Developing COX-2-targeted fluorescent probes for HClO/ONOO⁻ detection in cancer cells: an integrated experimental and computational study

Zhang Yonglin,Liu Jianxi,Song Shengfa,Hu Jiale,Chen Shulin,Zhu Lulu,Shen Li,Jiang Hao,Xia Yunqing,Yi Xuexue,Yue Ningning,Xue Ying,Liu Wanhui,Zhu Yanping,Xu Lixiao

Overexpression of cyclooxygenase-2 (COX-2) and associated reactive species, specifically hypochlorous acid (HClO) and peroxynitrite (ONOO⁻), in the tumor microenvironment are critical hallmarks for early cancer diagnosis. In this work, we designed and synthesized two fluorescent probes,  S1  and  S2 , for the targeted imaging of HClO and ONOO⁻, respectively. Both probes integrate a celecoxib ligand for COX-2 targeting with a naphthalimide fluorophore. The probes operate via a photoinduced electron transfer (PET) mechanism, producing a distinct fluorescence turn-on response with excellent linearity against analyte concentration ( R 2  = 0.9902 for  S1  with HClO;  R 2  = 0.9897 for  S2  with ONOO⁻). This mechanism was theoretically validated through DFT calculations. In vitro cellular imaging demonstrated that both probes exhibit low cytotoxicity and achieve superior tumor cell selectivity, showing significantly higher fluorescence intensity in COX-2 overexpressing MCF-7 cancer cells than in normal HUVEC cells. The molecular basis for targeting was rigorously investigated using molecular docking and 100 ns molecular dynamics simulations. MM/PBSA calculations revealed favorable binding enthalpies for both  S1  (−80.05 ± 7.19 kcal/mol) and  S2  (−83.76 ± 3.88 kcal/mol) with COX-2, confirming stable and specific complex formation. This integrated experimental and computational strategy yields highly sensitive and selective probes, positioning  S1  and  S2  as promising tools for studying cancer-related biomarkers at the cellular level. A novel lysosome-targeting fluorescent probe for intracellular HClO imaging and aqueous environment detection based on smartphone-assisted quantification

Jiayi Zhang,Qian Wang,Yashuang Ren,Honglu Xiang,Lamei Cao,Yu Li,Dan Li,Feiran Gao,Songyang Xiao,Yuping Tang,Weifang Luo

Hypochlorous acid (HClO), a reactive oxygen species (ROS), regulates multiple physiological and pathological processes in humans. However, its excessive accumulation induces diseases such as inflammatory bowel disease, rheumatoid arthritis, cardiovascular diseases, and neurodegenerative diseases. Besides, HClO is employed for public disinfection in infectious disease outbreaks to block pathogen transmission, but its over-discharge causes water pollution, endangering aquatic organisms and triggering severe human respiratory and skin diseases. To clarify HClO's complex roles in environmental and biological systems, developing accurate detection methods is imperative. This study synthesized a novel naphthalene-derived fluorescent probe Z via acylation and substitution reactions, using 4-bromo-1,8-naphthalimide as the fluorophore, morpholine as the lysosome-targeting group, and dimethylthiocarbamoyl chloride as the recognition group. Probe Z exhibits prominent visualization advantages: upon HClO addition, it turns from pale yellow to fluorescent yellow under 365 nm UV light within 1 min, with a 13-fold fluorescence enhancement, high selectivity, high sensitivity, and a low detection limit of 0.52 nM. It can detect HClO in water samples and achieve fluorescent imaging of lysosomal HClO in cells. Notably, combined with the data processing function of smartphones, it enables reliable quantification of HClO, providing a new tool for exploring HClO's biological roles and on-site high-throughput visual detection.

