Flaxseed extract induces apoptosis in human breast cancer MCF-7 cells

Tingyan Hu,Kegang Linghu,Siqi Huang,Maurizio Battino,Milen I. Georgiev,Gokhan Zengin,Defang Li,Yun Deng,Y.T. Wang,Hui Cao

Significant evidence indicated that flaxseed ( Linum usitatissimum ) possesses various positive health aspects such as reducing the risk of cancer and cardiovascular diseases. The fatty acids are considered to be responsible for these benefits of flaxseed. Herein, the in vitro effects of flaxseed extract on the growth and apoptosis of human breast cancer MCF-7 cells were investigated. The MCF-7 cells treated with flaxseed extract showed a dose-dependent decrease in cell viability. The flaxseed extract induced reactive oxygen species and the flow cytometric analysis demonstrated that flaxseed fatty acids triggered apoptosis of MCF-7 cells, which was also shown by the loss of mitochondrial membrane potential and caspase cascade reaction. Thus, the flaxseed extract regulated the growth of MCF-7 cells and induced apoptosis. Eventually, the flaxseed could be used as a dietary supplement to prevent breast cancer.

Kai Dong,Zhuang-Zhuang Zhao,Jian Kang,Lei-Ruo Lin,Wen-Ting Chen,Jin-Xi Liu,Xiang-Long Wu,Ting-Li Lu

Background Combined chemotherapy is often affected by the different physicochemical properties of chemotherapeutic drugs, which should be improved by the reasonable design of co-loaded preparations.Purpose A kind of simple but practical graphene oxide (GO) wrapped mesoporous silica nanoparticles (MSN) modified with hyaluronic acid (MSN@GO-HA) were developed for the co-delivery of cinnamaldehyde (CA) and doxorubicin (DOX), in order to enhance their combined treatment on tumor cells and reduce their application defects. Methods The MSNCA@GODOX-HA was constructed by MSNCA (loading CA via physical diffusion) and GODOX-HA (modified with HA and loading DOX via π–π stacking) through the electrostatic adsorption, followed by the physicochemical characterization, serum stability and in vitro release study. Cytotoxicity on different cells was detected, followed by the tumor cell uptake tests. The intracellular reactive oxygen species (ROS) changes, mitochondrial functions and activities of caspase-3/-9 in MCF-7 cells were also evaluated, respectively. Results The MSNCA@GODOX-HA nanoparticles kept stable in FBS solution and achieved pH-responsive release behavior, which was beneficial to increase the accumulation of CA and DOX in tumor cells to enhance the treatment. MSNCA@GODOX-HA exerted higher cytotoxicity to MCF-7 human breast cancer cells than H9c2 cardiac myocyte cells, which were not only attributed to the active targeting to tumor cells by HA, but also related with the activation of intrinsic apoptotic pathway in MCF-7 cells induced by CA, which was mediated by the specific ROS signal amplification and the interference with mitochondrial function. Moreover, the efficacy of DOX was also enhanced by the above process. Conclusion The establishment of the MSNCA@GODOX-HA nanoparticles played a role in promoting strengths and restricting shortcomings of CA and DOX, thereby exerting their function and achieving efficient treatment against cancer.

Screening Multidrug Resistance Reversal Agents in Traditional Chinese Medicines by Efflux Kinetics of D-Luciferin in MCF-7/DOXFluc Cells

Yue Zhao,Chaoyuan Tang,Jingyi Huang,Hongyan Zhang,Jingbin Shi,Shujun Xu,Lisha Ma,Chun Peng,Qi Liu,Yang Xiong

