Here, the inner filter effect (IFE) was applied to construct a “turn off” mode up-conversion fluorescence and colorimetric dual-reading detection platform consisting of the chromogenic substance p-phenylenediamine (PPD) and NaYF 4 :Yb/Er@NaYF 4 core–shell up-conversion nanoparticles (UCNPs) for the detection of glucose and hydrogen peroxide (H 2 O 2 ) with high sensitivity and selectivity. In this strategy, hydrogen peroxide is produced in the process of glucose oxidation by catalyzing glucose oxidase (GOx). PPD is oxidized by H 2 O 2 under the action of horseradish peroxidase (HRP), resulting in the transformation into its oxidation product PPDox, whose absorption peak coincides with the emission peak of UCNP at 540 nm, which can significantly quench the fluorescence of UCNP. The quenching efficiency is related to PPDox production, so the relationship between the glucose content and the fluorescence changes of UCNP can be established to realize the sensitive detection of glucose. UCNPs modified by –COOH and the oxidation product of PPD were negatively charged, which increased the distance between donor and recipient due to electrostatic repulsion. It was not conducive to the occurrence of distance-dependent fluorescence resonance energy transfer (FRET), which was also verified by fluorescence lifetime test. Under the optimal experimental conditions, the linear ranges of hydrogen peroxide and glucose were 2.5 ∼ 50 and 5 ∼ 80 μmol/L with the detection limit (LOD) of 0.32 and 0.83 μmol/L, respectively, which were lower than of some reported approaches. Furthermore, the system could be applied to the detection of glucose in actual serum sample and the results were consistency of commercial glucose meters which indicated that the protocol was reliable and potential applications for clinical diagnosis.

Matrix metalloproteinase-2 (MMP-2) is a key biomarker involved in various pathological processes, so its sensitive and selective detection is of great clinical significance. A fluorescent sensor for the detection of MMP-2 was successfully designed based on the inner filter effect (IFE) between carbon dots (CDs) and gold nanoparticles (AuNPs). In the presence of MMP-2, peptide chains are hydrolyzed, releasing short peptides that contain thiol groups and positive charges, thereby inducing the aggregation of AuNPs. This aggregation disrupts the IFE between AuNPs and CDs, leading to a significant enhancement of the fluorescence. By monitoring the change in fluorescence intensity of CDs, the concentration of MMP-2 was accurately determined. The sensor exhibited a linear response across the concentration range from 1.0 to 150 ng/mL, with a detection limit as low as 0.23 ng/mL, which is lower than those of previously reported methods. Furthermore, the sensor demonstrated minimal interference from coexisting biochemical substances for MMP-2 in human serum. Notably, the sensor was successfully applied to determine MMP-2 in human serum, showing satisfactory recovery rates ranging from 98.8 to 1106.0%. In conclusion, this fluorescent sensor offers high selectivity and sensitivity for MMP-2 detection, making it a promising tool for clinical diagnosis of related diseases.

Exploring high-performance fluorescence strategies for onsite monitoring of oxytetracycline (OTC) is of great significance for protecting human health. Herein, an innovative ratiometric fluorescence biomimetic sensor featured with high sensitivity and selectivity was constructed based on magnesium and nitrogen co-doped carbon dots embedded europium metal–organic framework and incorporating molecularly imprinted polymer (Mg,N-CDs/Eu-MOFs@MIP). In this protocol, MIP layer functioning as recognition units endowed Mg,N-CDs/Eu-MOFs@MIP with satisfactory binding efficiency and anti-interference ability. Mg,N-CDs/Eu-MOFs as both supporter and signal tag enabled the sensor to not only demonstrate a fast fluorescence response, but also yield reverse response signals depending on the synergistic effect of inner filter effect (IFE) and antenna effect (AE), achieving signal amplification. Benefiting from the efficient dual-channel strategy, Mg,N-CDs/Eu-MOFs@MIP exhibited a wide linear range (0.02–50 μg mL −1 ) with a sensitive detection limit (6.6 ng mL −1 ) and a marked color transformation (blue-to-red), which was highly desirable for monitoring of oxytetracycline. Simultaneously, aiming at attaining precise onsite analysis, a portable intelligent sensing platform was devised by integrating Mg,N-CDs/Eu-MOFs@MIP and a smartphone-assisted device. The smartphone with an installed color recognizer application as signal reader and analyzer realized oxytetracycline visual sensing by capturing and digitizing fluorescence images. More significantly, the portable platform was successfully applied to the onsite monitoring of OTC in water samples and milk samples with acceptable results. Accordingly, the as-developed platform demonstrated low-cost, portability, easy implementation, desirable specificity and sensitivity, presenting great potential for point-of-care testing.

