A 3D porous graphene aerogel@GOx based microfluidic biosensor for electrochemical glucose detection

Jian Xu,Kangkai Xu,Yu Han,Di Wang,Xiao Li,Tao Hu,Hong Yi,Zhonghua Ni

As a chronic disease, diabetes may result in serious complications that endanger the health and life of patients. Accurate and real-time detection of blood sugar levels is of great significance for the prevention and treatment of diabetes. In this paper, an enzymatic electrochemical microfluidic biosensor for glucose detection was developed based on a three-dimensional (3D) porous graphene aerogel and glucose oxidase (GOx). A graphene aerogel was prepared by freeze-drying a graphene hydrogel and has a high electrical conductivity, the 3D porous structure provided a good near-biological condition for GOx and the increased specific surface area allowed more GOx to be immobilized on the graphene aerogel. The microfluidic system greatly reduced the consumption of samples during tests. Amperometric measurements were carried out to test glucose concentrations, and the enzyme biosensor showed a linear range from 1 mM to 18 mM (R2 = 0.991). The limit of detection (LOD) was 0.87 mM (S/N = 3) and the sensor showed excellent selectivity and stability. Finally, monitoring glucose in serum samples was achieved by the proposed sensor and good recoveries were obtained. Due to its excellent performance, the proposed biosensor has a favorable application prospect in the prevention and clinical diagnosis of diabetes. Furthermore, our method, which is used to prepare a graphene aerogel modified electrode in a microfluidic chip, can be widely used in various electrochemical sensors.

Porous Fe₃O₄-GOx theranostic nanoplatform for tumor imaging and starvation/photothermal synergistic therapy

Xuan Shang,Mingyue He,Huiying Jiang,Fangying Jiang,Deping Wang,Jimin Cao,Yan Tan,Jigen Li,Yanlin Feng,Xin Zhou

Cancer remains a global health challenge, calling for integrated strategies that combine precise diagnosis with synergistic treatment. Although glucose oxidase (GOx)-mediated starvation therapy disrupts tumor metabolism by converting glucose into cytotoxic hydrogen peroxide (H 2 O 2 ), its clinical translation is hindered by inefficient delivery and insufficient monotherapy efficacy. To address these issues, we developed a multifunctional nanoplatform by encapsulating GOx within hollow porous superparamagnetic Fe 3 O 4 nanoparticles (Fe 3 O 4 -GOx NPs) to preserve its catalytic activity and enable high loading capacity. This system simultaneously enables magnetic resonance imaging (MRI), photothermal therapy, and enzyme-mediated catalytic intervention. In vitro studies demonstrated strong T 2 -weighted MRI contrast for precise tumor localization, together with near-infrared laser-triggered hyperthermia that enhance the efficacy of GOx-mediated starvation therapy, elevating oxidative stress and promoting apoptosis in 4T1 cells. In 4T1 tumor-bearing mice, a single administration of Fe 3 O 4 -GOx NPs combined with laser irradiation elicited potent tumor suppression through the synergistic effects of intratumoral ROS generation, glucose depletion, and localized photothermal ablation, outperforming either modality alone. Histopathological assessment revealed negligible systemic toxicity and preserved organ integrity. This work presents an integrated theranostic nanoplatform that unites tumor-targeted imaging, enzyme- driven metabolism intervention, and photothermal therapy into a single system, offering a promising strategy for precision cancer management.

Multifunctional Nanotherapeutic Platform: Metformin Potentiates Ce6/GOX-based Targeted Synergistic Therapy for Colorectal Cancer

