Effects of tea polyphenol and EDC/NHS cross-linking treatments on the storage quality of ready-to-eat sea cucumbers at room temperature: Insights into the underlying interaction mechanisms

Jiarun Gao,Le Yu,Xiang Wan,Yuxue Shan,Jingfang Zhang,Mingbo Li,Leilei Sun

Ready-to-eat sea cucumbers (RSC) cannot be preserved at room temperature due to severe autolysis. This study systematically examined the quality-preserving effects of tea polyphenol (TP) and N-(3-Dimethylaminopropyl)-N′-ethylcarbodiimide hydrochloride/N-hydroxysuccinimide (EDC/NHS), two cross-linking treatments, on RSC during storage at room temperature. The results demonstrated that both cross-linking treatments significantly delayed the deterioration of quality and exhibited antimicrobial effects, accompanied by enhanced the structural and thermal stability of collagen in RSC. Microstructural analysis further confirmed that TP and EDC/NHS strengthened the collagen fibers. Molecular docking simulations suggested that TP interacts with collagen primarily through non-covalent binding, whereas EDC/NHS achieves positional localization via electrostatic attraction, subsequently reinforcing the collagen structure through covalent amide bonds. Both strategies effectively slowed the deterioration of RSC during storage at room temperature, thereby providing a theoretical foundation for the development of advanced preservation technologies for RSC.

Impact of surface biofunctionalization strategies on key effector cells response in polyacrylamide hydrogels for bone regeneration

Yi Zhang,Jinjun Dai,Ruiyue Hang,Xiaohong Yao,Long Bai,Di Huang,Ruiqiang Hang

Despite the clinical prevalence of various bone defect repair materials, a full understanding of their influence on bone repair and regeneration remains elusive. This study focuses on poly(acrylamide) (PAAm) hydrogels, popular 2D model substrates, which have regulable mechanical properties within physiological. However, their bio-inert nature requires surface biofunctionalization to enhance cell-material interactions and facilitate the study of bone repair mechanisms. We utilized PAAm hydrogels of varying stiffness (18, 76 and 295 kPa), employed sulfosuccinimidyl-6-(4′-azido-2′-nitropheny-lamino) hexanoate (sulfo-SANPAH) and N-(3-dimethylaminopropyl)-N-ethylcarbodiimide hydrochloride/N-hydroxysuccinimidyl acrylate (EDC/NHS) as crosslinkers, and cultured macrophages, endothelial cells, and bone mesenchymal stem cells on these hydrogels. Our findings indicated that sulfo-SANPAH's crosslinking efficiency surpassed that of EDC/NHS, irrespective of pore size and stiffness. Importantly, we observed that the stiffness and surface biofunctionalization method of hydrogels significantly impacted cell adhesion and proliferation. The collagen-modified hydrogels by EDC/NHS strategy failed to support the normal biological behavior of bone mesenchymal stem cells and hindered endothelial cell spreading. In contrast, these modified hydrogels by the sulfo-SANPAH method showed good cytocompatibility with the three types of cells. This study underscores the critical role of appropriate conjugation strategies for PAAm hydrogels, providing valuable insights for hydrogel surface modification in bone repair and regeneration research.

Synthesis and structural characterization of pyrroloquinoline quinone grafted carboxymethyl chitosan copolymer with neuroprotective potential

Shengnan Zheng,Chuzhou Wen,Jiaxue Tang,Changkai Sun,Fengtao Wang,Shui Guan

Pyrroloquinoline quinone (PQQ) is a redox cofactor that has been shown to have strong antioxidant capacity and neuroprotective effects and has been extensively studied in recent years. However, PQQ small molecule compounds have poorly stability and low bioavailability in vivo, making it difficult to exert drug activity for long periods of time. In this study, we employed the EDC/NHS method to graft PQQ onto carboxymethyl chitosan (CMCS) to improve the utilization of PQQ. The obtained PQQ-g-CMCS product was a water-soluble copolymer, and the grafting rate could be controlled by adjusting the pH and the raw material feeding ratio. The PQQ-g-CMCS samples were characterized using FT-IR, UR-Vis, NMR, and XRD spectroscopy, suggesting a conjugation reaction between the amino groups of CMCS and the carboxyl groups of PQQ. Specifically, the in vitro antioxidant experiments showed that the Fe2+ chelating activity of the grafted copolymer was significantly enhanced. In addition, the PQQ-g-CMCS copolymer also exhibited good biocompatibility and significant protective effect against H2O2-induced oxidative damage in SH-SY5Y cells. These results preliminarily indicate that PQQ-g-CMCS may be a promising antioxidant material for applications such as drug release and tissue engineering.