In this study, we established a simple and rapid in vitro method for screening multidrug resistance (MDR) reversal agents in traditional Chinese medicines (TCMs), which could better correspond to the MDR reversing effect in vivo. Here, D-luciferin, a substrate for the enzyme firefly luciferase and also a substrate for ATP-binding cassette transporters (ABC transporters), was used as the probe to detect its efflux kinetics caused by ABC transporters. First, we established a stable doxorubicin (DOX)-resistant cell line (MCF-7/DOXFluc) that overexpressed luciferase. Then, some kinds of TCMs were chosen for the MDR reversal agents to measure its effect on inhibiting the D-luciferin outflow from MCF-7/DOXFluc, and the ideal reversal agent with the least D-luciferin efflux from MCF-7/DOXFluc was selected to further investigate its effect combined with DOX on MCF-7/DOXFluc tumor-bearing mice. The results indicated that quercetin (Qu) could remarkably increase the retention of D-luciferin in MCF-7/DOXFlucin vitro and in vivo. Also, the combination of Qu and DOX could exceedingly inhibit the tumor growth, which proved the feasibility of this in vitro screening method. The study proposed a feasible method for mass screening of MDR agents from TCMs in vitro.

Intracellular release of PluronicL64 unimers into MCF-7/ADR cells to overcome multidrug resistance by surface-modified PAMAM

Mengjun Zhang,Shasha Jing,Jie Zhang,Jiulong Zhang,Xinlong Zang,Mingxi Qiao,Xiuli Zhao,Haiyang Hu,Dawei Chen

Multidrug resistance (MDR) has been a major obstacle to tumor chemotherapy. Pluronic unimers have been reported to be promising copolymers to reverse MDR, and the intracellular delivery of Pluronic unimers is a problem worth thinking. To exert the excellent reversal effect of Pluronic unimers, DOX-loaded G4.0 PAMAM was modified with PluronicL64 via cis-aconitic acid as a pH-sensitive linkage (PCPAMAM/DOX), which could release DOX and Pluronic unimers into cytoplasm. The Pluronic-modified PAMAM (PCPAMAM) exhibited favorable biocompatibility and pH-sensitivity. PCPAMAM/DOX showed a nano-scale size and a sustained in vitro release profile. Compared with a control formulation, PCPAMAM/DOX showed a higher reversal effect on MCF-7/ADR cells and enhanced intracellular drug accumulation. The results of P-gp activity, subcellular distribution of PluronicL64, the ATP level and mitochondrial transmembrane potential all illustrated that free Pluronic unimers could be released by PCPAMAM functioning as reversal agents. In conclusion, PCPAMAM could be a promising vehicle to enhance DOX accumulation by overcoming MDR in MCF-7/ADR cells. This work also provided an effective method to deliver Pluronic unimers into MDR cells.

An electrochemiluminescence lab-on-paper device for sensitive detection of two antigens at the MCF-7 cell surface based on porous bimetallic AuPd nanoparticles

Min Su,Haiyun Liu,Shenguang Ge,Na Ren,Longhua Ding,Jinghua Yu,Xianrang Song

In this work, a novel paper-based electrochemiluminescence (ECL) immunosensor for ultrasensitive and selective detection of corresponding antigens on living cells was developed employing AuPd nanoparticles as a signal amplifier and macroporous Au-paper working electrodes (Au-PWE) for efficient cell capture. The porous AuPd nanoparticles are chosen as label nanomaterials due to their controllable three-dimensional porosity and peroxidase like activity, large surface area, superior conductivity, and favorable catalytic activity to H2O2 which could enhance the ECL intensity for signal amplification. CdTe QDs and luminol groups as ECL probes were linked with the antibodies to recognize the corresponding antigens on the cell surface and the wide potential range between them compared to conventional antigens sensors could prevent interfacing with each other. Using this strategy, human carcinoembryonic (CEA) and alphafetoprotein (AFP) antigens on living MCF-7 cancer cells as models were quantified serially through a potential scanning way. Significantly, this strategy may be quite promising in cancer early diagnosis due to the excellent analytical performance.

