Diafenthiuron is a novel derivative of thiourea and is highly toxic to non-target organisms, necessitating its efficient removal from wastewater before discharge. This study compared diafenthiuron removal efficiencies at a target concentration of 1 µM using three methods: a 4 mg/L ozone (O3) treatment; an ultraviolet (UV) light treatment, applying UV254radiation with a fluence of 60 mJ/cm2for 10 min; and a combined O3/UV treatment utilizing ozone and ultraviolet light. An acute toxicity assessment was conducted using a modeled zebrafish embryo (Danio rerio). The diafenthiuron removal efficiencies were 49.59%, 54.51%, and 68.90% for the UV light, O3, and O3/UV treatments, respectively. The treatments showed additional benefits of exerting no negative impacts on the survival rate, heart rate, or body length of the zebrafish larvae posttreatment. The survival and heart rates at 120 hpf, as well as the body length at 96 and 120 hpf, showed significant differences between the advanced oxidation and 1 μM diafenthiuron treatment groups. However, these parameters remained consistent with those of the control group. The three treatments alleviated the spatiotemporal downregulation of the liver-specific markerfabp10acaused by diafenthiuron exposure. The UV light and O3/UV treatments were efficient at degrading diafenthiuron, causing decreased reactive oxygen species levels and increasedpomcandprlexpression levels. The O3-treated diafenthiuron and 1 μM diafenthiuron treatments increased the reactive oxygen species levels and decreased thepomcandprlexpression levels. The combined O3/UV treatment showed the highest removal efficiency and the least toxicity, making it the most effective method for diafenthiuron degradation. This study provides valuable insights into the treatment of diafenthiuron-laden wastewater.

The surge in global plastic usage has exacerbated the pollution of nanoplastics and plasticizers in aquatic ecosystems, causing widespread concern about population health issues. We used a zebrafish model to investigate the combined toxicity of polystyrene nanoparticles (PS-NPs) and plasticizer metabolite mono-(2-ethylhexyl) phthalate (MEHP), revealing the synergistic neurotoxic effects and potential mechanisms of compound exposure. Co-exposure amplifies the bioaccumulation of PS-NPs ( p < 0.05), causing innate immune damage ( p < 0.0001), severe oxidative stress ( p < 0.0001), and embryonic apoptosis ( p < 0.05), while inducing larval neuromotor dysfunction and changes in neural markers. Further exploration revealed that oxidative stress-induced lipid peroxidation disrupts glycerophospholipid metabolism and cholinergic synthesis, which can be an important mechanism for neurotoxicity caused by the co-exposure of PS-NPs and MEHP. This hypothesis is supported by dysregulation of acetylcholinesterase ( ache ), docking protein 7 ( dok7 ), and heparan sulfate proteoglycan 2 ( hspg2 ) genes linked to neurotransmission. In summary, this study reveals new synergistic toxicity mechanisms between nanoplastics and plasticizers, advancing the risk assessment framework for complex environmental pollutant interactions.

Benzovindiflupyr is a broad spectrum succinate dehydrogenase inhibitor fungicide, and information about its risk to aquatic organisms is limited. The present study examined the developmental effects of benzovindiflupyr exposure to zebrafish under static conditions through fish embryo toxicity test. First, zebrafish hatchability and survivability were decreased. Edema, body deformation, and microcephaly were abnormalities observed in embryos. Secondly, in vivo fluorescence analysis showed that benzovindiflupyr induced the reactive oxygen species. Moreover, stress-related oxidative enzyme experiments showed that malondialdehyde, superoxide dismutase, glutathione S-transferase, peroxidase and glutathione level were affected, and gene experiments showed that catalase, glutathione peroxidase 1a, superoxide dismutase 1 and superoxide dismutase 2 gene transcription levels were upregulated. Fourth, succinate dehydrogenase activity and the associated gene transcription level were decreased. Taken together, this study suggested that benzovindiflupyr could induce oxidative stress and decrease succinate dehydrogenase activity of zebrafish and has the potential to affect embryonic development.

