 amino acids). Virtual screening with ToxinPred, PeptideRanker, StackDPPIV, and KarmaDock prioritized non-toxic, bioactive candidates, followed by molecular docking and 500 ns Gaussian accelerated molecular dynamics simulations using AMBER 22 to elucidate binding and conformational dynamics with DPP-4 (PDB: 4A5S). High-performance liquid chromatography validated the presence of 3 top peptides—MMSF (from α-lactalbumin), MPFPK, and GPFPIL (from β-casein)—in the < 3 kDa hydrolysate fraction, with retention times closely matching standards (MPFPK: 8.923 vs. 8.856 min; MMSF: 11.003 vs. 10.912 min; GPFPIL: 12.406 vs. 12.334 min). In vitro assays confirmed competitive inhibition, with IC 50 values of 0.69 mM (MMSF), 0.32 mM (MPFPK), and 2.83 mM (GPFPIL), and negligible cytotoxicity (>95% cell viability at 10 mM in Caco-2 cells). Docking revealed interactions with critical DPP-4 residues (H740, Y631, Y547), with MPFPK forming the most extensive hydrogen bond network (5 bonds). Molecular dynamics simulations indicated enhanced DPP-4 stability (reduced RMSD), increased radius of gyration and solvent-accessible surface area, and distinct flap region (H235–E255) perturbations: GPFPIL induced a stable open conformation, MPFPK caused dynamic open-closed transitions, and MMSF disrupted β-sheet stability. MM-PBSA analysis confirmed MPFPK's superior binding affinity. These findings establish goat milk as a valuable source of DPP-4 inhibitory peptides, with MPFPK demonstrating exceptional potency and stability. This integrated framework provides a scalable approach for discovering food-derived bioactives, advancing the development of natural therapeutics and functional foods for type 2 diabetes management.

To achieve efficient separation of emodin and rhein, a double-template Fe₃O₄-imprinted polymer (DT-MIP@Fe₃O₄) was successfully synthesized. The characterization (FT-IR, XRD, TGA, SEM, TEM, VSM) demonstrated that the synthesized DT-MIP@Fe₃O₄ exhibited a core-shell architecture, exhibiting thermal stability up to 270 °C and superparamagnetic characteristics, facilitating separation in 8 s. This material exhibited significant adsorption capacities (31.7823 mg/g and 15.8942 mg/g) and quick adsorption kinetics (equilibrium achieved within 35 min), coupled with the benefits of facile separation, high selectivity, and exceptional reusability. DT-MIP@Fe₃O₄ was utilized as an adsorbent for magnetic solid-phase extraction (MSPE) in conjunction with high-performance liquid chromatography (HPLC) for the simultaneous separation and detection of emodin and rhein in the Chinese herbal medicine SXD. The optimization of parameters influencing MSPE-HPLC was conducted, such as adsorbent dosage, type and volume of desorbent, desorption time, and salt effect. The established method demonstrated exceptional linearity (R² > 0.9999) across the 0.1–100 mg/L, exhibiting high sensitivity and stability. The limits of detection (LOD) and the limits of quantification (LOQ) were established at 0.0092–0.02 mg/L and 0.0307–0.0668 mg/L, respectively. Moreover, the separated fractions demonstrated remarkable hypoglycemic efficacy by DPP-4 inhibition. These findings confirmed the effectiveness of the strategy and indicated that it has considerable prospects in the rapid separation of rhein and emodin from complex Chinese herbal compound, providing a method for the search of natural DPP-4 inhibitors.

Antarctic krill (Euphausia superba) is a nutrient-rich marine resource. Although several terrestrial proteases have been used to prepare Antarctic krill peptides (AKPs), there has been no report on the preparation of AKPs using a marine protease. Here, marine bacterial protease A69 was used to prepare AKPs with multi-bioactivities. Through optimizing hydrolysis parameters, we established a process for AKPs preparation by hydrolyzing Antarctic krill powder with A69. In the prepared AKPs, peptides less than 3000 Da and 1000 Da accounted for 99.23% and 88.37%, respectively. The scavenging ratios of the AKPs to ABTS+, DPPH· and ·OH reached 93.23 ± 0.09%, 99.90 ± 0.15%, and 93.90 ± 0.47%, respectively. The AKPs also had high angiotensin-converting enzyme (ACE)-inhibitory activity, with an IC50of 0.22 ± 0.04 mg/mL. At 40 mg/mL, the AKPs inhibited α-glucosidase and dipeptidyl peptidase IV (DPP-IV) activities by 7.18% and 13.62%, respectively, and displayed antibacterial activity toEscherichia coli. Moreover, 14 antioxidant peptides, 24 ACE-inhibitory peptides, 2 α-glucosidase-inhibitory peptides, and 10 DPP-Ⅳ-inhibitory peptides were identified from the AKPs. These results demonstrate that the prepared AKPs contain diverse bioactive peptides and have multi-bioactivities. This study indicates that marine bacterial protease A69 has promising application potential in preparing AKPs with multi-bioactivities.

Natural compounds have tremendous potential to regulate glucose metabolism, but conventional methods for studying their bioactivities are usually labor intensive. Here, hypoglycemic properties in 22 selected food-derived compounds were examined using molecular docking. The results indicated that curcumin is an inhibitor of both α-glucosidase and dipeptidyl-peptidase 4 (DPP-4), which are important for glycemic control. These effects of curcumin were also confirmed by enzymatic determination in vitro. Furthermore, curcumin significantly improved diet-induced hyperglycemia (e.g., fasting plasma glucose levels and glycogen storage in muscle or liver) in mice. This might be attributed to its inhibitory effects on the activities of α-glucosidase and DPP-4 in vivo. Curcumin also upregulated the expression of genes (e.g., glucagon-like peptide 1) related to DPP-4 activity in the small intestine. In conclusion, curcumin is a potential ingredient of functional foods used for diet-induced hyperglycemia management. Practical applications Curcumin has been widely used as a colorant in the food industry. Moreover, a growing number of studies have described its diverse biological functions, such as anti-inflammatory, anti-oxidant, and anti-angiogenic activities. Thus, curcumin is regarded as a potential ingredient in functional foods. Our results highlighted the hyperglycemic effect of curcumin, suggesting that curcumin may be included in food products for hyperglycemic patients.
