Summary The effect of carnauba wax-based nanoemulsion coating-1 (NC-1) combined with 1-methylcyclopropene (1-MCP) treatment on the storage of honey peach fruit ‘Yulupantao’ for 28 days under 0 ± 0.5 °C, and 90% RH conditions was studied. The results showed that NC-1 or 1-MCP treatment alone and in combination were proved to be more effective in maintaining fruit qualities compared to the control group with no treatment. However, the application with the combination of NC-1 and 1-MCP treatment had the most significant effect on reducing mass loss and decay rate, maintaining firmness, total soluble solids (TSS), ascorbic acid (AsA) and titratable acid (TA) contents, suppressing malondialdehyde (MDA) accumulation and conductivity ascend. Moreover, the activities of peroxidase (POD) were maintained, and polyphenol oxidase (PPO) and phenylalanine ammonialyase (PAL) were effectively suppressed in NC-1, 1-MCP and combined treatment than that of control. Therefore, the combination of NC-1 and 1-MCP treatment could be an efficient way to maintain the qualities of peach fruit during low-temperature storage after harvest.

As a worldwide health issue, obesity is associated with the infiltration of monocytes/macrophages into the adipose tissue causing unresolved inflammation. Monocyte chemoattractant protein-1 (MCP-1) exerts a crucial effect on obesity-related monocytes/macrophages infiltration. Clinically, aspirin and salsalate are beneficial for the treatment of metabolic diseases in which adipose tissue inflammation plays an essential role. Herein, we investigated the effect and precise mechanism of their active metabolite salicylate on TNF-α-elevated MCP-1 in adipocytes. The results indicated that salicylate sodium (SAS) could lower the level of MCP-1 in TNF-α-stimulated adipocytes, which resulted from a previously unrecognized target phosphodiesterase (PDE), 3B (PDE3B), rather than its known targets IKKβ and AMPK. The SAS directly bound to the PDE3B to inactivate it, thus elevating the intracellular cAMP level and activating PKA. Subsequently, the expression of MKP-1 was increased, which led to the decrease in p-EKR and p-p38. Both PDE3B silencing and the pharmacological inhibition of cAMP/PKA compromised the suppressive effect of SAS on MCP-1. In addition to PDE3B, the PDE3A and PDE4B activity was also inhibited by SAS. Our findings identify a previously unrecognized pathway through which SAS is capable of attenuating the inflammation of adipocytes.Keywords:cAMP;MKP-1;p-ERK;p-p38;metaflammation

Carbon dioxide (CO2) emission and fresh water scarcity have become two huge global issues over the last decades, and CO2 hydrate-based technology was promoted and developed to address them. This study investigated the effects of methylcyclopentane (MCP) on the hydrate formation condition and CO2 capture efficiency. Five cases and 31 cycles experiments of different MCP/water molar ratios (ranging from 0 to 0.03) in salt solution and porous media were conducted using the isochoric method. The hydrate phase equilibrium data of CO2–MCP hydrates were measured, and meanwhile the CO2 capture efficiencies were calculated. The results indicated that the rising MCP concentrations increase both the hydrate equilibrium temperature and the gas uptake. After the comparisons of the interpolated hydrate gas uptakes at uniform hydrate phase equilibrium pressure of 3 MPa among CO2–MCP, CO2–CP (cyclopentane), and CO2–C3H8 (propane) hydrates, CO2-MCP hydrates are especially suitable for CO2 capture. It should be noted that MCP is likely to form s-H hydrate with CO2 gas, but further confirmation is needed. Anyway, MCP can be thought as a potential coguest for integrated hydrate-based CO2 capture and desalination process.

