Liposome-cyclodextrin synergistic system for enhanced chiral separation of Fmoc-amino acid enantiomers in capillary electrophoresis

Wang Yunan,Zhang Qiang,Cheng Yue,Shi Chenglong,Wang Yong,Ma Xiaofei

A micro-nano structure enhanced chiral separation strategy is proposed by integrating biomimetic liposome assemblies with cyclodextrin (CD) derivatives as composite chiral selectors in capillary electrophoresis (CE). The self-assembly of weakly chiral, poorly soluble phospholipids into liposomes markedly amplifies chiral discrimination when coupled with CD-based molecular recognition, enabling highly efficient separation of multiple 9-fluorenylmethoxycarbonyl (Fmoc)-amino acid enantiomers. This enhancement is proposed to be associated with the formation of interfacial pre-enrichment microzones on liposome surfaces, where locally concentrated selectors and analytes may engage in synergistic hydrogen bonding, hydrophobic interactions, and steric effects, collectively strengthening chiral recognition. The composite system exhibits good selectivity and compatibility in mixed-sample separations, supporting simultaneous enantioseparation of multiple amino acids with high reproducibility. Beyond establishing an effective CE-based approach for amino acid enantiomer separation, this work demonstrates a proof-of-concept for transforming poorly soluble, weakly chiral substances into efficient chiral selectors through micro-nano assembly, offering preliminary insights for synergistic chiral separation systems. Self-Assembling Peptide Artificial Enzyme as an Efficient Detection Prober and Inhibitor for Cancer Cells

Meiling Lian,Shuo Zhang,Jun Chen,Xuejiao Liu,Xu Chen,Wensheng Yang

A new hybrid nanoparticle (NP; fluorenylmethoxycarbonyl-arginine-glycine-aspartate and hemin, Fmoc-RGD/hemin NP) was developed for the simultaneous detection and inhibition of breast cancer cells. Hemin can regulate the reactive oxygen species (ROS), while Fmoc-RGD acts as a scaffold for hemin nanocrystallization. Fmoc groups interact with the porphyrin groups of hemin through hydrophobic and π–π interactions to form a hydrophobic core of the NPs. The hydrophilic RGD chains surround the core to maintain the stability of the nanoparticles in an aqueous medium. The RGD groups of Fmoc-RGD are also selective for tumor cells. This interaction can be exploited to enhance the selectivity of tumor detection. Based on enhanced peroxidase activity, Fmoc-RGD/hemin NPs were developed as signal transducers in a facile and fast point-of-care cancer diagnosis platform. This platform is sensitive to breast cancer cells and hydrogen peroxide (H2O2), a biomarker for breast cancer. In addition, these Fmoc-RGD/hemin NPs can be used as nanoscavengers for ROS and for regulating the redox status of cancer cells. They also exhibit a targeted inhibitory effect on the epithelial–mesenchymal transition (EMT). The peptide-tuned self-assembly of Fmoc-RGD/hemin NPs as functional artificial enzymes boasts simple preparation, biofriendliness, and the versatility required for on-demand therapeutics and diagnostics for metastatic cancer cells. These NPs can therefore be used as effective tools for potential applications in medicine and biotechnology.

Fluorescent quantification of amino groups on silica nanoparticle surfaces

Chen Yang,Zhang Yanqin

Functionalization of the surfaces of silica particles is often the first step in their various applications. An improved heterogeneous Fmoc-Cl fluorescent assay using an aqueous solution was developed to detect the number of amino groups on solid-phase supports. The fluorescent Fmoc-Cl method is 50-fold more sensitive than the current UV assay using an organic solvent. This method, together with the homogeneous fluorescamine and OPA assays, is used to detect amino groups on the silica particle surface. The accuracy and effect factors of these methods were examined and the assays were optimized. The results showed that the amine groups on silica particles can produce stronger fluorescence than small amine molecules in solution, because the porous structure of the particle surface is a more hydrophobic environment. The number of active amino groups that can be conjugated with biomolecules is much less than the total number of amino groups on the silica particle. Compared with physical methods, chemical assays involving direct reaction with amino groups would furnish the closest result to the number of active amino groups on the particle surface. Fluorene methoxycarbonyl-PEG-deferoxamine conjugates “hitchhike” with albumin in situ for iron overload therapy