A cytosensor based on NiO nanoparticle-enhanced surface plasmon resonance for detection of the breast cancer cell line MCF-7

Jia Shengsong,Li Pengfei,Koh Kwangnak,Chen Hongxia

The article describes the use of a designed histidine (His)-tagged arginine-glycine-aspartic acid (RGD) peptide as a linker to bind integrin to captured cancer cells, and how to amplify the surface plasmon resonance (SPR) signal after binding of NiO nanoparticles (NiO-NPs) via the His-tag on the peptide. Specifically, breast cancer cells were captured via the interaction between human mucin (MUC-1) and a gold surface modified with a MUC-selective aptamer. The resulting cytosensor exhibits specificity and high sensitivity which is due to the enhancement of the SPR signal by NiO-NPs. The breast cancer cell line MCF-7 can be easily distinguished from normal islet beta cells by using this biosensor. Implementation of the His-tagged RGD peptide modified with NiO-NPs resulted in 20-fold enhancement of the SPR signal at the limit of detection. Hence, the actual limit of detection is lowered to 136 cells per mL. In our perception, this cytosensor has a large clinical potential in that it may also be used to detect various other kinds of tumor cells. Histidine-tagged arginine-glycine-aspartic acid peptide carried by NiO interacts with the integrin of cancer cells and amplifies the SPR signal resulting from interaction between human mucin (MUC-1) and a gold surface modified with a MUC-selective aptamer. As little as 136 cells·mL −1 are detectable.

Effect of H2O2 induced oxidative stress (OS) on volatile organic compounds (VOCs) and intracellular metabolism in MCF-7 breast cancer cells

Yuanling Liu,Wenwen Li,Yixiang Duan

Oxidative stress (OS) refers to the process of oxidative damage caused by reactive oxygen species (ROS) and reactive nitrogen species (RNS) accumulated by the organisms or cells during the process of generating and scavenging oxygen free radicals. In this study, an OS model of MCF-7 cells was constructed through H2O2-treatment, which for the first time combined volatile organic compounds (VOCs) and intracellular metabolic analysis for a comprehensive and in-depth study of the OS effect on cell metabolism. The VOCs produced by cells in H2O2-treated groups and control groups were extracted with solid-phase microextraction (SPME) and analyzed by gas chromatography-mass spectrometer (GC-MS). Meanwhile, the intracellular metabolites were extracted by methanol and analyzed by GC-MS coupled with silyl-derivatization. The results indicated 15 VOCs and 29 intracellular metabolites were statistically different (p < 0.05) between the H2O2-treated groups and the control groups. The VOC biomarkers were mainly linear chain alkanes and branched chain alkanes, and they were up-regulated in cells treated by H2O2. The intracellular metabolites with significant changes were mainly fatty acids, amino acids, and organic acids. Most of them, however, were down-regulated under OS state. This study revealed volatile and non-volatile metabolites that influenced by elevated OS in MCF-7 cells, and provided clues for mechanism exploration of exhaled biomarkers, thus promoting the non-invasive prediction of BC.

Ultrasound-mediated destruction of vascular endothelial growth factor (VEGF) targeted and paclitaxel loaded microbubbles for inhibition of human breast cancer cell MCF-7 proliferation

Jilian Su,Junmei Wang,Jiamin Luo,Haili Li

Aims Vascular endothelial growth factor (VEGF) can promote cell division, proliferation and migration. In this study, we aimed to investigate roles of ultrasound-mediated destruction of VEGF-targeted and paclitaxel (PTX)-loaded lipid microbubbles (VTPLLM + US) in human breast cancer cells . Methods The activity of MCF-7 cells was determined by cell counting Kit-8. Flow cytometry was performed to detect the cells apoptosis and cell cycle. The expression of cell cycle-associated proteins, matrix metalloprotein-9 (MMP-9), VEGF and apoptosis-associated proteins were detected by qRT-PCR and Western blot . Results The obtained data suggested that VTPLLM + US promoted the G1 phase cell cycle arrest and suppressed the viability of MCF-7 cells. We also found that VTPLLM + US accelerated cells apoptosis. Cell cycle-associated proteins and VEGF expression were modulated by VTPLLM + US. Moreover, VTPLLM + US was found to regulate the expression levels of apoptosis-associated proteins in MCF-7 cells. Our findings suggested that VTPLLM + US suppressed the proliferation and accelerated the apoptosis of MCF-7 cells through regulating VEGF expression. Conclusions The potential effects of VTPLLM + US on apoptosis of MCF-7 cells suggest that applying VTPLLM + US might be an effective strategy in breast cancer therapies.

