Exposure scenarios modulate the combined toxicity of nanoplastics and Cd to Daphnia magna

Shuang Zhang,Zhongmiao Wang,Caijiao He,Daohui Lin

Nanoplastics (NPs) have extensively accumulated in aquatic environments and coexist with various contaminants including heavy metals. While the adverse effects of NPs and Cd on aquatic organisms have been widely reported, most studies focus on co-exposure scenarios, leaving the effect of different exposure sequences poorly understood. Here, we evaluated the combined toxicity of NPs and Cd to Daphnia magna under different exposure scenarios (single exposure, pre-exposure, and co-exposure). The bioaccumulation of NPs and Cd, intestinal histology, and gut microbiota were further analyzed to elucidate the underlying mechanisms of combined toxicity. Our findings reveal that environmentally relevant concentrations of NPs stimulated D. magna reproduction, increasing total number of offspring (hormesis). Pre-exposure to NPs enhanced tolerance to subsequent stressors, thus reducing the combined toxicity of NPs and Cd. In contrast, slow toxicological recovery of Cd caused persistent residual effects that amplified the toxicity of subsequent combined exposure to NPs and Cd. Specifically, compared to single-exposure groups, Cd pre-exposure led to a 5-fold and 3-fold increase in the bioaccumulation of NPs and Cd, respectively, accompanied by exacerbated intestinal damage and disruption of the gut microbiota. Our results highlighted the importance of evaluating combined toxicity under different exposure scenarios in natural environments, providing new insight into the potential risks of NPs in aquatic environments.

Effects of Daphnia magna on the Concentration and Size Distribution of Suspended Particles

Xiaoyan Zhang,Feng Ge,Lichao Tan,Fukai Tang,Keke Xu,Xiaorong Wu,Mingzhu Zhang,Peiyong Guo

Suspended particles are key components of aquatic ecosystems that modulate the biogeochemical processes of natural waters. The filter feederDaphnia magnacan alter suspended particle characteristics and aquatic habitats, while habitat variations in turn affect its growth and reproduction. This study primarily investigated the impacts ofD. magnaon the concentration and size distribution of suspended particles, and explored its particle size selectivity. Three particle types, namely kaolinite (KN), montmorillonite (MN) and natural particles (NP), were adopted for experiments, and natural particles were further divided into three size fractions (<150 μm, <96 μm, <75 μm). The results demonstrated thatD. magnasignificantly reduced suspended particle concentrations via ingestion, with the most prominent effect observed within the initial three hours of experimentation, verifying its practical value for wastewater remediation. Though classified as a non-selective filter feeder,D. magnaexhibited distinct particle size selectivity for identical substances. Fine particles (<8 μm) presented higher bioavailability and were preferentially ingested byD. magna. Additionally, the regulatory effects ofD. magnaon particle size distribution were closely associated with its particle digestion process, which depended on inherent particle properties, including composition and organic matter content.

Quantifying nanoplastic-bound chemicals accumulated in Daphnia magna with a passive dosing method

Ruifen Jiang,Wei Lin,Jiayi Wu,Yaxin Xiong,Fang Zhu,Lian-Jun Bao,Jing You,Gangfeng Ouyang,Eddy Y. Zeng

Because of their large surface area and high hydrophobicity, nanoplastics (NPs) possess a large sorption capacity for and strong affinity to hydrophobic organic chemicals, which may substantially alter the environmental behavior, especially the bioaccumulation of hydrophobic organic compounds. The present study aims to examine the effects of nano-size polystyrene (PS) on the bioaccumulation of polychlorinated biphenyls (PCBs) in Daphnia magna. A novel passive dosing method was developed to quantify the accumulation of NPs in D. magna in different exposure suspensions with various NP concentrations during 2 h and 24 h exposure. Addition of NPs enhanced the amount of PCBs accumulated in D. magna by 1.4–2.6 times compared to no presence of NPs during 24 h exposure. The relative amounts of NP-bound PCBs accumulated in D. magna were 18% to 81% for different PCB congeners at various NP concentrations. The significance of different uptake pathways in the accumulation of PCBs in D. magna with the presence of NPs was found to relate to the chemical properties of PCBs, the ingestion rate of plastics and the exposure conditions. These results have justified additional research efforts toward investigating the fate and toxic effects of NP-bound contaminants ingested by organisms.