Xinyu Mao,Shiwei Liu,Xinhao Li,Menghui Zhang,Peng Shen,Yaning Zhu,Xiaozhong Yang

Colorectal cancer (CRC) is the third most prevalent malignancy globally. Its tumor microenvironment (TME) heterogeneity and chemotherapy resistance compromise the efficacy of conventional treatments such as surgery and chemotherapy. This study developed a multifunctional nanotherapeutic platform by synthesizing mesoporous silica nanoparticles (MSNs) via a modified Stöber method. The carboxylated MSN surface was sequentially loaded with metformin (Me), chlorin e6 (Ce6), and glucose oxidase (GO X ), constructing an Me/Ce6@MSN-GO X (MCMG) nanosystem. The synthesized MSNs demonstrated excellent porosity, pore volume, and high loading capacity. In vitro and in vivo studies showed that MCMG reversed the Warburg effect via the AMPK/ACC/mTOR axis, reprogrammed tumor energy metabolism, and enhanced GO X -induced starvation therapy and Ce6-based photodynamic therapy (PDT). MCMG also alleviated TME hypoxia by suppressing HIF-1α expression, inducing an approximately 2-fold increase in intracellular reactive oxygen species levels and culminating in a 71% tumor inhibition rate in vivo. This platform integrated Me’s metabolic regulation with Ce6-mediated PDT and GO X -driven starvation therapy to achieve synergistic CRC eradication. MCMG enables multifunctional theranostics through synergistic mechanisms (metabolic modulation, oxidative damage, and nutrient deprivation) coupled with fluorescence imaging capabilities, presenting a novel strategy for CRC treatment.

Janus-structured GOx-Fe/cu Nanozymes for broad-range glucose detection and Milk analysis

Jiajie Xu,Yuhang Huang,Jiayu Xu,Siyu Hu,Xiangyang Tan,Liangchong Yu,Taolei Sun,Guanbin Gao

Glucose oxidase (GOx) is unstable under temperature/pH variations, limiting its biosensing applications. Existing nanozyme-based glucose sensors suffer from narrow detection ranges (0.2–12.0 mM) and poor stability. Herin a Janus-structured GOx-Fe/Cu nanozyme was synthesized via one-pot self-assembly with Fe₁₋ₓS and GOx partially-covered on Cu₃(PO₄)₂ nanoflowers. This nanozyme achieved an ultra-broad glucose detection range (0.5–20 mM, LOD = 34.18 μM), 40 times wider than existing nanozyme, with enhanced stability (>80% activity after 7 h at pH 3 and 5 h at 60 °C) and high specificity. When integrated into a chitosan/agarose hydrogel, it enabled visual glucose quantification through smartphone-based portable detection. In milk samples, it showed reliable glucose and H₂O₂ detection with recovery rates of 95–121% and relative standard deviations (RSD) of 0.31%–8.26%, demonstrating practical applicability. This Janus-structured GOx-Fe/Cu nanozyme improves GOx stability and sensing performance, and the hydrogel platform enables portable detection of glucose and hydrogen peroxide in food chemistry.

Colorimetric detection of blood glucose based on GOx@ZIF-8@Fe-polydopamine cascade reaction

Zhenghong Zhao,Tianran Lin,Wenren Liu,Li Hou,Fanggui Ye,Shulin Zhao

Zeolitic imidazolate framework-8 (ZIF-8) has become one of the most typical examples of nanostructures for multi-enzyme immobilization due to its economical, mild and easy synthesis process. However, ZIF-8 nanocrystals are easily decomposed under acidic conditions. To solve this problem, the Fe-polydopamine (Fe-PDA) was bonded with ZIF-8 surface to form [email protected] hybrid shell with good stability. Based on this, we developed glucose [email protected] @Fe-PDA ( [email protected] @Fe-PDA) integrated nanozymes (INAzymes) with cascade reactions via a mild and environmentally friendly method. In order to synthesize the INAzymes, GOx was first embedded in ZIF-8 by coprecipitation ( [email protected] ), and then [email protected] was bonded with Fe-PDA, which acted as a peroxidase mimic. The [email protected] hybrid shell protected the INAzymes nanostructure from degradation under acidic conditions, which results in good chemical stability of the [email protected] @Fe-PDA. In the INAzymes system, glucose is converted to gluconic acid by GOx in the presence of oxygen to produce H 2 O 2 as an intermediate. The H 2 O 2 reacts rapidly with Fe-PDA to generate OH, which oxidizes 3,3′,5,5′-tetramethylbenzidine (TMB). The UV absorbance of oxidized TMB is directly proportional to the glucose concentration, and has a good linear relationship in the range of 5.0–100.0 μM glucose with detection limit of 1.1 μM. The INAzymes system has been successfully applied to rapid colorimetric detection of blood glucose levels. The INAzymes system exhibits high catalytic activity, excellent sensitivity, and enhanced chemical stability, playing great promise in clinical diagnosis and biosensing.