To address the need for highly sensitive and reliable detection of trace ochratoxin A (OTA) in food matrices, we developed a ratiometric fluorescent aptasensor by integrating CRISPR/Cas12a, hybridization chain reaction (HCR), and horseradish peroxidase (HRP)-induced inner filter effect (IFE). The mechanism involves OTA releasing an activator that initiates CRISPR/Cas12a trans-cleavage, blocking HCR assembly. This reduces HRP levels, limiting the conversion of o -phenylenediamine (OPD) to fluorescent 2,3-diaminophenazine (DAP) (emitting at 562 nm) while maintaining strong emission from 2-amino terephthalic acid (BDC-NH 2 ) at 426 nm. The F 426 /F 562 ratio serves as a “signal-on” indicator, enabling sensitive OTA detection over 0.1 pM to 10 nM, with a detection limit of 0.0417 pM. The method exhibits excellent reproducibility, with intra-day and inter-day relative standard deviations (RSDs) of 1.91 %–3.87 % and 1.79 %, respectively, along with recovery rates of 90.1 %–110.6 % in real samples. These advantages highlight its significant potential for CRISPR/Cas-based OTA detection.

How to manage the potential environmental pollution of waste biomass while realizing the resource utilization of the waste is a topic worthy of in-depth investigation. In this paper, after successfully preparing a deep eutectic solvents, we constructed N, Cl-CDs fluorescent probes with high fluorescence yield and biocompatibility by utilizing waste soybean residue as a cheap nitrogen-rich carbon-sourced biomass precursor. The results showed that the fluorescence quenching of N, Cl-CDs has a good linear relationship with the concentration of p-nitrophenol in the range of 0.1–60 μM, with the detection limit of 30 nM, and the fluorescence quenching mechanism was the inner filter effect. This paper demonstrates that the carbon dots fluorescence system has a superior prospect for the highly selective detection of p-nitrophenol, and provides a reliable basis for the future exploration of efficient and environmentally friendly carbon dots synthetic method as well as the economic recycling of waste.

The level alterations of antioxidants are highly associated with various oxidative stress-related diseases, food quality and drug safety problems, which enables the reliable and accurate determination of total antioxidant capacity (TAC) become progressively significant. Herein, for the first time, we explored the catalytic effect of MnSiO 3 nanozyme with oxidase-like activity towards different fluorescent substrates, and introduced ratiometric fluorescence to MnSiO 3 -based biosensing. By harnessing MnSiO 3 -propelled catalytic oxidation of two fluorescent substrates with contrary responses (Scopoletin, SC and o-phenylenediamine, OPD), we constructed a facile, cost-effective ratiometric fluorescent (RF) platform for TAC measurement. In the absence of antioxidants, SC and OPD can be oxidized, and the synergistic effect of fluorescence resonance energy transfer (FRET) and inner-filter-effect (IFE) between SC and oxidized OPD (Ox-OPD) will result in the weak fluorescence of Ox-SC at 465 nm and the strong one of Ox-OPD at 562 nm, respectively. However, the addition of antioxidants will dissociate MnSiO 3 , generating the recovered high/low fluorescence values of both substrates. Through monitoring the FI 465 /FI 562 changes, the RF analysis of different antioxidants (Cys, AA, GSH) with satisfactory sensitivity is performed and the TAC measurements in diverse real samples (human serums, beverages, and drugs) are successfully accomplished. Moreover, the system demonstrates high selectivity and could compete with the commercial TAC test kits.

