Trehalose prevents glyphosate-induced kidney damage in roosters via inhibiting apoptosis

Liu Shuang,Qiu Bo,Li Yue,Wang Hai-Peng,Zhong Zhao-Jin,Wang Lin

Glyphosate (Gly) is a globally prevalent herbicide, which poses significant health risks to human and animals via the food chain. The kidney is the primary excretory organ and a site of residual accumulation during Gly exposure. Trehalose (Tre) has been shown to prevent kidney injury induced by various toxicants due to its anti-apoptotic properties. However, the potential protective effect of Tre on Gly-induced kidney damage in poultry remains unclear. Thus, 60 one-day-old healthy Hy-line Brown roosters were randomly divided into three groups ( n  = 20), i.e., control group (basal diet without Gly), Gly group (200 mg Gly/kg basal diet), and Gly + Tre group {(200 mg Gly plus 5 g Tre)/kg basal diet}. The experiment lasted for 120 days, and serum and kidney samples were finally collected for respective analysis. Data first showed that Tre treatment can significantly alleviate Gly-induced renal injury in roosters, as evidenced by serum levels of creatinine and blood urea nitrogen as well as renal histopathological observations. Tre also significantly inhibited the elevated abundance of renal injury marker proteins KIM-1 and NGAL induced by Gly. Meanwhile, Tre obviously alleviated Gly-induced ultrastructural abnormalities in the nuclei of renal tissues. Additionally, Tre markedly inhibited Gly-induced apoptotic assays in rooster kidneys, as demonstrated by the results from TUNEL staining and the protein abundance of apoptotic markers cleaved caspase-3 and cleaved PARP. In summary, these findings suggest that Tre exerts its protective effects against Gly-induced kidney damage in roosters via inhibiting apoptosis.

Insights Into Proliferation Effects of Low-Dose Glyphosate on Phytoplankton Communities

Su Jinzhu,Su YuPing,weng Yuxin,Ayub Gohar,She Chenxing,Xiao Yumei

Glyphosate-based herbicides are among the most widely used agricultural chemicals globally, and their widespread application presents risks to environmental health and aquatic ecosystems. Continuous glyphosate inputs disrupt phytoplankton communities, potentially triggering harmful algal blooms. This study examines the proliferation of microalgal species exposed to low glyphosate concentrations (0.05 mg/L) and various phosphorus sources, with a particular focus on C-P and C-O-P bond phosphonates, which have been insufficiently studied in previous research. We hypothesized that cyanobacteria might exhibit a competitive growth advantage over other algal species when exposed to C-P bond glyphosate, especially under phosphorus-limited conditions. In monoculture experiments, Microcystis aeruginosa and Peridinium umbonatum var. inaequale significantly increased their biomass when cultured with C-P bond phosphonates, whereas Scenedesmus bijuga failed to thrive under similar conditions. Peridinium umbonatum var. inaequale also displayed increased soluble protein content in response to glyphosate stress, indicating an adaptive stress response. In co-culture experiments, M. aeruginosa demonstrated greater tolerance to glyphosate than P. umbonatum var . inaequale , though biomass increases were not significantly correlated with soluble protein or APA. Sediment–water interface experiments revealed that glyphosate exposure significantly promoted cyanobacterial biomass, which was approximately five times greater than that of the inorganic phosphorus group. Notably, when cyanobacterial biomass exceeded 20% of the total, Cyanophyta replaced Chlorophyta as the dominant group, suggesting a potential competitive advantage under low-dose glyphosate exposure. These findings highlight that glyphosate may promote cyanobacterial dominance by altering phytoplankton community composition, potentially contributing to the increased frequency of harmful algal blooms in nutrient-limited aquatic environments.

Effective removal of aqueous glyphosate using CuFe2O4@biochar derived from phragmites

Dongmei Jia,Min Liu,Jiangbao Xia,Changhai Li

BACKGROUND The excess use of glyphosate in both agricultural and nonagricultural areas has led to tremendous health (e.g. cardiac and respiratory impacts) and environmental concerns worldwide. There is an urgent need to find efficient ways to remove glyphosate residue from the aqueous environment. RESULTS Herein, an effective method for the removal of aqueous glyphosate is developed by employing nano-CuFe 2 O 4 modified biochar (BC) as an adsorbent. The CuFe 2 O 4 @BC adsorbents were prepared through hydrolyzing phragmite powders at 500 °C, followed by a simple coprecipitation method. The influence of initial glyphosate concentration, temperature, pH, time and coexisting species on the adsorption behavior of CuFe 2 O 4 @BC to glyphosate were fully examined. CONCLUSION It was found that the adsorption of glyphosate onto CuFe 2 O 4 @BC is highly dependent on the initial glyphosate concentration, pH and temperature, as well as coexisting species. The Freundlich and Langmuir models fitted the equilibrium isotherm data well, indicating the presence of both physisorption and chemisorption. In addition, the adsorption kinetic is well described by the pseudo-second-order kinetic model, which indicated that chemical adsorption may dominate the adsorption process. The CuFe 2 O 4 @BC adsorption of glyphosate presented a maximum adsorption capacity of 269.4 mg g −1 within 240 min, under 600 mg L −1 of glyphosate solution at 298 K and pH 4; this performance exceeds all other biochar-derived adsorbents for the removal of aqueous glyphosate encountered in the open literature. Such a cost-effective, high-performance adsorbent shows a great potential for the removal of aqueous glyphosate in industry. © 2019 Society of Chemical Industry

