Fabrication of dendrite PtPd bimetallic nanocomposites modified carbon fiber microelectrodes for detecting superoxide anion released from living cells

Ziyue Sun,Hongli Zhao,Kaicha Chen,Fangfang Zhou,Minbo Lan

In this work, the carbon fiber microelectrode (CFME) coated by ERGO-CNT network was used as a substrate to support dendrite PtPd nanoclusters. An enzyme-free O 2 − electrochemical sensor (PtPd@ERGO-CNT-CFME) was prepared based on this strategy. Due to the brilliant loading and dispersing abilities of the substrate and the excellent electrochemical activity of the dendrite PtPd alloy, PtPd@ERGO-CNT-CFME demonstrated outstanding detection ability towards O 2 − . The as-constructed sensor showed a wide-linear range of 16–744 μM and 744–3064 μM with high sensitivity of 6967.3 μA mM −1 cm −2 and 4124.3 μA mM −1 cm −2 , and the detection limit was 5.07 μM. Furthermore, PtPd@ERGO-CNT-CFME was successfully applied to monitor O 2 − released from living cells. This phenomenon indicated the application potential for O 2 − detection in biological samples.

Sensitive Electrochemical Detection of Caffeic Acid using a Carboxyl-Functionalized Reduced Graphene Oxide-Modified Glassy Carbon Electrode (ERGO-COOH/GCE)

Ming Zhou,Yong Xu,Rong Zhong,Yurong Wang

A glassy carbon electrode modified by carboxyl-functionalized reduced graphene oxide (ERGO-COOH/GCE) was developed by electrochemical deposition method. The electrochemical behavior of caffeic acid (CA) on the ERGO-COOH/GCE was studied, and an electroanalytical method for the detection of CA in tablets was established. The effects of the modification parameters, the pH of the buffer solution and the scanning rate on the electrochemical behavior of CA were investigated by cyclic voltammetry (CV). The results revealed that modifying the GCE by electrochemical deposition facilitated a more sensitive response to CA on the modified electrode than modifying it by drop coating. The peak current of CA on the ERGO-COOH/GCE was significantly higher than that on the bare glassy carbon electrode (GCE) over five scan cycles, and the optimized the concentration of GO-COOH, the scanning cycles, and the pH were 4 mg·mL -1 , 5 cycles, and 2.0, respectively. The redox reaction of CA on the modified electrode was a quasi-reversible process controlled by adsorption and involved the transfer of two electrons and two protons. The ERGO-COOH/GCE showed a linear response to CA in the range of 0.05 ~ 20 μM with a limit of detection of 8 nM ( S/N =3). The modified electrode exhibited good repeatability and reproducibility and anti-interference performance. The ERGO-COOH/GCE was applied to the detection of CA in caffeic acid tablets with a recovery of 97.7% ~ 108%.

Silver nanoclusters-assisted triple-amplified biosensor for ultrasensitive methyltransferase activity detection based on AuNPs/ERGO hybrids and hybridization chain reaction

Xiaolun Peng,Jinlin Zhu,Wei Wen,Ting Bao,Xiuhua Zhang,Hanping He,Shengfu Wang

In the paper, a triple-amplified biosensor was built for the purpose of ultrasensitive methyltransferase (Dam MTase) activity detection and inhibitor screening based on in-situ synthesized silver nanoclusters (AgNCs) as signal probes to provide amplified current signal coupling with gold nanoparticles/electrochemical reduced graphene oxide (AuNPs/ERGO) hybrids and hybridization chain reactions (HCR) amplification strategy. For biosensor preparation, the AuNPs/ERGO hybrids were firstly generated on the electrode surface by electrochemical co-reduction method, then the ds-DNA structures which comprised the specific recognition sequences (5′-G-A-T-C-3′) of the Dam MTase and the restrictive endonuclease DpnI were formed on the electrode by the hybridization of the assistant DNA with the capture DNA. The presence of Dam MTase methylated partial ds-DNA structures on the electrode which could be digested by DpnI and could not undergo HCR process. The unmethylated ds-DNA structures underwent HCR process to further hybridize with DNA1 and DNA2 to form ds-DNA superstructures as effective template for AgNCs in-situ synthesis. Oxidation peak current of silver was obtained as signal output for Dam MTase detection. Integrated synthesis and detection in one as well as combined metal nanocluster with DNA superstructures, this biosensor showed a good performance for Dam MTase activity detection with a detection limit of 0.0073 U/mL. Compared with reported biosensor, the designed biosensor showed high sensitivity and had potential in clinical diagnosis as well as inhibitor screening.