The effects of 1-methylcyclopropene (1-MCP) treatment, storage time, and shelf life and temperature on phenolic concentrations and antioxidant activity in apple peel and flesh of ‘Jonagold’ apple ( Malus domestica Borkh) have been investigated. Hyperoside and chlorogenic acid were the most abundant phenolic compounds in the peel and flesh, respectively. The flesh had lower concentrations of individual phenolics, total phenolics, flavonoids and lower antioxidant activity (determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay) than the peel. In addition, the profiles of phenolics as well as the changing patterns of individual phenolics also differed between the peel and the flesh. After long-time storage (180 d), the greatest decrease of concentrations was the concentrations of hyperoside (27 %) in the peel and total phenolics (35 %) in the flesh. Also, during the 90–180 d storage period, hyperoside and rutin in the peel, chlorogenic acid in the flesh, and flavonoids and antioxidant activity decreased in both peel and flesh tissues. Individual phenolics exhibited different stabilities during the early stages after harvest. 1-MCP reduced the reduction in the concentrations of hyperoside in the peel, chlorogenic acid in the flesh, and total phenolics, flavonoids and antioxidant activity in both the peel and flesh tissues during storage and shelf life. However, for quercetin and rutin, the protective effect of 1-MCP was only observed during the shelf life period. Overall, phenolic compounds responded variously to storage, shelf life, shelf temperature and 1-MCP treatment.

Nanoemulsion has been shown to extend the postharvest life of horticultural produce. In this study, “Fuji” apple were treated with the combination of nanoemulsion coating-1 (NC-1) and 1-methylcyclopropene (1-MCP). Biochemical and nutritional values were assessed every 35 days during 280 days storage at 4°C and 90% RH. Results show that apples treated with the combination of NC-1 and 1-MCP has significantly reduced mass loss and decay, suppressed malondialdehyde (MDA) content. The combination treatment of NC-1 and 1-MCP also retained significantly higher ascorbic acid (ASC), titratable acid (TA), and total soluble solid (TSS) content. NC-1 and 1-MCP treatments alone or in combination produced higher peroxidase (POD) and catalase (CAT) activities but lower phenylalanine ammonia-lyase (PAL) and polyphenol oxidase (PPO) activities than control. Therefore, the combination of NC-1 and 1-MCP preserved better “Fuji” apple quality, which could be used to extend the postharvest life of “Fuji” apple during long-term storage. Practical applications This research work shows that the combined treatment of NC-1 and 1-MCP on “Fuji” apple reduced mass loss, decay and MDA level, retained higher ASC, TA, and TSS content, promoted POD and CAT activities but suppressed PAL and PPO activities, by which preserved better “Fuji” apple qualities during long-term storage. Moreover, the combination of NC-1 and 1-MCP could provide a potential alternative to extend the storage life, which helps in reducing postharvest losses in the supply chain.

Wolfberry, esteemed as a traditional Chinese medicinal material and functional food, is replete with nutrients and boasts a diverse array of health benefits, including hypoglycemic, antitumor, antioxidant, anti-inflammatory, and immune-enhancing properties. Notably, inflammation is a pivotal factor in the onset and progression of numerous diseases. Despite this, there is a paucity of research on the comprehensive evaluation of the components found in different wolfberries, and the exploration of their primary active components is limited. To address this issue, we conducted a comprehensive targeted metabolomics analysis, employing statistical methods such as principal component analysis (PCA), orthogonal partial least squares discriminant analysis (OPLS-DA), KEGG pathway analysis, and volcano plots to delineate the compositional differences among red, black, and yellow wolfberries. Furthermore, we investigated the anti-inflammatory effects of their primary components through in vitro experiments. Our analysis revealed a total of 1,104 chemical compositions in the three wolfberries, with alkaloids, phenolic acids, flavonoids, and lipids being the predominant nutritional components. KEGG enrichment analysis indicated that these compositions were primarily involved in the biosynthesis of secondary metabolites, ABC transport, and galactose metabolism pathway. Moreover, our study demonstrated that quercetin exhibited dose-dependent anti-inflammatory activity in LPS-stimulated HUVECs. It effectively inhibited the production of inflammatory factors such as TNF-α, MCP-1, and IL-1β, while also down-regulating the gene and protein expression levels of ICAM-1 and VCAM-1. In conclusion, our findings indicate that there are variations in compositions among the three wolfberries, with flavonoids being the most abundant, and in vitro studies also confirmed the anti-inflammatory potential of quercetin. It is worth noting that Lycium ruthenicum contains higher levels of antioxidant components and possesses greater nutritional value, providing valuable insights for the future development and utilization of the three wolfberries.

