Lactobacillus reuteri DSM 17938 alleviates d-galactosamine-induced liver failure in rats

Huiyong Jiang,Ren Yan,Kaicen Wang,Qiangqiang Wang,Xiaoxiao Chen,Lifeng Chen,Lanjuan Li,Longxian Lv

Liver failure is a serious hepatic dysfunction with high mortality. This work aimed to investigate the effect of a famous probiotic and drug, Lactobacillus reuteri DSM 17938, on liver failure in rats. Sprague-Dawley rats were gavaged with 3 × 10 9 CFU of DSM 17938 for 7 days. d -galactosamine was intraperitoneally injected to induce acute liver failure on the eighth day. Samples were collected to determine the liver function, serum cytokines levels, terminal ileum and liver histology, gut microbiota, metabolome and transcriptome. Our results showed that pretreatment with DSM 17938 not only reduced the elevation in serum alanine aminotransferase, aspartate aminotransferase, gamma-glutamyl transferase, IL-1α, IL-2, IL-18, M-CSF, and MIP-3α levels but also alleviated histological abnormalities of both the terminal ileum and liver induced by d -galactosamine. Additionally, DSM 17938 reduced d -galactosamine-induced enrichment of some taxa of gut Actinobacteria or Firmicutes, including abundant pathogens such as Actinomycetales , Coriobacteriaceae , Staphylococcaceae and Enterococcaceae . Furthermore, DSM 17938 reduced the d -galactosamine-induced increase in not only fecal metabolites such as trisaminol and lithocholic acid but also the transcription of liver inflammatory genes, such as Ccl2 , Ccl7 , Ccl11 , Ccl12 , Il6 , Il11 , Il20rb , Mmp3 and Mmp10 . Downregulation of retinol metabolism and PPAR signaling pathway as well as upregulation of viral protein interaction with cytokine and cytokine receptor and central carbon metabolism in cancer signaling pathway were involved in the mechanism of L. reuteri DSM 17938 alleviating liver failure. Our findings suggested that DSM 17938 is a potential probiotic for the prevention or treatment of liver failure.

Expression, characterization and structural profile of a heterodimeric β-galactosidase from the novel strain Lactobacillus curieae M2011381

Jiaying Zhu,Jiaqi Sun,YaJie Tang,Jingli Xie,Dongzhi Wei

A heterodimeric β-galactosidase was discovered in the novel strain Lactobacillus curieae M2011381. The gene encoding the enzyme was expressed in Escherichia coli BL21 (DE3). The specific enzyme activities of the recombinant holoenzyme (LacLM) and large subunit (LacL) measured 11.4 U/mg and 3.8 U/mg, respectively. The k cat /K m values of LacLM and LacL were 740 mM −1 s −1 and 1.40 mM −1 s −1 , respectively. LacLM showed maximum activity at pH 8.0 and 55 °C, and it could maintain its activity at a neutral pH and below 45 °C. LacLM displayed both hydrolysis and transgalactosylation activity on 200 g/L lactose. When LacLM was added to milk, the lactose was hydrolyzed after 6 h without galactooligosaccharide generation. The sequence alignment and homology modeling of the structures of the holoenzyme and subunits revealed that LacL has a catalytic domain with a catalytic dyad, Glu470 and Glu538, and small subunit LacM is a β-sheet domain with a conserved Trp294. The molecular docking of LacLM helped to illustrate the roles of both subunits in the reaction with lactose.

Lactobacillus fermentum CECT5716 Alleviates the Inflammatory Response in Asthma by Regulating TLR2/TLR4 Expression

Weifang Wang,Yunfeng Li,Guojing Han,Aimin Li,Xiaomei Kong

Background Asthma is a chronic disease, which is harmful to the health of the body and the quality of life. Supplementation of Lactobacillus can affect the immune environment of the lungs through the gut-lung axis. This study aimed to explore the potential regulatory targets of Lactobacillus to relieve inflammation in asthma and determine a new approach for improving asthma. Methods A mouse ovalbumin (OVA)-induced model was constructed. OVA mice were supplemented with Lactobacillus fermentum CECT5716 by gavage. The gut microbiota composition of normal and OVA mice was analyzed using 16S ribosomal DNA identification. BALF, serum, lung tissues, and duodenal tissues were collected. Wright’s staining was performed to determine the cell content of the alveolar lavage fluid. Hematoxylin-eosin staining, Masson staining, and periodic acid-Schiff staining were performed to observe the improvement in the lungs of OVA mice supplemented with Lactobacillus . Immunofluorescence was performed to measure the severity of the intestinal barrier leakage. Enzyme-linked immunosorbent assay was carried out to determine the expression levels of inflammatory cell factors, while quantitative reverse transcription-polymerase chain reaction and western blotting were performed to detect the levels of toll-like receptor 2 (TLR2)/TLR4 expression and cell adhesion factors. Results Compared with Control mice, OVA mice exhibited malignant conditions, such as intestinal leakage and lung edema. After supplementation with Lactobacillus , the inflammatory cell content in the bronchoalveolar lavage fluid decreased, and the inflammatory response was alleviated. The level of TLR2/TLR4 expression was reduced. The inflammatory cell infiltration in the airway mucosa of OVA mice was improved, alveolar swelling was reduced and the basement membrane appeared thinner. Conclusion The Lactobacillus inhibited the TLR2/TLR4 expression in OVA mice. Supplementation with Lactobacillus can alleviate the inflammatory response in OVA mice, inhibit pulmonary fibrosis, and treat asthma.

