Involvement of cytochrome P450 gene CYP333B119v2 in the resistance of Cydia pomonella to lambda-cyhalothrin

Yu-Ze Li,Chao Hu,Yu-Xi Liu,Yun-Tong Lv,Xue-Qing Yang

Extensive studies have demonstrated that cytochrome P450 enzymes play a critical role in the metabolic detoxification and resistance development of pests against insecticides; however, relative to P450 genes in Clan 3, knowledge on mitochondrial Clan members remains limited. Here, we report the involvement of a CYP333 family member, CYP333B119v2 , in conferring lambda -cyhalothrin resistance in Cydia pomonella . Phylogenetic analysis shows that CYP333B119v2 clusters with CYP333 family members of Lepidoptera, it containing three highly conserved P450 domains: the Heme-binding region, C-Helix, and K-helix. Spatiotemporal expression profiling indicates that CYP333B119v2 transcription levels increase progressively during larval development, peaking at the fourth instar, with significant expression in the midgut, the primary detoxification tissue. Relative to susceptible population (SS), CYP333B119v2 expression is 2.73-fold higher in lambda -cyhalothrin moderate resistant population (LCR) and is significantly induced by sublethal doses of lambda -cyhalothrin . Knockdown of CYP333B119v2 enhances larval susceptibility to lambda -cyhalothrin. Molecular docking based on AlphaFold models revealed strong binding interactions between CYP333B119v2 and lambda -cyhalothrin, with diverse non-covalent interactions involved in the binding process. These findings suggest that CYP333B119v2 may contribute to lambda -cyhalothrin resistance in C. pomonella . This study is the first report implicating a mitochondrial CYP333B subfamily member in the resistance mechanisms of Lepidoptera, advancing our understanding of P450-mediated metabolic resistance and providing new insights into pest management strategies.

Preparation of zein-grafted mesoporous silica nanoparticles for improving Lambda-cyhalothrin insecticidal activity and biosafety

Shaoyang Sun,Shengfu Zhang,Zhenxin Lv,Ning Wei,Ze Lv,Xiaohan Meng,Jianguo Feng

Currently, stimuli-responsive pesticide carriers can effectively enhance the insecticidal effectiveness against target pests and safety to nontarget organisms , and further reduce environmental pollution. In this work, a pesticide delivery system (LCN@MSN-SS-Zein) based on pH and redox dual-stimuli-responsive was prepared by loading Lambda-cyhalothrin (LCN) onto biodegradable disulfide-functionalized mesoporous silica nanoparticles and combining with gatekeeper zein. The results showed that LCN@MSN-SS-Zein had a high pesticide loading (24.7%, w/w). The nanoparticles have excellent pH and redox dual response behavior and have significant controlled release characteristics. Under the stimulation of pH and glutathione , the disulfide bond and zein structure of the nanoparticles are decomposed to release LCN on demand. Compared with LCN technical, the stability of LCN@MSN-SS-Zein nanoparticles under ultraviolet light was significantly enhanced, and the adhesivity of these nanoparticles on the cabbage leaf surface was significantly improved. Biological activity tests showed that LCN@MSN-SS-Zein nanoparticles had higher insecticidal activity against Spodoptera exigua than Lambda-cyhalothrin emulsion oil in water (LCN@EW), and the control effect was more lasting, while the toxicity to zebrafish was significantly reduced. In addition, MSN-SS-Zein nanoparticles were safe for the growth of cabbage and had good biocompatibility. Therefore, LCN@MSN-SS-Zein nanoparticles provide a new strategy for effective pest control and have broad application prospects.