Covalent grafting of corn stover cellulose nanofibers with zein for pickering emulsion stabilization and controlled quercetin delivery

Xinling Wu,Qiang Zhang,Deyi Zhou,Jianpeng Dou,Qing Liang,Li Zhang,Haigen Zhou,Xiaodi Gao,Xuelin Zhao,Jinsong Zhang

Enhancing hydrophobic bioactives' bioaccessibility remains challenging in functional foods due to instability and insufficient controlled-release ability in conventional protein-polysaccharide carriers. We pioneer a new interaction model by covalently grafting corn stover cellulose nanofibers (CNF) with Zein using N-(3-dimethylaminopropyl)-N′-ethyl carbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS), creating conjugates with gradient grafting degrees (CNF/Zein 0.5, CNF/Zein 1, and CNF/Zein 2). Excitingly, compared to the non-covalent complex (CNF-Zein), conjugates exhibited a stable network-like microstructure, lower crystallinity, and enhanced thermal stability. Furthermore, the conjugates exhibited excellent amphiphilicity, forming denser and more stable emulsions. Confocal laser scanning microscope (CLSM) revealed a dual stabilization mechanism of “interface adsorption-network encapsulation”. Additionally, CNF/Zein-stabilized emulsions demonstrated controlled quercetin release and improved bioaccessibility during in vitro digestion tests, with the highest grafted CNF/Zein 2 showing 43.65 % bioaccessibility. This study first demonstrates the effectiveness of the EDC/NHS technique in alcohol-solution systems, providing novel strategies for conjugate design and lipophilic nutraceutical delivery.

Dual-network hydrogel based on ionic nano-reservoir for gastric perforation sealing

Yuan Yuehui,Wu Hao,Ren Xueyang,Wang Jianwu,Liu Ruiqing,Hu Benhui,Gu Ning

In recent years, remarkable progress has been made in the research of injectable hydrogel for internal tissue healing. However, the therapeutic outcome is usually limited when the hydrogel is used for the treatment of gastric perforation due to the high acidic gastric juice and violent deformation of the gastric wall. Regarding these challenges, we proposed an ionic nano-reservoir (INR)-based dual-network hydrogel, which has excellent adhesion and mechanical properties, and can be easily applied to the perforation site to block the perforation while promoting tissue repairing. The results showed that the first network made of polyacrylamide had cross-linked on the stomach tissue within 5 s under blue light, and enhanced the adhesion performance through mechanical interlock. The nano-hydroxyapatite acted as ionic INR, which can gradually release Ca2+ under acid environments to form the second network with sodium alginate and inhibit the swelling of hydrogel in gastric juice. Meanwhile, the adhesion was further enhanced through amide covalent bonds at the hydrogel-tissue interface with the presence of 1- ethyl-3-(3-dimethylaminopropyl)carbodiimide/N-hydroxysuccinimide (EDC/NHS). The dual network hydrogels obtained by the INR strategy could be employed as a potential therapeutic option for gastric perforation and other similar biomedical prolems.

Preparation, characterization and antioxidant activity of protocatechuic acid grafted carboxymethyl chitosan and its hydrogel

Chao Xu,Shui Guan,Jianqiang Xu,Weitao Gong,Tianqing Liu,Xuehu Ma,Changkai Sun

In this study, protocatechuic acid (PCA) was grafted onto carboxymethyl chitosan (CMCS) via EDC/NHS to improve the antioxidant effect. The grafting ratio of PCA-g-CMCS conjugates could be controlled by adjusting the pH value and feed ratio of raw materials. The conjugates exhibited similar pH sensitivity to CMCS and showed dramatic enhancements of DPPH and ABTS radicals scavenging activities, total antioxidant capacity, reducing power, and Fe 2+ -chelating activity. Three-dimensional porous PCA-g-CMCS hydrogel was prepared by lyophilization and secondary cross-linking. The shaped hydrogel preserved its antioxidant activity, and the sustained release of PCA-containing degraded fragment from biodegradable hydrogel could be achieved with the aid of lysozyme in vitro (15 days). PCA-g-CMCS hydrogel also showed excellent biocompatibility and protective effect on H 2 O 2 -induced oxidative damage in SH-SY5Y cells. These results suggested that PCA-g-CMCS conjugates and its hydrogel would appear to be a promising oxidation-resistant material for applications such as drug release and tissue engineering.