Linyi Xu,Rou Guan,Bohong Yu,Yicheng Li,Hongzhuo Liu,Yiguo Jiang

Although deferoxamine (DFO) has been approved for the treatment the iron overloaded diseases, its clinical application is impeded by very short circulation time and its relating toxicity. In this work, the fluorene methoxycarbonyl (FMOC) for “albumin hitchhiking” was used to prolong the plasma circulation time of DFO and reduce toxicity. The designed FMOC-PEG-DFO conjugates were found to reversible bind to albumin and gradually release DFO in vivo . Herein, the FMOC-PEG 1000 -DFO conjugates could increase 30 times the blood circulation time of DFO with the improvement of the iron elimination efficacy. Meanwhile, the conjugates markedly reduced the cytotoxicity of DFO. Taken together, the result demonstrated the FMOC-PEG 1000 -DFO conjugates could be a potential therapeutic choice for iron-overload-related diseases.

A supramolecular metalloenzyme possessing robust oxidase-mimetic catalytic function

Xu Shichao,Wu Haifeng,Liu Siyuan,Du Peidong,Wang Hui,Yang Haijun,Xu Wenjie,Chen Shuangming,Song Li,Li Jikun,Shi Xinghua,Wang Zhen-Gang

Enzymes fold into unique three-dimensional structures to distribute their reactive amino acid residues, but environmental changes can disrupt their essential folding and lead to irreversible activity loss. The de novo synthesis of enzyme-like active sites is challenging due to the difficulty of replicating the spatial arrangement of functional groups. Here, we present a supramolecular mimetic enzyme formed by self-assembling nucleotides with fluorenylmethyloxycarbonyl (Fmoc)-modified amino acids and copper. This catalyst exhibits catalytic functions akin those of copper cluster-dependent oxidases, and catalytic performance surpasses to date-reported artificial complexes. Our experimental and theoretical results reveal the crucial role of periodic arrangement of amino acid components, enabled by fluorenyl stacking, in forming oxidase-mimetic copper clusters. Nucleotides provide coordination atoms that enhance copper activity by facilitating the formation of a copper-peroxide intermediate. The catalyst shows thermophilic behavior, remaining active up to 95 °C in an aqueous environment. These findings may aid the design of advanced biomimetic catalysts and offer insights into primordial redox enzymes.

A self-assembled amino acid-based hydrogel with broad-spectrum antibacterial activity

Xie Yan-Yan,Qin Xiao-Tong,Feng Jia-Yuan,Zhong Cheng,Jia Shi-Ru

Abstract Amino acid-based hydrogels with broad-spectrum antibacterial properties are increasingly popular alternatives to existing treatments in the fight against bacterial infection. Here, an amino acid-based hydrogel formed by the self-assembly of N -(9-fluorenylmethoxycarbonyl)- L -Leucine (Fmoc-L) and Fmoc- L -Lysine (Fmoc-K) is reported. The Fmoc-L/Fmoc-K hydrogel, which consisted of twisted nanobelts, displayed antibacterial activity against Gram-positive S. aureus , B. subtilis , L. monocytogenes , and Gram-negative E. coli , Sh. sonnei . Specifically, inhibition rates of 97%, 100%, 44%, 45%, and 32% were achieved against S. aureus , B. subtilis , L. monocytogenes , E. coli , and Sh. sonnei after incubation with 4 mM of an Fmoc-L/Fmoc-K hydrogel with the molar ratio of 3:1 for 2 h, respectively. The inhibition rates against E. coli and Sh. sonnei reached 80% and 55% after treatment with 10 mM hydrogel, respectively. The antibacterial activity of hydrogels was time-dependent and enhanced with the prolongation of time. Additionally, the antibacterial activity was easily tailored by modifying the molar ratio of Fmoc-L and Fmoc-K. It was found that Fmoc-L/Fmoc-K hydrogel targeted and destroyed the bacterial cell membrane, causing cytoplasmic leakage and eventually cell death. Moreover, the hydrogel also displayed antibacterial action by inducing the agglomeration of S. aureus . This work demonstrates the potential of amino acid-based hydrogels as a new class of effective antibacterial materials in biomedical applications. Graphical abstract A novel amino acid-based hydrogel formed by the self-assembly of Fmoc-L leucine and Fmoc-L lysine , which consisted of twisted nanobelts, displayed antibacterial activity against Gram-positive and Gram-negative bacteria.

