In this study, we encapsulated epigallocatechin gallate (EGCG) in emulsified oil droplets of protein-stabilized emulsions for the first time, with the participation of peppermint essential oil (PEO). Gum arabic (GA) and sodium caseinate (SC) were sufficiently crosslinked to construct the oil-water interface of the emulsion, improving the stability of EGCG in the emulsified oil droplets. When SC and 4 g/kg GA were mixed for 12 h before emulsification, the protein adsorption at the oil-water interface was significantly enhanced. Compared with the SC-stabilized emulsion, the retention of EGCG in the emulsion stabilized by mixing SC and 4 g/kg GA for 12 h increased about 2 and 33 times, respectively, after thermal treatment (30 min at 90 °C) and storage 10 weeks at 4 °C. Moreover, the emulsion stabilized by SC and 4 g/kg GA was resistant to the ionic strength of 0–0.6 mol/L and had predominantly elastic characteristics in the rheological properties. This indicates that the stability of active ingredients in emulsified oil droplets can be improved by regulating the rheological properties of the oil-in-water emulsion. The study is of great value for the efficient encapsulation and stability improvement of hydrophilic active ingredients in emulsified oil droplets based on emulsion systems.

This study aimed to synthesize and characterize L-epigallocatechin gallate (EGCG) complexed Mn2+ nanoparticle (L-EGCG-Mn), a proof-of-concept pH-sensitive manganese core nanoparticle (NP), and compare its magnetic resonance (MR) properties with those of Gd-DTPA, both in vitro and in vivo. Reverse microemulsion was used to obtain the L-EGCG-Mn NPs. The physicochemical properties of L-EGCG-Mn were characterized using dynamic light scattering, transmission electron microscopy, and near-infrared fluorescence small animal live imaging. The in vitro relaxivity of L-EGCG-Mn incubated with different pH buffer solutions (pH = 7.4, 6.8, 5.5) was evaluated. The T1-weighted MR imaging (MRI) properties were evaluated in vitro using hypoxic H22 cells as well as in H22 tumor-bearing mice. Cytotoxicity tests and histological analysis were performed to evaluate the safety of L-EGCG-Mn. L-EGCG-Mn showed good biocompatibility, stability, pH sensitivity, and tumor-targeting ability. Moreover, when the pH was decreased from 7.4 to 5.5, the r1 relaxivity of L-EGCG-Mn was shown to gradually increase from 1.79 to 6.43 mM−1·s−1. Furthermore, after incubation with L-EGCG-Mn for 4 h, the T1 relaxation time of hypoxic H22 cells was significantly lower than that of normoxic H22 cells (1788 ± 89 vs. 1982 ± 68 ms, p=.041). The in vivo analysis showed that after injection, L-EGCG-Mn exhibited a higher MRI signal compared to Gd-DTPA in H22 tumor-bearing mice (p < .05). Furthermore, L-EGCG-Mn was found to have a good safety profile via cytotoxicity tests and histological analysis. L-EGCG-Mn has a good safety profile and pH sensitivity and may thus serve as a potential MRI contrast agent.

Although different delivery systems have been developed to overcome the limits of Epigallocatechin-3-gallate (EGCG) and quercetin in food application, none have referred to their simultaneous encapsulation. In this study, these two polyphenols were successfully co-loaded into liposomes. Under the optimal conditions (lecithin-total polyphenols ratio 25:1, lecithin-cholesterol ratio 6:1, lecithin-Tween 80 ratio 8:1 and ultrasonic time 2 min), the mean size, polydispersity index (PDI) and zeta potential of liposomes were 111.10 ± 0.52 nm, 0.259 ± 0.006 and –19.83 ± 0.45 mV, with an encapsulation efficiency of 64.05 ± 1.56% and 61.73 ± 2.55% for EGCG and quercetin, respectively. After 30-day storage, an increase of 4.05% was observed in the mean size with no significant change ( P ≥ 0.05) in the PDI and zeta potential. Moreover, 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay revealed a synergistic antioxidant effect of the two compounds in liposomal system. These results demonstrated that co-encapsulation of EGCG and quercetin enhances their effectiveness. Practical Application EGCG and quercetin are natural polyphenols abound in the human diet with diverse biological activities. These two polyphenols were successfully co-encapsulated into a homogeneous and stable liposomal system. Interestingly, a synergistic antioxidant effect of the two polyphenols was observed due to co-encapsulation. This indicated that the simultaneous delivery of EGCG and quercetin was an attractive approach to improve their functionality for expanding their application in food, cosmetic and pharmaceutical industries.

