Carbapenem-Resistant E. coli Adherence to Magnetic Nanoparticles

Oznur Caliskan-Aydogan,Chloe Zaborney Kline,Evangelyn C Alocilja

Carbapenem-resistant Enterobacterales (CRE) is an emerging global concern. Specifically, carbapenemase-producing (CP) E. coli strains in CRE have recently been found in clinical, environmental, and food samples worldwide, causing many hospitalizations and deaths. Their rapid identification and characterization are paramount in control, management options, and treatment choices. Thus, this study aimed to characterize the cell surface properties of carbapenem-resistant (R) E. coli isolates and their interaction with glycan-coated magnetic nanoparticles (gMNPs) compared with carbapenem-susceptible (S) E coli . This study used two groups of bacteria: The first group included E. coli (R) isolates harboring carbapenemases and had no antibiotic exposure. Their initial gMNP–cell binding capacity, with cell surface characteristics, was assessed. In the second group, one of the E. coli (R) isolates and E. coli (S) had long-term serial antibiotic exposure, which we used to observe their cell surface characteristics and gMNP interactions. Initially, cell surface characteristics (cell morphology and cell surface charge) of the E. coli isolates were evaluated using confocal laser scanning microscope (LSCM) and a Zetasizer, respectively. The interaction of gMNPs with the E. coli isolates was assessed through LSCM and transmission electron microscope (TEM). Further, the gMNP–cell attachment was quantified as a concentration factor (CF) through the standard plating method. The results showed that the CF values of all E. coli (R) were significantly different from those of E. coli (S), which could be due to the differences in cell characteristics. The E. coli (R) isolates displayed heterogeneous cell shapes (rod and round cells) and lower negative zeta potential (cell surface charge) values compared to E. coli (S). Further, this research identified the differences in the cell surface characteristics of E. coli (S) under carbapenem exposure, compared to unexposed E. coli (S) that impact their attachment capacity. The gMNPs captured more E. coli (S) cells compared to carbapenem-exposed E. coli (S) and all E. coli (R) isolates. This study clearly found that differences in cell surface characteristics impact their interaction with magnetic nanoparticles. The gained insights aid in further understanding adhesion mechanisms to develop or improve bacterial isolation techniques and diagnostic and treatment methods for CRE.

Surface display of metal binding domain derived from PbrR on Escherichia coli specifically increases lead(II) adsorption

Hui Chang-Ye,Guo Yan,Yang Xue-Qin,Zhang Wen,Huang Xian-Qing

Objectives To improve the Pb 2+ biosorption capacity of the potential E. coli biosorbent, a putative Pb 2+ binding domain (PbBD) derived from PbrR was efficiently displayed on to the E. coli cell surface. Results The PbBD was obtained by truncating the N-terminal DNA-binding domain and C-terminal redundant amino acid residues of the Pb 2+ -sensing transcriptional factor PbrR. Whole-cell sorbents were constructed with the full-length PbrR and PbBD of PbrR genetically engineered onto the surface of E. coli cells using Lpp-OmpA as the anchor. Followed by a 1.71-fold higher display of PbBD than PbrR, the presence of PbBD on the surface of E. coli cells enabled a 1.92-fold higher Pb 2+ biosorption than that found in PbrR-displayed cells. Specific Pb 2+ binding via PbBD was the same as Pb 2+ binding via the full-length PbrR, with no observable decline even in the presence of Zn 2+ and Cd 2+ . Conclusions Since surface-engineered E. coli cells with PbBD increased the Pb 2+ binding capacity and did not affect the adsorption selectivity, this suggests that surface display of the metal binding domain derived from MerR-like proteins may be used for the bioremediation of specific toxic heavy metals.