Chronic glyphosate exposure compromises male reproductive health: Golgi dysfunction mediating spermatogenesis impairment

Yulong Ren,Kai Chen,Bo Qiu,Haijing Zhang,Qing Liang,Lin Wang

Growing evidence links global male infertility to environmental endocrine disruptors, particularly pesticides. As the world's most extensively used herbicide, glyphosate (GLY) poses significant concerns for human reproductive health. While epidemiological studies suggest associations between GLY exposure and male infertility, conclusive evidence elucidating its reproductive toxicity remains limited and controversial. Thus, this study established an 18-week rat model of chronic GLY exposure to investigate its impact on male reproductive health. Data demonstrated that chronic dietary exposure to GLY reduced sperm quality and impaired acrosome integrity. Transcriptomic analysis revealed that GLY impairs sperm acrosome by disrupting acrosome assembly. Ultrastructural pathology and immunofluorescence observations further indicated that GLY disrupted acrosome biogenesis by reducing the accumulation and fusion of golgi associated PDZ and coiled-coil motif containing (GOPC)-decorated proacrosomal vesicles. Mechanistically, oxidative stress-mediated Golgi damage was identified as the primary driver of GLY-impaired proacrosomal vesicle release. Significantly, GLY-induced Golgi damage and the resultant sperm acrosomal malformation were effectively mitigated by supplementing with the antioxidant puerarin. Collectively, this study first establishes a mechanistic link between Golgi apparatus dysfunction and defective acrosome formation, underscoring redox imbalance as a pivotal mediator of GLY-induced reproductive toxicity. These findings provide novel mechanistic insights into environmental pollutant-driven oxidative stress as a risk factor for male reproductive health.

SERS detection of glyphosate by Suspended sandwich Ag/SiO2/Ag nanopore array

Haiyu Tian,Haidong Qi,Yongjun Zhang,Yaxin Wang,Xiaoyu Zhao,Renxian Gao

Surface-enhanced Raman scattering (SERS) is a powerful surface analysis technique that significantly amplifies the Raman scattering signals of molecules adsorbed onto metallic nanostructures, enabling highly sensitive detection of the analytes. The development of low-cost, high-performance SERS substrates through facile and straightforward fabrication methods represents a challenging and highly rewarding endeavor. Here, we develop a suspended sandwich Ag/SiO 2 /Ag nanopore array structure based on template-assisted method with significant plasma coupling effects, which shows an ultra-low detection limit (LOD of 2.91 × 10 −11  mol/L) for 4-MBA molecules, excellent sensitivity (EF = 5.76 × 10 8 ) and high uniformity and stability. COMSOL simulation confirms the electromagnetic coupling in the sandwich structure in addition to the hotspots along the nanopore brims. The proposed SERS substrate enables quantitative detection of glyphosate with the concentration down to 4.66 × 10 −9  mol/L and fulfills the requirements for monitoring pesticide residues.

Influence of Seed Properties and Operating Conditions on Secondary Nucleation of Glyphosate

Chenchen Zhang,Yixin Ma,Zhihao Wang,Shuo Jia,Yan Zhang,Li Yang,Hongxiao Yang,Jingcai Cheng,Chao Yang

Secondary nucleation critically governs crystal size distribution (CSD), yet its precise control remains challenging. Here, in situ process analytical technologies (PAT) were used to investigate the secondary nucleation of glyphosate. A method based on a defined nucleation core-interval was established to determine the average secondary nucleation rate, enabling quantitative assessment of seed size, seed loading, seed shape, cooling rate, and agitation rate. To clarify morphology effects, an additive-free approach combining isoelectric-point and cooling crystallization was developed to prepare spheroidized seeds. Under identical size conditions, spheroidized seeds markedly suppressed secondary nucleation relative to prismatic seeds, yielding larger and narrower crystals. Thus, a seed-shape factor was incorporated into the averaged secondary nucleation model, improving performance (R2: 0.54 to 0.95; ARD: 49.94% to 18.51%) and yielding more physically consistent exponents. These results highlight seed shape as a key regulator of secondary nucleation kinetics and offer an effective pathway for tailoring CSD.