Pathogenic bacteria and biofilm formation have become the urgent threats to global public health. Herein, metal-epigallocatechin gallate (EGCG) network with improved bactericidal effect and pathogenic biofilm eradication ability via trivalent cationic Fe 3+ catalyzed hydrogen peroxide (H 2 O 2 ) decomposition is developed. EGCG, the predominant catechin from tea, is predicted to be a good chelator for eight metal ions (i.e., Al 3+ , Cr 3+ , Cu 2+ , Co 2+ , Zn 2+ , Mn 2+ , Ni 2+ , Fe 3+ ). 3,3',5,5'-tetramethylbenzidine oxidation experiment using UV–vis spectrometer demonstrated that Zn 2+ , Mn 2+ , Ni 2+ are the potential candidates to accelerate the generation of H 2 O 2 by EGCG. Compared with the other two metal-EGCG network (Zn 2+ -EGCG or Mn 2+ -EGCG), only Ni 2+ -EGCG network has better bactericidal property and plays a synergistic role in the network system. Furthermore, by introducing Fe 3+ to Ni 2+ -EGCG network, the complex exhibits the best bactericidal property for gram-negative bacteria Escherichia coli ( E. coli ) and Pseudomonas aeruginosa (PAO1), which can be explained by the rapid H 2 O 2 decomposition and associated production of hydroxyl radicals. With SYTO 9/PI staining assay, Fe 3+ as a catalytic mediator in Ni 2+ -EGCG network shows the most significant bactericidal effect and pathogenic biofilm eradication ability. According to the above analysis, Ni 2+ -EGCG network presents a promising nanoplatform as an alternative to antibiotics for eradication of pathogenic bacteria and biofilms.

Tea polyphenol of epigallocatechin-3-gallate (EGCG) has been verified to possess multiple biological activities. Interleukin-23 (IL-23) is a heterodimeric cytokine consisting of two subunits of IL-23p19 and IL-12p40, with the functionality in regulating the production of cytokines under physiological or pathological conditions. By serendipity, the raised expression of IL-23 was observed after treating cells with EGCG, whereas the detailed mechanism remains poorly understood. This study was proposed to investigate the signaling related to EGCG-induced IL-23. The raised expression of IL-23 was confirmed primarily by intraperitoneally injecting with different concentrations of EGCG (0, 20, 50, 80 mg/kg) into BALB/c mice, and the raised expression was confirmed by quantitative real-time polymerase chain reaction (qRT-PCR) and western blot. Results from enzyme-linked immunosorbent assay (ELISA) revealed the increase of IL-23 in serum from 116.09 to 153.90 pg/mL after treating with EGCG. The same results were also observed in RAW264.7 and peritoneal macrophages after treating with EGCG (0, 1, 5, 10, 25 μM) with the increased tendency of IL-23 in cultural medium (7.98 to 25.38 pg/mL for RAW264.7; 3.64 to 260.93 pg/mL for peritoneal macrophages). After preliminary exploration of the signaling related to the increased IL-23, the classical signaling pathways and key transcription factors, such as nuclear factor kappa-B (NF-κB), mitogen-activated protein kinase (MAPK) signaling pathways, and interferon regulatory factor 5 (IRF5), were demonstrated with no relevant contribution. A further study revealed the involvement of the key transcription factor of BATF2, which could antagonistically modulate the transcription and translation of IL-23. The signaling of STAT3-BATF2-c-JUN/ATF2-IL-23 has been further verified in RAW264.7 macrophages using the STAT3 inhibitor of AG490 and the activator of Colivelin TFA. The results indicated that EGCG inhibits the phosphorylation of STAT3 to facilitate the decreased level of BATF2, which contributed to the increased level of IL-23 by the enhancing heterodimerization of c-JUN and ATF2.

