Silanized iminodiacetic acid (GLYMO–IDA) modified mesoporous silica (G-SBA) was prepared following a co-condensation method. G-SBA/Cu2+, G–SBA/Zn2+ and G-SBA/Cu2+–Zn2+ were obtained through the impregnation method with coordination by GLYMO–IDA. SBA/CuO, SBA/ZnO, and SBA/CuZnO were generated after calcination of G-SBA/Cu2+, G-SBA/Zn2+ and G-SBA/Cu2+–Zn2+. After modification, the amount of metal ion was 6 to 70 times higher than that of the blank mesoporous silica. The diameter of nano-copper oxide particles in mesoporous silica was 4.8 nm. The bacteriostatic rates of SBA/CuO at a copper oxide concentration of 250 ppm against E. coli and S. aureus were 85.87% and 100%, respectively. SBA/CuO and SBA/CuZnO with {111} lattice planes exhibited better antibacterial effects compared to the commercial-grade nano-zinc oxide and nano-copper oxide. When exposed to ultraviolet light, SBA/CuZnO displayed the highest photocatalytic activity and optimal antimicrobial effect. Therefore, SBA/CuZnO can be an alternative to antibiotics because of its non-toxic nature and good antibacterial properties.
Coated microneedles (CMNs) are a minimally invasive platform for immediate-release transdermal drug delivery. However, the practical applications of CMNs have been significantly hindered by the challenges associated with complex formulations, single function, and limited drug loading capacity. This study has developed a spiderweb-shaped iron-coordinated polymeric nanowire network (Fe-IDA NWs). The resulting Fe-IDA NWs are endowed with a certain viscosity due to the synergy of multiple supramolecular interactions. This allows them to replace traditional polymeric thickeners as microneedle coatings. The Fe-IDA NWs-coated microneedles (Fe-IDA MNs) display rapid disintegration in the skin model, which also enables the swift diffusion of Fe-IDA NWs and their payloads into the deeper skin layers. Additionally, Fe-IDA MNs exhibit desirable enzymatic activity and potential antibacterial ability. Thus, Fe-IDA MNs can enhance the therapeutic efficacy against wound infection through synergistic effects, and avoid the overly complicated formulation and the release of nontherapeutic molecules of conventional CMNs. As a proof-of-concept, Fe-IDA MNs loaded with chlorin e6 showed a synergistic chemodynamic-photodynamic antibacterial effect in a methicillin-resistant Staphylococcus aureus-infected wound model in mice. Collectively, this work has significant implications for the future of CMNs-based transdermal drug delivery systems and expands the application fields of metal coordination polymer (MCP) materials.
In this paper, an efficient and convenient Fe 3 O 4 /PMG/IDA-Ni 2+ nanoparticles that applied to purify and immobilize his-tagged β-glucosidase was synthesized, in which, Fe 3 O 4 /PMG (poly ( N, N’- methylenebisacrylamide-co-glycidyl methacrylate) core/shell microspheres were synthesized firstly using distillation-precipitation polymerization, then iminodiacetic acid (IDA) was used to open epoxy rings on the shell of microspheres to the combination of Ni 2+ . The gene of β-glucosidase that was from Coptotermes formosanus Shiraki was amplified, cloned into the expression vector pET28a with an N-terminal His-tag, and expressed in E.coli BL21. The nanoparticles showed the same purification efficiency as commercial nickel column which was a frequently used method in the field of purifying his-tagged proteins from crude cell lysates. The results indicated that Fe 3 O 4 /PMG/IDA-Ni 2+ nanoparticles can be considered as an excellent purification material. β-glucosidase was immobilized on the surface of Fe 3 O 4 /PMG/IDA-Ni 2+ to form Fe 3 O 4 /PMG/IDA-β-glucosidase by means of covalent bound with imidazolyl and Ni 2+ . The immobilized β-glucosidase exhibited excellent catalytic activity and stabilities compared with free β-glucosidase. In addition, immobilized β-glucosidase can be recycled for many times and retain more than 65% of the original activity. The materials display enormous potential in the aspect of purifying and immobilizing enzyme.
