Influenza virus detection using an electrochemical biosensor based on DSN and RCA

Yuqi Yang,Shengjun Bu,Xiaoying Zhang,Qiqing Duan,Heyuan Meng,Zhuo Hao,Xiuxia He,Jiayu Wan

Infectious diseases caused by the influenza virus are the primary global concern. This study aimed to develop an electrochemical biosensor for detecting influenza A H1N1 virus based on duplex-specific nuclease (DSN) and rolling circle amplification (RCA) technologies. A single-stranded DNA (ssDNA) probe, comprising a recognition sequence for a target sequence of influenza virus H1N1-specific RNA sequence and primer sequence for initiating RCA, was designed and synthesized. A specific sequence of target H1N1 RNA triggered cyclical DSN cleavage of the probe. The released primer sequence then initiated RCA to generate detectable ssDNA-labeled multiple biotins. ssDNA was captured by streptavidin (SA) on the electrode surface and labeled with free Streptavidin-Horseradish Peroxidase (SA-HRP). The H1N1 influenza virus was quantitatively analyzed based on the changing current signal generated on the electrode surface. A linear range from 10 pM to 100 nM and a limit of detection of 0.44 pM were recorded. In addition, the biosensor demonstrated excellent selectivity, repeatability, and stability for analyzing H1N1 in real samples. This biosensor could be applied for the clinical diagnosis of influenza A H1N1 virus.

Yimeng An,Xiao Ding,Yabin Tian,Mengyi Zhang,Yun Ma,Xi Wu,Meina Cai,Lan Huang,Yifei Wang,Jincheng Tong,Yuanling Yu,Chenyan Zhao,Weijin Huang,Aiping Wu,Youchun Wang

Background The continuous mutations in the haemagglutinin gene in A/H1N1 influenza viruses drives antigenic drift, necessitating frequent vaccine updates. Pseudovirus systems, with their flexible HA protein presentation and high-throughput luciferase-based reporter assays, provide a versatile approach for mapping the antigenic landscape of A/H1N1 viruses. Methods This study constructed a pseudovirus library comprising 123 representative strains and 21 vaccine strains to analyse the antigenic evolution of human A/H1N1 from 1918 to 2023. Additionally, a machine learning-based antigenicity evaluation model, PN-AgEvaH1, was developed using pseudovirus neutralisation assay data. Findings The pseudovirus neutralisation experiments identified eight antigenic clusters, each associated with distinct epidemiological characteristics. All vaccine strains were found to align closely with the prevalent circulating strains of their respective years. Key amino acid residues contributing to pdm09 antigenic clusters were also identified. Furthermore, the PN-AgEvaH1 model achieved a high antigenicity evaluation accuracy with a receiver operating characteristic area under the curve (ROC AUC) score of 0.990, providing a reliable tool for characterising A/H1N1 antigenic clusters and guiding vaccine selection. Interpretation The pseudovirus neutralisation (PN) assay demonstrated high accuracy in identifying antigenic matches between vaccine and circulating strains, highlighting its value in vaccine strain selection. This study represents the large-scale antigenicity evaluation of human A/H1N1 viruses using the PN assay and underscores its potential as a complementary or alternative approach to the HI assay in both experimental and computational applications. Dual-Mode Lateral Flow Immunoassay Based on “Pompon Mum”-Like Fe3O4@MoS2@Pt Nanotags for Sensitive Detection of Viral Pathogens

Meimei Xu,Shuai Zhao,Chenglong Lin,Yanyan Li,Weida Zhang,Yusi Peng,Rui Xiao,Zhengren Huang,Yong Yang

Lateral flow immunoassay (LFIA) has been widely used for the early diagnosis of diseases. However, conventional colorimetric LFIA possesses limited sensitivity, and the single-mode readout signal is easily affected by the external environment, leading to insufficient accuracy. Herein, multifunctional Fe3O4@MoS2@Pt nanotags with a unique “pompon mum”-like structure were triumphantly prepared, exhibiting excellent peroxidase (POD)-like activity, photothermal properties, and magnetic separation capability. Furthermore, the Fe3O4@MoS2@Pt nanotags were used to establish dual-mode LFIA (dLFIA) for the first time, enabling the catalytic colorimetric and photothermal dual-mode detection of severe acute respiratory syndrome coronavirus 2 nucleocapsid protein (SARS-CoV-2 NP) and influenza A (H1N1). The calculated limits of detection (cLODs) of SARS-CoV-2 NP and H1N1 were 80 and 20 ng/mL in catalytic colorimetric mode and 10 and 8 ng/mL in photothermal mode, respectively, demonstrating about 100 times more sensitive than the commercial colloidal Au-LFIA strips (1 ng/mL for SARS-CoV-2 NP; 1 μg/mL for H1N1). The recovery rates of dLFIA in simulated nose swab samples were 95.2–103.8% with a coefficient of variance of 2.3–10.1%. These results indicated that the proposed dLFIA platform showed great potential for the rapid diagnosis of respiratory viruses.

