Procyanidin C1 (PCC1) is a biologically relevant oligomeric small molecule and a trimeric epicatechin with the potential to modulate gut microbiota. However, how it reshapes microbiota-linked phosphatidylcholine homeostasis in the context of insulin resistance remains insufficiently defined, limiting the establishment of mechanism-informed design principles for microbiota-directed metabolic modulation. Here, PCC1 improved systemic glucose and insulin sensitivity, promoted hepatic glycogen accumulation, reduced hepatic steatosis, attenuated skeletal muscle fibrosis, and decreased lipid deposition in adipose tissues. Notably, 16S rRNA sequencing revealed that PCC1 treatment enriched Lactobacillus johnsonii and depleted Leptogranulimonas caecicola , which were positively associated with elevated GPCho (16:0/20:5) and circulating eicosapentaenoic acid (EPA) identified by untargeted metabolomics, indicative of enhanced α-linolenic acid metabolism and a gut microbiota-driven contribution to EPA biosynthesis. Meanwhile, lipidomics revealed that PCC1 reduced diacylglycerols, triacylglycerols, ceramides, and lysophosphatidylcholines, while restoring EPA-containing phosphatidylcholines. Transcriptomics further demonstrated that PCC1 induced tissue-specific metabolic reprogramming, activating PPAR signaling in liver, enhancing oxidative phosphorylation in brown adipose tissue (BAT), and stimulating AMPK signaling in skeletal muscle. Importantly, PCC1 reversed HFD-induced phospholipase A2 dysregulation, upregulating Pla2g2d in skeletal muscle and downregulating Pla2g7 in liver and BAT, two genes implicated in EPA release and phospholipid remodeling. Together, integrated multi-omics analyses identify PCC1 as an oligomeric small-molecule modulator associated with improved insulin sensitivity across multiple tissues and implicate an EPA-linked phosphatidylcholine signaling axis that connects microbiota remodeling with host metabolic responses.

Introduction Intramuscular fat, known as “snowflake” pattern in pork, critically determines meat tenderness, juiciness, flavor, and overall consumer preference. Cultured meat has emerged as a promising solution to address future protein demands sustainably. However, producing cultured meat with authentic marbling remains challenging, as myocytes and adipocytes require distinct microenvironments for differentiation. Objective This study aimed to develop a co-differentiation strategy independent of conventional hormonal adipogenic cocktails (IBMX and dexamethasone) to simultaneously induce myogenesis and adipogenesis in a single culture system, thereby enabling the production of structured cultured meat with microscale muscle-fat patterning. Methods An edible κ-carrageenan-konjac glucomannan (CK) hydrogel was developed as a versatile platform for cultured meat. A 3D co-culture system was established by encapsulating porcine muscle satellite cells (pMuSCs) and fibro-adipogenic progenitors (pFAPs) within the CK scaffold, followed by differentiation induction using an eicosapentaenoic acid (EPA, C20:5n-3)-based strategy. The physicochemical properties of the resulting cultured meat induced with EPA (CM-EPA) were rigorously characterized using texture profile analysis and nutritional composition analysis, with comparisons made against horse serum-induced controls (CM-HS) and commercial streaky pork. Results The CK hydrogel exhibited tunable, biomimetic mechanical properties and a porous microstructure that supported the proliferation and differentiation of both cell lineages. Notably, the EPA-based induction strategy significantly enhanced lipid accumulation while simultaneously supporting myotube formation from pMuSCs, effectively mitigating the myogenesis inhibition associated with conventional hormonal adipogenic cocktails. The co-cultured meat exhibited significantly improved textural properties, such as hardness and chewiness, which closely resembled those of real streaky pork. Its nutritional profile also showed elevated protein and fat content relative to the CM-HS control. Conclusion This work establishes a versatile scaffold-based platform for producing structured cultured meat with integrated muscle and fat components, offering a promising strategy to replicate the textural and nutritional qualities of traditional meat.