Identification of G Protein-Coupled Receptors (GPCRs) Associated with Lambda-Cyhalothrin Detoxification in Cydia pomonella

Yu-Xi Liu,Chao Hu,Yu-Ting Li,Ping Gao,Xue-Qing Yang

While previous studies have reported G protein-coupled receptor (GPCR)-mediated insecticide resistance in various arthropods, the understanding of GPCR-associated resistance mechanisms in Cydia pomonella remains limited. In this study, a total of 95 CpGPCR genes categorized into four families were identified in C. pomonella. Results revealed high expression levels of the majority of the CpGPCRs during the first larval stage and in the head of C. pomonella. Exposure to lambda-cyhalothrin significantly increased the expression of 15 CpGPCRs, including CpGPCR70, which is highly expressed in all larval stages and shows the highest expression in the midgut. RNA interference (RNAi) demonstrated that downregulation of CpGPCR70 leads to reduced expression of key resistance-related genes and a decreased tolerance of larvae to lambda-cyhalothrin. These findings indicate that CpGPCR70 plays a crucial role in regulating the expression of detoxifying genes involved in lambda-cyhalothrin resistance, offering valuable insights for the development of more effective pest control strategies.

Transcription Factor AhR Regulates Glutathione S-Transferases Conferring Resistance to lambda-Cyhalothrin in Cydia pomonella

Chao Hu,Yu-Xi Liu,Shi-Pang Zhang,Ya-Qi Wang,Ping Gao,Yu-Ting Li,Xue-Qing Yang

Aryl hydrocarbon receptor (AhR) enhances insect resistance to insecticides by regulating the detoxification network. Our previous studies have confirmed that overexpressions of cytochrome P450 monooxygenases (P450s) and glutathione S-transferases (GSTs) are involved in lambda-cyhalothrin resistance in Cydia pomonella. Here, we report that CpAhR regulates the expression of GST and P450 genes, thus conferring resistance. Expression patterns indicated that the expression of CpAhR was highly induced by lambda-cyhalothrin exposure and upregulated in a lambda-cyhalothrin-resistant population. RNA interference (RNAi) of CpAhR decreases the expression of key resistance-related genes (CpGSTe3, CpCYP9A121, and CpCYP9A122) and the activity of the GST enzyme, reducing the tolerance to lambda-cyhalothrin. Furthermore, β-naphthoflavone, a novel agonist of AhR, was first proven to be effective in increasing CpAhR expression and larval tolerance to lambda-cyhalothrin. These results demonstrate that CpAhR regulates the expression of key detoxifying genes and GST activity, resulting in the development of resistance to lambda-cyhalothrin in C. pomonella.

Mesoporous silica size, charge, and hydrophobicity affect the loading and releasing performance of lambda-cyhalothrin

Shuting Xiao,Ali Shoaib,Jiang Xu,Daohui Lin

Nanopesticides are attracting increasing attention as a promising technology in agriculture to improve insecticidal efficacy, decrease pesticides uses, and reduce potential environmental impacts. We synthesized mesoporous silica nanoparticles , i.e., Mobil Composition of Matter No.48 (MCM-48), with different sizes (63–130 nm), charges (−22 to 12 mV), and hydrophobicity (water contact angle 29–103°) to assess their loading amount and release of a typical poorly soluble halogenated pyrethroid (i.e., lambda-cyhalothrin particles, LCNS). The smallest MCM-48 displayed relatively higher loading amount of LCNS (~16%) compared to the larger MCM-48 nanoparticles, likely because of its higher pore volume (1.46 cm 3 g −1 ) and pore size (3.56 nm). LCNS loading amount was further improved to ~26% and ~36% after –NH 2 (positively charged) and –CH 3 (hydrophobic) functionalization, respectively, probably due to hydrogen bonding, electrostatic, and hydrophobic interactions with LCNS. Loading LCNS in MCM-48 nanoparticles also significantly improved its dispersion in water and ultraviolet (UV) light stability, with a 3–7 times longer half-life than that of free LCNS. Although the –NH 2 and –CH 3 modifications of MCM-48 slightly decreased the UV stability of LCNS, they significantly decreased the release efficiency of LCNS, possibly because of their stronger interactions with LCNS. In addition, the insecticidal effects of LCNS-loaded MCM-48 were more efficient and longer than those of free LCNS. The findings clarify the relationships between physicochemical properties and performance of mesoporous silica nanoparticles, and will inform the rational design of materials for controlled release of pesticides and sustainable control of pests.