Entropy-driven amplification strategy-assisted lateral flow assay biosensor for ultrasensitive and convenient detection of nucleic acids

Shasha Li,Hong-Min Meng,Hong Zong,Juan Chen,Jianjun Li,Lin Zhang,Zhaohui Li

Accurate, sensitive and rapid nucleic acid tests are important to implement timely treatment measures and control the spread of disease. Herein, we developed a novel portable platform for highly sensitive and specific detection of nucleic acids by integrating an entropy-driven amplification strategy into lateral flow assay (LFA) biosensor. We find that introducing an entropy-driven amplification strategy yields bright intensities on the test line of LFA stirp, which results in improved sensitivity for targeted nucleic acid detection. The developed LFA biosensor showed good reproducibility, specificity and sensitivity for target DNA and H1N1-RNA detection with a low detection limit of 1.43 pM and 2.02 pM, respectively. Its practical potential was also verified by detecting the target nucleic acid in human serum. More importantly, the design of an entropy-driven amplification strategy in this portable platform retained the convenient, rapid and low-cost characterizations of LFA biosensor due to the compact amplification principle and the elimination of enzyme use. Thus, we believe that this assay biosensor will certainly report its own position in the timely detection of nucleic acid, especially when the medical environment and resources are fewer.

TRIM21 reduces H1N1-induced inflammation and apoptosis by regulating the TBK1–IRF3 signaling pathway in A549 cells

Yuan Juan,Pan Jianli,Zhang Xiaofang,Gao Rui

Triple motif protein 21 (TRIM21) has an antiviral function that inhibits various viral infections. However, its role in the progress of influenza A virus (IAV) infection is unclear. In this study, we investigated the role and molecular mechanism of TRIM21 in IAV infection. RT-qPCR was used to determine the level of TRIM21 mRNA, and ELISA was used to measure the levels of IFN-α, IFN-β, IL-6, and TNF-α. The levels of the TRIM21, NP, TBK1, IRF3, p-TBK1, and p-IRF3 proteins were estimated by Western blot. The results showed that, after IAV infection, TRIM21 was upregulated in clinical patient serum and A549 cells, and this was correlated with the IFN response. Overexpression of TRIM21 reduced IAV replication and transcription in in vitro cell experiments. TRIM21 also increased IFN-α and IFN-β levels and decreased IL-6 and TNF-α levels in A549 cells. In addition, overexpression of TRIM21 inhibited IAV-induced apoptosis. Further experiments demonstrated that TBK1-IRF3 signaling was activated by TRIM21 and was involved in the inhibitory effect of TRIM21 on virus replication. In summary, our study suggests that TRIM21 inhibits viral replication by activating the TBK1-IRF3 signaling pathway, further inhibiting the infection process of IAV.

Label-free and ultra-sensitive H1N1 influenza virus immunosensor based on a triple-enhanced LIG/PB/PEDOT/Au microelectrode