Kinetics study of chloride-activated peracetic acid for purifying bisphenol A: Role of Cl2/HClO and carbon-centered radicals

Zhao Song,Yu Zhang,Xue Zhang,Xu Zhou,Yidi Chen,Xiaoguang Duan,Nanqi Ren

Peracetic acid is an emerging oxidant and disinfectant for wastewater purification. In this study, we first developed a comprehensive and accurate model to elucidate the reaction mechanisms and simulate reaction kinetics of peracetic acid (PAA, CH 3 C(=O)OOH) activated by chloride (Cl – ) based on experimental results and literature. A diversity of experiments methods (e.g., quenching experiments, probe compounds degradation, electron paramagnetic resonance (EPR) measurements) and kinetic modeling were used to determine the reactive species. As a result, carbon-centered radicals and free chlorine reactive species (Cl 2 and HClO) were devoted to BPA degradation in the PAA/Cl − system. The carbon-centered radicals CH 3 C(=O)OO • , CH 3 C(=O)O • , CH 3 OO • , and • CH 3 greatly accelerated BPA degradation with their corresponding kinetics of k CH 3 C(=O)OO•, BPA  = 2 × 10 8 M − 1 s −1 , k CH 3C(= O)O• , BPA  = 2 × 10 7 M − 1 s −1 , k •CH 3 , BPA  = 2 × 10 6 M − 1 s −1 and k CH 3 OO•, BPA  = 2 × 10 4 M − 1 s −1 . Dissolved Cl 2 (l) species was also important for BPA degradation with k Cl 2 , BPA of 2 × 10 7 M − 1 s −1 , much higher than HClO/ClO − of k HClO, BPA  = 1.2 × 10 1 M − 1 s −1 and k ClO − , BPA  = 9 × 10 −3 M − 1 s −1 . While free chlorine tends to transform BPA to estrogenic chlorinated organic products, the primary degradation of BPA by carbon-centered radicals results in chlorine-free products, reducing the production of disinfection byproducts during the treatment of saline wastewater. This study improves the knowledge of reaction kinetics and mechanism and reactive species generation in the PAA/Cl – system.

Design, synthesis and application of dual-channel fluorescent probes for ratiometric detection of HClO and H2S based on phenothiazine coumarins

Zixuan Teng,Huimin Shangguan,Longfei Liu,Shihu Zhang,Guanlin Li,Zishi Cheng,Fengpei Qi,Xingjiang Liu

The ubiquity of diverse material entities in environmental matrices renders the deployment of unifunctional fluorescent indicators inadequate. Consequently, this study introduces a ratiometric dual-emission fluorescent sensor ( Probe CP ), synthesized by conjugating phenothiazine coumarin to hydroxycoumarin through a piperazine linker for concurrent detection of HClO and H 2 S. Upon interaction with HClO, the phenothiazine unit's sulfur atom undergoes oxidation to sulfoxide, facilitating a shift from red to green fluorescence in a ratiometric manner. Concurrently, at the opposite terminus of Probe CP , 2,4-dinitroanisole serves as the reactive moiety for H 2 S recognition; it restores the blue emission characteristic of 7-hydroxycoumarin while maintaining the red fluorescence emanating from phenothiazine coumarin as an internal standard for ratio-based assessment. Exhibiting elevated specificity and sensitivity coupled with minimal detection thresholds (0.0506 μM for HClO and 1.7292 μM for H 2 S) alongside rapid equilibration periods (3 min for HClO and half an hour for H 2 S), this sensor was efficaciously employed in cellular environments and within zebrafish models as well as imaging applications pertaining to alcohol-induced hepatic injury in murine subjects.

Chitosan/silk fibroin nanofibers-based hierarchical sponges via a HClO self-producing cascade catalytic reaction accelerating infected diabetic wound healing

Suya Deng,Yinggui Huang,Enling Hu,Liang-Ju Ning,Ruiqi Xie,Kun Yu,Fei Lu,Guangqian Lan,Bitao Lu

The diabetic chronic wound healing is extremely restricted by issues such as hyperglycemia, excessive exudate and reactive oxygen species (ROS), and bacterial infection, causing significant disability and fatality rate. Herein, the chitosan/silk fibroin nanofibers-based hierarchical 3D sponge (CSSF-P/AuGCs) with effective exudate transfer and wound microenvironment modulation are produced by integrating cascade reactor (AuGC) into sponge substrates with parallel-arranged microchannels. When applied to diabetic wounds, the uniformly parallel-arranged microchannels endow CSSF-P/AuGCs with exceptional exudate absorption capacity, keeping the wound clean and moist; additionally, AuGCs efficiently depletes glucose in wounds to generate H 2 O 2 , which is then converted into HClO via cascade catalytic reaction to eliminate bacterial infection and reduce inflammation. Experiments in vitro demonstrated that the antibacterial activity of CSSF-P/AuGCs against S. aureus and E. coli was 94.27 and 92.7 %, respectively. Experiments on animals indicated that CSSF-P/AuGC could cure wounds in 11 days, displaying superior wound-healing abilities when compared to the commercial medication Tegaderm™. This versatile CSSF-P/AuGCs dressing may be an attractive choice for expediting diabetic wound healing with little cytotoxicity, providing a novel therapeutic method for establishing a favorable pathological microenvironment for tissue repair.

