Developmental toxicity of functionalized polystyrene microplastics and their inhibitory effects on fin regeneration in zebrafish

Qian Li,Wenjing Cheng,Hongyu Wang,Jun Chen,Yueyun Qin,Ruozhu Sheng,Shaohong Peng,Zaitian Li,Tao Lu,Liwei Sun

The hazardous effects of microplastics on aquatic biota have been extensively documented. However, the impact of surface functionalization on the toxicological profiles of microplastics, particularly in fish, requires further investigation. In the present study, the effects of polystyrene microplastics (50–100 nm in diameter), either non-functionalized or functionalized with carboxyl (-COOH) or amino (−NH 2 ) groups, on zebrafish were explored. Exposure to all three types of microplastics (0.1, 1, and 10 mg/L) resulted in developmental toxicity in zebrafish at early life stages. This included increased mortality, reduced larval body length, decreased heart rate, impaired swimming ability, as well as disruptions in redox homeostasis, nervous system development, and immune responses. Moreover, all types of microplastics (1 mg/L) inhibited the regenerative capacity of larval zebrafish after caudal fin amputation, both morphologically and functionally. This inhibition was likely due to alterations in immune response, excessive production of reactive oxygen species, and disruptions in the signaling networks that regulate fin regeneration. Among the tested microplastics, amino-modified particles generally exhibited greater developmental toxicity and stronger inhibitory effects on fin regeneration. Our findings confirm that the functionalization of microplastics plays a crucial role in their interactions with biological systems and highlight concerns regarding the potential effects of functionalization, whether intentional or resulting from natural weathering processes.

Teardrop-like micro pin fin coated nanotube arrays chip for enhancement of flow boiling electronics cooling

Hongqiang Chen,Quan Gao,Xiang Ma,Kai Li,Wangfang Du,Caifeng Li,Yonghai Zhang,Jinjia Wei

Phase change flow boiling heat transfer in microchannel is a very efficient thermal management mode for high-power electronics/devices cooling. However, achieving comprehensive enhancement of flow boiling heat transfer performance at low power consumption is still challenging. Herein, we devised and manufactured a teardrop-like micro-pin-fin coated stable copper hydroxide nanotubes array chip surfaces (S-Nanotube), demonstrating their exceptional enhancement in flow boiling heat transfer efficiency. A series of experiments were conducted using HFE-7100 as a working fluid within a semi-open microchannel. Compared to the smooth surface, the critical heat flux (CHF) and the maximum boiling heat transfer coefficient (HTC) of the S-Nanotube is increased by 82.1 % (from 45.1 to 72.9 and then to 82.1 W/cm 2 ) and 140.5 % (from 5955 to 11,325 and then to 14,316 W/m 2 ·K) at extremely low-pressure drop (≤4 kPa), showing a high coefficient of performance (COP). The temperature of the onset of nucleate boiling on the S-Nanotube surface is reduced by 26.4 %, and the heat flux is greatly increased in a small wall temperature variations (Δ T  ≤ 10 °C). In situ observation and analysis of the surface properties and the bubble dynamics, the S-Nanotube chip promotes the phase change heat transfer process by providing massive nucleation sites, reducing bubbles size and residence time, and enhancing the wicking wetting capacity. These findings provide guidance for the rational design of boiling heat transfer-enhanced surfaces and heat sinks and point the way to achieving efficient thermal management of power devices.

Digestive properties of half-fin anchovy hydrolysates/glucose Maillard reaction products and modulation effects on intestinal microbiota

