Highly Sensitive Electrochemical Detection of ERα Based on AuNPs-SWCNT Composites and Hemin@HKUST-1

Mengli Wang,Lele Ma,Le Kang,Mingsan Miao,Huaixia Yang

Estrogen receptor alpha (ERα) is an important biomarker for breast cancer which is positive in over 75% of breast cancer cases. In this study, an electrochemical biosensor was constructed for sensitive detection of ERα based on the signal amplification of AuNPs-SWCNT nanocomposites and hemin@HKUST-1. AuNPs-SWCNT was served as a conductive substrate, and be used to immobilize the aptamer (Apt) by an Au–S bonding. Then, Apt, ERα and antibody (Ab) formed a sandwich composite structure through specific recognition and immobilized on the electrode. The hemin@HKUST-1 was introduced into the biosensor via an amide bond with Ab and acted as the signal molecule. AuNPs-SWCNT could significantly enhance the conductivity of the electrode and the large specific surface area of HKUST-1 enabled it to load more hemin, and greatly improved the sensitivity of the biosensor. The detection limit (LOD) of the biosensor could reach 27.8 fg·ml−1 after optimization of the conditions, with the current intensity and the logarithm of the ERα concentration exhibited a good linear relationship between 100 fg·ml−1 and 100 ng·ml−1. In addition, the biosensor possessed excellent selectivity, stability, reproducibility and Interference immunity. More importantly, it shows great application potential in practical detection.

Z/Ce@hemin enzymes with enhanced peroxidase activity for monitoring and screening the oxidative stress models of Parkinson’s disease

Lei Li,Chen Xiuli,Long Xincheng,Tu Qiuxia,Zhao Liping,Huang Dan,Song Lingli,Wang Jiaojiao,Zhai Suzhen,Chen Xiaozhong,Zhang Chunlin

Inspired by natural enzymes, artificial enzymes have garnered significant interest due to their simplicity of production, robustness under harsh conditions, and enhanced stability. This study introduces, for the first time, a novel Z/Ce@hemin composite enzyme, constructed by anchoring hemin onto zeolitic imidazolate framework-8 (ZIF-8)-encapsulated ceria (CeO 2 ) nanoparticles. This innovative design overcomes the challenges of hemin dimerization, enhances substrate affinity, and accelerates mass transport, leading to significantly improved peroxidase-like activity. The enzyme also demonstrates remarkable stability and resistance to environmental interference. Utilizing this Z/Ce@hemin composite, a sensitive and selective colorimetric detection system was developed for rapid and accurate quantification of hydrogen peroxide (H 2 O 2 ), a key oxidative stress marker. This approach was successfully applied in cellular models and Caenorhabditis elegans models of Parkinson’s disease, enabling precise monitoring of H 2 O 2 . The study provides a groundbreaking platform for oxidative stress modeling and expands possibilities in neurodegenerative disease research and therapeutic screening.

Dual Functions of Fluorescence and Peroxidase Mimics for Hemin@NH2-UiO-66 and Ratiometric Fluorescence Sensing to l-Cysteine

Jin Chai,Chunpeng Liu,Hongtian Yang,Yide Han,Yufeng Liu,Ying Zhang,Xia Zhang

The bifunctions of fluorescence activity and peroxidase mimics were well-integrated by a hemin chloride-modified Zr-based metal–organic framework (Hemin@NH2-UiO-66), which can catalyze the oxidation of o-phenylenediamine (OPD) to generate 2,3-diaminophenazine (DAP), presenting typical peroxidase mimic properties. The influence of the introduction of hemin chloride upon the enhanced peroxidase mimic activity was clarified, and the nanozyme catalytic parameters were optimized. Interestingly, the oxidized product of DAP presents strong fluorescence emission at 564 nm; combining it with the intrinsic fluorescence emission of NH2-UiO-66 at 452 nm, a dual-emission system could be built up by the resulting Hemin@NH2-UiO-66 in the presence of definite OPD and H2O2. Moreover, the fluorescence quenching effect (564 nm) was observed by adding l-cysteine (l-Cys); based on that, a straight-line dependence of the fluorescence intensity ratio (I452/I564) upon the concentrations of l-Cys was established. The detection limit was 0.21 μmol, and the analytical selectivity for l-Cys was also demonstrated. The work highlights the idea of combining the intrinsic fluorescence property and nanozyme catalytic activity in a functional MOF, and its special usability is found in the ratiometric fluorescence sensing analyses.