Mitochondria-Targeted Chemosensor to Discriminately and Continuously Visualize HClO and H2S with Multiresponse Fluorescence Signals for In Vitro and In Vivo Bioimaging

Xiaojun He,Ziman Zheng,Feifan Zhang,Chuchu Xu,Wei Xu,Lisong Ye,Xiaoshuai Sun,Zhan Zhou,Jianliang Shen

Bioactive molecules play a vital role in the process of regulating the redox balance in the intracellular environment, especially in maintaining the function of organelles. To explore the association and function of bioactive molecules in organelles, it is essential to develop a chemosensor tool that uses multiresponse fluorescence signals to distinguish between and track two related bioactive molecules in organelles. However, the development of sensors with multiresponse functions is still a challenging task. Herein, we present a unique and practical single chemosensor (Mito-CTC) that can monitor HClO (as an oxidative substance) and H2S (as a reductive substance) in mitochondria (organelle targeting) with multiresponse fluorescence signals. The response of the sensor to HClO and H2S changes from red to green and blue channel emission simultaneously, respectively, thereby providing a specific signal response to reductive/oxidative substances in the mitochondria. Using a single chemosensor, we have realized multichannel bioimaging of the exogenous and endogenous HClO and H2S in cellular mitochondria. Additionally, the excellent properties of the sensor Mito-CTC can be used to reveal the relationship between HClO and H2S in mitochondria. Meanwhile, Mito-CTC has been endowed with the ability to image in bacteria and zebrafish attributed to the good permeability and low cytotoxicity. Expectantly, drug-induced liver injury (DILI) caused by fluoxetine (an antidepressant drug) and the degree of drug-induced toxicity to the liver were evaluated using Mito-CTC through discriminating and imaging HClO, indicating that Mito-CTC has the potential function of evaluating the toxicity of the drug to the liver.

Evaluating Drug-Induced Liver Injury and Its Remission via Discrimination and Imaging of HClO and H2S with a Two-Photon Fluorescent Probe

Xiaoyun Jiao,Yongsheng Xiao,Yong Li,Muwen Liang,Xilei Xie,Xu Wang,Bo Tang

Drug-induced liver injury (DILI) has aroused wide concern. Finding new markers or indicators as well as detoxification molecules for DILI is of great significance and good application prospect, which can help develop effective preclinical screening methodology and corresponding treatment protocols. Herein, in this article, DILI caused by antidepressant drugs of duloxetine and fluoxetine and its remission were evaluated by a two-photon fluorescent probe, RPC-1, through discriminating and imaging HClO and H2S simultaneously. By being applied both in vitro and in vivo, RPC-1 revealed slight up-regulation of HClO and negligible liver damage after administration of either of the two drugs. In contrast, an apparent up-regulation of HClO and obvious liver damage was observed after combined administration of the drugs. Meanwhile, the pretreatment of N-acetyl cysteine (NAC) resulted in the increasing of endogenous H2S level, which contributed to the remission of DILI. The histological analysis and serological test both gave good consistency with the imaging results. These findings demonstrate that HClO may be an appropriate indicator of DILI, and H2S plays an important role in the antidotal effect of NAC. We envision that RPC-1 can be used as a powerful tool to predict clinical DILI and study the effect of antidote, as well as explore the molecular mechanisms involved.