Macroscopic assembly behavior of silk/lignin-based adhesive hydrogels: elementary macroscopic assembly

Qinfu Lu,Xinyue Wang,Miaoya Zhang,Zhen Wang,Xianan Qin,Baochang Sun,Shanshan Lv

Macroscopic assembly has been investigated to construct appealing materials. But it seems a question remains to be answered, i.e., is macroscopic assembly always one by one? In this study, a new kind of silk-based hydrogels were developed through incorporating lignin-related compounds via the 1-ethyl-3-(3-pyl)-carbodiimide/N-hydroxysuccinimide (EDC/NHS)-mediated coupling chemistry. Thanks to the hydrogen bond, hydrophobic and π-π interactions, the hydrogels processed capability of adhesive and macroscopic assembly, which could be further tuned by introducing iron ions, as Fe^(3+) can form coordination bonds with lignin. Examination on the three-body macroscopic assembly behavior of the silk/lignin-based hydrogels was conducted as an approach towards answering the above question. Under experimental conditions tested in this study, the answer is yes. Yet it would be reckless to draw a definitive conclusion to the question. This study intended to draw attention to this matter and evoke more interests and investigations using various macroscopic assembly systems.

Preparation and evaluation of an innovative antibacterial bi-layered composite dressing for skin wound healing

Leilei Sun,Mingbo Li,Tengfei Gong,Jianling Feng

Aim of the study The aim of the current study was to develop collagen-based bi-layered composite dressings with antibacterial property and evaluate the efficiency for wound healing. Materials and methods A bi-layered composite wound dressing was fabricated using two marine biomacromolecules (collagen and chitosan or carboxymethyl chitosan). Non-crosslinked and N-Ethyl-N'-(3-dimethylaminopropyl) carbodiimide/N-Hydroxy succinimide (EDC/NHS) cross-linked collagen sponges fabricated by vacuum freeze-drying technology was used as the inner layer. The medical spun-laced nonwoven coated with chitosan and carboxymethyl chitosan was used as the outer layer. The antibacterial activities against E. coli and S. aureus were evaluated by the inhibition zone assay. Deep second-degree scald model was performed to evaluate the efficiency of bi-layered composite dressings for wound healing. Results In view of comprehensive evaluation of appearance and in vitro antibacterial activity, medical spun-laced nonwoven coated with 3% of chitosan solution was chosen to be used as the optimized preparation conditions to produce the outer layer of composite dressing, which acted as a barrier against microorganisms and provided mechanical support. Furthermore, the results of wound closure and histopathological analysis indicated that EDC/NHS cross-linked collagen-based bi-layered composite dressing was superior to non-crosslinked and commercial products, which stimulated the wound healing process and accomplished deep second-degree scalded skin healing within a time span of 28 days. Conclusion The EDC/NHS cross-linked collagen-based bi-layered composite dressing had immense potential to be applied for an ideal wound dressing for more efficient and faster wound healing. Therefore, the findings provided the essential theoretical basis for great potential of collagen-based composite dressing used in wound healing applications.

Preparation and Optimization of a Biomimetic Triple-Layered Vascular Scaffold Based on Coaxial Electrospinning

Hu Qingxi,Wu Chuang,Zhang Haiguang

Electrospinning is a promising method for preparing bionic vascular scaffolds. In particular, coaxial electrospinning can encapsulate polymer materials in biological materials and provide vascular scaffolds with good biomechanical properties. However, it is difficult to produce a stable Taylor cone during the coaxial electrospinning process. Moreover, glutaraldehyde cross-linked natural biomaterials are cytotoxic. To address these issues, a novel electrospinning process is proposed in this report. A non-ionic surfactant (Tween 80) was added to poly(lactic-co-glycolic acid) electrospinning solution and gelatin-collagen electrospinning solution, which prevented the interfacial effect of coaxial electrospinning due to different core/shell solutions. The as-prepared materials were then cross-linked with the non-toxic coupling agents N-(3-dimethylaminopropyl)-N′-ethylcarbodiimide/N-hydroxysuccinimide (EDC/NHS). By comparing the biomechanical properties of EDC/NHS cross-linked vascular scaffold with glutaraldehyde vapor–cross-linked vascular scaffold, it was found that the fracture strain and biological performance of EDC/NHS cross-linked vascular scaffold were better than those of the glutaraldehyde cross-linked scaffold. Finally, a three-layer bionic vascular scaffold was prepared by the proposed electrospinning process. Biomechanical performance tests were carried out and the prepared scaffold was found to meet the requirements of tissue-engineered blood vessels. The research in this paper provides a useful reference for the preparation and optimization of vascular scaffolds.