Combined toxic effects of nitrite and ammonia on life history traits of Daphnia pulex

Yu Bo,Lyu Kai,Li Jiajia,Yang Zhou,Sun Yunfei

Nitrite and ammonia are two of the most common toxic nitrogenous pollutants in aquatic ecosystem, which can pose a serious threat to the health of aquatic organisms. Cladocerans, as an important part of freshwater ecosystem, will inevitably be harmed by these pollutants. To evaluate the combined toxic effects of these nitrogenous pollutants on cladocerans, we simulated 15 combinations of five nitrite concentrations (0, 0.5, 2, 4, 8 mg L−1) and three ammonia concentrations (0, 0.1, 1 mg L−1) to study the changes of life history traits of Daphnia pulex exposed to these combinations for 21 days. Results showed that under the combined stress of nitrite and ammonia, the survival time of D. pulex was shortened, the time to first batch of eggs and time to first brood were delayed, the body size and tail spine length at maturation were reduced, and the total offspring and the number of broods were decreased. There were some synergistically negative effects between the two nitrogenous pollutants. The presence of ammonia reduced the tolerance of D. pulex to nitrite, and vice versa. These findings provided new insights into the combined toxic effects of nitrite and ammonia on the life history traits of cladocerans, and were of great significance for understanding the population dynamics of specific species in cladocerans community under the condition of compound nitrogenous pollutants.

Stereoselective Impacts of Flusulfinam on Daphnia magna: Stability, Behavior, Oxidative Stress, and Metabolomics Insights

Ling Wu,Mengting Liang,Yanxi Sun,Shanshan Fu,Yuping Zhang

Flusulfinam, a novel chiral herbicide, has attracted considerable attention because of its high efficacy against common paddy weeds and its favorable crop safety profile. However, information regarding its enantioselective toxicity toward aquatic organisms remains scarce. In this study, single-crystal X-ray diffraction was employed to determine the absolute configurations of flusulfinam enantiomers. The results indicated that the enantiomers can stably persist in the aquatic environment of Daphnia magna , with no detectable configurational transformation or degradation. Both racemic flusulfinam and its individual enantiomers exhibited low toxicity to D. magna . At sublethal concentrations (0.95 and 9.5 mg/L), R-flusulfinam and S-flusulfinam induced short-term inhibition (24 h) followed by long-term stimulation (48–72 h) of swimming distance and speed in D. magna . Oxidative stress assessments showed increased activities of superoxide dismutase (SOD) and catalase (CAT), with R-flusulfinam producing more pronounced effects than S-flusulfinam. Metabolomic analyzes further revealed that both enantiomers interfered with nucleotide and purine metabolism in D. magna , while R-flusulfinam caused more severe metabolic disturbances. Overall, these findings demonstrate that R-flusulfinam exhibits greater toxicity to D. magna than S-flusulfinam, providing important insights for the environmental risk assessment of flusulfinam.

Study on acute toxicity and structure–activity relationship of Daphnia magna exposed to naphthoquinones

Wenhua Song,Jing Guo,Feng Ding,Weixuan Hu,Zhen Li,Minling Gao

The acute toxicities of the six naphthoquinone compounds on Daphnia magna ( D. magna ) at 48   h were classified as harmful, toxic, and very toxic. The results indicated that log   P played an important part in the toxicity of compounds to organism. And 1-carbonyl or the other hydrophobic substituents of the naphthoquinone compounds are likely to mediate the binding of the compound to the target via hydrogen bonding and hydrophobic interactions. Our results provided a foundation for further investigation using 3D-QSAR and HQSAR to evaluate the aquatic ecological risk and the mechanism of toxicity of naphthoquinones.

Multiple Screening of Pesticides Toxicity in Zebrafish and Daphnia Based on Locomotor Activity Alterations

Akhlaq Hussain,Gilbert Audira,Nemi Malhotra,Boontida Uapipatanakul,Jung-Ren Chen,Yu-Heng Lai,Jong-Chin Huang,Kelvin H.-C. Chen,Hong-Thih Lai,Chung-Der Hsiao

Pesticides are widely used to eradicate insects, weed species, and fungi in agriculture. The half-lives of some pesticides are relatively long and may have the dire potential to induce adverse effects when released into the soil, terrestrial and aquatic systems. To assess the potential adverse effects of pesticide pollution in the aquatic environment, zebrafish (Danio rerio) andDaphnia magnaare two excellent animal models because of their transparent bodies, relatively short development processes, and well-established genetic information. Moreover, they are also suitable for performing high-throughput toxicity assays. In this study, we used both zebrafish larvae and water flea daphnia neonates as a model system to explore and compare the potential toxicity by monitoring locomotor activity. Tested animals were exposed to 12 various types of pesticides (three fungicides and 9 insecticides) for 24 h and their corresponding locomotor activities, in terms of distance traveled, burst movement, and rotation were quantified. By adapting principal component analysis (PCA) and hierarchical clustering analysis, we were able to minimize data complexity and compare pesticide toxicity based on locomotor activity for zebrafish and daphnia. Results showed distinct locomotor activity alteration patterns between zebrafish and daphnia towards pesticide exposure. The majority of pesticides tested in this study induced locomotor hypo-activity in daphnia neonates but triggered locomotor hyper-activity in zebrafish larvae. According to our PCA and clustering results, the toxicity for 12 pesticides was grouped into two major groups based on all locomotor activity endpoints collected from both zebrafish and daphnia. In conclusion, all pesticides resulted in swimming alterations in both animal models by either producing hypo-activity, hyperactivity, or other changes in swimming patterns. In addition, zebrafish and daphnia displayed distinct sensitivity and response against different pesticides, and the combinational analysis approach by using a phenomic approach to combine data collected from zebrafish and daphnia provided better resolution for toxicological assessment. Keywords:zebrafish;daphnia;pesticide;locomotion;behavior;phenomics