Hyaluronic acid and cholecalciferol conjugate based nanomicelles: Synthesis, characterization, and cytotoxicity against MCF-7 breast cancer cells

Ning Gao,Yuhan Fu,Hexin Gong,Haotian Liu,Weinan Li

Recently, growing and conclusive evidences showed the particularly close ties between the vitamin D (VD) and breast cancer (BC). However, few researches were focused on the delivery system against breast cancer which based on cholecalciferol (VD 3 ). Herein, we designed a series of self-assembled micelles. The conjugate (HA-VD) of VD 3 with hyaluronic acid (HA) actively targeting breast cancer can efficiently delivery doxorubicin (DOX). The HA-VD conjugate was successfully synthesized, which was confirmed using 1 H nuclear magnetic resonance (NMR) and Fourier transform infrared (FITR). Preliminary studies of its physical and chemical properties indicated that VD 3 was successfully grafted with the HA (HAVD). When the molar ratio was 1:2 and the degree of substitution (DS) was 18.6%, the minimum critical micelle concentration (CMC) value was 0.0137 mg/mL. For DOX-loaded HAVD micelles (DOX/HAVD), the drug loading (DL) was 6.2% and drug encapsulation efficiency (EE) was 79.5%. The DOX/HAVD micelles remain stable in serum for 48 h. The in vitro release rate of DOX decreased after encapsulating compared with that without HAVD encapsulation. The decrease with IC 50 of DOX/HAVD micelle was significantly stronger than that of free DOX ( p  < 0.05) and blank micelles ( p  < 0.01). At 50% cell inhibition rate, VD 3 and DOX acted synergistically on BC cells. Hence, the HAVD micelles can significantly improve the therapeutic effect of DOX against BC cells. In a word, the DOX/HAVD micelles have great potential in treating breast cancer.

Biodegradable Polymersomes as Nanocarriers for Doxorubicin Hydrochloride: Enhanced Cytotoxicity in MCF-7/ADR Cells and Prolonged Blood Circulation

Chao Yanhui,Liang Yuheng,Fang Guihua,He Haibing,Yao Qing,Xu Hang,Chen Yinrong,Tang Xing

Purpose DOX is one of the most potent anticancer drugs. But its short half-life and the occurrence of multi-drug resistance (MDR) markedly limit its clinical application. To solve these problems, we develop DOX loaded polymersomes (DOX polymersomes). Methods An methoxy poly(ethylene glycol)-b-poly(epsilon-caprolactone) (mPEG-b-PCL) copolymer was synthesized and used to prepare DOX polymersomes. The pharmaceutical properties of DOX polymersomes were characterized. The in vitro release profile of DOX from polymersomes was investigated. The in vitro cytotoxicity and cell uptake studies were performed on MCF-7 and MCF-7/ADR cells. The in vivo pharmacokinetic profiles were investigated on Sprague–Dawley rats. Results DOX polymersomes had a nano-scale particle size of about 60 nm with a hydrophobic membrane about 10 nm in thickness. Release of DOX from the polymersomes took place in a sustained manner. Cell experiments showed DOX polymersomes enhanced the cytotoxicity and the intracellular accumulation of DOX in MCF-7/ADR cells, compared with free DOX. In vivo pharmacokinetic study showed the DOX polymersomes increased the bioavailability and prolonged the circulation time in rats. Conclusions The entrapment of DOX in biodegradable polymersomes could enhance cytotoxicity in MCF-7/ADR cells and improve its in vivo pharmacokinetic profile.

