Occult blood testing is a routine screening method for evaluating the function of the urinary system and digestive tract. The hemoglobin (Hb) content in human excreta is an important indicator of the presence of occult blood. In this study, a surface plasmon resonance imaging (SPRi)-based immunosensor with high sensitivity was developed for the detection of Hb. The immunosensor was constructed by physically immobilizing anti-Hb antibodies on the detection channels. Samples for evaluation were pre-incubated with the polymeric microsphere-antibody conjugate. The shift in resonance wavelength induced by the specific binding of microsphere-antibody-antigen and immobilized antibodies was monitored in real time. Owing to the amplification effect of microspheres, the detection limit of the immunosensor was reduced 42-fold to 4.8 ng/mL under the optimized concentration of immobilized antibody (10 μg/mL) with microsphere-antibody conjugate dilution ratio of 1:200 and incubation time of the microsphere-antibody conjugate and samples (10 min). The immunosensor displayed a good linear response to Hb ranging from 10 ng/mL to 500 ng/mL in both test solutions and in 1:30 urine sample, as well as a good specificity toward Hb and an acceptable recovery rate for spiked Hb solution in urine sample. The immunosensor was also exploited to detect the Hb concentration in 50 clinical urine samples. Therefore, the proposed SPRi immunosensor developed in this study provides a simple, rapid, high-throughput, and sensitive immunology strategy for the detection of occult blood.

The detection of β-hydroxybutyrate (β-HB) is crucial for the rapid diagnosis of diabetic ketoacidosis and health management. However, daily monitoring of dynamically changing β-HB concentrations is challenging and still limited to laboratory settings or painful and invasive measurements. Herein, we report a wearable electrochemical sensor integrated with iontophoresis for non-invasive monitoring of sweat β-HB. This device can actively stimulate the skin to induce sweat by iontophoresis, and the sweat is controllably collected and transported to the sensing area through the microfluidic channel. The sweat β-HB was measured through an electrochemical oxidation reaction mediated by the generated nicotinamide adenine dinucleotide, which can achieve fast response analysis of sweat β-HB within 40 seconds and a low detection limit of 10 μM. We also used this sensor to monitor the sweat β-HB levels in a volunteer experiencing the ketogenic diet and the sweat and blood β-HB levels exhibited high correlation. We believe this wearable sensor capable of on-demand and noninvasive detection of sweat β-HB shows a wide application prospect in clinical medicine and home healthcare.

Effective molecularly imprinted membranes(MIMs) were developed as an efficient adsorbent for the selective removal of p -hydroxybenzoic acid( p -HB) from acetylsalicylic acid(ASA, aspirin). The MIMs were grafted successfully from poly(vinylidene fluoride) microfiltration membranes via reversible addition-fragmentation chain transfer(RAFT) polymerization. The graft copolymerization of acrylic acid(AA) in the presence of template p -hydroxybenzoic acid led to molecularly imprinted polymer(MIP) film coated membranes. The obtained MIMs were characterized by scanning electron microscopy(SEM), Fourier transform infrared spectrophotometer(FTIR) and Raman spectra, and batch mode adsorption studies were carried out to investigate the specific adsorption equilibrium, kinetics and selective recognition properties of different MIMs. The kinetic properties of the MIMs could be well described by the pseudo-second-order rate equation. Selective permeation experiments were performed to evaluate the permeation selectivity of the p -HB imprinted membranes. The observed performances of the MIMs are applicable to the further purification of aspirin.