Rapid Detection and Antimicrobial Susceptibility Testing of Pathogens Using AgNPs-Invertase Complexes and the Personal Glucose Meter

Zheng Laibao,Shen Yunqiu,Dong Wenjia,Zheng Chaochuan,Zhou Ruolan,Lou Yong-Liang

Rapid detection of pathogens and assessment of antimicrobial susceptibility is of great importance for public health, especially in resource-limiting regions. Herein, we developed a rapid, portable, and universal detection method for bacteria using AgNPs-invertase complexes and the personal glucose meter (PGM). In the presence of bacteria, the invertase could be released from AgNPs-invertase complexes where its enzyme activity of invertase was inhibited. Then, the enzyme activity of invertase was restored and could convert sucrose into glucose measured by a commercially PGM. There was a good linear relationship between PGM signal and concentration of E. coli or S. aureus as the bacteria model with high sensitivity. And our proposed biosensor was proved to be a rapid and reliable method for antimicrobial susceptibility testing within 4 h with consistent results of Minimum Inhibitory Concentrations (MICs) testing, providing a portable and convenient method to treat infected patients with correct antibiotics and reduce the production of antibiotic-resistant bacteria, especially for resource-limiting settings.

Efficient photocatalytic hydrogen production by E. coli-ZnIn2S4 biohybrid system

Feng Zhu,Guopeng Zhu,Shaoqing Zhang,Haibin Niu,Xiutao Yang,Xiaoming Li,Hongwen Yu

Constructed an E. coli -ZnIn 2 S 4 biohybrid for photocatalytic H 2 production. • Achieved a high H 2 yield of 335 μmol under optimized conditions. • Demonstrated a synergistic effect surpassing individual component performance. • Revealed improved charge separation at the bio-abiotic interface. • Identified light-induced redirection of microbial metabolic flux. Semiconductor-microorganism hybrid systems exhibit considerable potential for light-driven hydrogen production via water splitting. However, their performance is limited by the efficiency of electron transfer at the bio-abiotic interface and the stability of the hybrid system. In this study, we constructed a novel light-driven hybrid hydrogen production system utilizing Escherichia coli ( E. coli ) and ZnIn 2 S 4 (ZIS). E. coli interacts with ZIS surface through specific interfacial interactions facilitated by electrostatic adsorption and forms robust connections via metal-nonmetal chemical bonds. The E. coli -ZIS hybrid demonstrated a significantly enhanced hydrogen evolution activity compared to pure ZIS or E. coli systems alone, achieving a hydrogen yield of 335 μmol within 3 h. Metabolic analysis revealed that ZIS can comprehensively regulate the microbial metabolic network to accelerate substrate decomposition and drive the reaction toward hydrogen production. By leveraging the light-enhanced synergistic mechanism of E. coli metabolic hydrogen production, the E. coli -ZIS composite system achieves efficient and stable photocatalytic hydrogen production.

Metabolic engineering of E. coli for producing phloroglucinol from acetate

Yu Shengzhu,Guo Liwei,Zhao Luyao,Chen Zhenya,Huo Yixin

Phloroglucinol is a three-hydroxyl phenolic compound and has diverse physiological and pharmacological activities such as antivirus and anti-inflammatory activities. Chemical synthesis of phloroglucinol suffered from many drawbacks such as high cost and environmental pollution. To avoid the above issues, microbial phloroglucinol biosynthesis was successfully accomplished in this study, while the abundant and low-cost acetate was used as the main carbon source. Firstly, the toxicity of phloroglucinol was tested, and E. coli BL21(DE3) could tolerate 5 g/L phloroglucinol. The ability of phloroglucinol synthase (PhlD) for catalyzing malonyl-CoA to phloroglucinol was confirmed, and E. coli BL21(DE3) expressing PhlD and acetyl-CoA carboxylase (ACCase) could produce 1107 ± 12 mg/L phloroglucinol from glucose. Then, E. coli BL21(DE3) was engineered to utilize acetate to produce 228 ± 15 mg/L phloroglucinol. Then, the endogenous citrate synthase (GltA) which could catalyze oxaloacetate and acetyl-CoA generated from acetate to citrate was knocked down by CRISPRi system in order to enhance the carbon flux for phloroglucinol production, and the titer was improved to 284 ± 8 mg/L. This work demonstrated that acetate could be used as low-cost substrate to achieve the biosynthesis of phloroglucinol and provided an example of effective utilization of acetate.