Highly sensitive fluorescent sensor based on electrosynthesized poly(Fmoc-l-serine) enables ultra-trace analysis of Cr2O72− in water and agro-product samples

Lie Zou,Yangping Wen,Hui Zhang,Jingdang Chai,Xuemin Duan,Liang Shen,Ge Zhang,Jingkun Xu

A fluorescent sensor based on the electrosynthesized conjugated polymer poly(Fmoc- l -serine) ( PFOLS ) for highly sensitive analysis of Cr 2 O 7 2− at ultra-trace levels in water and agro-product samples was reported. PFOLS was obtained by potentiostatical electropolymerization of corresponding monomer Fmoc- l -serine in CH 2 Cl 2 -boron trifluoride diethyl etherate system. The introduction of serine group in PFOLS efficiently avoided the interference of anions and other acid substances in agro-products. Although metal ion Fe 3+ could also quench the fluorescence of PFOLS , EDTA could recover the fluorescence of PFOLS -Fe 3+ system. So in the presence of EDTA, PFOLS had a high-selectivity to Cr 2 O 7 2− and it could detect Cr 2 O 7 2− in linear ranges of 80 pM to 1.95 mM with a low detection limit of 72 pM. Then PFOLS was used to detect Cr 2 O 7 2− in water samples (tap water, lake water, and wastewater) and agro-product samples (white rice, red rice, and black rice) with satisfactory results, revealing that PFOLS could be as a practical sensing platform for ultra-trace analysis of Cr 2 O 7 2− in environment and agro-product samples.

Self-Assembled G-fold DNA/Amino Acid Amphiphiles-Based Oxidase-Mimetic Materials Exhibiting Drug-Degrading and Photoswitchable Capabilities

Shichao Xu,Haifeng Wu,Yuanxi Liu,Zhen-Gang Wang

Recreating the intricate enzymatic sites within in artificial materials poses a significant challenge due to the complex folding characteristics of enzymes. In this study, we designed a supramolecular catalyst by assembling intramolecularly folding G-quadruplex DNA with Fmoc-amino acid aggregates and Cu2+. This supramolecular material possesses active sites and catalytic functions that rely on copper clusters, mirroring the functionality of catechol oxidase. Experimental and theoretical simulations showed that Fmoc-amino acids interact with G-quadruplex DNA through groove binding, facilitating Cu2+ coordination to both components, thereby enhancing the oxidative catalysis of Cu2+ upon assembly. Our catalyst exhibited excellent tolerance to high temperature, varying ionic strength, and extended room-temperature storage in aqueous solutions. Moreover, our supramolecular complex effectively catalyzed the degradation of doxorubicin, a drug with a high affinity for the DNA component, demonstrating its potential for removing pharmacological pollutants. Additionally, by integrating a photoisomerizable azobenzene derivative as a ligand to the G-quadruplex DNA, we can switch the activity of the enzyme-mimetic catalyst between on and off states by adjusting the irradiation wavelength. This multicomponent approach offers a promising avenue for the design and engineering of active, adaptable supramolecular catalysts.

Aggregation-induced emission-active amino acid/berberine hydrogels with enhanced photodynamic antibacterial and anti-biofilm activity

Yan-Yan Xie,Yan-Wen Zhang,Xiao-Zhi Liu,Xiao-Fang Ma,Xiao-Tong Qin,Shi-Ru Jia,Cheng Zhong

Bacterial infections are a great threat to public health. The development of materials which prevent biofilm formation, and exhibit antibacterial properties, but are otherwise biocompatible, is a major research goal. In this study, a novel hybrid hydrogel with aggregation-induced emission (AIE) behavior was prepared through the self-assembly of commercial N -(9-fluorenylmethoxycarbonyl)- L -phenylalanine (Fmoc-F) and berberine chloride (BBR). BBR self-assembled into nanoparticles which dispersed throughout the gel network, and endowed the hydrogel with AIE functionality. Fmoc-F/BBR hydrogel displayed enhanced antibacterial and anti-biofilm activity against both Gram-negative ( Escherichia coli ) and Gram-positive ( Staphylococcus aureus ) bacteria when used as the active agent in the photodynamic therapy (PDT) process. Additionally, negligible cytotoxicity and low hemolytic activity were displayed. In vivo infection studies showed that Fmoc-F/BBR hydrogel facilitated the repair of wounds in rats infected with S. aureus . Therefore, Fmoc-F/BBR hydrogel provides potential applications in biomedicines, and especially in wound healing.