Comprehensive assessment of Nile tilapia skin collagen sponges as hemostatic dressings

Leilei Sun,Bafang Li,Wenkui Song,Kai Zhang,Yan Fan,Hu Hou

Nile tilapia skin collagen sponge was fabricated by lyophilization and cross-linked with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide in the presence of N-hydroxysuccinimide (EDC/NHS). The physicochemical properties were examined. The EDC/NHS cross-linked collagen sponge presented an enhanced water absorption capacity. In addition, biocompatibility and hemostatic efficiency were evaluated by acute systemic toxicity assay, dermal irritation test, intradermal reaction test, sensitization test, cytotoxicity, blood clotting assay in vitro , and liver and femoral artery hemorrhage models in vivo . Results showed that the produced collagen sponges before and after EDC/NHS cross-linking had excellent biocompatibility. Furthermore, EDC/NHS cross-linking promoted fibroblast cells viability and proliferation reflected by the MTT reduction assay. Meanwhile, EDC/NHS cross-linked collagen sponge exhibited the best blood clotting ability and hemostatic efficiency in rat femoral artery hemorrhage model in comparison with non-crosslinked and commercial collagen sponges. Our results demonstrated that the fabricated collagen sponges could be used as perfect hemostatic dressings.

A novel detergent-based decellularization combined with carbodiimide crosslinking for improving anti-calcification of bioprosthetic heart valve

Yu Luo,Shenyu Huang,Lie Ma

Currently, valve replacement surgery is the only therapy for the end-stage valvular diseases because of the inability of regeneration for diseased heart valves. Bioprosthetic heart valves (BHVs), which are mainly derived from glutaraldehyde (GA) crosslinked porcine aortic heart valves or bovine pericardium, have been widely used in the last decades. However, it is inevitable that calcification and deterioration may occur within 10–15 years, which are still the main challenges for the BHVs in clinic. In this study, N-Lauroylsarcosine sodium salt (SLS) combined with N-(3-Dimethylaminopropyl)-N'-ethylcarbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS) were utilized to decellularize and crosslink the heart valves instead of GA treatment. The obtained BHVs exhibited excellent extracellular matrix stability and mechanical properties, which were similar with GA treatment. Moreover, the obtained BHVs exhibited better in vitro biocompatibilities than GA treatment. After subcutaneous implantation for 30 d, the obtained BHVs showed mitigated immune response and reduced calcification compare with GA treatment. Therefore, all the above results indicated that the treatment of SLS-based decellularization combined with EDC/NHS crosslink should be a promising method to fabricate BHVs which can be used in clinic in future.

Construction of an enzyme-based all-fiber SPR biosensor for detection of enantiomers

Zhuoyue Zhou,Zhao Yang,Li Xia,Houjin Zhang

Chiral analysis of amino acids (AAs) is of great importance in medical science due to the distinctive effect of AA isomers on human health. Although various chiral recognition techniques have been developed, the quantitative chiral recognition of low-level AA isomers remains challenging. Here, we combined the fiber optic SPR with an enzyme-substrate recognition mechanism to construct a direct-assay-type chiral AA biosensor. As a proof-of-concept attempt, a recently discovered Rasamsonia emersonii D-amino acid oxidase (ReDAAO) with a wide substrate spectrum and high stability was immobilized on the graphene oxide and gold nanorods composites (GO-AuNRs), using both EDC/NHS coupling and the gold-binding peptide (GBP) method. Such a biosensor can distinguish two AA isomers at the same concentration. It achieved specific detection of D-amino acids (D-AAs) with a linear range from 5x10 -4 mM to 30 mM. Furthermore, it showed good resistance to enantiomeric interference. When the percentage of D-AA increases in the isomer mixture, a good linear relationship between the D/(D + L)-AA ratio and SPR spectral shift was obtained. This unique combination of the enzyme , nanocomposite , and SPR taps into the rich reservoir of proteins for chiral receptors. It lays the foundation for protein-based chiral recognition of other clinically important small molecules in future biosensor designs.