Nanoreactor activated in situ for starvation-chemodynamic therapy of breast cancer

Linyu Gao,Xiangyang Xuan,Mingli Sui,Jingjing Wang,Yaping Wang,Huijuan Zhang

The nano-drug delivery system activated by the tumour microenvironment (TME) can effectively treat tumours with low toxicity. Based on a high level of reductive GSH in TME and the different coordination properties of Fe ions, this project intended to prepare a GSH-activated cascade catalytic nanoreactor for breast cancer treatment using Fe3+/Fe2+ as the molecular switch. In this study, the glucose oxidase (GOx) loaded iron alginate nano hydrogel (FeAlg/GOx) was prepared by the simple one-step titration method. Results showed that FeAlg/GOx could remain stable during in vivo circulation to avoid hypoglycaemia. When it reached the targeted tumour site, reductive GSH can reduce Fe3+ to Fe2+. Thereafter, FeAlg/GOx nanogel was broken and GOx was released to consume the essential nutrient glucose (Glu) to achieve tumour starvation therapy. Next, the substrate H2O2 generated by the reaction between GOx and Glu can be catalysed by Fe2+ to produce highly cytotoxic •OH in situ, which could further kill tumour cells. The in vivo pharmacodynamics results demonstrated that compared with the control group (V/V0 = 8.36 ± 1.73), FeAlg/GOx group showed the most significant anti-tumour effect with V/V0 of 3.08 ± 1.06. In conclusion, this "inactivated" FeAlg/GOx nanogel can be converted into "activated" therapeutic substances in situ to achieve starvation-chemodynamic combined treatment for breast cancer.

Temperature-Sensitive Nano-GOx Combined with Downregulation of Tumor Stemness to Initiate Robust Antitumor Efficacy

Panpan Gu,Jinhu Liu,Tong Gao,Qingping Ma,Shuying Gao,Nan Li,Weihan Zhang,Zhenxing Xia,Qinglin Yang,Weiwei Mu,Shuang Liang,Shunli Fu,Shijun Yuan,Suyun Wei,Jie Liu,Yuxin Yang,Xiaoxin Yan,Yongjun Liu,Cong Wang,Na Zhang

GOx-mediated glucose depletion offers an alternative noninvasive strategy for tumor therapy, but its lower catalytic activity in vivo limits its clinical application. Herein, we designed a temperature-sensitive nano-GOx (NG) that was constructed by gold nanorods chemically modified with GOx (AuNRs-GOx) and coated with temperature-sensitive lipids. The chemical linkage could maintain the natural conformation of GOx, ensuring that NG exerted powerful catalytic activity within the tumor and initiated antitumor immune response through moderate starvation and mild photothermal therapy (mPTT) to coregulating dendritic cells (DCs) and tumor-associated macrophages (TAMs). Ulteriorly, VTNG was obtained by NG coloading with verteporfin (VP) and evofosfamide (TH-302). VTNG demonstrated temperature-sensitive triggered drug release when exposed to near-infrared laser irradiation. NG exacerbated the degree of TME hypoxia and facilitated the activation of TH-302. Meanwhile, VP enhanced tumor cell sensitivity by decreasing the stemness of the tumor cells, thus realizing the effective killing of tumor cells and further enhancing the therapeutic effect of NG. Notably, VTNG had a significant antitumor effect in melanoma models compared with first-line melanoma therapy and formed an immune memory effect. In conclusion, VTNG provided an effective approach to enhance the therapeutic effect of GOx for tumor treatment.