Yanxia Li,Fangqing Yu,Wen Li,Liping Liu

Laser-induced 3D porous graphene enables scalable, low-cost biosensing. • PB/PEDOT/Au composite synergistically enhances stability, antifouling, and electroactivity. • The immunosensor achieves ultrasensitive detection of H1N1 and a wide dynamic range. • Label-free and pretreatment-free detection is successfully demonstrated in complex biological matrices. As a major cause of seasonal influenza, H1N1 virus requires advanced monitoring. Custom-patterned conductive graphene microelectrodes are essential for building relevant sensing platforms. In this study, a label-free H1N1 electrochemical immunosensor was developed based on a sequentially engineered laser-induced graphene (LIG)/Prussian blue (PB)/poly3,4-ethylenedioxythiophene (PEDOT)/Au nanocomposite microelectrode. Unlike previous studies that focused on single-component modifications, our approach integrates multiple functional layers to synergistically enhance signal stability, sensitivity, and antifouling performance. The LIG microelectrode was fabricated using a low-cost intelligent semiconductor laser, which directly patterned a three-electrode system on a polyimide (PI) film. Subsequently, PB, PEDOT, and gold nanoparticles (Au NPs) were sequentially electrodeposited onto the LIG electrode. PB served as an ultrasensitive electrochemical signal reporter, while the encapsulating PEDOT polymer layer not only stabilized the electrical signal of PB in neutral environments but also significantly enhanced the electrode's antifouling properties. To construct the label-free immunosensor, Au NPs were introduced to improve conductivity and were functionalized via Au–S–COOH bonding for the immobilization of H1N1 antibodies. Under optimal conditions, the LIG/PB/PEDOT/Au composite electrode exhibited excellent electrochemical activity and conductivity. The fabricated immunosensor demonstrated ultrahigh sensitivity with a wide concentration range from 0.1 fg mL −1 to 100 ng mL −1 , and an extremely low detection limit of 0.063 fg mL −1 . The results confirmed that the immunosensor possesses excellent selectivity, precision, and stability, and is thus suitable for the analysis of complex biological samples without the need for labeled probes and sample pretreatment. Therefore, this environmentally friendly and label-free immunosensing platform represents a significant advancement in point-of-care biosensing and demonstrates strong potential for rapid on-site screening and prevention of infectious disease outbreaks. A label-free immunosensor based on laser-induced graphene microelectrode modified with PB/PEDOT/Au nanocomposite enables ultra-sensitive detection of H1N1 virus in complex biological samples without pretreatment. A Versatile catalytic and photothermal lateral flow immunoassay Based on ultrathin Fe-MoS2 nanosheets for sensitive and accurate detection of Influenza A

Meimei Xu,Chenglong Lin,Shuai Zhao,Weida Zhang,Dan Li,Fanghao Fang,Zheng Teng,Yusi Peng,Ming Liu,Zhengren Huang,Jianlin Shi,Yong Yang

Influenza A virus (H1N1) poses a significant threat to global human health that imperative demands the development of sensitive and accurate point-of-care testing (POCT) methods. Here, for the first time, Fe-MoS 2 nanosheets were employed as a multifunctional nanotag in the development of catalytic colorimetric-photothermal dual-mode lateral flow immunoassay (dLFIA) strips for the sensitive detection of H1N1 inactivated virus. The Fe-MoS 2 nanosheets featuring large size, high specific surface area, and ultrathin structure could flow smoothly on the strips and thus quickly produce an ideal colorimetric signal for qualitative analysis. Both the limit of detection (LOD) of catalytic colorimetric and photothermal signals reached 1000 copies/mL and the corresponding calculated LOD was 550 and 691 copies/mL, respectively, which were about 50–90-fold more sensitive than traditional gold nanoparticles based-LFIA (5 × 10 4 copies/mL). The developed assay could correctly identify eight positive clinical samples with Ct values less than 35 and 10 negative actual samples, proving significant promise for rapid, sensitive, and accurate detection of H1N1, especially in resource-limited areas.

Nicotinic acid from Bubali Cornu alleviates H1N1-induced acute lung injury via dual antiviral and anti-inflammatory actions: A bioactivity-driven discovery