Ru Song,Min Shi,Luo Gu

BACKGROUND The consumption of dietary Maillard reaction products (MRPs) might lead to positive or negative effects on health. The digestibility of half-fin anchovy hydrolysates/glucose MRPs (HAHp(9.0)-G MRPs) was therefore determined. The intestinal microbiota modulation of HAHp(9.0)-G MRPs in mice was also evaluated after administration for 14 days (1 g kg −1 •bodyweight). RESULTS Different levels of digestibility of MRPs of fructosamine and advanced glycation products of N ε -carboxymethyllysine were detected in HAHp(9.0)-G MRPs during simulated gastrointestinal digestion. An increased relative proportion of soluble fluorescent melanoidins (SFMs) was observed during gastric digestion as compared to that in the original HAHp(9.0)-G MRPs, followed by decreases in SFMs in intestinal digestion. After feeding with HAHp(9.0)-G MRPs for 14 days, increased goblet cells were observed in the ileum regions of female and male mice. High-throughput 16S ribosomal RNA gene sequencing of fecal samples revealed that HAHp(9.0)-G MRPs administration increased the density of the phylum Bacteriodetes and reduced the density of the phylum Firmicutes in male mice. By comparison, a relatively higher density of members of the phylum Saccharibacteria was observed in female mice. A consistent increase in the abundance of Bacteroidales_S24-7_group_norank was found in female and male groups fed with HAHp(9.0)-G MRPs. Female and male mice treated with HAHp(9.0)-G MRPs also showed higher levels of propionic and butyric acids in feces than their corresponding controls. CONCLUSION Half-fin anchovy hydrolysates/glucose MRPs can be partly hydrolyzed in the simulated gastrointestinal digestion system. Treatment with HAHp(9.0)-G MRPs induced sex-related differences in bacterial abundance and diversity in mice; however, the up-regulation of anti-inflammatory activity was predicted in both female and male mice. © 2021 Society of Chemical Industry.

Characterization and antibacterial activity of the nanocomposite of half-fin anchovy (Setipinna taty) hydrolysates/zinc oxide nanoparticles

Ru Song,Qingqing Shi,Ghada Adel A. Abdrabboh,Rongbian Wei

Half-fin anchovy ( Setipinna taty ) hydrolysates (HAHp) was conjugated with zinc oxide nanoparticles (ZnO NPs) using a hydrothermal method to develop a novel antibacterial nanocomposite. The generated supernatants of the conjugate, designated as HAHp(3.0)/ZnO NPs, were characterized by transmission electron microscopy, high resolution transmission electron microscopy, and inductively coupled plasma-optical emission spectrometer. Results showed that HAHp(3.0) was absorbed on the surface of the ZnO NPs. The total content of zinc element was 9127.4   mg/kg in HAHp(3.0)/ZnO NPs. The increased antibacterial effects were observed for the HAHp(3.0)/ZnO NPs with the minimal inhibitory concentration of 3.5   μgprotein/mL against Escherichia coli ( E. coli ), Pseudomonas fluorescens , Salmonella and Staphylococcus aureus , compared to the bare HAHp(3.0). The antibacterial activity of HAHp(3.0)/ZnO NPs was further evaluated using E. coli as the model strain. The incubation of HAHp(3.0)/ZnO NPs increased the outer and inner membrane permeability in E. coli cells, and the leakages of potassium ions and the cytoplasmic β-galactosidase were detected during the process. Furthermore, porous structures were observed on the membrane of E. coli cells by scanning electron microscopy. In addition, the formation of intracellular reactive oxygen species was detected using fluorescence microscopy. The results suggested that the HAHp(3.0)/ZnO NPs could be a promising antibacterial nanocomposite.

mTOR mutation disrupts larval zebrafish tail fin regeneration via regulating proliferation of blastema cells and mitochondrial functions

Xiao Gongyi,Li Xiangwei,Yang Huiping,Zhang Ruobin,Huang Junlan,Tian Yu,Nie Mao,Sun Xianding

Background The larval zebrafish tail fin can completely regenerate in 3 days post amputation. mTOR , the main regulator of cell growth and metabolism, plays an essential role in regeneration. Lots of studies have documented the role of mTOR in regeneration. However, the mechanisms involved are still not fully elucidated. Materials and results This study aimed to explore the role and mechanism of mTOR in the regeneration of larval zebrafish tail fins. Initially, the spatial and temporal expression of mTOR signaling in the larval fin was examined, revealing its activation following tail fin amputation. Subsequently, a mTOR knockout ( mTOR -KO) zebrafish line was created using CRISPR/Cas9 gene editing technology. The investigation demonstrated that mTOR depletion diminished the proliferative capacity of epithelial and mesenchymal cells during fin regeneration, with no discernible impact on cell apoptosis. Insight from SMART-seq analysis uncovered alterations in the cell cycle, mitochondrial functions and metabolic pathways when mTOR signaling was suppressed during fin regeneration. Furthermore, mTOR was confirmed to enhance mitochondrial functions and Ca 2 +  activation following fin amputation. These findings suggest a potential role for mTOR in promoting mitochondrial fission to facilitate tail fin regeneration. Conclusion In summary, our results demonstrated that mTOR played a key role in larval zebrafish tail fin regeneration, via promoting mitochondrial fission and proliferation of blastema cells.