Periodontitis is a chronic inflammatory disease characterized by oxidative stress, immune system imbalance, and the progressive destruction of periodontal tissues. Traditional treatment approaches are limited by the incomplete eradication of pathogens, the risk of antibiotic resistance, inadequate control of inflammation and oxidative stress, and restricted tissue regeneration capacity. Therefore, this study proposes an injectable multifunctional Laponite/gelatin hydrogel loaded with epigallocatechin gallate (EGCG)-copper ion (Cu 2+ ) complexes as a localized therapy for periodontitis. EGCG exhibits antioxidant, anti-inflammatory, and antimicrobial properties; however, its clinical application is hindered by poor stability and bioavailability. Cu 2+ coordination enhances EGCG stability and antioxidant capacity while improving its antimicrobial efficacy. Experimental results demonstrate that the Laponite/gelatin hydrogel is adaptable for the localized delivery of EGCG-Cu, with bioactive metal ions such as Li + , Mg 2+ , and Si 4+ contained in Laponite, which promote osteogenesis and periodontal tissue regeneration. In vitro and in vivo studies confirm that this hydrogel exhibits excellent biocompatibility, effectively inhibits Porphyromonas gingivalis , suppresses M1 polarization while promoting M2 polarization, and facilitates periodontal tissue repair. Therefore, this study provides promising insights into a localized therapeutic strategy for periodontitis.

The activation of M1-type macrophages are dominant cells secreting proinflammatory present within the inflamed synovium in the progression of osteoarthritis (OA). Increased oxidative stress, such as redundant ROS and hydrogen peroxide (H 2 O 2 ), are important factors in driving macrophages to polarize into M1 type. In this study, metal-polyphenol nanoformulations (Cu-Epigallocatechin-3-gallate (Cu-EGCG) nanosheets) were synthesized through the coordination interaction between EGCG and copper ions, which possessed the antioxidant effect of EGCG and anti-inflammatory of Cu 2+ . Results showed that Cu-EGCG nanosheets were biocompatible and the Cu 2+ could be sustained released from the nanoparticles. Cu-EGCG nanosheets with multienzyme-like antioxidative activity could effectively scavenge the excessive intracellular ROS, leading to significantly decreased expression of the pro-inflammatory cytokines, which could reduce the expression of M1-type macrophages and exhibit excellent promotion on shifting macrophages to M2 phenotypes. Moreover, the secreted factor from the cell supernatant of Cu-EGCG treated macrophages exhibited anti-inflammatory potential in chondrocytes of inflamed synovial joints. This study suggests a novel strategy for OA therapy by using metal-polyphenol nanoformulations targeting macrophages.

The concept of “synergy” and its applications has rapidly increased in the food industry as a practical strategy to preserve and improve health-promoting effects of the functional ingredients. In this study, hydrophilic epigallocatechin-3-gallate (EGCG) and lipophilic lycopene (LYC) were loaded separately (EGCG loaded PDE (PE), and LYC loaded PDE (PL), and also co-delivered (PEL) within a pickering double emulsion (PDE) system to induce synergistic hypolipidemic effect. Their effects on the serum lipid profile, weight of tissue fats, liver lipid droplet accumulation, and liver steatosis in a high-fat diet rat model were investigated. Moreover, different pathways (HMG-CoR, LDL-R, PPARγ and AMPK) involved in triggering hypolipidemic effects were verified at both mRNA and protein levels. The study's findings showed that inclusion of EGCG improved while LYC negatively affected the physical stability of PDE. The contents of total cholesterol and triacylglyceroles in serum and liver as well as the weight of tissue fats were substantially reduced in all groups (PE/PL/PEL). The alteration in absorption site of EGCG and enhanced bioaccessibility of LYC when delivered by PDE strengthened the hypolipidemic properties of PE and PL, mainly through triggering the pathways of HMG-CoR, LDL-R and PPARγ. Furthermore, the synergistic hypolipidemic effect of PEL was achieved mainly through the activation of the AMPK pathway.

Oxidative stress induced by reactive oxygen species (ROS) can adversely affect tissue repair, whereas endowing biomaterials with antioxidant activity can improve the in vivo microenvironment, thereby promoting angiogenesis and osteogenesis. Accordingly, this study utilized epigallocatechin-3-gallate (EGCG), a material known for its reducing properties, oxidative self-polymerization capability, and strong binding characteristics, to modify a bioactive core–shell fibrous membrane (10RP-PG). Compared to the 10RP-PG fibrous membrane, the EGCG-modified fibrous membrane (E/10RP-PG) exhibited superior hydrophilicity, excellent cell adhesion, and compatibility. Moreover, the EGCG-modified fibrous membrane can effectively scavenge free radicals, ameliorate the local microenvironment, and foster angiogenesis (enhancing the expression of angiogenic genes in human umbilical vein endothelial cells (HUVECs) by 1.58 times and promoting vascular generation area upon subcutaneous implantation by 4.47 times). The enhancement of angiogenic activity of the E/10RP-PG fibrous membrane further promoted cartilage degeneration and absorption, as well as new bone formation, thus facilitating the repair of bone defects. This study provides a new strategy for promoting bone defect repair through the surface modification of biomaterials with an antioxidant agent, and the fabricated E/10RP-PG fibrous membranes show promise for guiding vascularized bone regeneration.