The effects of storage conditions and cooking methods on chlorophyll, glucosinolate (GSL), and sulforaphane content in broccoli florets were investigated in this study. For the storage experiment, fresh broccoli florets were stored for 2, 4, and 6 days at 10, 4, and 0 °C with or without 1-methylcyclopropene (1-MCP) treatment. For the cooking experiment, fresh broccoli florets were cooked for 1, 3, 5, 7, and 9 min under three cooking methods, namely, steaming, microwaving, and boiling. Results showed that the contents of chlorophyll, aliphatic GSL, indole GSL, and sulforaphane in broccoli florets of two cultivars decreased with prolonged storage time. The retained contents of chlorophyll, GSLs, and sulforaphane under 0 °C storage condition were significantly higher than those under 10 °C storage condition after 6 days of storage. The sulforaphane content was increased by 1-MCP treatment but differed among varieties. The cooking experiment showed that aliphatic GSL content decreased with increased cooking time under three cooking methods, and indole GSL and sulforaphane contents had a fluctuating trend with increasing cooking time after steaming and microwaving. Sulforaphane content increased by 17.15–50.16% relative to that in fresh broccoli florets and was considerably affected by cooking time. The highest level of sulforaphane content was retained for 7 min during steaming or 5 min during microwaving. Therefore, the combination of 1-MCP treatment and 0 °C storage condition had the best performance in preserving chlorophyll, GSLs, and sulforaphane. Moreover, steaming for 7 min or microwaving for 5 min is a more effective method for preserving the quality and increasing the sulforaphane content of broccoli florets than boiling. Keywords:broccoli;storage;cooking;glucosinolate;sulforaphane

In this study, two homogeneous polysaccharides (APS-A1 and APS-B1) were isolated from Astragalus membranaceus by DEAE-52 cellulose and Sephadex G-100 column chromatography. Their chemical structures were characterized by molecular weight distribution, monosaccharide composition, infrared spectrum, methylation analysis, and NMR. The results revealed that APS-A1 (2.62 × 10 6 Da) was a 1,4-α-D-Glc p backbone with a 1,4,6-α-D-Glc p branch every ten residues. APS-B1 (4.95 × 10 6 Da) was a heteropolysaccharide composed of glucose, galactose, and arabinose (75.24:17.27:19.35). Its backbone consisted of 1,4-α-D-Glc p , 1,4,6-α-D-Glc p , 1,5-α-L-Ara f and the sidechains composed of 1,6-α-D-Gal p and T-α/β-Glc p . Bioactivity assays showed that APS-A1 and APS-B1 had potential anti-inflammatory activity. They could inhibit the production of inflammatory factors (TNF-α, IL-6, and MCP-1) in LPS-stimulated RAW264.7 macrophages via NF-κB and MAPK (ERK, JNK) pathways. These results suggested that the two polysaccharides could be potential anti-inflammatory supplements.

Objective In this study, we investigated the effect and mechanism of action of eugenol on oxidized low-density lipoprotein (ox-LDL)-induced abnormal proliferation and migration of human vascular smooth muscle cells (HVSMCs).Methods HVSMCs were treated with 100 ug/mL ox-LDL for 24 hours to establish a cell model. After 1-hour pretreatment, eugenol at concentrations of 5, 25, and 50 uM was added. Cell viability was assessed using an MTT assay, PCNA expression was detected using Western blot, cell cycle distribution was analyzed using flow cytometry, and cell migration ability was evaluated using wound healing and Transwell migration assays. To investigate the mechanisms, Ang II receptors were inhibited by 1000 nM valsartan, MFG-E8 was knocked down by shRNA, MCP-1 was inhibited by siRNA, and MFG-E8 was overexpressed using plasmids. Results The findings from this study elucidated the stimulatory impact of ox-LDL on the proliferation and functionality of HVSMCs. Different concentrations of eugenol effectively mitigated the enhanced activity of HVSMCs induced by ox-LDL, with 50 uM eugenol exhibiting the most pronounced inhibitory effect. Flow cytometry and Western blot results showed ox-LDL reduced G1 phase cells and increased PCNA expression, while 50 uM eugenol inhibited ox-LDL-induced HVSMC proliferation. In wound healing and Transwell migration experiments, the ox-LDL group showed larger cell scratch filling and migration than the control group, both of which were inhibited by 50 uM eugenol. Inhibiting the Ang II/MFG-E8/MCP-1 signaling cascade mimicked eugenol’s effects, while MFG-E8 overexpression reversed eugenol’s inhibitory effect. Conclusion Eugenol can inhibit the proliferation and migration of ox-LDL-induced HVSMCs by inhibiting Ang II/MFG-E8/MCP-1 signaling cascade, making it a potential therapeutic drug for atherosclerosis.