Nanoparticle TiO2 size and rutile content impact bioconcentration and biomagnification from algae to daphnia

Xiangjie Chen,Ya Zhu,Kun Yang,Lizhong Zhu,Daohui Lin

Little information is available about effect of particle size and crystal structure of nTiO 2 on their trophic transfer. In this study, 5 nm anatase , 10 nm anatase, 100 nm anatase, 20 nm P25 (80% anatase and 20% rutile), and 25 nm rutile nTiO 2 were selected to investigate the effects of size and crystal structure on the toxicity, bioconcentration, and trophic transfer of nTiO 2 to algae and daphnia. In the exposed daphnids, metabolic pathways affected by nTiO 2 and nTiO 2 -exposed algae (nTiO 2 -algae) were also explored. The 96 h IC 50 values of algae and the 48 h LC 50 values of daphnia were 10.3, 18.9, 43.9, 33.6, 65.4 mg/L and 10.5, 13.2, 37.0, 28.4, 60.7 mg/L, respectively, after exposed to nTiO 2 -5A, nTiO 2 -10A, nTiO 2 -100A, nTiO 2 -P25, and nTiO 2 -25R, respectively. The bioconcentration factors (BCFs) for 0.1, 1, and 10 mg/L nTiO 2 in daphnia ranged from 21,220 L/kg to 145,350 L/kg. The nTiO 2 biomagnification factors (BMFs) of daphnia fed with 1 and 10 mg/L nTiO 2 -exposed algae were consistently greater than 1.0 (5.7–122). The results show that the acute toxicity, BCF, and BMF all decreased with increasing size or rutile content of nTiO 2 . All types of nTiO 2 were largely accumulated in the daphnia gut and were not completely depurated within 24 h. At the molecular level, 22 Kyoto Encyclopedia of Genes and Genomes (KEGG) metabolic pathways of daphnia were impacted by the nTiO 2 and nTiO 2 -algae treatments, including glutathione metabolism, aminoacyl-tRNA biosynthesis, among others. Six and four KEGG metabolic pathways were significantly disturbed in daphnids exposed to nTiO 2 and nTiO 2 -algae, respectively, indicating the presence of algae partially alleviated the negative impact of nTiO 2 on metabolism. These findings increase understanding of the impacts of physicochemical properties of nTiO 2 on the food chain from molecular scale to that of the whole organism, and provide new insight into the ecological effect of nanomaterials .

Protective effects of small heat shock proteins in Daphnia magna against heavy metal exposure

Muyi Li,Ting Tang,Fengyu Yuan,Yuming Zhang,Fengchao Li,Fengsong Liu

Daphnia magna is one of the most commonly used model organisms to assess toxicity of heavy metal and other xenobiotics. However, the lack of knowledge about important stress-resistant molecules limits our understanding of the alteration of phenotypic and physiological traits of D. magna upon stress exposures. In this study, we focused on a chaperone family of small heat shock protein (sHSP) that has been found in archaea, bacteria and eukaryotes and plays an important role in stress tolerance. A total of eleven sHSP genes (termed DmsHSP1 - DmsHSP11) were identified from the D. magna genome, whose expression profiles during exposure to heavy metal (Cd 2+ , Cu 2+ and Zn 2+ ) and a few other potential pollutants were evaluated via qRT-PCR and RNA-Seq analysis. The results highlighted the predominant role of DmsHSP1 with the highest basal expression level in adults and robust upregulation upon exposure to heavy metals (Cu 2+ > Cd 2+ > Zn 2+ ). In vivo , recombinant protein rDmsHSP1–21 and rDmsHSP11–12.8 could not only prevent model substrates agglutination induced by heavy metals or reducer dithiotreitol (DTT), but also protect tissue proteins and enzymes from denaturation and inactivation caused by heavy metals or high temperature. Ectopically expression of DmsHSP1–21 or DmsHSP11–12.8 in E. coli conferred host enhanced resistance against various abiotic stresses including Cd 2+ , Cu 2+ and phenazine methosulfate (PMS). Knockdown of DmsHSP1–21 by RNAi, but not for DmsHSP11–12.8, significantly increased the vulnerability of D. magna to heavy metal exposure. Our work provides systematic information on the evolution and function of sHSPs in D. magna and leads to important insights into the mechanisms by which D. magna survive in adverse environments.