Integrated substrate-nanozyme-adsorbent tri-functional sensor: sensitive detection of lambda-cyhalothrin residues in agricultural products

Yu Su,Liwen Wei,Xinjie Yin,Xiaofeng Wei,Yanhua Chen,Lan Ding,Daqian Song

Lambda-cyhalothrin (LC), a persistent pesticide with health risks, requires rapid and reliable monitoring methods. Herein, multifunctional materials (MIPs@MIL-88-Cu) were developed by incorporating molecular imprinting technology into Cu-doped MIL-88 nanozyme synthesis, with terephthalic acid serving as both structural backbone and self-supplying substrate. This method successfully realized the integration of substrate, nanozyme and adsorbent functionalities within a single system. LC specifically occupied the imprinted cavities, which inhibited the nanozyme activity and quenched the fluorescence through electron transfer, thus enabling specific recognition of LC. The developed method exhibited a linearity (50.0–1000 ng L −1 ), with a limit of detection of 15.0 ng L −1 . Furthermore, it also simplified both the synthesis and detection processes. The practical application potential of the developed method was validated using spiked vegetable and fruit sample matrix, achieving satisfactory recoveries and low relative standard deviations. This study offered innovative insights into constructing sensitive recognition sensors.

Molecularly imprinted polymers derived from lignin-based Pickering emulsions and their selectively adsorption of lambda-cyhalothrin

Mengying Gan,Jianming Pan,Yunlei Zhang,Xiaohui Dai,Yijie Yin,Qin Qu,Yongsheng Yan

Lignin particles were used as stabilizers to establish a stable Pickering oil–water emulsion, and then the molecularly imprinted polymers (MIPs) were fabricated by radical polymerization in the presence of functional and cross-linked monomers in the oil phase. As-prepared MIPs were observed with a diameter from 10   μm to 65   μm when oil/water ratio increased from 0.11 to 0.68, and exhibited thermal stability and hydrophobic surface. A series of static adsorption experiments were conducted to analyze their adsorption performance for lambda-cyhalothrin (LC). The preliminary results showed that MIPs possessed good recognition toward LC , that is, the adsorption equilibrium time was about 3.0   h and the equilibrium adsorption amount was 72.25   μmol   g − 1 for MIPs and 43.10   μmol   g − 1 for nonimprinted Polymers (NIPs), respectively, indicating the enhanced selectively adsorption of LC. The adsorption process followed pseudo-second-order model by the kinetic analysis and Freundlich equation by the isothermal analysis, respectively. The selective recognition experiments displayed the outstanding selective adsorption effect of the MIPs for target LC. Moreover, the obtained MIPs could be effectively regenerated and recycled at less three times without significant loss of adsorption capacity.

Functional Analysis of Two Carboxylesterase Genes AsCarE13 and AsCarE15 Associated with Lambda-Cyhalothrin Resistance in Aphis spiraecola

You Yi,Zhang Shuo,Zhang Shengyuan,Liu Yunxaing,Li Yongqiang,Yan He,Ma Zhiqing,Kang Wang

Carboxylesterases (CarEs) are critical enzymes involved in insecticide resistance, yet their specific roles in pyrethroid resistance of Aphis spiraecola remain unclear. In this study, enzymatic activity and synergism bioassays initially implicated CarE involvement in lambda-cyhalothrin resistance of A. spiraecola . RT-qPCR revealed that AsCarE13 and AsCarE15 were significantly upregulated in the lambda-cyhalothrin resistant strain (LC-R), with predominant expression in the gut and hemolymph, and marked inducibility upon insecticide exposure. RNAi of AsCarE13 and AsCarE15 significantly increased the susceptibility of A. spiraecola to lambda-cyhalothrin by 45.99% and 29.86%, respectively. In contrast to AsCarE15, recombinant AsCarE13 displayed strong α-NA hydrolytic activity and was competitively inhibited by lambda-cyhalothrin. HPLC and GC–MS/MS analyses further confirmed that AsCarE13 efficiently hydrolyzed lambda-cyhalothrin to yield 3-phenoxybenzaldehyde, whereas no obvious metabolism was detected for AsCarE15. These results suggest that AsCarE13 plays a key role in the metabolic detoxification of lambda-cyhalothrin in A. spiraecola .