Inactivation mechanism of E. coli O157:H7 under ultrasonic sterilization

Lin Lin,Xinlei Wang,Changzhu Li,Haiying Cui

Ultrasonic sterilization (US), as a promising non-thermal sterilization method, exhibits unique superiorities than traditional sterilization methods. In this study, the inactivation mechanism of E. coli O157:H7 under US was investigated in cucumber and bitter gourd vegetable juices. Results revealed that the US treatment showed good antibacterial ability in countering E. coli O157:H7. Through determinations of conductivity and β -galactosidase activity, significant augmentation in membrane permeability of the bacteria was confirmed after the US treatment. The morphologies of the US treated E. coli O157:H7 demonstrated that the integrity of the cell membrane was disrupted by US treatment. SDS-PAGE and LSCM data further proved the disruptive action of US, leading to the leakage of proteins and DNA through the breakage on cell membrane. The decrease of metabolic-related enzyme activity was verified through investigation of bacterial metabolism. The antibacterial mechanism analysis indicated that the US can generate free radicals which resulted in the rise of intracellular oxidative stress, attenuation of energy metabolism and inhibition of hexose monophosphate pathway. As the application verification, the US treatment can cause the deprivation of E. coli O157:H7 cell viability in vegetable juices without obvious impact on the sensory quality.

The hydrophobicity of lignin enhances E. coli survival during NaClO exposure

Weihong Xu,Haibei Li,Zhongwei Yang,Tianjiao Chen,Shuqing Zhou,Yue Zhao,Danyang Shi,Dong Yang,Junwen Li,Min Jin

High turbidity, attributed to suspended solids and colloidal particles, can lead to microbiological control failure during water disinfection. Lignin, a naturally occurring particle suspended in water, may contribute to turbidity. However, its ability to adsorb bacteria and protect them from chlorination remains poorly understood. In this study, we investigated Escherichia coli inactivation by NaClO in the presence and absence of natural particles. Among the five tested microparticles, lignin significantly enhanced E. coli survival under NaClO exposure, diminishing its bactericidal effect. Furthermore, surviving bacteria showed upregulation of gene expression related to energy metabolism and oxidative stress, as indicated by transcriptional analysis and qPCR measurements. These findings were further supported by increased ATP content, decreased ROS levels, and a prolonged lag phase in growth curves, enabling bacteria to rapidly restore their growth activity. Further analysis revealed that E. coli binds to the surface of lignin through hydrophobic interactions involving hydroxyl, methyl, aromatic rings, and ether groups. Lignin exhibited a strong bacterial adsorption capacity, as described by pseudo-second-order (PSO) kinetics and the Freundlich model, leading to reduced membrane permeability to NaClO. Importantly, the protective effect of lignin on bacteria was positively correlated with its hydrophobicity, with greater hydrophobicity resulting in enhanced bacterial protection. These findings suggest that lignin significantly increases E. coli resistance to chlorination, potentially contributing to disinfection failure in water treatment. This study highlights the role of lignin as a potential hazard that compromises microbiological control, offering new insights into chlorination failure caused by turbidity.