Lactobacillus paracasei fermented goji juice improves constipation in mice through inhibiting inflammation and regulating intestinal microecology

Xiaoyu He,Yu Zhang,Haitian Fang,Ni Ma,Xueni Yan,Huabing Yang,Lu Cao,Lu Xu,Haiming Hu,Hongtao Liu

This study investigated the effects of Lactobacillus paracasei ( L. paracasei )-fermented Goji juice supernatant (FGS) on constipation. GC–MS data showed that the critical metabolites of FGS were nicotinamide, squalene, L-proline, melatonin, and sarcosine. FGS intervention significantly promoted intestinal peristalsis. Meanwhile, FGS ameliorated intestinal inflammation and mucosal barrier damage by inhibiting the expressions of pro-inflammatory factors (TNF-α, IL-1β, COX-2), promoting the expressions of Mucin 1, Mucin 3, ZO-1, AQP4, and ENac-γ. In addition, FGS treatment modulated gut microbiota composition, reduced the abundance of harmful bacteria ( Faecalibaculum , Bacteroides , Alloprevotella and Alistipes ), induced a bloom of probiotics, including Marvinbryantia , Eubacterium _ xylanophilum _group, Colidextribacter , and Candidatus _ Soleaferrea . Conclusively, this study demonstrated that FGS significantly reduced inflammatory response, prevented intestinal barrier damage, and modulated microbiota dysbiosis to improve constipation. Compared to unfermented Goji juice (UFGJ), FGS has shown a more effective anti-constipation effect, indicating that FGS could relieve constipation as a dietary therapeutic.

Physicochemical properties and volatile components of pea flour fermented by Lactobacillus rhamnosus L08

Mengqi Pei,Zifu Zhao,Song Chen,E.I. Reshetnik,S.L. Gribanova,Chun Li,Guofang Zhang,Libo Liu,Lin Zhao

Pea ( Pisum sativum L.) is an important source of plant proteins . However, the use of pea flour as a food ingredient is greatly limited due to its strong beany flavor. In this work, we investigated the effects of Lactobacillus rhamnosus L08 ( L. rhamnosus L08) fermentation on the physicochemical characteristics, amino acids composition and volatile flavor compounds of pea flour. We found that L. rhamnosus L08 had the probiotic potential of utilizing pea flour as fermentation substrate. L. rhamnosus L08 fermentation significantly increased the content of most amino acids , and total amino acid content increased from 222.18 mg/g at 0 h to 262.35 mg/g at 6 h. A total of 44 volatile compounds were identified in pea flour. L. rhamnosus L08 fermentation increased the variety of acids and esters, while reduced the unpleasant flavor such as nonanal , decanal , octanal , 1-hexanol and 2-ethyl-1-hexanol. The emulsifying stability and foam stability of pea flour were slightly enhanced during fermentation. Our results illustrate that lactic acid bacteria fermentation is a promising approach to eliminate undesirable aroma of pea flour. Moreover, this study provides theoretical support for the application of peas as high-quality protein source.

Insight into the mechanism of decreasing N-nitrosodimethylamine by Lactobacillus pentosus R3 in a model system

Xuefei Shao,Baocai Xu,Hui Zhou,Conggui Chen,Peijun Li

The mechanism of decreasing N- nitrosodimethylamine (NDMA) by Lactobacillus pentosus R3 isolated from Chinese dry sausage was assessed in a model system. The model system was devised using a DeMan-Rogosa and Sharp-ferrous sulfate broth supplement with NDMA or its precursors. The results showed that inoculating L. pentosus R3 could significantly decrease the content of NDMA ( P  < 0.05). The NDMA precursors (dimethylamine and putrescine) were found to be decreased by L. pentosus R3 ( P  < 0.05). Nitrite was degraded to nitrosonium cation by acidolysis, which may promote the formation of NDMA. Direct degradation of NDMA by L. pentosus R3 was also observed. The strain exhibited antioxidant activities , including Fe 2+ chelating activity, which may be responsible for inhibiting the formation reaction and decreasing the precursors of NDMA. Thus, L. pentosus R3 decreased the content of NDMA possibly by three ways: inhibition of the formation reaction, decrease in precursors (DMA and putrescine) and direct degradation. It is interesting to understand the mechanism of decreasing NDMA by L. pentosus R3 to improve the safety of fermented meat products .