Curcumin (Cur) and (−)-Epigallocatechin gallate (EGCG) have synergistic bioactivity in several terms, but their polar differences and environmental sensitivity make co-utilization challenging. Soy protein fibrils (UF) have sparked broad attention as outstanding carriers for bioactive substances. In this study, EGCG was applied to remodel the structure of Cur-loaded UF complex (UFC) to form novel UF-based hydrogels co-encapsulated Cur and EGCG (UFCE). The fluorescence spectroscopy, ITC , and FTIR analysis demonstrated that EGCG absorbed and deposited on the surface of UFC through hydrophobic and electrostatic interactions. The binding of EGCG led to the formation of UFCE, which constructed a stable and uniform gel network during heat treatment. UFCE gels formed with varying EGCG/protein concentration ([E]/[F]) ratios showed different structural properties with different encapsulation effects. At 2.5% protein concentration, UFCE gels formed at [E]/[F] ratio of 0.08 (UFCE 0.08 ) exhibited better gelling properties. The encapsulation efficiency of Cur and EGCG in UFCE 0.08 gel was up to 97.71% and 91.02%, respectively. Moreover, UFCE 0.08 gel exhibited significantly enhanced stability of Cur (95.27%) and EGCG (70.27%) than that of free Cur (53.13%) and free EGCG (13.32%) after thermal treatment at 85 °C for 120 min. UFCE 0.08 gel was also effective in improving the UV light stability of Cur (88.70%) and EGCG (72.96%). Furthermore, the binding of EGCG had pronouncedly boosted the anticancer activity of Cur in the UFCE 0.08 gel against Caco-2 cells. This study provides new perspectives into the fibril-polyphenol hydrogel systems for the co-encapsulation of EGCG and Cur, which has substantial potential for health-promoting applications.

Nano-based immunotherapy of therapeutic biomolecules is attractive but tremendously hampered by the poor delivery efficiency. This study reports a novel delivery system of fluorinated-coordinative-epigallocatechin gallate (EGCG), referring as FEGCG/Zn, through the integration of fluorination and zinc ions (Zn 2+ ) into EGCG. The robust therapeutics of FEGCG/Zn are measured in terms of the regulating effect on programmed cell death ligand 1 (PD-L1), the effective delivery of diverse biomolecules, and the hitchhiking ability using living cells. Taking small interfering RNA of PD-L1 (siPD-L1) and erythrocytes as an example, the fabricated biomimetic system achieves excellent siPD-L1 delivery and further improves siPD-L1 accumulation in tumors. Finally, the combination of FEGCG/Zn and siPD-L1 promotes antitumor immunotherapy through alleviation of T cells exhaustion by regulating PD-L1 expression in tumor cells. The results demonstrate that FEGCG/Zn substantially regulates PD-L1 expression and improves immune-biomolecule delivery by forming biomimetic nanoassemblies, offering a versatile platform for cancer immunotherapy.

Bone defect repair relies on the reconstruction of the maxilla and alveolar bone. Magnesium and its alloys offer great potential as implant materials due to their biocompatibility and mechanical properties, but their rapid degradation limits clinical use. This study developed an MgF 2 -polydopamine-Epigallocatechin gallate (MgF 2 -PDA-EGCG) composite coating on AZ31B magnesium alloy using hydrofluoric acid fluorination and layer-by-layer self-assembly. The coating’s corrosion resistance, biocompatibility, antibacterial properties, and osteogenic potential were evaluated. Characterization showed a dense, uniform coating (∼5.6 ± 0.2 μm thick) that significantly improved corrosion resistance. In a seven-day hydrogen evolution test and long-term immersion study, the P2E0.5 coating reduced hydrogen release (0.57 ± 0.16 mL/cm 2 ) and mass loss (7.31 ± 0.83%). Biocompatibility tests (CCK-8 and live/dead staining) confirmed no cytotoxicity. Antibacterial tests showed inhibition rates of 75.8% for Escherichia coli and 83.43% for Staphylococcus aureus. ALP activity was 188.18% ± 37.56% of the control, indicating enhanced early osteogenesis. In summary, the MgF 2 -PDA-EGCG coating improves the corrosion resistance and biological properties of AZ31B magnesium alloy, supporting its potential for biomedical implants.