Emerging contaminants refer to chemical substances that have not been widely regulated but possess the potential to cause adverse effects on both the environment and human health. Antibiotics, as emerging contaminants, pose significant threats to ecosystems and human health due to their widespread use and persistence in the environment. Levofloxacin, a broad-spectrum fluoroquinolone antibiotic, is commonly employed in the treatment of bacterial infections, and has been frequently detected in environmental matrices and freshwater systems. In this study, we assessed the effects of levofloxacin on hatchability, mortality rates, malformations, behavioral changes, and cardiac development in zebrafish embryos by exposing them to varying concentrations of levofloxacin (0, 0.5, 1, 2, 4, and 8 mM). Our results demonstrate that levofloxacin exposure significantly impaired the growth and development of zebrafish larvae, particularly at higher concentrations. Notable effects included reduced body length, abnormal yolk sac and swim bladder development, pericardial edema, prolonged distances between the sinus venosus and arteriolar bulb (SV-BA), and disruptions in heart rate. Quantitative PCR analysis further revealed that levofloxacin exposure significantly upregulated the expression of key cardiac development genes in zebrafish larvae, includingnppa,myh6,cacna1ab,myl7,gata4,nkx2.5,tbx2b, andtbx5b. These findings indicate that levofloxacin exposure exerts significant toxic effects on both embryonic and larval growth as well as heart development and gene expression in zebrafish. This study provides critical insights into the potential ecological risks posed by levofloxacin along with other antibiotics while laying a foundation for further investigation into their toxicological mechanisms.

Spring viraemia of carp virus (SVCV) is a critical pathogen of cyprinids. In this study, we investigated the effects of mannan on the antiviral resistance of zebrafish and elucidated the underlying mechanisms. Zebrafish were fed with a control diet or diet supplemented with 0.2%, 0.4%, 0.6% and 0.8% of mannan for 2 weeks, and were challenged by SVCV. The intestinal microbiota was evaluated by 16S rRNA gene pyrosequencing . Zebrafish embryonic fibroblast (ZF4) cells were treated with mannan and were infected by SVCV, and the expression of antiviral genes was investigated. In addition, the effect of the in vitro culture supernatant of mannan-associated microbiota on SVCV infection was evaluated in ZF4 cells model. Results showed that dietary 0.2%, 0.4% and 0.6% mannan significantly improved the survival rate of zebrafish compared with the control group ( P < 0.05). Consistent with in vivo results, mannan significantly inhibited the replication of SVCV in ZF4 cells ( P < 0.05). Moreover, mannan inhibited the adsorption of SVCV to ZF4 cells ( P < 0.05). The expression of type I interferon (IFN) signaling pathway related genes, including IRF3 , IFNφ1, IFNφ2, IFNφ3 , and PKZ, was significantly increased in mannan treated cells versus control ( P < 0.05). Dietary mannan modulated gut microbiota, with enrichment of Proteobacteria and reduced abundance of Firmicutes , Actinobacteriota, and Fusobacteriota. Furthermore, the culture supernatant of mannan-associated microbiota significantly inhibited SVCV replication in ZF4 cells versus control microbiota counterpart ( P < 0.05). Initial evaluation suggests that the antiviral effect of culture supernatant of mannan-associated microbiota involves a heat stable component derived from the commensal bacteria. To sum up, our results indicate that the mannan enhanced the resistance of zebrafish against SVCV infection. The mechanism underlying the antiviral effect of mannan include enhancement of IFN signaling and inhibition of viral adsorption. Dietary addition of Mannan altered the structure of the intestinal microbiota of zebrafish, and the intestinal microbiota also contributed to the anti-viral effect of mannan.

Decabromodiphenyl ethane (DBDPE), a chemical compound widely detected in aquatic ecosystems, has shown a range of toxicological effects in zebrafish ( Danio rerio ). However, its effects on zebrafish reproduction remain underexplored. In this study. Adult zebrafish were exposed to 0, 1, 10, and 100 μg/L of DBDPE for 21 days. DBDPE accumulated in the gonads, leading to a marked decrease in both cumulative spawning and gonadosomatic index in female zebrafish. To investigate the mechanisms underlying reproductive abnormalities induced by DBDPE, we measured serum hormone levels, oocyte development, and gene expression in the hypothalamic–pituitary–gonadal (HPG) axis. Our results showed that exposure to DBDPE significantly reduced estradiol (E2) levels in female zebrafish, along with abnormal oocyte development and disrupted gene expression in the HPG axis. Further metabolomic and lipidomic analyses revealed considerable changes in taurine and hypotaurine metabolism, glutathione metabolism, and the FoxO signaling pathway, along with significant disturbances in lipid metabolism. These disruptions in metabolic pathways may be an important underlying cause of delayed oocyte development. Our findings show that exposure to DBDPE delays oocyte development by altering lipid metabolism, reducing sex hormone levels, and impairing HPG axis regulation, ultimately leading to reduced fecundity in female zebrafish. This study highlights the repercussions of DBDPE exposure on the reproductive health of zebrafish, emphasizing the need for stricter regulation of its production and use, a well as a comprehensive reassessment of the risks it poses to fish.