Oleanolic acid inhibits the tumor progression by regulating Lactobacillus through the cytokine-cytokine receptor interaction pathway in 4T1-induced mice breast cancer model

Kan He,Xia Meng,Jinxing Su,Shangquan Jiang,Min Chu,Bei Huang

The therapeutic mechanism of oleanolic acid (OA) in breast cancer has been widely reported, but little has been known about the combined effects of transcriptome and gut microbiome. In this study, the phenotypic effect of oleanolic acid on mice was tested at the end of the administration cycle, and RNA sequencing on murine tumor tissue and 16S-rRNA sequencing on intestinal contents were conducted to analyze gene expression profiles and microbial diversity between the control group and OA treated group using 4T1-induced mice breast cancer model. As a result, it has been confirmed that oleanolic acid would play a significant inhibitory effect on the development of breast tumors in mice. Based on the integrative analysis of the transcriptomic and metagenomic data, it was found that the abundance of Lactobacillus in the intestinal flora of mice significantly increased in the OA group. Moreover, the up-regulation of Il10 had a significant effect on inhibiting the tumor progression, which played a role through cytokine-cytokine receptor interaction pathway.

Attenuation of MEHP-induced hepatotoxicity and gut microbiota dysbiosis by Lactobacillus brevis Lb-0401 in Carassius auratus

Yao Yao,Yunshan Zhang,Binqing Xue,Weihua Chu

Mono-2-ethylhexyl phthalate (MEHP) is one of the major biologically active metabolites of phthalate plasticizers that pose a health threat to aquatic animals. In this study, we investigated the ecotoxicological effects of MEHP on liver damage and gut microbiota in crucian carp ( Carassius carassius ) and the mitigating effects of Lactobacillus brevis Lb-0401 as a dietary supplement. The results showed that MEHP induced hepatotoxicity and increased serum AST, ALT and triglyceride levels, while L. brevis Lb-0401 alleviated abnormalities in serum metabolic parameters and increased antioxidant enzymes (T-SOD, CAT, GSH-Px). Further transcriptome and microbiota analysis revealed that MEHP exposure caused hepatic glucose, lipid and purine metabolism disorders, inhibited energy production and disrupted gut microbial homeostasis. Administration of L. brevis Lb-0401 reversed all these changes. In conclusion, this study not only provides new mechanistic insights into the ecotoxicological effects of MEHP, but also suggests a potential strategy using intestinal autochthonous probiotic bacteria to mitigate the harmful effects of phthalate plasticisers on aquatic organisms.

Colonization and immunoregulation of Lactobacillus plantarum BF_15, a novel probiotic strain from the feces of breast-fed infants

Na Zhang,Chen Li,Zhihua Niu,Hongyan Kang,Miaoshu Wang,Bo Zhang,Hongtao Tian

Immunosuppression is a manifestation imbalance in the immune system, often during unhealthy states. In recent years, lactic acid bacteria (LAB) have been found to be important components of the body's innate immune system, and indispensable to maintaining normal immune function. Lactobacillus plantarum BF_15, a novel strain isolated from the feces of breast-fed infants, which has shown potential as an immunomodulator in vitro. In the present study, with the Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis (PCR-DGGE) based on RNA-polymerase beta subunit encoding gene (rpoB) to analyze the colonization of L. plantarum BF_15 in the intestine of mice. In addition, Lactobacillus rhamnosus GG (LGG) as a positive control strain, by measuring immune-related indexes and the diversity of intestinal microbiota, the effects of BF_15 on immunoregulation and intestinal microbiota dysbiosis were investigated to elucidate whether the attenuation of immunosuppression is related to the modulation of intestinal microbiota. Results did indeed support this notion that BF_15 did colonize murine intestines well, in which it could still be detected in mice feces 14 days after stopping the probiotic administration. Moreover, BF_15 found to protect mice against reduction in the levels of several immune-related indicators, including the thymus and spleen indexes, splenic lymphocyte proliferation, toe swelling degree, serum hemolysin-antibody level, and macrophage phagocytosis index, triggered by high-dose (200 mg kg−1) intraperitoneal administration of cyclophosphamide (CTX). In addition, the strain was also found to effectively balance intestinal microbiota dysbiosis in the mice. Collectively, these results indicated that L. plantarum BF_15 can not only successfully colonize murine intestines, but also can effectively alleviate CTX-induced immunosuppression, once established, by rebalancing the intestinal microbiota. This, therefore, provides strong evidence for the view that BF_15 has the potential to become a highly effective immunomodulating probiotic in human microbiota as well.