The development of dual-mode biosensors for the accurate detection of acid phosphatase (ACP) activity and the screening of its inhibitors is of vital importance in clinical diagnostics and environmental monitoring. In this work, we report the synthesis of Mn–Ce co-doped carbon dots (Mn&Ce@CDs) exhibiting both oxidase-mimetic catalytic activity and intrinsic fluorescence. The nanozymes efficiently catalyze the oxidation of colorless o -phenylenediamine (OPD) to produce a yellow chromogen, 2,3-diaminophenazine (DAP), characterized by a distinct absorption peak. Significantly, the excitation spectrum of DAP overlaps with the emission wavelength of Mn&Ce@CDs, resulting in an inner filter effect (IFE) that quenches the blue fluorescence of the carbon dots while simultaneously enhancing DAP's emission. This mechanism establishes a robust ratiometric fluorescence signal. The enzymatic hydrolysis of ascorbyl-2-phosphate by ACP yields ascorbic acid, which inhibits the nanozyme-catalyzed oxidation of OPD. Consequently, this leads to a decrease in DAP formation, diminishing both its characteristic absorption and the IFE-induced fluorescence quenching, thus producing a dual-signal response. Quantitative ACP detection exhibited a linear range of 0.5–15 U/L with limits of detection of 0.037 U/L in fluorescence mode and 0.49 U/L in colorimetric mode. The platform demonstrated high selectivity and reliability, showing recovery rates of 97.7–109.3 % (fluorescence) and 97.9–107.5 % (colorimetric) in spiked serum samples. Additionally, smartphone-based RGB analysis enables rapid on-site detection. The system was successfully applied for screening ACP inhibitors, with chlorpyrifos exhibiting IC 50 values of 50.45 μg/mL (fluorescence) and 43.77 μg/mL (colorimetric). This integrated dual-mode approach offers a powerful, reliable tool for clinical diagnostics and environmental assessment.

Tetracycline antibiotics (TCs), widely used as broad-spectrum bacteriostatic agents in veterinary and medical applications, had cause serious harm to human beings, and thus, on-site and portable quantification methods for TCs evaluation are urgently necessary. In this study, through integrating inner filter effect (IFE) into antenna effects, a response approach with fluorescence and colorimetric dual signals was constructed for portable sensing of TCs based on metal-organic framework (MOF)-based nanoprobe, i.e., NH 2 -MIL-101(Fe)-Eu 3+ . With this strategy, TCs first quenches the blue fluorescence of nanoprobe by IFE and subsequently excites sharp red fluorescence of Eu 3+ through antenna effect, achieving the bimodal response in one fluorescence spectrum. Notably, the complexation of TCs and nanoprobe promotes electron transfer between Fe(Ⅲ) and Fe(Ⅱ), enhancing the peroxidase-like activity of the nanoprobe for specific colorimetric evaluation of TCs with assistance of 3,3',5,5'-tetramethylbenzidine (TMB) indicator. The ratiometric fluorescence-colorimetric dual-mode sensing strategy demonstrates an ultra-high sensitivity with low limits of detection (LODs) of 8.74×10 -5 ppm, and provides reliable recoveries from 89.74% to 108.25% in the honey samples. Additionally, a rapid “two-solution” extraction strategy proposed in this study, as demonstrated by statistical comparisons (Two one-sided tests (TOST), Deming regression, and Bland-Altman analysis), yields result comparable to laboratory extraction methods across various food and environmental samples, adapting to different matrices to minimize preparation steps and time. Expectedly, this simple and responsive dual-mode sensing approach, which can be combined with smartphone recognition due to the chromaticity changes of MOF probe, provides a new direction for advancing on-site detection of contaminants in food and environmental samples.