Guoqi Yuan,Yang Liu,Zhou Hong,Haiyan Zhu,Yan Lu,Daofeng Chen

Ethnopharmacological relevance Bubali Cornu (BC) has a long history of use in traditional Chinese medicine for the treatment of various conditions, including high fever, hemoptysis, epistaxis, and infections. Despite its historical use, the active components responsible for its therapeutic effects, particularly in viral pneumonia, remain poorly understood. Aim of the study The objective of this study was to identify the active compounds in BC with antiviral activity against H1N1 and to evaluate their therapeutic potential in alleviating inflammation in H1N1-infected acute lung injury (ALI) mice. Materials and methods The antiviral components of BC were isolated using silica gel column chromatography and high-performance liquid chromatography (HPLC) according to a bioactive guided separation strategy. Then, the protective effects of these components in H1N1-induced ALI mice were evaluated. To explore the potential mechanisms, network pharmacology (NP) analysis and qPCR were used to screen the related signaling pathways or proteins. Finally, the functions of the most effective compound were verified in H1N1-infected RAW264.7 macrophages, focusing on its regulatory effects on key proteins. Results The ethyl acetate fraction (EAF) of BC had a protective effect against ALI induced by H1N1 in mice. Subsequently, 13 compounds were isolated from EAF of BC, of which nicotinic acid (NA) showed the best antiviral activity in vitro . In vivo studies showed that NA treatment of ALI mice infected with H1N1 significantly reduced lung index and inflammation, and improved lung tissue morphology. Flow cytometry analysis revealed that NA reduced the over-recruitment of macrophages and neutrophils to alleviate the inflammatory storm. As verified by NP and qPCR, NA reduced the mRNA levels of NOS2 and NOX1 in the lungs of mice with viral pneumonia, but Western blot analysis only showed that NA inhibited the expression of NOS2. Finally, NA significantly inhibited the expression of NOS2 in RAW264.7 cells infected with H1N1, suggesting that NA might alleviate ALI induced by H1N1 by inhibiting NOS2 expression in pulmonary macrophages. Conclusion The present study demonstrated that NA isolated from BC exhibited significant antiviral and anti-inflammatory properties in H1N1-induced ALI. NA might alleviate the progression of H1N1 pneumonia by inhibiting virus replication, reducing the over-recruitment of macrophages and neutrophils, and inhibiting the expression of NOS2. These findings provided valuable insights into the potential of BC in the treatment of viral infections, and highlighted the importance of exploring traditional animal-derived TCM in modern antiviral applications.

Artemisia Extracts Suppress H1N1 Influenza A Virus Infection by Targeting Viral HA/NA Proteins and Modulating the TLR4/MyD88/NF-κB Signaling Axis

Zhongnan Hu,Hui Liu,Weihua Wu,Tayyab Ali,Adam Junka,Farukh S. Sharopov,Xuan Zou,Shisong Fang,Yanfang Sun

Background: Influenza A virus is an acute respiratory virus that spreads quickly, affects a broad range of populations, and can lead to many complications and mortality.ArtemisiaL. species are widely used in traditional medicine, but their antiviral potential against H1N1 remains uncertain.Methodology: Network pharmacology and molecular docking were used to computationally explore their potential function in this domain, and to investigate how their invasion mechanisms and adsorption occur. UPLC-MS/MS analysis identified the main components of the extracts. The anti-H1N1 mechanism ofArtemisiaL. extracts was studied in vitro. Results: Network pharmacology identified 95 key targets betweenArtemisiaL. and IAV, with quercetin and luteolin as core active compounds. Molecular docking predicted strong binding affinities between these compounds and influenza virus proteins. UPLC-MS/MS analysis identified 75, 100, and 64 chemical components inACBE,AALE, andACTE, respectively, mainly flavonoids and terpenoids.ArtemisiaL. extracts exhibited both preventive and therapeutic effects against H1N1, reducing progeny virus NP mRNA and protein levels. In vitro experiments showed that higher concentrations of the extracts prevent virus attachment to MDCK cells by denaturing the HA protein. NA plays an essential role in progeny virus release. We found that a high concentration of ACTE can inhibit NA up to 85%, and ACBE showed a low inhibitory effect on NA.Conclusions: In terms of therapeutic effects,ArtemisiaL. extracts can regulate intracellular inflammatory factors via the TLR4/NF-κB/MyD88 signaling pathways and reduce the expression of IL-1β, IL-6, TNF-α, TLR4, NF-κB, p65, and MyD88 at the mRNA level, thereby inhibiting H1N1 virus replication. These results suggest that bioactive components inArtemisiaL. extracts may inhibit H1N1, potentially leading to the development of natural-product-based anti-influenza agents.