Vision serves as the primary sensory function for most animals and is essential for their survival. Benzothiazoles (BTs) are widely used in various industrial applications and can be frequently detected in various environmental media. However, current understanding of their visual toxicity remains limited. In the present study, three typical BTs, 2-mercapto-benzothiazole (MBT), 2-hydroxy-benzothiazole (2-OH-BTH), and 2-amino-benzothiazole (2-ABTH), were chosen to explore the visual toxicity and mechanisms in zebrafish larvae. The results showed that all of the three BTs inhibited eye development and suppressed optokinetic response in larvae. The damages induced by 2-ABTH and 2-OH-BTH could be attributed to the oxidative stress and the over activation of GABAergic signaling, respectively, while MBT exerted toxic effects through both pathways. Density functional theory calculation showed that MBT and 2-ABTH possessed high reactivity than 2-OH-BTH, which provided a potential explanation for their higher potency to induce oxidative stress. In addition, molecular docking and molecular dynamics simulation showed that MBT and 2-OH-BTH can form stable complexes with Gabrb3, which could be responsible for the enhanced GABAergic signaling. These findings provide new insights into the distinct visual toxic mechanisms of BTs underlying their subtle structural differences.

Tire wear particles (TWPs) are a significant source of microplastics (MPs) and N -(1,3-dimethylbutyl)- N ′-phenyl- p -phenylenediamine (6PPD) in aquatic environments. However, the combined toxicity of MPs and 6PPD to aquatic organisms remains unclear. Here, we investigated the bioaccumulation of 6PPD, developmental toxicity, oxidative stress, and metabolic alterations in zebrafish larvae following single and combined exposure to polystyrene MPs (PSMPs) and 6PPD. Our results indicated that co-exposure to PSMPs and 6PPD induced synergistic toxicity in zebrafish larvae, significantly elevating levels of reactive oxygen species (ROS) and malondialdehyde (MDA), along with enhancing antioxidant enzyme activities. PSMPs did not influence the bioaccumulation of 6PPD, but increased the concentration of 6PPD quinone (6PPD-Q) in zebrafish larvae. PSMPs and 6PPD altered the metabolomic profiles of zebrafish larvae, affecting organic acids and their derivatives, amino acids and carbohydrates. The primary distinctions between single and combined exposures were observed in organic acids (citric acid and 9-Octadecenoic acid), sugars (D-Allose, D-Arabinose, and d-Mannose), and cholesterol. These findings imply PSMPs alter the toxicity of 6PPD to zebrafish larvae, providing valuable data for assessing the potential risks of single and combined pollution of MPs and 6PPD to aquatic creatures.

Flocoumafen (FCF), a second-generation anticoagulant rodenticide (SGAR), has limited toxicological data regarding its effects on aquatic organisms. In this study, we exposed zebrafish embryos to FCF solutions at concentrations of 0.2, 0.4, and 0.8 mg/L until 120 h post-fertilization (hpf). The results revealed a decrease in survival rates. Notably, FCF exposure significantly reduced the frequency of spontaneous tail coils at 24 hpf, while shortened body length and induced spinal curvature at 120 hpf. Furthermore, zebrafish larvae exhibited craniofacial abnormalities and incomplete bone mineralization at 120 hpf following FCF exposure. In Tg( HuC:EGFP ) transgenic strains, neuronal loss was observed. Additionally, FCF-exposed zebrafish larvae showed a marked reduction in locomotor ability, activity levels, and turning capacity. The qPCR and enzyme activity assays revealed significant changes in gene expression associated with the Notch signaling pathway, accompanied by increased levels of reactive oxygen species (ROS), superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA). Astaxanthin (ASTA) partially alleviated the toxicities induced by FCF. These findings suggest that FCF may induce skeletal and neurological toxicities by affecting oxidative stress, disrupting the normal expression of skeletal and nervous system-related genes in the Notch signaling pathway, and ultimately leading to behavioral abnormalities. Our findings may provide new insights into a comprehensive evaluation of FCF toxicology in aquatic organisms, and may assist the government in formulating and implementing regulatory policies regarding the application of FCF.