Construction of ERGO film modified electrode and its combination with HPLC in determination of five active components in Osmanthus fragrans fruits

Ting-Ting ZHANG,Xiao-Yan LIAO,Jing REN,Chen CHEN,Yu-Ying WAN,Fang WANG,Zi-Lin CHEN

Neonuezhenide, nuezhenide, acteoside, isoacteoside and salidroside are common phenylethanoid glycosides in Osmanthus fragrans ( O. fragrans ) fruits and have various physiological and pharmacological activities. The electrochemical determination of the five components in O. fragrans fruits was researched at electrochemically reduced graphene oxide film modified glassy carbon electrode (ERGO/GCE). ERGO film was prepared by one-step deposition and showed a honeycombed network structure and provided good conductivity, controllability and stability. Compared with bare GCE or chemically reduced graphene oxide film modified electrode (RGO/GCE), the voltammetric behaviours of all five active components were improved at ERGO/GCE in terms of peak current and potential due to excellent electrical properties of ERGO. The numbers of protons and electrons transferred in electrochemical redox reaction were calculated and the possible electrode processes of five components have been estimated herein. The number of electrons involved was equal to that of protons transferred in the electrode processes for each compound and it was 2 for neonuezhenide, 1 for nuezhenide, 4 for acteoside, 4 for isoacteoside, and 1 for salidroside, respectively. Finally, a simple electrochemical method was developed for determination of the five components. This method has wide linear range with low detection limit of 3 μM for neonuezhenide, 6 μM for nuezhenide, 4 μM for acteoside, 3 μM for isoacteoside, and 3 μM for salidroside, respectively. Moreover, this method could be coupled with high performance liquid chromatography (HPLC) and applied to determination of the five active components in extractive from O. fragrans fruits. Compared with high performance liquid chromatography-mass spectrometry (HPLC-MS), this method would be competitive and alternative for the analysis of active components in real samples.

2D Leaf-Like Structured ZIF-L Embedded Electrochemically Reduced Graphene Oxide Composite as an Electrochemical Sensing Platform for Sensitively Detecting Benomyl

Min Shi,Guanwei Peng,Shuya Xue,Jingkun Xu,Yansha Gao,Shuwu Liu,Xuemin Duan,Limin Lu

In this work, a two-dimensional leaf-like framework-L embedded electrochemically reduced graphene oxide (ERGO@ZIF-L) was proposed as an outstanding electrode material for the sensitive electrochemical sensing of benomyl (BM). ZIF-L is surrounded by ERGO, which could effectively ensure the stability and dispersion of ZIF-L. With this unique combination, the prepared ERGO@ZIF-L displayed excellent synergistic characteristics with a large surface area, excellent conductivity, plentiful active sites, and high electrocatalytic properties, thus endowing it with high sensitivity for BM determination. The experimental parameters, such as solution pH, material volume, and accumulation time, were optimized. Under optimal conditions, the BM sensor showed a wide linear range (0.009–10.0 μM) and low-limit detection (3.0 nM). Moreover, the sensor displayed excellent stability, repeatability, and reproducibility, and good anti-interference capability. The method was successfully applied to detect BM in real-world samples.Keywords:two-dimensional MOFs;zeolitic imidazolate framework;ERGO;benomyl;electrocatalysts;electrochemical sensor

Multimodal Voltammetric sensor using Pd@ErGO nanocomposite for selective or simultaneous determination of Tetrandrine and Fangchinoline in traditional Chinese medicine and biological matrices

Huanhuan Zhu,Yong Xu,Jiafen Huang,Yiheng Zhang,Manman Sun,Zhiming Wang,Yurong Wang

In this work, a sensitive glassy carbon electrode modified with palladium-doped electrochemically reduced graphene oxide (Pd@ErGO/GCE) was developed for the selective and simultaneous determination of tetrandrine (TET) and fangchinoline (FAN). The electrochemical behavior was investigated by cyclic voltammetry (CV) and a multimodal voltammetric platform was established: (1) differential pulse voltammetric (DPV) quantification of TET (LOD = 55 nM), (2) linear sweep voltametric (LSV) determination of total content (LOD = 45 nM), and (3) simultaneous determination of both analytes via DPV. The methods have great reproducibility and repeatability, great interference tolerance, and satisfactory accuracy in serum (recovery: 95.7% ~ 104%) and urine (97.4% ~ 103%). Comprehensive validation against HPLC-UV ( t  = 0.32) confirmed method accuracy for pharmaceutical quality control in Radix Stephaniae tetrandrae. This methodology offers a reliable framework for alkaloid monitoring in herbal and biological matrices.

Indirect Electrochemical Determination of Ribavirin Using Boronic Acid-Diol Recognition on a 3-Aminophenylboronic Acid-Electrochemically Reduced Graphene Oxide Modified Glassy Carbon Electrode (APBA/ERGO/GCE)

Xiaoyan Wang,Weiyang Shen,Xiaolei Zhang,Siyan Guo,Ye Gao,Xiaotong Li,Fang Feng,Gongjun Yang

A novel method for the indirect electrochemical determination of ribavirin based on boronic acid-diol recognition was developed using the platform as the sensing element. The device was constructed using a 3-aminophenylboronic acid (APBA)-electrochemically reduced graphene oxide (ERGO) modified electrode. When the electrode was immersed in a solution of ribavirin, complexation of boronic acid groups of APBA with ribavirin occurred at the surface of the electrode and simultaneously caused steric hindrance, resulting in a current decrease because the ferricyanide redox probe was unable access the surface. Under the optimized conditions, a linear relationship was obtained between the relative change in current (%Δi) of [Fe(CN)6]3−/4−and the concentration of ribavirin at levels from 10.0 to 7.50 × 102 ng mL−1. The proposed electrochemical sensor performed with acceptable sensitivity and reproducibility and was successfully used to determine the content of ribavirin in an injection with satisfactory results.
