Electrochemical biosensor using SnO2 colloidal quantum wire for monitoring the interaction of microcystin antigen–antibody

Qing Huang,RuiQin Gu,Yunong Zhao,Huibing Fu,Huan Liu

Electrochemical sensors that incorporate immunoassay principles have the ability to monitor dynamic processes of antigen–antibody interactions in real time. In this study, a gold electrode was modified with tin dioxide colloidal quantum wire (SnO 2 QWs) and then coated with the leucine/arginine subtype microcystin (MC-LR) antibody. The active site of SnO 2 QWs that was not bound by MC-LR antibody was then passivated with bovine serum protein (BSA). When the MC-LR antigen binds specifically to the antibodies on the electrode's surface, it triggers electrochemical reactions and generates electrical signals at specific voltage conditions. The SnO 2 QW exhibits excellent electron transport ability, and its ability to form a loose and porous microstructure on the gold electrode surface, which is conducive to the receptor function of the biosensor. The results show a high affinity between the MC-LR antigen and antibody, ranging from 1 pg/mL to 10 ng/mL of MC-LR antigen concentration. The kinetic characteristics of the immune reaction between MC-LR antigen and antibody were elucidated, obtaining a binding constant of 1.399 × 10 11 M −1 and a dissociation constant of 7.147 pM, demonstrating the potential of electrochemical biosensing technology in biomolecular interactions.

Congener-specific fate and impact of microcystins in the soil-earthworm system

Bai-Lin Liu,Peng-Fei Yu,Jing-Jie Guo,Li-Si Xie,Xiang Liu,Yan-Wen Li,Lei Xiang,Hai-Ming Zhao,Nai-Xian Feng,Quan-Ying Cai,Ce-Hui Mo,Qing X. Li

Microcystins (MCs) have a significant influence on aquatic ecosystems, but little is known about their terrestrial fate and impact. Here, we investigated the fate of two MCs (MC-LR and MC-RR) in the soil-earthworm system, with consideration of their congener-specific impact on earthworm health, soil bacteria, and soil metabolome. Although MCs had little acute lethal effect on earthworms, they caused obvious growth inhibition and setae rupture. Relative to MC-RR, MC-LR exhibited higher bioaccumulation and the resulting dermal lesions and deformation of longitudinal muscles. While the incorporation of both MCs into soils stimulated pathogenic bacteria and depressed oxidative stress tolerant bacteria, the response among soil nitrification and glutathione metabolism differed between the two congeners. The dissipation kinetics of MCs obeyed the first-order model. Earthworms stimulated soil N-cycling enzyme activities, increased the abundance of MC-degrading bacteria, and promoted bacterial metabolic functions related to glutathione metabolism, xenobiotics biodegradation, and metabolism of amino acids that comprise MCs, which accelerated the dissipation of MC-LR and MC-RR by 227% and 82%, respectively. These results provide evidence of significant congener differences in the terrestrial fate and impact of MCs, which will enable a better understanding of their role in mediating soil functions and ecosystem services.

Electrochemical elimination of Microcystis aeruginosa with boron-doped diamond anode in different electrolyte systems: chemical and biological mechanisms

Long Yujiao,Li Hongna,Jin Hongmei,Ni Jinren

The chemical and biological mechanisms of electrochemical elimination of Microcystis aeruginosa ( M. aeruginosa ) using boron-doped diamond (BDD) anode were comparatively explored in three different electrolytes (chloride, sulfate, and phosphate solutions). The most efficient elimination of M. aeruginosa was observed in chloride solution, which was attributed to the greatest total long-lived oxidants from the favorable formation of active chlorine. Moreover, the high permeability of active chlorine resulted in profound intracellular damages to chlorophyll- a , microcystin-LR (MC-LR), superoxide dismutase (SOD) enzyme, and DNA in the chloride system. The change of membrane permeability and degradation of the released MC-LR induced by active chlorine were further confirmed by the increase of extracellular MC-LR in the initial 5 min and a complete decay in the subsequent 15 min, while the change in morphology of algae cells was insignificant from SEM images. In sulfate and phosphate electrolytes, membrane damages were much more pronounced based on lipid peroxidation observation, although changes in cell morphology was found more significant in phosphate system. The higher concentrations of oxidants (·OH, O 3 , H 2 O 2 , S 2 O 8 2− ) generated in sulfate than in phosphate solution explained the greater efficiency of electrochemical elimination of M. aeruginosa in the sulfate electrolyte in terms of changes of cell density, OD 680 , chlorophyll- a , MC-LR, lipids, SOD enzyme, and DNA.

Elucidating environmental factors and their combined effects on CKDu in Sri Lanka using zebrafish

Pan-Pan Jia,Rohana Chandrajith,Muhammad Junaid,Tian-Yun Li,Yong-Zhi Li,Xing-Yi Wei,Li Liu,De-Sheng Pei

Chronic kidney disease with uncertain etiology (CKDu) in Sri Lanka has attracted much attention as a global health issue. However, how environmental factors in local drinking water induce kidney damage in organisms is still elusive. We investigated multiple environmental factors including water hardness and fluoride (HF), heavy metals (HM), microcystin-LR (MC-LR), and their combined exposure (HFMM) to elucidate their toxic effects on CKDu risk in zebrafish. Acute exposure affected renal development and inhibited the fluorescence of Na, K-ATPase alpha1A4:GFP zebrafish kidney. Chronic exposure influenced the body weight of both genders of adult fish and induced kidney damage by histopathological analyses. Furthermore, the exposure significantly disturbed differential expression genes (DEGs), diversity and richness of gut microbiota, and critical metabolites related to renal functions. The transcriptomic analysis revealed that kidney-related DEGs were linked with renal cell carcinoma, proximal tubule bicarbonate reclamation, calcium signaling pathway, and HIF-1 signaling pathway. The significantly disrupted intestinal microbiota was closely related to the environmental factors and H&E score, which demonstrated the mechanisms of kidney risks. Notably, the Spearman correlation analysis indicated that the changed bacteria such as Pseudomonas , Paracoccus , and ZOR0006 , etc were significantly connected to the DEGs and metabolites. Therefore, the assessment of multiple environmental factors provided new insights on “bio-markers” as potential therapies of the target signaling pathways, metabolites, and gut bacteria to monitor or protect residents from CKDu.