Replication of DNA Containing Mirror-Image Thymidine in E. coli Cells

Yuhe Kan,Lu Chen,Dao Lin,Xinya Bu,Mengwu Mo,Liang Yan,Zhenjun Yang,Longfei Yuan,Li Wu,Yujian He

DNA damage can occur naturally or through environmental factors, leading to mutations in DNA replication and genomic instability in cells. Normally, natural d-nucleotides were selected by DNA polymerases. The template l-thymidine (l-T) has been shown to be bypassed by several types of DNA polymerases. However, DNA replication fidelity of nucleotide incorporation opposite l-thymidine in vivo remains unknown. Here, we constructed plasmids containing a restriction enzyme (PstI) recognition site in which the l-T lesion was site-specifically located within the PstI recognition sequence (CTGCAG). Further, we assessed the efficiencies of nucleotide incorporation opposite the l-T site and l-T lesion bypass replication in vitro and in vivo. We found that recombinants containing the l-T lesion site inhibited DNA replication. In addition, A was incorporated opposite the l-T lesion by routine PCR assay, whereas preference for nucleotide incorporation opposite the l-T site was A (13%), T (22%), C (46%), and G (19%), and no nucleotide insertion and deletions were detected in E. coli cells. In particular, a novel restriction enzyme-mediated method for detection of the mutagenic properties of DNA lesion was established, which allows us to readily detect restriction–digestion of the l-T-bearing plasmids. The study provided significant insight into how mirror-image nucleosides perturb the fidelity of DNA replication in vivo and whether they elicit mutagenic effects, which may help to understand both how DNA damage interferes with the flow of genetic information during DNA replication and development of diseases caused by gene mutation.

Sonophotocatalytic inactivation of E. coli using ZnO nanofluids and its mechanism

Lingling Zhang,Huan Qi,Zhengxu Yan,Yu Gu,Weiqiang Sun,Abraham Amenay Zewde

The present study evaluated inactivation efficiency of a sonophotocatalytic process using ZnO nanofluids including ultrasonic parameters such as power density, frequency and time. The result showed that inactivation efficiency was increased by 20% when ultrasonic irradiation was combined with photocatalytic process in the presence of natural light. Comparison of inactivation efficiency in photocatalytic, ultrasonic and sonocatalytic processes using Escherichia coli as a model bacteria identified that inactivation efficiencies are shown in the following order: ultrasonic irradiation   <   sonocatalysis   <   photocatalysis   <   sonophotocatalysis. Furthermore, inactivation mechanism of sonophotocatalysis was proposed. Studies of reactive oxygen species (ROS) and zinc ions (Zn 2+ ) release evaluation revealed that ROS play a key role in bacterial inactivation rather than Zn 2+ . Permeability of outer membrane (OM) and inner membrane (IM) of E. coli bacterial cells were studied and exhibited that sonophotocatalysis increased the permeability of OM and IM significantly. The enhanced bacterial inactivation effect in sonophotocatalytic process contributed to acoustic cavitation, sonocatalysis of ZnO and sonoporation phenomenon.

NaBiF4 upconversion nanoparticle-based electrochemiluminescent biosensor for E. coli O157 : H7 detection

Danqing Liu,Xingxing Lv,Chaoyue Zhao,Jiayue Li,Jinmei Huang,Ling Weng,Liangcan He,Shaoqin Liu

Foodborne or water-borne pathogens pose great threats to human beings and animals. There is an urgent need to detect pathogens with cheap, rapid and sensitive point-of-care diagnostic assays. Herein, we report the electrochemiluminescent (ECL) behaviors of NaBiF4 : Yb3+/Er3+ upconversion nanoparticles (UCNPs) which were synthesized via a fast and environment-friendly method at room temperature for the first time. The UCNPs together with K2S2O8 exhibit high ECL intensity and stable cathodic signals. Further, the Au nanoparticles (Au NPs) and Anti-E. coli O157 : H7 antibody were assembled on the surface of UCNPs successively to construct a novel ECL immunosensor for the detection of deadly E. coli O157 : H7. The as-prepared ECL immunosensor reveals high sensitivity to E. coli O157 : H7 in a linear range of 200–100 000 CFU mL−1, and the minimum detection limit could reach up to 138 CFU mL−1. The designed UCNP-based biosensor demonstrates high specificity, good stability and remarkable repeatability, and the strategy will provide a sensitive and selective method for rapid detection of E. coli O157 : H7 in food safety and preclinical diagnosis.