Amplified colorimetric sensor for detecting radon by its daughter lead based on the free-fixed auto-assembly structure of Duplex-hemin/G-quadruplex

Guiying Yang,Chunli Song,Qiwei Shi,Hongwen Liu,Shiya Li,Ran Liu,Shimeng Liu,Changyin Lv

A novel signal amplification strategy based on a Pb 2+ -dependent DNAzyme is proposed for detecting Pb 2+ and radon through Pb 2+ -induced DNAzyme cleavage and the auto-assembly of a duplex-hemin/G-quadruplex with two loop-stem structures and enzyme-mimicking catalytic activity. First, the Pb 2+ -specific DNAzyme cleaves a primer sequence, which induces assembly of the hairpin probe Hp1/Hp2 into a double helix structure. Subsequently, a G-quadruplex forms after the insertion of hemin into the free G-rich sequences; this G-quadruplex possesses the catalytic activity of horseradish peroxidase and changes colorless TMB to its deeply colored oxidized state. lead-dependent DNAzymes were constantly sheared by Pb 2+ , and the free primer strands were continuously assembled into double chains by hybridization with hairpin probes, providing amplification for the detection of lead (II) and radon. Under the optimum conditions, there was a good linear relationship between ΔA and the lead concentration for Pb 2+ concentrations ranging from 2.58 to 18 nM, and the detection limit was 0.77 nM. Analysis of actual samples indicated that when the radon concentration was in the range of 5.41 × 10 3– 1.65 × 10 5 Bq·h/m 3 , the radon concentration and the absorbance were linearly correlated with a detection limit of 1.62 × 10 3 Bq·h/m 3 . During the process of radon sampling and detection, radiation damage from the radioactive gas radon can be avoided. In this study, the primer dissociated by the DNAzyme was in a free state. Compared with the fixed-state primer chain, this system will be more convenient for the biological analysis of ultratrace metal ions. Therefore, this strategy has good application prospects for biosensors and is expected to become a novel platform for the amplification and detection of metal ion signals.

Label-free detection of microRNA by polymerization and isomerization cyclic amplification coupled with G/Hemin DNAzyme

Xinlan Zhu,Ziyan Zhang,Ruiyang Ma,Huachu Chen,Yi Zheng,Su Zeng,Zheyong Li,Sheng Cai

A label-free detection technique for microRNA (miRNA) was developed based on target replacement cycling, polymerization and isomerization cyclic amplification (PICA), and G/Hemin DNAzyme signal output. The designed hairpin probe undergoes cyclic alternation of polymerization and isomerization catalyzed by Bst DNA polymerase, enabling target replacement cycling and continuous self-extension of single-stranded DNA (ssDNA) strands. This process generates abundant G-quadruplex sequences that bind to Hemin, achieving label-free detection of miRNA through catalytic reactions. Its key advantages include a specific primer self-extension mechanism that avoids spurious amplification, one step isothermal design simplifies workflow and enhances system robustness, adapting to diverse detection scenarios, and label-free detection strategy that substantially reduces cost. Therefore, it is poised to become a reliable tool for clinical miRNA analysis, offering potential guidance for the early diagnosis, treatment, and prognosis of tumor biomarkers.