Colorimetric sensing based on silver nanoparticle-functionalized copper porphyrin nanozyme for glyphosate

Li-Hong Yu,Rui Jiang,Cheng-Lin Yang,Yue-Hong Pang,Xiao-Fang Shen

Glyphosate , a widely used herbicide due to the broad-spectrum and high-efficiency properties, has the potential to accumulate in the human body via agricultural food and water sources throughout the food chain. Its safety for both human health and aquatic ecosystems remains a topic of significant debate. In this study, the presence of glyphosate could be directly detected through a notable colorimetric change in an aqueous solution. For this purpose, a silver nanoparticle-functionalized copper porphyrin metal-organic framework (AgNPs/Cu-TCPP) nanozyme was conveniently prepared for colorimetric sensing of glyphosate. The as-synthesized nanozyme that modified AgNPs with Cu-TCPP metal-organic framework as the precursor, has excellent peroxidase mimetic activity and specific recognition performance of glyphosate. The nanozyme demonstrated the ability to catalyze the oxidation of 3,3,5,5-tetramethylbenzidine (TMB) using H 2 O 2 , resulting in a marked change in color. When glyphosate was added, which bonded with Cu 2+ , led to the occupation of the surface-active sites of the nanozyme, thereby making its catalytic performance ineffective. The concentration of glyphosate exhibited a linear range spanning from 2 to 400 μmol L −1 , and the limit of detection was 0.67 μmol L −1 (S/N = 3). The colorimetric sensing approach employing AgNPs/Cu-TCPP nanozyme exhibits remarkable selectivity and reproducibility, enabling the detection of real samples with recovery rates of 98.3%–103.5%, indicating its potential applicability in safeguarding water and food quality.

Green Glyphosate Treatment with Ferrihydrite and CaO2 via Forming Surface Ternary Complex

Nuanqin Zhang,Hongwei Sun,Guangming Zhan,Junning Zu,Lizhi Zhang

Glyphosate (PMG) is a globally used broad-spectrum herbicide and receives environmental concerns because of its moderate persistence and potential carcinogenicity. Traditional PMG treatment methods often suffer from the generation of a more toxic and persistent aminomethylphosphonic acid (AMPA) intermediate. Herein, we develop a green method with ferrihydrite (FH) and CaO2 (FH/CaO2) via regulating the coordination of PMG with FH and Ca2+, where the phosphonate group of PMG preferentially binds to FH and its carboxylate side complexes with Ca2+ released by CaO2, forming a FH-PMG-Ca ternary surface complex. This unique ternary complex can redistribute electrons within the PMG molecule for its C–P activation and C–N bond stabilization, favoring the selective C–P bond attack of superoxide radical produced by the Fenton reaction between CaO2-derived H2O2 and FH, thus generating environment-friendly glycine instead of AMPA. The FH/CaO2 process realizes over 99% PMG degradation in industrial wastewater within 1 h, with residual PMG < 0.1 ppm and AMPA < 40 ppb. More importantly, the CaO2 consumption was as low as 3.1 mg of CaO2/mg of PMG, one-fifth those of previously reported CaO2-based counterparts. This study provides an effective and environment-friendly PMG treatment strategy and highlights the importance of surface coordination modes on the degradation pathway of PMG.

Glyphosate Disorders Soil Enchytraeid Gut Microbiota and Increases Its Antibiotic Resistance Risk

Qi Zhang,Chaotang Lei,Mingkang Jin,Guoyan Qin,Yitian Yu,Danyan Qiu,Yan Wang,Ziyao Zhang,Zhenyan Zhang,Tao Lu,W. J. G. M. Peijnenburg,Michael Gillings,Ziang Yao,Haifeng Qian

Pesticides promote the stable development of intensive global agriculture. Nevertheless, their residues in the soil can cause ecological and human health risks. Glyphosate is a popular herbicide and is generally thought to be ecologically safe and nontoxic, but this conclusion has been questioned. Herein, we investigated the interaction among soil fauna (Enchytraeus crypticus) exposed to glyphosate and found that glyphosate induced oxidative stress and detoxification responses in E. crypticus and disturbed their lipid metabolism and digestive systems. We further demonstrated that glyphosate disordered the gut microbiota of E. crypticus and increased the abundance of resistance determinants with significant human health risks. Empirical tests and structural equation models were then used to confirm that glyphosate could cause E. crypticus to generate reactive oxygen species, indirectly interfering with their gut microbiota. Our study provides important implications for deciphering the mechanisms of the ecotoxicity of pesticides under the challenge of worldwide pesticide contamination.