Enabling Tunable Stiffness, Adhesive Grasping, and Interaction-Driven Reconfiguration: A Shape-Memory-Polymer-Enhanced Fin-Ray Gripper

Haotian Guo,Hao Wu,Yanzhe Wang,Yaoting Xue,Tuck-Whye Wong,Tiefeng Li,Huixu Dong

Soft grippers offer a compelling solution for handling tasks in diverse environments due to their inherent safety and adaptability. However, enhancing their versatility, particularly in load capacity and grasping range, while minimizing actuation, remains a persistent challenge. To address this, we propose a soft gripper with reconfigurable morphology, combining structure (Fin Ray Effect [FRE] gripper), and intelligent material (shape memory polymers [SMPs]) as a union, to achieve tunable stiffness, adhesive grasping, and interaction-driven reconfiguration. First, SMPs are integrated into both the front and back beams of the FRE fingers, enabling adhesion grasping and grasping force modulation through phase transition, respectively. Additionally, by leveraging its shape-locking capabilities through intentional environmental interactions, the gripper achieves versatile reconfiguration with a single motor. Besides, inspired by humans interacting with tools and grasping in constrained spaces, we demonstrate three extra grasping modes, including precision pinching, hooking, and corner grasping. Experimental results validate its ability to handle diverse objects, from thin sheets and small nuts to items up to 50 times its own weight. This passive reconfigurable design allows for effective handling of disparate surfaces and contours, guaranteeing safe grasping in constrained spaces. This work opens new possibilities for soft robotic hands, balancing system simplicity with versatility for a wider range of real-world applications.

Lacticaseibacillus rhamnosus GG enhances fin regeneration under oxytetracycline exposure via activating Wnt signaling and modulating gut microbiota

Ju Wang,Lei Wang,Qi Wang,Changhong Liu,Lei Zheng

Zebrafish possesses robust caudal fin regeneration which depends on multiple factors to maintain body integrity. However, it is uncertain whether the caudal fin regeneration is related to gut microbiota . Here, we investigated the effect of Lacticaseibacillus rhamnosus GG (LGG) on the regeneration of caudal fin under oxytetracycline (OTC) exposure. The results demonstrated that 1000 μg/L OTC exposure for 4 days decreased reactive oxygen species (ROS) production at 1 and 3 h post amputation (hpa), increased neutrophil recruitment at 6 hpa, enhanced the number of apoptotic cells at 1, 3, 6 and 12 hpa and inhibited Wnt signaling pathway at 48 hpa in wound site. Furthermore, OTC exposure caused dysbacteriosis by elevating level of Proteobacteria and decreasing the abundance of Firmicutes , particularly Lacticaseibacillus , thereby negatively impacting wound healing and repair. Additionally, the administration of 10 6  CFU/mL of LGG for 48 h could improve intestinal environment through increasing the colonization rate of LGG in OTC-treated larvae intestines. The regenerative process restored by LGG was accompanied with increased ROS production at 1, 3 and 6 hpa, inhibited neutrophil recruitment at 6 hpa, decreased the number of apoptotic cells at 1 hpa, and activated Wnt signaling pathway at 48 hpa in OTC-treated fish. LGG is a promising bacterium for restoring fin regeneration and provides new insights regarding the correlation among the gut microbiota and fin regeneration.

Multipotent anti-aging recombinant human elastin: Counteracting zebrafish caudal fin shrinkage and D-galactose-induced intrinsic skin aging in mice