Glucose oxidase immobilization on Hemin@PCN-222 (Mn): Integrated biomimetic and bioenzyme activities in cascade catalytic process

Peide An,Defeng Hu,Yide Han,Hao Meng,Xia Zhang

On accounting of the structure advantages of MOFs in the enzyme mimics and enzyme immobilization, in this work, the Hemin@PCN-222 (Mn) is constructed through an in-situ hydrothermal route by covalently bonded the –COOH of Hemin with the Zr(IV) of PCN-222 (Mn). With the help of the synergistic effects, the resulting Hemin@PCN-222 (Mn) represents excellent peroxidase-like activities in comparison with the single Hemin and PCN-222 (Mn) with a special enzyme activity of 6.71 U mg −1 . The Hemin@PCN-222 (Mn) was then employed as a carrier to immobilize the glucose oxidase (GOx), the immobilization conditions were optimized. Based on that, the optimized GOx@hemin@PCN-222 (Mn) was applied in the following cascade reaction: the glucose oxidation in the presence of oxygen catalyzed by the immobilized GOx firstly, and then the produced H 2 O 2 worked as an oxidant to oxidize ABTS. The quantity of produced ABTS + was monitored to assess the catalytic efficiency, and the contrast experiments were conducted over the mixture of the natural GOx and HRP. The above cascade process could also be used to detect the glucose, and the detection limit is determined. The high efficiency and the stability of the resulting GOx@hemin@PCN-222 (Mn) highlights the prospect applications of the MOFs-based enzyme mimics in the bio-catalysis and biosensing.

Target-inspired Pb2+-dependent DNAzyme for ultrasensitive electrochemical sensor based on MoS2-AuPt nanocomposites and hemin/G-quadruplex DNAzyme as signal amplifier

Renyue Ji,Wuceng Niu,Shuai Chen,Wei Xu,Xingduo Ji,Lingyue Yuan,Hangyu Zhao,Minghao Geng,Jingfu Qiu,Chaorui Li

In this work, a novel Pb 2+ electrochemical DNAzyme sensor was developed for ultrasensitive detection of lead ions (Pb 2+ ) in water environment by coupling with the MoS 2 –AuPt nanomaterials and hemin/G-quadruplex DNAzyme, which acting as the electrocatalytic signal tag. Streptavidin (SA) modified tin dioxide-functionalized reduced graphene oxide (rGO-SnO 2 ) /gold nanoparticles (AuNPs) served as a sensor platform for enhancing conductivity and immobilizing more Pb 2+ -specific DNAzyme. In the presence of Pb 2+ , the Pb 2+ -dependent DNAzyme specifically reacted with Pb 2+ , cleaving the substrate strand (SS) into two free fragment and releasing the biotin-modified enzyme strand (Bio-ES) on the electrode. Connecting MoS 2 –AuPt nanocomposites labeled with G-rich DNA (G-DNA) strand and exposure of Bio-ES through the Helper DNA, as well as adding hemin to form a hemin/G-quadruplex, the biosensor achieved signal amplification . Chronoamperometry was used to record the current signal, which was primarily derived from the cocatalysis reduction of H 2 O 2 by MoS 2 –AuPt nanocomposites and the hemin/G-quadruplex. Under optimal conditions, the designed biosensor exhibited sensitive detection of Pb 2+ from 0.1 pg mL −1 to 1000 ng mL −1 , with a lower detection limit of 38 fg mL −1 (based on 3σ). This proposed biosensor is ultrasensitive and specific, representing a potential application for the detection of Pb 2+ in a water environment.

WS2/Bi/BiOBr Nanostructures for Photoelectrochemical Sensing of 5-Formyluracil-2′-deoxyuridine-5′-triphosphate through Hemin/G-Quadruplex Double Signal Amplification