A self-supplied O2 versatile nanoplatform for GOx-mediated synergistic starvation and hypothermal photothermal therapy

Bo Zhang,Xinyu Li,Weibin Shu,Yu-Shun Yang,Hai-Liang Zhu,Chenwen Shao

Hypothermal photothermal therapy, as a non-invasive therapy method, is becoming ever more prevalent. However, due to cellular heat resistance from the generation of heat shock proteins (HSPs) and the monotonicity of therapeutic modality, the antitumor efficacy is severely restricted. Herein, a self-supplied O 2 multifunctional nanoplatform mediated by glucose oxidase (GOx) is developed, which may combine starvation therapy (ST) and hypothermal photothermal therapy (HPTT) for tumor therapy. The obtained nanoplatform, [email protected] @ [email protected] (UMIGH), was assembled by the UIO-66 core, MnO 2 , indocyanine green (ICG) and hyaluronic acid (HA). GOx was introduced to decrease the ATP level thus alleviating the HSPs-mediated heat tolerance, while the loaded MnO 2 could decompose intratumoral H 2 O 2 into O 2 , which enhanced the effect of HPTT and ST. Due to the coated HA, UMIGH could target the high expressed CD44 in CT26 tumor cells. The active targeting and NPs-associated passive targeting promoted the uptake of nanoparticles. Furthermore, ICG encapsulated in UMIGH NPs could also accurately image tumors. This synergistic treatment strategy of HPTT and ST exhibited an excellent anti-proliferation effect on colon cancer cells in vitro and mouse xenograft model in vivo , which might bring an informatic strategy for future investigations.

A nanostructured p-NiO/n-Bi4Ti3O12 heterojunction for direct GOx electrochemistry and high-sensitivity glucose sensing

Weiwei Mao,Bin Cai,Zhizhen Ye,Jingyun Huang

A glucose oxidase (GOx) based p-NiO/n-Bi 4 Ti 3 O 12 sandwich heterojunction was fabricated and explored as a glucose electrochemical sensor. The constructed GOx/p-NiO/n-Bi 4 Ti 3 O 12 sandwich structure effectively prevents the leakage of GOx. Dandelion-like Bi 4 Ti 3 O 12 nanowires were directly grown on the titanium foil substrates, and dense NiO nanoparticles were deposited by employing pulsed laser deposition at room temperature. Amperometric measurements towards glucose were carried out, and efficient direct electron transfer was successfully achieved. The enzyme biosensor showed a linear range from 20 μM to 3.55 mM (R = 0.999) with an ultrahigh sensitivity of 215 μA mM −1  cm −2 , response time within 2 s, low detection limit of 1.26 μM (S/N = 3) and high specificity. The apparent Michaelis-Menten constant (Km app) was estimated to be 0.22 mM. Intercrossed nanowires and interconnected pores favor the electrons transfer and the massive GOx loading. Binder-free feature leads to a seamless connection between Bi 4 Ti 3 O 12 nanowires and the substrate. Co-catalysis of GOx and p-NiO/n-Bi 4 Ti 3 O 12 heterojunction plays a synergetic role in gaining performance boosts. The GOx based p-NiO/n-Bi 4 Ti 3 O 12 heterojunction biosensor remedies the limitation of enzyme-based glucose sensor and be potentially applied in practice.

Glucose-triggered self-perpetuating catalytic cascade in GOx@CuO2 nanoreactors for treating bacteria-infected diabetic wounds

Jun Wang,Yi Huang,Yan Cao,Wenjiao Zhu,Yuhui Chen,Yan Chen,Yuanyuan Zhang,Jianfeng Cui,Yuanxiang Wang,Kai Lan,Yanxia Lin