Lactobacillus intestinalis facilitates tumor-derived CCL5 to recruit dendritic cell and suppress colorectal tumorigenesis

Yong Sun,Qiwen Wang,Yao Jiang,Jiamin He,Dingjiacheng Jia,Man Luo,Wentao Shen,Qingyi Wang,Yadong Qi,Yifeng Lin,Ying Zhang,Lan Wang,Liangjing Wang,Shujie Chen,Lina Fan

Gut microbes play a crucial role in regulating the tumor microenvironment (TME) of colorectal cancer (CRC). Nevertheless, the deep mechanism between the microbiota-TME interaction has not been well explored. In this study, we for the first time discovered that Lactobacillus intestinalis (L. intestinalis) effectively suppressed tumor growth both in the AOM/DSS-induced CRC model and the ApcMin/+ spontaneous adenoma model. Our investigation revealed that L. intestinalis increased the infiltration of immune cells, particularly dendritic cells (DC), in the TME. Mechanically, the tumor-derived CCL5 induced by L. intestinalis recruited DC chemotaxis through the NOD1/NF-κB signaling pathway. In clinical samples and datasets, we found positive correlation between L. intestinalis, CCL5 level, and the DC-related genes. Our study provided a new strategy for microbial intervention for CRC and deepened the understanding of the interaction between tumor cells and the immune microenvironment modulated by gut microbes.

Enhancement in the physicochemical properties, antioxidant activity, volatile compounds, and non-volatile compounds of watermelon juices through Lactobacillus plantarum JHT78 fermentation

Feifei Shi,Li Wang,Shurong Li

In this study, the influences of Lactobacillus plantarum JHT78 fermentation on the physiological properties, antioxidant activities , and volatile/non-volatile metabolites of watermelon juices were comprehensively investigated. The results indicated that total polyphenols flavonoids and anthocyanin in the watermelon juices remarkably increased through L. plantarum JHT78 fermentation. L. plantarum JHT78 fermentation enhanced the antioxidant activities , lipase inhibition, and α-glucosidase activities of watermelon juices. A total of 62 volatile compounds were detected using HS-SPME-GC-MS, mainly including 11 acids, 8 aldehydes, 7 ketones, and 7 alcohols. The abundance of 19 volatile compounds especially for acids remarkably increased for the fermentated watermelon juice. Furthermore, non-volatile compounds detected by UHPLC-QTOF-MS revealed that L. plantarum JHT78 significantly altered the non-volatile compounds of watermelon juices, especially increased indole-3-lactic acid. The results confirmed that L. plantarum JHT78 enhanced the functionality of watermelon juices thus providing a theoretical basis for the development of LAB on plant-based beverages.

Lactobacillus helveticus LZ-R-5 Ameliorates DSS-Induced Colitis in Mice by Modulating Gut Microbiota and Enhancing Intestinal Barrier Function

Xiaogan Zhao,Tao Lin,Wenkai Jiang,Yihan Lin,Luyao Xiao,Yufang Tian,Kai Ma,Changliang Zhang,Feng Ji,Ghahvechi Chaeipeima Mahsa,Xin Rui,Wei Li

Lactobacillus helveticus LZ-R-5 (R-5), a strain with high epithelial adhesion and bioactive exopolysaccharide production, was isolated from Tibetan kefir grains. This study investigated its potential to alleviate intestinal inflammation using a DSS-induced colitis model in BALB/c mice. We integrated microbial diversity and serological analyses to assess changes in gut flora and cytokines following the R-5 treatment. Pathological assessments showed that R-5 reduced crypt distortion in the proximal colon and mitigated hepatic immune challenges by enhancing gut barrier function. The increased relative expression of TGF-β1 and the downregulation of NLRP3-related inflammatory factors were conducive to preventing organ damage in the thymus and spleen of mice with colitis. Additionally, R-5 stimulated GPR43 expression and improved epithelial nutrition, promoting mucin production to prevent enterotoxin leakage. It also modulated the gut microbiota by suppressing Bacteroides and Erysipelatoclostridium, leading to a microbiota composition more akin to that of normal flora.