Lijun Zhu,Wenjie Huang,Jingjing Shi,Fei Li,Xiuxia Sun,Jianxi Xiao

The degradation of elastin in aged skin results in the loss of elasticity, the formation of wrinkles, and even a reduced ability for wound healing after injury. However, previous studies about elastin material have been limited by high immunogenicity of animal-derived elastin, the use of single elastin peptide sequences, and residual crosslinking agents. In this study, we designed a multipotent anti-aging recombinant human elastin (rElastin) and utilized it to counteract caudal fin shrinkage in zebrafish and D-galactose-induced intrinsic skin aging in mice. The rElastin demonstrated thermoresponsive reversible phase-transition closely resembling that of natural elastin. In vitro , cell experiments demonstrated that rElastin effectively promoted the proliferation, adhesion, and spreading of HFF-1 cells. In a D-galactose-induced intrinsic aging mouse model, rElastin significantly restored dermal density, skin hydration, and TEWL to levels approaching those of healthy skin. Histological analysis further confirmed that rElastin facilitated the deposition of collagen and regeneration of elastic fibers, thereby promoting structural and functional repair of aging skin. Additionally, rElastin obviously inhibited the tail fin shrinkage after UV radiation. rElastin effectively targets skin symptoms associated with intrinsic aging, providing fresh insights into anti-aging therapies and showcasing strong potential for applications in regenerative medicine and cosmetic dermatology.

Identification of antibacterial peptides from Maillard reaction products of half-fin anchovy hydrolysates/glucose via LC-ESI-QTOF-MS analysis

Ru Song,Qingqing Shi,Peiyu Yang,Rongbian Wei

We aimed to identify antibacterial peptides from Maillard reaction products of half-fin anchovy hydrolysates/glucose. Seven peptides from the active fraction of P3-2 derived from DEAE-Sepharose™ CL-6B chromatography and PL aquagel-OH 30 on HPLC system were identified by liquid chromatography-electrospray ionization/multi-stage mass spectrometry, and five peptides were selected for further synthesis on the basis of peptide sequences. The anionic peptide FFTQATDLLSR adopted an α-helical structure, as predicted by Network Protein Sequence @ analysis and helical wheel projection, and further determined using circular dichroism assay. The cationic peptide WLPVVR exhibited stronger inhibition effects against Escherichia coli than other peptides at similar concentration. Furthermore, differential sizes of hydrodynamic diameters of anionic 1,2-dimyristoyl-sn-glycero-3-phosphocholine liposome vesicles treated with peptides were observed, suggesting the clustering and decomposition of peptide on liposome vesicles. To the best of our knowledge, this is the first study that reports antibacterial peptides derived from the MRPs of half-fin anchovy hydrolysates.

In vitro membrane damage induced by half-fin anchovy hydrolysates/glucose Maillard reaction products and the effects on oxidative status in vivo

Ru Song,Qingqing Shi,Peiyu Yang,Rongbian Wei

Membrane damage via the Maillard reaction products of half-fin anchovy hydrolysates and glucose (designated as HAHp(9.0)-G MRPs) was evaluated using an in vitro Escherichia coli (E. coli) model. The incubation of HAHp(9.0)-G MRPs led to a significant increase in outer and inner membrane permeability. Furthermore, membrane integrity was seriously destroyed, as observed by scanning electron microscopy and indicated by the decreased fluorescence signal of 4′,6-diamidino-2-phenylindole stain. Interestingly, HAHp(9.0)-G MRPs induced significant hydrogen peroxide (H2O2) production in E. coli after 3 h of incubation, which contributed to the antibacterial activity of HAHp(9.0)-G MRPs. As one of the reactive oxygen species, excess H2O2in vivo might impair the antioxidant balance. Therefore, the oxidative status of healthy mice after short-term intake of HAHp(9.0)-G MRPs was investigated. After the administration of HAHp(9.0)-G MRPs at low dose (0.1 g per kg per day body weight) and high dose (1.0 g per kg per day body weight) for 14 days, the body weight of female mice decreased, and the body weight of male mice increased. However, the administration of HAHp(9.0)-G MRPs did not affect the hepatic antioxidant defense in either female or male mice. Some positive responses, like increased superoxide dismutase (SOD) and glutathione peroxidase (GPx) as well as decreased lipid peroxidation (LPO) content, were detected, especially in liver. Though the decrease in catalase activity indicated that the glycation compounds from HAHp(9.0)-G MRPs might be absorbed in vivo, the lower SOD/GPx ratios of HAHp(9.0)-G MRPs-fed groups than those of the normal groups also suggested that the administration of HAHp(9.0)-G MRPs could reduce LPO stress in mice. Our results suggest that a higher antioxidant status could be generated in the healthy animals after fed with HAHp(9.0)-G MRPs for 14 days.