Sodium caseinate (SC) binds to sodium hyaluronate (SH) by non-covalent interactions. • SC/SH complex effectively encapsulates EPA-rich algal oil. • SC/SH emulsions show better salt, and storage stability with uniform droplets. • The SC/SH complex shows potential as a functional lipid stabilizer. Eicosapentaenoic acid (EPA)-rich algal oil is widely used in functional dairy products due to its favorable functional properties. However, its limited restricts broader application in the food industry. In this work, sodium hyaluronate (SH) was combined with sodium caseinate (SC) to form an aqueous network as an emulsifier for EPA. The binding mechanism between SC and SH was investigated via fluorescence spectroscopy and thermodynamic analysis, revealing that van der Waals forces and hydrogen bonds dominate the SC-SH interaction. When used as an emulsifier, the SC/SH complex effectively encapsulated EPA-rich algal oil. The resulting emulsions exhibited significantly enhanced stability against environmental stresses—including storage, ionic, and oxidative stresses—with more uniform droplet size distribution. The SC/SH complex thus demonstrates potential as a stabilizer for functional lipid-delivery systems. Eicosapentaenoic acid (EPA)-rich algal oil is commonly used in functional dairy products due to its excellent functional properties, but its poor stability limits its wide application in the food industry. Lipase catalysis has become the most promising technique for enrichment of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) due to its mild and green conditions. In this study, Trichosporon sp. F1-2 was newly isolated from oil-contaminated sludge, and its extracellular lipase was purified using ammonium sulfate precipitation and DEAE-Sepharose chromatography. The molecular weight of the lipase was estimated to be 32.6 kDa. The optimal temperature and pH for lipase activity were 50 °C and 8.0, respectively, and the optimal storage conditions were found to be temperatures below 45 °C and pH 7.0–9.0. The lipase did not require metal ions as cofactors and was significantly inhibited by Zn 2+ . It was stable in various organic solvents with different log P values, and n-hexane, toluene, dichloromethane, diethyl ether and glycerol even enhanced its activity. It had sn -1,3 regioselectivity and the highest activity towards a medium-chain fatty acid (C8) ester. This study demonstrated the lipase has good potential as a catalyst in enrichment of EPA and DHA in fish oil. The content of EPA and DHA reached 22.9 and 25.4 mol%, respectively, using 100 U of the lipase within 24 h, and the enrichment efficiency of DHA was higher than that of EPA.

Fish oil is beneficial to health and its quality is significantly impacted by the extraction method. To determine the effect of various methods used to extract the oil on its chemical composition and thermal properties, tilapia oil extracted by dilute alkali hydrolysis, enzymatic hydrolysis, and microwave-assisted methods were investigated in this study. Nutrient analyses showed that freeze-dried tilapia contained fat (36.69 g/100 g freeze-dried tilapia), protein (49.95 g/100 g freeze-dried tilapia), ash (12.04 g/100 g freeze-dried tilapia) and moisture (1.21 g/100 g freeze-dried tilapia). Chemical analysis showed that the fish oil extracted by dilute alkali hydrolysis had the lowest acid value (AV) and peroxide value (POV) (AV: 0.77 mg KOH/g oil and POV: 3.76 mmol/kg oil) compared to the oil extracted using the other methods. The main fatty acid components were palmitic acid (22.78–23.16%), oleic acid (25.73–26.19%), and linoleic acid (15.69–15.98%). Furthermore, the oil extracted by dilute alkali hydrolysis showed significantly higher content of DHA+ EPA (2.30%). The thermal behaviors indicated that the aforementioned oils had significantly different crystal structures. The main endothermic peaks of fish oil extracted by dilute alkali hydrolysis, enzymatic hydrolysis and microwave-assisted methods were located at −2.03℃, −3.20℃ and −4.37℃, respectively. However, infrared spectroscopic analyses indicated them had the same characteristic functional groups. Consequently, different extraction methods significantly affect the oil’s physical and chemical properties without affecting spectral characteristics. Furthermore, these results advocated the use of alkali hydrolysis for fish oil extraction.

Objective: Dietary n-3 polyunsaturated fatty acids (PUFAs), especially eicosapentaenoic acids (EPA) and docosahexaenoic acid (DHA), are proved to be effective in obesity reduction. Microalgal oil (MO) is an important alternative source of n-3 PUFAs that effectively alleviates obesity. The aim of the present study was to explore the anti-obesity effects of microalgal oil from Schizochytrium sp. (SMO) and to compare the effects of 2 SMOs (SMO1 and SMO2) with different levels of purity of n-3 PUFAs on high fat diet (HFD)-induced obesity in male C57BL/6J mice. Methods: Mice were randomly divided into 5 groups: (1) regular chow (RC); (2) HFD; (3) HFD + fish oil (FO); (4) HFD + SMO1; and (5) HFD + SMO2. Body weight and food intake were weekly monitored. After 16 weeks of treatment, a glucose tolerance test (GTT) and an insulin tolerance test (ITT) were performed. Serum lipid profile, morphological changes in the liver and epididymal white adipose tissue (eWAT), and the mRNA expression of lipid metabolism–related genes were also examined. Results: SMO treatment significantly decreased HFD-induced abdominal fat accumulation, lowered the levels of triglycerides, cholesterol, and low-density lipoprotein, as did the positive control treated with FO. Morphological examination revealed a remarkable reduction in lipid droplet formation in the liver tissue and the particle size of eWAT. An alleviation of inflammation infiltration in eWAT caused by a high-fat diet was also observed. Real-time reverse transcription–polymerase chain reaction analysis examination confirmed that microalgal oil inhibited the gene expression of fatty acid synthase, sterol responsive element-binding protein-1c, and acetyl-CoA carboxylase but promoted that of hormone-sensitive lipase and lipoprotein lipase, carnitine palmitoyltransferase-1, and uncoupling proteins in the liver and eWAT. Moreover, similar anti-obesity effects were obtained with the same dosage but different purity of n-3 PUFAs.Conclusions: As an alternative n-3 PUFAs resource, dietary intake of SMO might be beneficial to prevent HFD-induced abdominal fat accumulation.