Qian Wang,Huanshun Yin,Jia Ding,Yunlei Zhou,Shiyun Ai

5-Formyluracil (5fU) is considered an important epigenetic modification agent involved in gene regulation, cell differentiation, and development. 5fU exists in many cells and tissues but is highly similar to the structure of 5-formylcytosine (5fC) and is less abundant in genome. Therefore, the development of a rapid, sensitive, and highly selective method to detect 5fU is desired. In this study, a photoelectrochemical biosensor based on WS2/Bi/BiOBr nanocomposites, black TiO2 (b-TiO2), and hemin/G-quadruplex was described, which was found to be highly sensitive and selective for 5-formyluracil-2′-deoxyuridine-5′-triphosphate (5fdUTP) detection. WS2/Bi/BiOBr was first employed as a photosensitive material, and 3,4-diaminobenzoic acid (DABA) was adopted as a 5fdUTP recognition and capture reagent, where DABA was captured by polyethylene polyamine through covalent reaction to form a stable amid bond. Then, a benzimidazole bond was formed between DABA and −CHO of 5fdUTP, and 5fdUTP was selectively captured on the electrode. Using b-TiO2 as a cross-linker, a guanine-rich ssDNA sequence was further immobilized on the electrode surface by covalent reaction with the 5fdUTP phosphate group. With the assistance of K+ and hemin, the hemin/G-quadruplex structure was formed on the electrode. The hemin molecules in the structure of hemin/G-quadruplex obviously enhanced the PEC response of the biosensor for detecting 5fdUTP, with a linear range of 0.005–500 nM and a detection limit of 1.53 pM (S/N = 3). In addition, this method exhibits optimistic selectivity. The precision and accuracy of this method were evaluated by studying the content of 5fdUTP in genome DNA of crop tissues with standard addition recovery experiments.

A Non-Enzymatic Electrochemical Sensing Platform Based on Hemin@MOF Composites for Detecting Hydrogen Peroxide and DNA

Dan Cheng,Xuelian Xiao,Xi Li,Chen Wang,Yani Liang,Zhengsong Yu,Chenglong Jin,Nan Zhou,Ming Chen,Yulin Dong,Yawei Lin,Zhizhong Xie,Chaocan Zhang

In this work, Hemin@MOF was prepared by doping hemin into Cu-MOF-199 and applied in the electrochemical sensing of H2O2 and DNA. The Hemin@MOF modified electrode (Hemin@MOF/GCE) exhibited an excellent electrocatalytic activity toward H2O2 and the detection limit of H2O2 was as low as 0.07 μmol·L−1 with the range of 0.1 to 2200 μmol·L−1. Based on the eminent electrochemical performance, Hemin@MOF was further employed as an electrochemical signal indicator to construct a novel electrochemical DNA biosensor. Here, via amidation reaction, the stem-loop-structure probe DNA (pDNA) carrying 3'-amino and 5'-biotin was anchored on the carboxyl functionalized graphene. Then the pDNA was unfolded by the hybridization of the matched target DNA (tDNA). The following recognition between streptavidin (SA) on the Hemin@MOF-SA and biotin at the pDNA led the Hemin@MOF-SA to access the electrode surface. In this way, the electrode modified with Hemin@MOF-SA and matched tDNA could display remarkable catalytic activity toward the reaction of H2O2. The proposed electrochemical DNA biosensor showed an excellent performance with 9 orders of magnitude in the range from 1.0 × 10−15 to 1.0 × 10−6 mol·L−1, the low detection limit of 6.9 × 10−16 mol·L−1 and the one-base mismatch differentiation ability. In addition, the two as-prepared electrochemical sensors also exhibited good stability, showing a potential application in the practical analysis.

A high-sensitivity label-free electrochemical aptasensor for point-of-care measurements of low-density lipoprotein in plasma based on aptamer and MXene-CMCS-Hemin nanocomposites

Jintao Liang,Shuaikang Dong,Xiaohong Tan,Junyan Gong,Xuyang Chen,Zhide Zhou,Guiyin Li

Cardiovascular disease (CVD) remains a significant worldwide health challenge, with mortality rates rising rapidly. Low-density lipoprotein (LDL) is a crucial serum biomarker for the early diagnosis of CVD, which can significantly improve outcomes and reduce mortality. Herein, a label-free electrochemical aptasensor for rapid detection of LDL was developed based on the titanium carbide–carboxymethyl chitosan–hemin (MXene-CMCS-Hemin) nanocomposites as the electrochemical signal probe. Firstly, gold nanoparticles (Au NPs) were electrodeposited onto a screen-printed carbon electrode (SPCE) to form a conductive substrate. Subsequently, the MXene-CMCS-Hemin nanocomposites were anchored onto the Au NPs/SPCE surface. Then LDL Apt was immobilized on the surface of MXene-CMCS-Hemin/Au NPs/SPCE to construct the electrochemical aptasensor. When LDL specifically bound with the LDL Apt to form LDL-LDL Apt complexes, hindering the electron transfer and reducing the Hemin oxidation current, LDL detection can be achieved via differential pulse voltammetry (DPV). Under optimal circumstances, the changes of Hemin's oxidation current showed a good linear response with LDL concentration in the range of 0.1–4.0 μmol/L with a detection limit of 0.095 μmol/L (S/N = 3). The aptasensor demonstrated good performance with the relative errors of 0.60 % to 6.58 % for the direct detection of LDL in human serum samples, which offers a novel tool for the clinical diagnosis of CVD.