Nanodiamond mediated co-delivery of doxorubicin and malaridine to maximize synergistic anti-tumor effects on multi-drug resistant MCF-7/ADR cells

Hao Zhu,Yun Wang,Abid Hussain,Zhipeng Zhang,Yuanyuan Shen,Shengrong Guo

Co-delivery of two or more chemotherapeutic agents via nanocarriers can provide an effective approach to maximize synergistic or additive effects. In this work, we developed novel nanodiamond (ND) based nanoparticle co-loading of doxorubicin (DOX) and malaridine (MAL) for treatment of MCF-7/ADR cells by virtue of the good biocompatibility, high drug loading capacity and pH-responsive drug release properties of NDs. The combination index (CI) was the smallest, 0.17, for DOX and MAL against MCF-7/ADR cells, when their ratio was 5 : 1, showing the greatest synergistic effects. (DOX + MAL)@NDs or FA-PEG–DOX/(DOX + MAL)@NDs with different ratios of DOX to MAL (4 : 1, 5 : 1 and 6 : 1) were prepared by simple physical adsorption of DOX, MAL and FA-PEG–DOX. Among them, (DOX + MAL)@NDs with the mass ratio of 5 : 1 had the smallest CI of 0.08. The concentrations of DOX and MAL for (DOX + MAL)@NDs producing 50% growth inhibition were 1.90 and 0.35 μg mL−1, respectively, smaller than the 50% inhibitory concentrations for DOX@NDs and MAL@NDs (10.75 and 4.38 μg mL−1) and for free DOX and free MAL (49.65 and 9.03 μg mL−1). (DOX + MAL)@NDs were not stable in water, FA-PEG–DOX/(DOX + MAL)@NDs could stay stable in water for 24 h due to steric repulsion effects offered by PEG, which also compromised their cytotoxicities to some extent. (DOX + MAL)@NDs or FA-PEG–DOX/(DOX + MAL)@NDs presented faster DOX and MAL co-release at pH 5.5 than 7.4. The high anti-tumor efficacy suggested that the ND mediated DOX and MAL co-delivery system could be promising for the treatment of multi-drug resistant cancer.

MCF-7 cell - derived exosomes were involved in protecting source cells from the damage caused by tributyltin chloride via transport function

Yiren Xiong,Guoqiang Guo,Hongyi Xian,Zuqing Hu,Di Ouyang,Jiayi He,Shanshan He,Renyi Liu,Zhenjie Gao,Meilin Tang,Ying Chen,Suqin Tan,Xiaoqi Zhu,Abudumijiti Abulimiti,Sujin Zheng,Hehai Huang,Dalin Hu

Tributyltin chloride (TBTC) is a ubiquitous environmental pollutant with various adverse effects on human health. Exosomes are cell - derived signaling and substance transport vesicles. This investigation aimed to explore whether exosomes could impact the toxic effects caused by TBTC via their transport function. Cytotoxicity, DNA and chromosome damage caused by TBTC on MCF-7 cells were analyzed with CCK-8, flow cytometry, comet assay and micronucleus tests, respectively. Exosomal characterization and quantitative analysis were performed with ultracentrifugation, transmission electron microscope (TEM) and bicinchoninic acid (BCA) methods. TBTC content in exosomes was detected with Liquid Chromatography-Mass Spectrometry (LC-MS). The impacts of exosomal secretion on the toxic effects of TBTC were analyzed. Our data indicated that TBTC caused significant cytotoxicity, DNA and chromosome damage effects on MCF-7 cells, and a significantly increased exosomal secretion. Importantly, TBTC could be transported out of MCF-7 cells by exosomes. Further, when exosomal secretion was blocked with GW4869, the toxic effects of TBTC were significantly exacerbated. We concluded that TBTC promoted exosomal secretion, which in turn transported TBTC out of the source cells to alleviate its toxic effects. This investigation provided a novel insight into the role and mechanism of exosomal release under TBTC stress.