Isofenphos-methyl (IFP), an organophosphorus pesticide widely used in agriculture, has raised public health concerns because of its potential risks to human health. Although recent studies have linked IFP exposure to developmental toxicity, the underlying toxicological mechanisms remain insufficiently understood. To address this gap, this study employed a zebrafish model to investigate the developmental toxicity of IFP through an integrated approach combining behavioral, histopathological, biochemical, and transcriptomic analyses. Exposure to IFP from 2 to 96 hours post-fertilization (hpf) adversely affected early zebrafish embryonic development, with a calculated 96-h LC₅₀ of 6.407 mg/L. Embryos exposed to 0.1, 2, 4, 8 mg/L IFP diminished responsiveness to light/dark stimulation, and disrupted circadian rhythms. Moreover, IFP exposure impaired immune function, significantly reducing in macrophage and neutrophil numbers. Transcriptomic analysis revealed disruptions in neuroactive ligand–receptor interaction, leading to dysfunctional synaptic signalling and neuronal communication; and it also disturbed protein digestion and absorption, thereby limiting the availability of essential amino acid precursors (e.g., tryptophan and tyrosine) required for the synthesis of key neurotransmitters such as serotonin and dopamine (DA). These transcriptomic alterations were supported by altered neurotransmitter levels, brain mitochondrial damage, and histopathological abnormalities in both brain and ocular structures. RNA-sequencing results were further validated using quantitative real-time reverse transcription polymerase chain reaction (RT-qPCR), confirming the robustness of the data. Collectively, these findings demonstrate that IFP exerts pronounced neurotoxic and immunotoxic effects on developing zebrafish, underscoring potential risks during early developmental stages and the urgent need for stricter regulation of IFP contamination in aquatic environments.

Nanomaterials have attracted much attention in the biomedical field. Black phosphorus quantum dots (BPQDs) have shown great potential in biomedical applications, but their potential risks to biosafety and environmental stability have not been fully evaluated. In the present study, zebrafish ( Danio rerio ) embryos were exposed to 0, 2.5, 5 and 10 mg/L BPQDs from 2 to 144 h post-fertilization (hpf) to explore developmental toxicity. The results showed that exposure to BPQDs for 96 h induced developmental malformations (tail deformation, yolk sac edema, pericardial edema, and spinal curvature) in zebrafish embryos. ROS and antioxidant enzyme activities (CAT, SOD, MDA and T-AOC) were substantially altered and the acetylcholinesterase (AChE) enzyme activity was significantly decreased in the BPQDs exposed groups. Locomotor behavior was inhibited after BPQDs exposure for 144 h in zebrafish larvae. A significant increase in 8-OHdG content indicates DNA oxidative damage in embryos. In addition, obvious apoptotic fluorescence signals were detected in the brain, spine, yolk sac and heart. At the molecular level, the mRNA transcript levels of key genes related to skeletal development (igf1, gh, MyoD and LOX), neurodevelopment (gfap, pomca, bdnf and Mbpa), cardiovascular development (Myh6, Nkx2.5, Myl7, Tbx2b, Tbx5 and Gata4) and apoptosis (p53, Bax, Bcl-2, apaf1, caspase-3 and caspase-9) were abnormal after BPQDs exposure. In conclusion, BPQDs induced morphological malformations, oxidative stress , locomotor behavior disorders, DNA oxidative damage and apoptosis in zebrafish embryos. This study provides a basis for further study on the toxic effects of BPQDs.

Paclobutrazol is a widely used chiral plant growth regulator and its enantioselective toxicity in aquatic organisms is less explored till now. Herein, the enantioselective neurotoxicity of paclobutrazol mediated by oxidative stress in zebrafish were investigated. The oxidative stress parameters and neurotoxic biomarkers changed significantly in each exposure group, and paclobutrazol showed enantioselective toxicity in zebrafish. Firstly, (2 R , 3 R )-paclobutrazol exhibited a stronger oxidative stress in zebrafish than (2 S , 3 S )-enantiomer ( P < 0.05). Then, activities of acetylcholinesterase , calcineurin , and total nitric oxide synthase in (2 R , 3 R )-paclobutrazol treatments were 0.61–0.89, 1.24–1.53, and 1.21–1.35-fold stronger ( P < 0.05) than those in (2 S , 3 S )-enantiomer treatments, respectively. Next, the content variations of four neurotransmitters in zebrafish exposed to (2 R , 3 R )-paclobutrazol were significantly larger than those in (2 S , 3 S )-enantiomer treatments ( P < 0.05). Moreover, (2 R , 3 R )-paclobutrazol had stronger binding with the receptors than (2 S , 3 S )-enantiomer through molecular docking . The integrated biomarker response values further demonstrated that (2 R , 3 R )-paclobutrazol showed stronger toxicity to zebrafish than (2 S , 3 S )-enantiomer. Furthermore, the neurotoxicity of paclobutrazol can be interpreted as the mediating effect of oxidative stress in zebrafish through correlation analysis, and an adverse outcome pathway for the nervous system in zebrafish induced by paclobutrazol was proposed. This work will greatly extend our understanding on the enantioselective toxic effects of paclobutrazol in aquatic organisms.