Photocurrent-polarity-switching photoelectrochemical and electrochemical dual-mode sensing platform for the highly selective detection of trenbolone

Feng Jiang,Shanghua Liu,Mengxiao Mo,Xinyu Liu,Dongyu Han,Qing Liu,Yueyuan Li,Yueyun Li,Yaqi Li,Qin Wei

As an emerging contaminant in the environment, trenbolone (TBE) has aroused a global health crisis and caused the huge impact on ecosystem. Its sensitive detection is critical to safeguard human health and ecosystem. Herein, a photocurrent polarity switching photoelectrochemical (PEC)-electrochemical (EC) dual-mode sensing platform was presented for highly selective and sensitive detection of TBE by ingeniously tailor signal probe. The tailored Cu 2 O@CuO-hemin acting as both photocurrent polarity reversal and electroactivity factor were cross-linked with TBE-BSA to obtain competitive conjugates for TBE. When the competitive response was initiated, numerous of Cu 2 O@CuO-hemin nanocomposites are introduced on the CdS-modified indium tin oxide electrode, serving as the reporting agent of EC/PEC dual-mode assay. The photocurrent polarity of the CdS-modified ITO can be effectively switched, enabling PEC analysis of TBE with distinguishing photocurrent changes. The linear response range is 1 pg/mL to 500 ng/mL, and the detection limit is 15.0 fg/mL. Meanwhile, the Cu 2 O@CuO could triggered the amplified current variations via effectually catalyzing the H 2 O 2 decomposition compared to the CdS NPs, leading to the sensitive electrochemical detection of TBE with a detection limit 200 fg/mL and a linear response range from1 pg/mL to 500 ng/mL. Additionally, the developed sensing platform enabled mutual authentication of PEC/EC detection results, being conducive to increasing the assay reliability and accuracy. Such the dual-mode sensor has great potential for detecting environmental pollutants due to its improved selectivity and mutual authentication of results.

Biomimetic-Based Design of Tandem Catalysts for Efficient Nitrate to Ammonia Conversion

Longlong Huang,Ling Ye,Hui Wang,Ting Wei,Tianle Xia,Yiwen Du,Xuhai Zhang,Yongjie Ge

Electrocatalytic NO3– synthesis of NH3 (NO3RR) presents a promising alternative to the traditional Haber–Bosch process. However, The NO3RR is a complex process that converts NO3– to NH3 through a series of nitrogen-containing intermediates, such as NO2–, NO–, NOH, etc. This transformation involves 8-electron and 9-proton couplings, resulting in a slow overall reaction rate. Herein, we mimic the dual active-center mechanism found in nature for NH3 synthesis from NO3– and design an Ag-GO-Hemin-His tandem catalyst to optimize the kinetic rate of the NO3RR intermediate reaction. In situ infrared and activation energy studies revealed that Ag serves as the first active center, rapidly promoting the conversion of NO3– to NO2–, while FeN5 in GO-Hemin-His acts as the second active center, efficiently facilitating the reduction of NO2– to NH3. This dual active-center catalyst lowered the energy barrier of the rate-determining step in the NO3RR process from 11.7 to 8.29 kJ mol–1, achieving a high Faraday efficiency (FE) of 91.9% and an impressive NH3 yield rate of 3.80 mg h–1 cm–2 in a 1 M KOH and 0.1 M KNO3 solution at −0.5 V versus RHE. Furthermore, the zinc nitrate battery, assembled with Ag-GO-Hemin-His as the cathode material, exhibited a high power density of 3.28 mW cm–2 and an impressive FE of 88.6% for NH3 generation.