A non-PCR SPR platform using RNase H to detect MicroRNA 29a-3p from throat swabs of human subjects with influenza A virus H1N1 infection

Jacky F. C. Loo,S. S. Wang,F. Peng,J. A. He,L. He,Y. C. Guo,D. Y. Gu,H. C. Kwok,S. Y. Wu,H. P. Ho,W. D. Xie,Y. H. Shao,S. K. Kong

As in all RNA viruses, influenza viruses change and mutate constantly because their RNA polymerase has no proofreading ability. This poses a serious threat to public health nowadays. In addition, traditional pathogen-based detection methods may not be able to report an infection from an unknown type or a subtype of virus if its nucleotide sequence is not known. Because of these factors, targeting host microRNA signatures may be an alternative to classify infections and distinguish types of pathogens as microRNAs are produced in humans shortly after infection. Although this approach is in its infant stage, there is an urgent need to develop a rapid reporter assay for microRNA for disease control and prevention. As a proof of concept, we report herein for the first time a non-PCR MARS (MicroRNA-RNase-SPR) assay to detect the microRNA miR-29a-3p from human subjects infected with influenza virus H1N1 by surface plasmon resonance (SPR). In our MARS assay, RNase H is employed to specifically hydrolyze the RNA probes immobilized on the gold surface where they hybridize with its cognate target cDNAs miR-29a-3p, where it was formed from reverse transcription with mature miR-29a-3p specific stem-looped primers. After the digestion of the RNA probe by RNase H, the intact cDNA was released from the RNA–DNA hybrid and bound to a new RNA probe for another enzymatic reaction cycle to amplify signals. With assay optimization, the detection limit of our MARS assay for miR-29a-3p was found to be 1 nM, and this new assay could be completed within 1 hour without thermal cycling. This non-PCR assay with high selectivity for mature microRNA provides a new platform for rapid disease diagnosis, quarantine and disease control.

Bidirectionally Regulating Viral and Cellular Ferroptosis with Metastable Iron Sulfide Against Influenza Virus

Xinyu Miao,Yinyan Yin,Yulian Chen,Wenhui Bi,Yuncong Yin,Sujuan Chen,Daxin Peng,Lizeng Gao,Tao Qin,Xiufan Liu

Influenza virus with numerous subtypes and frequent variation limits the development of high-efficacy and broad-spectrum antiviral strategy. Here, a novel multi-antiviral metastable iron sulfides (mFeS) against various influenza A/B subtype viruses is developed. This work finds that mFeS induces high levels of lipid peroxidation and •OH free radicals in the conservative viral envelope, which depends on Fe 2+ . This phenomenon, termed as a viral ferroptosis, results in the loss of viral infectibility and pathogenicity in vitro and in vivo, respectively. Furthermore, the decoction of mFeS (Dc(mFeS)) inhibits cellular ferroptosis-dependent intracellular viral replication by correcting the virus-induced reprogrammed sulfur metabolism, a conserved cellular metabolism. Notably, personal protective equipment (PPE) that is loaded with mFeS provides good antiviral protection. Aerosol administration of mFeS combined with the decoction (mFeS&Dc) has a potential therapeutic effect against H1N1 lethal infection in mice. Collectively, mFeS represents an antiviral alternative with broad-spectrum activity against intracellular and extracellular influenza virus.

Copper based metal–organic frameworks triggered chemically driven redox-cycling system for ultrasensitive magnetic separated-type fluorescent DNA assay

Sun Lu,Xu Qian-Yue,Wang Hai-Bo

Being enlightened by Fenton-like reaction, a novel chemically driven redox-cycling system has been developed without the participation of additional reducing agents. In this chemically driven cycling system, terephthalic acid (TA) was selected as fluorescence substrate to trap the generated hydroxyl radicals (·OH) to produce 2-hydroxyterephthalic acid (TAOH), which possessed high fluorescence intensity. On one hand, Cu + triggered a Fenton-like reaction with H 2 O 2 to yield Cu 2+ and ·OH. But on the other hand, the generated Cu 2+ could react with H 2 O 2 to produce Cu + and O 2 ·− (further converted into ·OH). The regenerated Cu + could take turns reacting with H 2 O 2 to produce more ·OH and form more TAOH. As a result, a kind of chemical redox-cycling signal amplification reaction was touched off. On the basis of copper based metal–organic frameworks (Cu-MOFs) and signal amplified strategy, a magnetic separated-mode fluorescent biosensor was designed for the sensitive determination of DNA. The method allowed ultrasensitive detection of target DNA H1N1 with a linear range from 5 pM to 10 nM, and with a limit of detection of 2 pM. This magnetic separated-type system could effectively minimize the background signals by reducing the mutual interferences between nucleic acids hybridization and fluorescence signal conversion.