Dual-channel ratiometric sensing of hypochlorous acid based on organic cage with red and green fluorescence

Ying Xu,Cong Dai,Zhifeng Xu

The high-performance detection of hypochlorous acid (HClO) is crucial for life and environmental health, but still challenging for the single fluorescence signal and single sensing channel of probes. Organic cage with strong adsorption ability and cage structure-related fluorescence property, is a type of potential fluorescent probe. However, the single signal and short emission of organic cage hinders its sensing performance. The design of dual emitting organic cage with long wavelength for dual-channel ratiometric sensing of HClO is of great significance. Herein, we reported an organic cage (ROX) as a probe for accurate quantification of HClO. The prepared ROX exhibited capsule topology-facilitated red/green dual emission around 660/502 nm. The cage-like structure was disrupted by the rapid specific redox interaction between imine bond in ROX and HClO. As a result, the fluorescence at 660 nm quenched in conjunction with the enhancement at 502 nm in PBS (pH 4). Besides, the fluorescence increased at 582 nm with constant emission at 480 nm in DMSO/PBS (pH 7) ( V / V , 9/1). The ratio signal in PBS and DMSO/PBS respectively gave the linearity range of 1–30 and 20–45 μM, the limit of detection (3  s ) of 14 and 49 nM, and the relative standard deviation of 1.04 % and 0.54 % for 10 replicate determinations. Based on the mutual validation of dual ratio, the dual-channel ratiometric sensing possesses good accuracy, sensitivity, selectivity, and precision. Moreover, ROX can be applied to HClO monitoring in actual water and disinfectant samples with the recoveries of spiked HClO ranged from 95.0 % to 104.2 %. The proposed sensing method highlights the great potential of organic cage in fluorescence sensing.

A ratiometric and rapid-response fluorescent nanoprobe based on modification of carbon quantum dots by naphthalimide for HClO bioimaging in living cells and mice

Kedian Xu,Ruiqing Liang,Ziqing Ye,Yunling Gao

A ratiometric and rapid-response fluorescent nanoprobe (NA-CQDs) for the detection of hypochlorous acid based on naphthalimide (NA) modified carbon quantum dots (CQDs) has been prepared. The nanoprobe NA-CQDs showed blue fluorescence with a unique yellow fluorescence in the presence of HClO, accompanying the emission wavelength change from 457 nm to 538 nm. The fluorescence detection limit for HClO was 99 nM with a linear range from 0.1 to 32 μM. The nanoprobe exhibited a rapid response to HClO within 20 s. The sensing mechanism might be attributed to the oxidation and cleavage of p -aminophenylether group on the nanoprobe NA-CQDs, leading to the FRET process from the CQDs to the NA due to enhanced ICT of the NA portion. A series of real applications were conducted including spiked real water samples, test paper strips, and fluorescence bioimaging in RAW 264.7 and HepG2 cells and nude mice, highlighting the potential application of NA-CQDs for future environmental sensing and biosensing in living things.

Efficiently removing dissolved organic pollutants in landfill leachate concentrate using dual-anode Fe2+/HClO system: The significance of insolubilization based on oxidative coupling of organics

Sheng Liang,Jinling Wang,Zhizhi Shen,Wei Yan,Lichao Nengzi,Chunhua Feng,Xin Lei,Li Yu,Junqing Hu

The strategy to mineralize dissolved organics for the decontamination of landfill leachate concentrate (LLC) is expensive. Because mineralization requires high stoichiometric ratio of oxidants (e.g., •OH) or electron transfer (i.e., energy) to the pollutant molecules. Whereas, transferring the dissolved organic pollutants into separable insoluble matters is an energy-saving strategy. Herein, we developed a dual-anode Fe 2+ /HClO system for removing dissolved organic pollutants in LLC through insolubilization based on oxidative coupling. In this system, the majority of dissolved organic pollutants were transferred into insoluble substances, which could be removed easily. As a result, 89.8 % of the dissolved organic carbon (DOC) in LLC was fast removed in 3 h with a low specific energy consumption (SEC, 4.0 kWh kg −1 COD and 17.6 kWh kg −1 DOC), and the formation of toxic by-products can be well controlled to total organic chlorine (TOCl) < 0.1 mg/L. The formation of insoluble substances was attributed to the oxidative coupling of organic small molecules into the macromolecular ones. This was evidenced by the generation of insoluble compounds with the larger molecular mass, and the formation of coupling or polymerization products of the model pollutant (guaiacol). The predominant mechanism was proposed that Fe(IV) was generated in the dual-anode Fe 2+ /HClO system, and Fe(IV) oxidized pollutants to produce organic radicals, then the organic radicals coupled and polymerized to form larger molecules and insoluble compounds. This work offers a novel electrochemical method for efficient removal of organic pollutants in LLC with minimal toxic by-products generated, and revealed the significance of oxidative coupling process in the electrochemical Fe 2+ /HClO system.