Solubility enhancement has been a priority to overcome poor solubility with optoelectronic molecules for solution-processable devices. This study aims to obtain experimental data on the effect of particle sizes on the solubility properties of several typical optoelectronic molecules in organic solvents, including the solubility results of 1,3-bis(9-carbazolyl)benzene (mCP), 1,3,5-tris(1-phenyl-1H-benzimidazol-2-yl)benzene (TPBi) and 2-(4- tert -butylphenyl)-5-(4-biphenyl)-1,3,4-oxadiazole (PBD) in ethanol and acetonitrile , respectively. Nanoparticles of mCP, TPBi and PBD with sizes from dozens to several hundred nanometers were prepared by solvent antisolvent precipitation method and their solubility were determined by using isothermal saturation method. The saturation solubility of nanoparticles of three kinds of optoelectronic molecules exhibited increase of 12.9%−25.7% in comparison to the same raw materials in the form of microparticles. The experimental evidence indicates that nanonization technology is a feasible way to make optoelectronic molecules dissolve in liquids with enhanced solubility.

Persistent inflammation, characterized by the intense interplay of inflammatory chemokine secretion and immune cell infiltration, is a hallmark of many skin disorders including diabetic wounds and psoriasis with inadequate therapeutic interventions. Monocyte chemotactic protein-1 (MCP-1) is an inflammatory chemokine that plays a key role in recruiting and polarizing monocytes into pro-inflammatory macrophages to establish a vicious cycle that worsens the inflamed tissue microenvironment. Here, the sponge-like microneedles (HPMN) technology is described to alleviate inflammatory skin disorders. Heparin/4-arm PEG-NH 2 network crosslinked onto microneedle surface spatially attracted and sequestered multiple inflammatory chemokines including MCP-1. Enrichment of MCP-1 on microneedles recruits and traps inflammatory monocytes within the porous structure of microneedles. Subsequent removal of microneedles not only depletes inflammatory chemokine, MCP-1, but also its cellular source. As a result, HPMN treatment facilitates 47.1% smaller open wound area in mice and 27.2% shorter wound length in pigs. To demonstrate the versatility of the HPMN technology, it is also shown that combining the method with standard-of-care immunosuppressants reduces 45.1% epidermis thickening and attenuated immune cell influx in a mouse psoriasis model. Overall, the HPMN technology is a novel demonstration of employing inflammatory chemokine and cell extraction to treat a broad range of inflammatory skin disorders.

Objective: To investigate the cellular mechanism that sinomenine (SIN) inhibits inflammation in macrophages induced by LPS through α7 nicotinic acetylcholine receptor (α7nAChR).Materials and methods: RAW264.7 cells were stimulated with LPS and treated by SIN or nicotine (Nic). A selective antagonist of α7nAChR, α-bungarotoxin (BTX) was used to block α7nAChR. AG490 was used to inhibit JAK2 activation. ELISA was performed to detect the levels of TNF-α and MCP-1. Western blotting was used to analyze the expression of MIF, MMP-9, CD14, TLR4, STAT3 and p-STAT3. Intracellular-free calcium level was measured by Fluorescent probe fluo-3/AMResults: SIN inhibited the production of TNF-α, MCP-1, MIF, and MMP-9, decreased the expression of CD14 and TLR4, and inhibited the release of intracellular-free calcium from intracellular stores in RAW 264.7 cells stimulated by LPS. JAK-specific inhibitor AG490 attenuated the inhibitory effect of SIN on TNF-α. SIN increased the phosphorylation of STAT3. And the above effects of SIN were attenuated by antagonist of α7nAChR.Conclusions: SIN can decrease the expression of CD14/TLR4 and intracellular free calcium level, activate JAK2/STAT3 pathway to inhibit inflammatory response through α7nAChR in macrophages.