Characterization of the thioredoxin peroxidase gene and its role in lambda-cyhalothrin tolerance in Agrotis ipsilon (Lepidoptera: Noctuidae)

Yu Gao,Dong-Yang Liu,Quan Deng,Wen-Long Feng,Jia-Min Yu,Mao-Ye Li,Su Liu

Exposure to insecticides can cause abnormally high accumulation of reactive oxygen species (ROS) and oxidative stress in insects. Thioredoxin peroxidases (TPXs) play critical roles in protecting insects from insecticide-induced oxidative injury. The black cutworm, Agrotis ipsilon (Hufnagel) (Lepidoptera: Noctuidae), is an important agricultural pest. Lambda-cyhalothrin (LCT) is a pyrethroid insecticide used to control A. ipsilon . However, whether LCT causes oxidative stress and whether TPXs are involved in the antioxidant defense in A. ipsilon remain unknown. In this study, a TPX gene, AiTPX6 , was identified from A. ipsilon . AiTPX6 consists of five exons and four introns and encodes a protein of 227 amino acid residues. The AiTPX6 protein has characteristic features of TPXs, including the peroxidase active site motif PVCTTE with a conserved catalytic cysteine residue. Phylogenetic analysis revealed that AiTPX6 belongs to the “1-Cys” TPX group. The mRNA transcripts of AiTPX6 were most abundant in the larval midgut and at the fifth- and sixth-instar larval stages. Exposure to a sublethal concentration (LC 50 ) of LCT significantly increased the H 2 O 2 and malondialdehyde (MDA) concentrations and elevated the mRNA level of AiTPX6 , suggesting that LCT-induced oxidative stress activates AiTPX6 transcription. Recombinant AiTPX6 expressed in Escherichia coli displayed potent antioxidant activity, protecting the cells against oxidative stress. Knockdown of AiTPX6 via RNA interference (RNAi) significantly increased the mortality rate of A. ipsilon following LCT exposure. These findings revealed that AiTPX6 contributes to LCT tolerance via antioxidant defense and could therefore be used as a potential target for RNAi-based control of A. ipsilon .

Development of biodegradable starch-based lambda-cyhalothrin microcapsules for sustainable long-term pest control and enhanced utilization efficacy

Liang Li,Long Chen,Qian Du,Xiquan Ding,Hongyan Chen,Fei Gao,Bo Cui,Haixin Cui,Zhanghua Zeng

The inefficiency of traditional pesticides leads to significant resource waste, severe environmental pollution, and potential threats to human health. Pesticide microcapsules present a promising strategy for developing environmentally friendly, safe, and sustained-release formulations. In this study, we produced degradable starch nanocrystals (SNCs) via acid hydrolysis and employed octenyl succinic anhydride-modified SNCs (O-SNCs) to fabricate pesticide microcapsules. Specifically, O-SNCs were used to emulsify molten lambda-cyhalothrin (LC), forming a Pickering emulsion. Subsequent interfacial polymerization of this emulsion yielded microcapsules, whose size can be adjusted by varying the O-SNCs content. These microcapsules exhibit high encapsulation efficiency and excellent storage stability. Furthermore, they demonstrate superior sustained pest control efficacy while significantly reducing acute toxicity to zebrafish and earthworms. This study provides valuable insights for designing sustainable, environmentally friendly, and highly effective pesticide formulations.