Starvation therapy kills tumor cells via consuming glucose to cut off their energy supply. However, since glucose oxidase (GOx)-mediated glycolysis is oxygen-dependent, the cascade reaction based on GOx faces the challenge of a hypoxic tumor microenvironment. By decomposition of glycolysis production of H 2 O 2 into O 2 , starvation therapy can be enhanced, but chemodynamic therapy is limited. Here, a close-loop strategy for on demand H 2 O 2 and O 2 delivery, release, and recycling is proposed. The nanoreactor (metal-protein-polyphenol capsule) is designed by incorporating two native proteins, GOx and hemoglobin (Hb), in polyphenol networks with zeolitic imidazolate framework as sacrificial templates. Glycolysis occurs in the presence of GOx with O 2 consumption and the produced H 2 O 2 reacts with Hb to produce highly cytotoxic hydroxyl radicals (•OH) and methemoglobin (MHb) (Fenton reaction). Benefiting from the different oxygen carrying capacities of Hb and MHb, oxygen on Hb is rapidly released to supplement its consumption during glycolysis. Glycolysis and Fenton reactions are mutually reinforced by oxygen supply, consuming more glucose and producing more hydroxyl radicals and ultimately enhancing both starvation therapy and chemodynamic therapy. This cascade nanoreactor exhibits high efficiency for tumor suppression and provides an effective strategy for oxygen-mediated synergistic starvation therapy and chemodynamic therapy.

Sonodynamic therapy (SDT) has attracted great interest in the field of cancer therapy because of its non-invasiveness, deep penetration, and spatiotemporal controllability. However, the sonodynamic effect is severely hindered by hypoxia and a high glutathione (GSH) level in tumor microenvironment. In this work, a new type of nanohybrid sonosensitizer is designed, which incorporates several prominent advantages of organic, inorganic, and natural sonosensitizers for imaging-guided SDT and ferroptosis induction. As an endogenous transporter, hemoglobin (Hb) has dual functions of oxygen and iron supplement. Apparently, Hb can transport iron to the tumor site for Fe-dependent ferroptosis, yet this oxygen carrier is able to enhance the sensitivity of tumor cells to oxygen-driven SDT. We innovatively hybridized Hb, sinoporphyrin sodium (DVDMS), and titanium dioxide (TiO 2 ) to prepare an integrated nano-oxygen carrier (designated as HDT) for multifarious cancer theranostics, which not only attained satisfactory sonosensitization of DVDMS/TiO 2 but also achieved oxygen-boosted SDT and potent ferroptosis via Hb. Under ultrasound irradiation, the oxygenation of HDT markedly amplified the generation of reactive oxygen species (ROS) in hypoxic tumors, promoted the valence transition of Hb to accelerate Fenton reaction, and led to ferroptosis. This strategy virtually eradicated tumors in situ and exhibited immunoregulatory potential for augmenting anti-tumor effects in vivo and in vitro through ROS formation, O 2 self-supplement, GSH depletion, and lipid peroxidation. Collectively, HDT demonstrated excellent dual-modal imaging performance, providing a valuable platform for photoacoustic/fluorescence-guided SDT and ferroptosis induction in tumor cells.

Chemotherapy plays a crucial role in triple-negative breast cancer (TNBC) treatment as it not only directly kills cancer cells but also induces immunogenic cell death. However, the chemotherapeutic efficacy was strongly restricted by the acidic and hypoxic tumor environment. Herein, we have successfully formulated PLGA-based nanoparticles concurrently loaded with doxorubicin (DOX), hemoglobin (Hb) and CaCO 3 by a CaCO 3 -assisted emulsion method, aiming at the effective treatment of TNBC. We found that the obtained nanomedicine (DHCaNPs) exhibited effective drug encapsulation and pH-responsive drug release behavior. Moreover, DHCaNPs demonstrated robust capabilities in neutralizing protons and oxygen transport. Consequently, DHCaNPs could not only serve as oxygen nanoshuttles to attenuate tumor hypoxia but also neutralize the acidic tumor microenvironment (TME) by depleting lactic acid, thereby effectively overcoming the resistance to chemotherapy. Furthermore, DHCaNPs demonstrated a notable ability to enhance antitumor immune responses by increasing the frequency of tumor-infiltrating effector lymphocytes and reducing the frequency of various immune-suppressive cells, therefore exhibiting a superior efficacy in suppressing tumor growth and metastasis when combined with anti-PD-L1 (αPD-L1) immunotherapy. In summary, this study highlights that DHCaNPs could effectively attenuate the acidic and hypoxic TME, offering a promising strategy to figure out an enhanced chemo-immunotherapy to benefit TNBC patients.