Glyphosate Induces Liver Macrophage Pyroptosis via Mitochondrial Damage-Mediated cGAS-STING Activation

Xiangyu Yu,Jiawen Ren,Ying Kang,Shizhi Wang,Jianrui Dou,Yongquan Yu

Glyphosate, the most widely used herbicide worldwide, is now ubiquitous in the environment, posing a growing threat to human health. While accumulating evidence has linked glyphosate exposure to liver injury, the underlying mechanisms remain unclear. In this study, based on data from NHANES 2013–2018, we identified significant associations between glyphosate exposure and abnormal liver function parameters in the general US population. A glyphosate-exposed mouse model was further established, and the results showed that hepatic accumulation of glyphosate induced direct histopathological damage and increased serum AST, ALT, and ALP levels in mice. Combined network toxicology and gene set analyses revealed that glyphosate activated liver macrophages, upregulating genes related to lipid metabolism, inflammation, and pyroptosis. The activation of the pyroptosis pathway was further confirmed by Western blot analysis of NLRP3 inflammasome-associated proteins. Mechanistically, glyphosate disrupted mitochondrial membranes and compromised mitochondrial function, leading to the release of mtDNA, which subsequently activated the cGAS-STING pathway in mouse livers and RAW264.7 macrophages. Moreover, glyphosate-induced NLRP3 activation in RAW264.7 cells was attenuated by the cGAS inhibitor. These findings provide a novel mechanistic insight into glyphosate-induced hepatotoxicity and reinforce the growing concern over its association with liver injury in humans.

Leaf metabolic influence of glyphosate and nanotubes on the Arabidopsis thaliana phyllosphere

Mingjing Ke,Yizhi Ye,Yan Li,Zhigao Zhou,Nuohan Xu,Lan Feng,Jinfeng Zhang,Tao Lu,Zhiqiang Cai,Haifeng Qian

Chemical exposure can indirectly affect leaf microbiota communities, but the mechanism driving this phenomenon remains largely unknown. Results revealed that the co-exposure of glyphosate and multi-carbon nanotubes (CNTs) caused a synergistic inhibitory effect on the growth and metabolism of Arabidopsis thaliana shoots. However, only a slight inhibitory effect was induced by nanotubes or glyphosate alone at the tested concentrations. Several intermediate metabolites of nitrogen metabolism and fatty acid synthesis pathways were upregulated under the combined treatment, which increased the amount of energy required to alleviate the disruption caused by the combined treatment. Additionally, compared with the two individual treatments, the glyphosate/nanotube combination treatment induced greater fluctuations in the phyllosphere bacterial community members with low abundance (relative abundance (RA) <1%) at both the family and genus levels, and among these bacteria some plant growth promotion and nutrient supplement related bacteria were markable increased. Strikingly, strong correlations between phyllosphere bacterial diversity and metabolites suggested a potential role of leaf metabolism, particularly nitrogen and carbohydrate metabolism, in restricting the range of leaf microbial taxa. These correlations between phyllosphere bacterial diversity and leaf metabolism will improve our understanding of plant-microbe interactions and the extent of their drivers of variation and the underlying causes of variability in bacterial community composition.

Citric acid-assisted ultrasmall CeO2 nanoparticles for efficient photocatalytic degradation of glyphosate

Hao Wu,Qian Sun,Jingyi Chen,Guan-Yu Wang,Dan Wang,Xiao-Fei Zeng,Jie-Xin Wang

Degradation of glyphosate has attracted much attention all over the world, since glyphosate has been demonstrated to affect the human health through the accumulation in food chain. Herein, we use citric acid as capping agent to synthesize highly-stable aqueous dispersions of monodispersed CeO 2 nanoparticles (NPs) with ultrasmall controllable size of 2.1–4.8 nm, high surface areas and rich oxygen defects, which were firstly used as photocatalyst for efficient degradation of glyphosate. The obtained CeO 2 NPs display a significantly size-dependent catalytic activity and a good stability performance. Ultrasmall 2.1 nm CeO 2 NPs have 6 times (UV) and 20 times (visible light) degradation rates of 4.8 nm counterpart, which can rapidly remove glyphosate from water at pH = 4 in 5 min (UV) and 20 min (visible light). And the related degradation kinetics data at pH = 4 are well related to the pseudo-first-order kinetic model. In this highly-efficient photocatalytic degradation process, the grafted citric acid can initiate the electron transfer to accelerate the regeneration of Ce 3+ sites as electron donors and enhance photocatalytic performance via ligand-to-metal charge transfer. This work provides a new method to prepare high-performance CeO 2 photo-nanocatalysts for the removal of glyphosate and emphasizes the important role of modifier, oxygen defects and particle size in seeking efficient photocatalytic materials.