Efficient immobilized lipase systems are essential for industrial lipid modification, yet the combined effects of enzyme loading, support properties, and post-immobilization stabilization on catalytic performance remain underexplored. In this study, Thermomyces lanuginosus lipase (TLL), Candida antarctica lipase B (CalB), and their mixture (T + C) were respectively immobilized on two structurally distinct commercial supports (A & B)—differing in hydrophobicity, surface area, and pore size—at varied loading densities. Adsorption followed monolayer kinetics, with support B enabling higher enzyme up to 44.1 mg/g wet for T + C. Co-immobilized T + C systems exhibited enhanced catalytic activity compared to individual lipases, particularly in esterification and transesterification, benefiting from the complementary substrate specificities of TLL and CalB. Chemical crosslinking with chitosan and glutaraldehyde significantly reduced lipase desorption and improved thermal stability. The optimized system Cr-B-T + C-M (crosslinked support B-TLL + CalB with medium enzyme loading) efficiently catalyzed fish oil transesterification at 60 °C for 18 h, increasing the EPA + DHA content from 30.5% to 54.8%. All immobilized lipases retained over 80% of their initial catalytic activity after three reuses (60 °C and 18 h). These results highlight the synergistic benefits of co-immobilization and crosslinking on tailored supports, offering a versatile strategy for developing stable and efficient lipase biocatalysts for high-value lipid processing.

A novel calcium-based sorbent was synthesized from calcium acetate and mesoporous silica through chemical impregnation method. The Brunauer-Emmett-Teller (BET) and scanning electron microscope (SEM) analysis revealed that the sorbent possessed a good surface structure as the calcium salt decomposed to calcium oxide (CaO) through multiple calcination steps. The mercury adsorption performance of the sorbent was investigated in a fixed bed system, the results showed that the sorbent has an obvious penetration behavior for gaseous element mercury (Hg 0 (g)) while completely adsorbed reactive gaseous mercury (Hg 2+ (g)) under the pure nitrogen and simulated flue gas atmosphere. On-site sampling test of the calcium-based sorbent traps was ultimately conducted on a 600 MW coal-fired power plant. The Ontario Hydra Method (OHM) was chosen as the reference method to evaluate the accuracy of the sorbent traps sampling results. The measured data obtained by sorbent traps was well consistent with OHM under the selected operational load and all the sorbent traps passed the QA/QC criteria, show that the sorbent traps performed with almost the same accuracy as OHM. The mercury emission and removal characteristics across the existing air pollution control devices (APCDs) were reported based on the sorbent traps sampling results. With the help of SCR for the Hg 0 oxidation, ESP for the removal of Hg 2+ and Hg p , and the good water solubility of Hg 2+ in WFGD, the combination of SCR + ESP + WFGD achieved an excellent mercury removal efficiency with the value of 88.54%. The final mercury emissions from the exhaust were 3.75 μg/m 3 and meet the emission requirements of China.

Determination of mercury species over solid waste from natural gas processing plants is a prerequisite for solid waste decontamination and mercury recycling. The temperature-programmed decomposition and desorption method (TPDD) is a promising technique to determine the mercury species over solid waste. Mercury species over solid wastes with complex components and similar desorption temperature, however, cannot always be determined using only the TPDD method. In the present study, the EPA 3200 method was applied to optimize the TPDD process for speciation of the mercury compound over solid wastes collected from a natural gas processing plant, including four spent adsorbents and three sludges. The mercury species with similar desorption temperatures over solid wastes were completely separated by EPA 3200 method optimized TPDD process. The mercury species over spent adsorbents and sludges could be determined based on the fingerprints of the mercury compounds. The mercury compounds over spent adsorbent were specified to be Hg0 and/or M-Hg amalgam, HgS(Black) and HgO(Red), of which HgS(Black) was the primary mercury compound. However, the mercury species over sludge were analyzed to be Hg0 and/or M-Hg amalgam, HgCl2(minor), HgS(Black), HgO(Yellow) and HgS(Red). The mercury recovery rates of solid wastes ranged from 73% to 120%. With the above results, it was considered that the EPA 3200 method optimized TPDD process was a promising method to specify and semi-quantify the mercury compounds in solid waste.