Nanofiber-based colorimetric platform for point-of-care detection of E. coli

Jiaqi Zhang,Christopher Hurren,Zhentan Lu,Dong Wang

Accurate and rapid detection of bacteria in complex environmental samples using simple and portable devices is still a major challenge. This research presents a simple nanofiber-based platform for highly sensitive colorimetric/fluorometric detection of Escherichia coli ( E. coli ). Nanofiber membranes (NFM) were loaded with target molecules (fluorescent and chromogenic substrate) via chemical modification to prepare functional NFM (NFM-MUG and NFM-XG). The β -glucuronidase secreted by E. coli during the metabolic process triggered the functionalized NFM to produce biological signals and color change. The intensity of bio-signals and color was shown to enable quantitative and qualitative detection of E. coli . The nanofiber-based platforms exhibited high stability and a wide detection range (10 2 -10 7 CFU/mL). The limit of detection (LOD) of NFM-MUG and NFM-XG sensors for E. coli were 26 and 69 CFU mL −1 , respectively. The sensing time required for NFM-MUG and NFM-XG was 15 mins and 30 mins respectively. The nanofiber-based platforms also exhibited high specificity and low interference from ionic compounds and pH changes. Notably, this assay was easily combined with a smartphone app as a portable device for on-site detection of E. coli , showing a great promise in the field of environmental and food safety testing.

A Novel Postbiotic From Lactobacillus rhamnosus GG With a Beneficial Effect on Intestinal Barrier Function

Gao Jie,Li Yubin,Wan Yu,Hu Tongtong,Liu Liting,Yang Shaojie,Gong Zelong,Zeng Qing,Wei Yi,Yang Weijun,Zeng Zhijie,He Xiaolong,Huang Sheng-He,Cao Hong

It has long been known that probiotics can be used to maintain intestinal homeostasis and treat a number of gastrointestinal disorders, but the underlying mechanism has remained obscure. Recently, increasing evidence supports the notion that certain probiotic-derived components, such as bacteriocins, lipoteichoic acids, surface layer protein and secreted protein, have a similar protective role on intestinal barrier function as that of live probiotics. These bioactive components have been named ‘postbiotics’ in the most recent publications. We previously found that the Lactobacillus rhamnosus GG (LGG) culture supernatant is able to accelerate the maturation of neonatal intestinal defense and prevent neonatal rats from oral Escherichia coli K1 infection. However, the identity of the bioactive constituents has not yet been determined. In this study, using liquid chromatography-tandem mass spectrometry analysis, we identified a novel secreted protein (named HM0539 here) involved in the beneficial effect of LGG culture supernatant. HM0539 was recombinated, purified, and applied for exploring its potential bioactivity in vitro and in vivo. Our results showed that HM0539 exhibits a potent protective effect on the intestinal barrier, as reflected by enhancing intestinal mucin expression and preventing against lipopolysaccharide (LPS)- or tumor necrosis factor α (TNF-α)-induced intestinal barrier injury, including downregulation of intestinal mucin (MUC2), zonula occludens-1 (ZO-1) and disruption of the intestinal integrity. Using a neonatal rat model of E. coli K1 infection via the oral route, we verified that HM0539 is sufficient to promote development of neonatal intestinal defense and prevent against E. coli K1 pathogenesis. Moreover, we further extended the role of HM0539 and found it has potential to prevent dextran sulfate sodium (DSS)-induced colitis as well as LPS/D-galactosamine-induced bacterial translocation and liver injury. In conclusion, we identified a novel LGG postbiotic HM0539 which exerts a protective effect on intestinal barrier function. Our findings indicated that HM0539 has potential to become a useful agent for prevention and treatment of intestinal barrier dysfunction- related diseases.