Chronic toxicity of broflanilide in Daphnia magna: changes in molting, behavior, and gene expression

Wang Kai,Liang Yaping,Duan Manman,Che Wunan,He Lu

Broflanilide is a novel pesticide used in agriculture that binds to unique receptors on pests; however, the widespread use of broflanilide has led to toxicity in Daphnia magna . At present, little information on the potential threats broflanilide imposes on D. magna is available. Therefore, the present study examined the chronic toxicity of broflanilide in D. magna by comparing changes in molting, neurotransmitter function, and behavior. The results showed that broflanilide caused chronic toxicity in D. magna at a concentration of 8.45 μg/L, and growth, development, reproduction, and the development of offspring were affected. In addition, broflanilide affected the molting of D. magna by significantly inhibiting the expression of chitinase, ecdysteroid, and related genes. Broflanilide also affected the expression of γ-glutamic acid, glutamine, gamma-aminobutyric acid, 5-hydroxytryptamine, 5-hydroxytryptophan, dopa, and dopamine. Furthermore, the swimming distance and speed of D. magna were reduced . Taken together, the results demonstrate the chronic toxicity and exposure risk of broflanilide in D. magna .

Effects of triadimefon fungicide on Daphnia magna: Multigenerational effect and population-level ecological risk

Lin Hou,Xiaowei Jin,Na Liu,Ying Luo,Jianhua Liao,Changsheng Guo,Jian Xu

Triadimefon is ubiquitous in various environmental media. Although toxicity of triadimefon to individual of aquatic organisms has been confirmed, its effect on organisms at population level remain poorly understood. In this study the long-term effect of triadimefon on individual and population of Daphnia magna were studied using multi-generational experiments and matrix model. Development and reproduction of three generations of F1 and F2 were significantly inhibited with the triadimefon concentration of 0.1 mg/L ( p  < 0.01). Toxicity of triadimefon to the offspring was stronger than to the parent ( p  < 0.05). When triadimefon concentration was higher than 0.1 mg/L, both population number and intrinsic rate of increase showed a decreasing trend with the increasing exposure concentration. Age structure of the population also tended to decline. Toxicity threshold derived on population-level was between mortality-based LC 50 and reproduction-based NOEC of Daphnia magna , and also between acute toxicity and chronic toxicity derived from species sensitivity distribution (SSD). The risk of population level derived from risk quotient was low for most areas, and the results derived from probability risk showed that the expected loss of intrinsic rate of increase of population was 0.0039 without considering other factors. Compared to the individual-level, the ecological risks at the population level were closer to the actual situation of the ecosystem response to the chemical pollution.

Using DeepLabCut for markerless cardiac physiology and toxicity estimation in water fleas (Daphnia magna)

Ferry Saputra,Michael Edbert Suryanto,Gilbert Audira,Cao Thang Luong,Chih-Hsin Hung,Marri Jmelou Roldan,Ross D. Vasquez,Chung-Der Hsiao

Daphnia magna is one species of water flea that has been used for a long time for ecotoxicity studies. In addition, Daphnia has a myogenic heart that is very useful for cardiotoxicity studies. Previous attempts to calculate the cardiac parameter endpoints in Daphnia suffer from the drawback of tedious operation and high variation due to manual counting errors. Even the previous method that utilized deep learning to help the process suffer from either overestimation of parameters or the need for specialized equipment to perform the analysis. In this study, we utilized DeepLabCut software previously used for animal pose tracking and demonstrated that ResNet_152 was the best fit for training the network. The trained network also showed comparable results with ImageJ and Kymograph, which was mostly done manually. In addition to that, several macro scripts in either Excel or Python format were developed to help summarize the data for faster analysis. The trained network was then challenged to analyze the potential cardiotoxicity of imidacloprid and pendimethalin in D. magna , and it showed that both pesticides cause alteration in their cardiac performance. Overall, this method provides a simple and automatic method to analyze the cardiac performance of Daphnia by utilizing DeepLabCut . The method proposed in this paper can contribute greatly to scientists conducting fast and accurate cardiotoxicity measurements when using Daphnia as a model.