Introduction: Sheep placenta extract (SPE) is a representative traditional medicinal substance that exhibits multiple experimentally validated physiological properties, including anti-aging effects, wound healing acceleration, antioxidant activity, and anti-inflammatory mechanisms. However, the mechanism by which SPE influences the delay of aging is still not yet clear. Methods: Exploring the effects of sheep placenta extract on D-gal induced senescence in a mouse model of aging by macrogenomics and metabolomics. Results: In the serum of aging mice treated with SPE, the levels of antioxidant function such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase (CAT) were notably higher compared to those in the blank group, whereas malondialdehyde (MDA) levels decreased. We revealed that SPE alleviated the changes in gut microbiota caused by aging in mice, with a significant decrease in the Firmicutes/Bacteroidetes (F/B) ratio in the gut. Furthermore, Akkermansia muciniphila (A. muciniphila), which is known for its regulating immune response and potential anti-aging effects, showed a significant increase of 1177.94%. The analysis of UHPLC-QE-MS combined with orthogonal partial least squares discriminant analysis (OPLS-DA) screening of differential metabolites in mouse serum metabolic profiles revealed a significant upregulation of cis-5,8,11,14,17-eicosapentaenoic acid (EPA) and triptolide in serum metabolites, following SPE treatment, which are commonly believed to have immunosuppressive, anti-inflammatory, anti-proliferative, and anti-tumor effects.Discussion: The role of SPE in ameliorating aging may be associated with the increased abundance of A. muciniphila in the gut microbiota and the accumulation of two metabolites, EPA and triptolide, in the serum.

Hot water pretreatment of sea cucumbers potentially changes nutritional benefits. This study aimed to quantify hot water pretreatment-induced changes in metabolite profiles of sea cucumber body walls. ICP-OES, GC-MS, and LC-MS analyses of untreated- (UT-BW), hot water-treated body walls (HW-BW) of Apostichopus japonicus , and the hot water extract (HW-E) determined significant losses of minerals (25–50% w/w), protein (~11% w/w), carbohydrate (33% w/w), saponins (~41% w/w), and spermidine (100%), a potential antipsychotic from hot water-treated samples. Multivariate comparisons of HW-BW with UT-BW and HW-BW with HW-E showed increases in amino acids and fatty acids, suggesting hot water-induced degradation or transformation or easier extraction of protein, lipid or other components. Presence of 80 to 88.5% of compounds in the HW-E and lower DHA, EPA and glycerophospholipids levels in HW-BW suggested extraction of these metabolites. These data indicate that novel processing technologies are required to preserve the full nutritional benefits of sea cucumbers.

Here we investigate the effects and mechanisms of eicosapentaenoic acid (EPA) in regulating chondrocyte mechanics during inflammation in the progression of osteoarthritis (OA). Primary porcine chondrocytes and human OA chondrocytes were isolated and cultured. Cell mechanical properties were measured using atomic force microscopy. RNA sequencing, immunocytochemistry, quantitative PCR and western blotting were used to elucidate associated signaling mechanisms. Porcine cartilage and human OA cartilage explants were collected. Human articular cartilage samples were obtained from ten donors. Anterior cruciate ligament transection surgery was performed to induce OA in male C57BL/6J mice. The therapeutic effects of EPA and activation of associated signaling were evaluated using histology, immunohistochemistry and micro-computed tomography. EPA reduced F-actin intensity and Young’s modulus in IL-1α-treated porcine chondrocytes and in human OA chondrocytes. Mechanistically, EPA inhibited IL-1α-induced increase in CD44 expression in porcine chondrocytes by suppressing phosphorylation of NF-κB subunits p65. In addition, EPA alleviated articular cartilage degeneration and decreased the expression of p-p65 and CD44 in IL-1α-treated porcine and human OA cartilage explants. Moreover, EPA suppressed the increase in Young’s modulus induced by CD44 ligands (A6 peptide and low-molecular-weight hyaluronic acid) in porcine chondrocytes. Finally, intraarticular injection of EPA emulsion-integrated hyaluronic acid injection attenuated OA-associated alterations in articular cartilage and subchondral bone and decreased CD44 expression in mice. Our data not only provide new insights into EPA’s chondroprotective actions and underlying mechanisms but also offer new evidence supporting the therapeutic efficacy of a novel EPA emulsion-integrated hyaluronic acid injection for OA treatment.