The hyperglycemic microenvironment of diabetic wounds is highly susceptible to bacterial infection, persistent inflammation, and impaired angiogenesis, severely delaying the healing process. To overcome the limitations of current therapeutic strategies, this study innovatively developed an intelligent cascade nanoreactor, GOx@CuO 2 , which utilizes excess glucose (Glu) as a driving force. The key innovation of this system is its exploitation of the excess glucose at the lesion site. Catalyzed by glucose oxidase (GOx), the Glu is converted into gluconic acid and hydrogen peroxide (H 2 O 2 ) which simultaneously accomplishes the tripartite microenvironmental modulation of Glu depletion, acid production, and H 2 O 2 supply. Under acidic conditions, GOx@CuO 2 exhibits efficient peroxidase-like (POD-like) activity, catalyzing the generation of hydroxyl radicals (·OH) to achieve potent antibacterial effects. As glucose is gradually consumed and the pH increases, GOx@CuO 2 intelligently switches to a catalase-like (CAT) mode, decomposing residual H 2 O 2 into oxygen (O 2 ). This alleviates oxidative stress and promote angiogenesis, forming an adaptive synergistic therapeutic cycle of “antibacterial-promoting healing”. In vivo experiments confirmed that the nanoreactor significantly improved the wound microenvironment, achieving a wound closure rate of 93.4 % within 14 days. This study not only proposes a novel non-antibiotic therapeutic strategy responsive to the lesion microenvironment but also provides new insights for designing intelligent enzyme-catalyzed systems for tissue repair related to metabolic diseases.

MnO2/Au-Ag nanozyme-GOx cascade system for sensitive colorimetric glucose detection and test strip applications

Qiao Yuchun,Wu Yiying,He Jiahui,Chen Wen,Fan Jiaqi,Zhang Zhicong,Shen Jialu,Su Mengping,Wang Qidong,Li Shuai,Shen Yingjie,Shang Jizhen,Wang Hua

Glucose is a vital energy source, and its accurate monitoring is crucial for the diagnosis and management of metabolic diseases. Herein, we report a simple colorimetric sensing platform for glucose detection based on MnO 2 /Au-Ag nanozymes, glucose oxidase (GOx), and 3,3′,5,5′-tetramethylbenzidine (TMB). In this cascade system, GOx catalyzes the oxidation of glucose to generate H 2 O 2 , which is subsequently decomposed by MnO 2 /Au-Ag to produce reactive species that oxidize TMB, yielding a visible blue signal. The incorporation of Au-Ag nanoparticles provides additional catalytic sites and promotes interfacial electron transfer, thereby enhancing the catalytic performance of MnO 2 . The platform exhibits a linear response over 0.025-0.20 mM with a detection limit of 0.012 mM. It enables reliable glucose detection in artificial sweat and 80-fold diluted serum samples. In addition, portable nitrocellulose test strips were developed for visual and smartphone-assisted glucose detection. This strategy provides a simple and portable approach for low-concentration glucose analysis and offers potential for point-of-care applications after further validation. Graphical

Metal-to-Ligand Charge Transfer Chirality Sensing of d-Glucose Assisted with GOX-Based Enzymatic Reaction

Junjie Hao,Yiwen Li,Xiaoqian Xu,Fenghuan Zhao,Ruikun Pan,Junzi Li,Haochen Liu,Kai Wang,Jiagen Li,Xi Zhu,Marie-Hélène Delville,Ming Zhang,Tingchao He,Jiaji Cheng

Chiral transition metal oxides nanoparticles (NPs) with tunable optical properties are widely accepted as promising toolbox for chiral recognition, stereoselective synthesis, and chiroptical devices. Herein, chirality-based strategy is presented for discrimination of d -glucose from its enantiomer through a cooperative synergy between chiral cysteine capped MoO 2 NPs and the glucose oxidase nanosystem. The valence-state-dependent chirality induced by metal-ligand charge transfer effect is found to be ultrasensitive to its redox environment such as the presence of hydrogen peroxide, which is a key indicator of the stereoselective enzymatic reaction between glucose oxidase and d -glucose. With this know-how, glucose enantiomers can be precisely quantified with a limit of detection of 0.446 µ m . Such a chiral bio-nanosystem would be an ideal platform for a rational design of new types of biosensors, photocatalysts, and chirality-based nanodevices.