Metabolic functional redundancy of the CYP9A subfamily members leads to P450-mediated lambda-cyhalothrin resistance in Cydia pomonella

Pei-Rong Li,Yu Shi,Di Ju,Yu-Xi Liu,Wei Wang,Ying-Shi He,Yu-Yun Zhang,Xue-Qing Yang

BACKGROUND The evolution of insect resistance to pesticides poses a continuing threat to sustainable pest management. While much is known about the molecular mechanisms that confer resistance in model insects and few agricultural pests, far less is known about fruit pests. Field-evolved resistance to synthetic insecticides such as lambda -cyhalothrin has been widely documented in Cydia pomonella , a major invasive pest of pome fruit worldwide, and the increased production of cytochrome P450 monooxygenases (P450s) has been linked to resistance in field-evolved resistant populations. However, the underlying molecular mechanisms of P450-mediated insecticide resistance remain largely unknown . RESULTS Here we found that functional redundancy and preference of metabolism by P450s genes in the CYP9A subfamily confer resistance to lambda -cyhalothrin in Cydia pomonella . A total of four CYP9A genes, including CYP9A61 , CYP9A120 , CYP9A121 , and CYP9A122 , were identified from Cydia pomonella . Among these, CYP9A120 , CYP9A121 , and CYP9A122 were predominantly expressed in the midgut of larvae. The expression levels of these P450 genes were significantly induced by a lethal dose that would kill 10% (LD 10 ) of lambda -cyhalothrin and were overexpressed in a field-evolved lambda -cyhalothrin resistant population. Knockdown of CYP9A120 and CYP9A121 by RNA-mediated interference (RNAi) increased the susceptibility of larvae to lambda -cyhalothrin. In vitro assays demonstrated that recombinant P450s expressed in Sf9 cells can metabolize lambda -cyhalothrin, but with functional redundancy and divergence through regioselectivity of metabolism. CYP9A121 preferred to convert lambda -cyhalothrin to 2′-hydroxy- lambda -cyhalothrin, whereas CYP9A122 only generated 4′-hydroxy metabolite of lambda -cyhalothrin. Although possesses a relatively low metabolic capability, CYP9A120 balanced catalytic competence to generate both 2′- and 4′-metabolites. CONCLUSION Collectively, these results reveal that metabolic functional redundancy of three members of the CYP9A subfamily leads to P450-mediated lambda -cyhalothrin resistance in Cydia pomonella , thus representing a potential adaptive evolutionary strategy during its worldwide expansion. © 2022 Society of Chemical Industry.

Fabrication and evaluation of Lambda-Cyhalothrin nanosuspoemulsion with pH- and temperature-responsive based on polyethylene wax as carrier

Yanmin Huang,Xiangying Li,Dongmei Mo,Haiyan Yuan,Qiang Hu,Yang Jiang,Chunfang Gan,Yong Chen,Weiguo Li,Rui Lu,Jianguo Cui

Using polyethylene wax (PW) as the coating matrix, the lambda-cyhalothrin-PW nanosuspoemulsion (LC-PW) with a particle size of 80-150nm was prepared through high-speed stirring, hot melt emulsification and ultrasonic dispersion. The formulation and composition of the LC-PW were optimized, the morphology of the LC-PW was analyzed by dynamic light scattering (DLS) and TEM, and the structure of the LC-PW was characterized by UV and IR. The anti-photolysis test showed that LC-PW had a good anti-photolysis performance. Furthermore, LC-PW could sustainably release Lambda-cyhalothrin, which was pH- and temperature dependent. The insecticidal activity analysis in the greenhouse indicated that the toxic strength between LC-PW and LC-SC (lambda-cyhalothrin-suspension concentrate) to Mythimna separata was similar within the same concentration ranges tested, but the insecticidal duration of LC-PW was significantly longer than LC-SC. Thus, the new type of LC-PW with the properties of anti-photolysis and controlled release is suitable for application in the field as a better insecticide.