Film-Spotting chiral miniPEG-γPNA array for BRCA1 gene mutation detection

Bo Dong,Kaixuan Nie,Huanhuan Shi,Lemeng Chao,Mingyang Ma,Fengxiao Gao,Bo Liang,Wei Chen,Mengqiu Long,Zhengchun Liu

Peptide nucleic acids array technology is a method of greatly increasing the throughput of laboratory processes to efficiently perform large-scale genetic tests. Diethylene glycol-containing chiral γPNA (miniPEG-γPNA) is considered to be the best PNA derivative and one of the best candidates for gene detection, because it can hybridize DNA with greater affinity and sequence selectivity than DNA and ordinary aminoethylglycyl PNA ( aeg PNA). Herein, miniPEG-γPNA probes are synthesized by 9-fluorenylmethyloxycarbonyl (Fmoc) solid phase peptide synthesis (SPPS) in a mild condition, and a new biochip fabrication method “Film-Spotting” is invented, by which γPNA arrays with regular pattern, uniform luminance, and very low fluorescence background are obtained easily and cheaply. The miniPEG-γPNA array can effectively distinguish the full matched and mismatched targets in SSarc buffer, serum and urine, and the detection limit of complementary DNA is less than 5.97 nM. A miniPEG-γPNA array for BRCA1 gene mutation (3099delT) detection is also fabricated with a very good detection performance. This work provides an effective avenue for the diagnosis of breast cancer biomarker and expands the application of miniPEG-γPNA in the field of biochip.

Colorimetric Sensor Based on the Oxidase-Mimic Supramolecular Catalyst for Selective and Sensitive Biomolecular Detection

Junhong Liu,Haifeng Wu,Yuanxi Liu,Zhen-Gang Wang

We have engineered a colorimetric sensor capable of selective and sensitive detection of amino acids. This sensor employs a supramolecular copper-dependent oxidase mimic as the probe, stemming from our prior research. The oxidase mimic is constructed through the self-assembly of commercially available guanosine monophosphate (GMP), Fmoc-lysine, and Cu2+. It catalyzes the formation of a red product with a maximum absorbance at 510 nm. The changes in color and absorbance are responsive to both the concentrations and types of amino acids present. This effect is most pronounced in the presence of histidine, with a detection limit (LOD) of 6.4 nM. Furthermore, the catalytic probe can distinguish histidine from histamine and imidazole propionate, as well as 1-methyl-histidine from 3-methyl-histidine, based on their distinct coordination capacities with copper. This underscores the high selectivity of the sensing platform. Both theoretical simulations and experimental results (including UV–vis spectra, fluorescence, and EPR) indicate that the amino acids may engage in copper center coordination, thereby impeding O2 access to copper─a pivotal aspect of the oxidase catalysis. This sensing platform, characteristic of its swift response, simple fabrication, and exceptional sensitivity and selectivity, can also be applied to detect other biological analytes such as nucleotides. It holds potential for use in environmental and biochemical analyses.

Oxidase-Mimetic Nanocatalyst Based on Geometry-Dependent Biomolecular Self-Assembly

Shan Li,Haifeng Wu,Yuanxi Liu,Baoli Zhang,Shichao Xu,Zhen-Gang Wang

Enzymes rely on protein folding to create functional sites essential for their catalytic activity. Reproducing enzymatic active sites in artificial systems has been difficult due to the intricate folding patterns that enzymes demonstrate. Inspired by natural systems, we developed a supramolecular copper-cluster-dependent catechol oxidase. This system is constructed through the self-assembly of flavin adenine dinucleotide (FAD) with fluorenylmethyloxycarbonyl-modified amino acids (e.g., Fmoc-lysine) and Cu2+ ions. Fmoc-amino acids self-associate through fluorenyl stacking, facilitating the formation of oxidase-mimetic copper clusters. FAD self-assembles with Fmoc-amino acids via aromatic stacking, enabling the incorporation of adenine and amino acid groups to form a coordination sphere around Cu2+. The self-assembly of FAD with Fmoc-amino acids enhances the oxidase-like activity of Cu2+ by over 100-fold. It is estimated that each copper can catalyze the transformation of ca. 80 substrate molecules within 15 min. The catalyst demonstrated thermophilic properties and excellent storage stability. When exposed to 460 nm light, the activity of the FAD/Fmoc-K/Cu2+ complex is further enhanced, potentially due to cooperative interaction between FAD and lumichrome generated from FAD photodegradation. These findings have implications for the design of advanced enzyme-mimetic catalysts.
