Synergistic Effect of a Combined 10-MDP/γ-MPTS Primer on Glass-Ceramic Bonding: A Multimodal Study

Jiawen Liu,Yidan Xu,Zhiyuan Lin,Suqing Tu,Chen Chen,Haifeng Xie

Introduction and aims This study investigated the synergistic effect of combining 10-methacryloyloxydecyl dihydrogen phosphate (10-MDP) and γ-methacryloxypropyl trimethoxysilane (γ-MPTS) in one primer on bonding to lithium disilicate ceramics. Methods A 5% ethanol solution with 10-MDP and γ-MPTS (1:1 by volume) was prepared. The mixed solutions after 24 hours, 72 hours, 7 days, and 6 months storage were characterized by Fourier transform infrared spectroscopy (FTIR). Ultraviolet-visible (UV-Vis) spectrophotometric titration monitored the intermolecular interactions and their dynamics within 30 minutes of mixing. 84 IPS e.max CAD specimens were etched with 5% HF acid and assigned to four surface pretreatment groups: control, 10-MDP, γ-MPTS, and 10-MDP+γ-MPTS. After applying RelyX Ultimate resin cement, 1-mm composite resin cylinders were bonded and light-polymerized. The bonded assemblies were then stored in water for 24 hours or aged via 6 months storage plus 5000 thermocycles before micro-shear bond strength testing. The data were analysed using multivariate analysis of variance ( P < .05). Six machine learning models including Logistic Regression, Decision Tree, Random Forest, Extra Trees, Gradient Boosting, and Extreme Gradient Boosting were employed to evaluate the feature importance influencing bond strength. Results FTIR and UV-Vis analyses indicated that 10-MDP promoted the hydrolysis and siloxane condensation of γ-MPTS, forming a stable organic-inorganic hybrid network. The combined group showed the highest initial (24.22 MPa) and aged (16.07 MPa) bond strength, with more cohesive failures. Machine learning confirmed the combined primer as the most critical factor. Conclusions The coexistence of 10-MDP and γ-MPTS in a single solvent environment provides a synergistic effect, significantly enhancing both the bonding strength and durability of glass-based ceramics. Clinical Relevance A combined 10-MDP/γ-MPTS primer stabilizes the adhesive interface and increases the durability of ceramic restorations for better long-term success.

The roles of 10-methacryloyloxydecyl dihydrogen phosphate and its calcium salt in preserving the adhesive–dentin hybrid layer

Xin Jin,Fei Han,Qi Wang,Xiaojun Yuan,Qiyue Zhou,Haifeng Xie,Lina Niu,Chen Chen

Objectives 10-Methacryloyloxydecyl dihydrogen phosphate (MDP) has been regarded as the most effective dentin-bonding monomer for more than 20 years. Although the dentin-bonding promoting effect of MDP has been well demonstrated, the mechanisms by which it benefits the stably of collagen within the adhesive–dentin hybrid layer are not currently fully understood. The objective of this study was to investigate the roles of MDP and its calcium salt in preserving the adhesive–dentin hybrid layer. Methods MDP-conditioned collagen was investigated by Fourier-transform infrared spectroscopy, Ultraviolet–visible spectroscopy, and molecular docking. The structural changes to the dentin surface upon acid-etching and MDP-conditioning were observed by SEM. X-ray diffraction and nuclear magnetic resonance were used to investigate the chemical interactions between MDP and HAp. The collagen-protecting effects of MDP and its Ca salt were investigated using in-situ zymography, rhMMP-9 colorimetric assay, hydroxyproline assay, and molecular docking. Results MDP forms a stable collagen-phosphate complex through hydrogen bonding with the collagen in dentin. Furthermore, it generates MDP-Ca salts that are deposited on the dentin collagen scaffold, protecting it from degradation. Moreover, both free MDP and the MDP-Ca salt inhibit matrix metallopeptidase and exogenous proteases, with the inhibitory effect of the calcium salt being significantly stronger than that of the free form. Significance MDP-based adhesives preserve the collagen within the hybrid layer by simultaneously improving collagen’s resistance to exogenous enzymes and inhibiting MMP activity, both of which contribute to the longevity of dentin–resin bonding.

A derivative of tanshinone IIA and salviadione, 15a, inhibits inflammation and alleviates DSS-induced colitis in mice by direct binding and inhibition of RIPK2

Hu Cheng-hong,Chen Yue,Jin Tian-yang,Wang Zhe,Jin Bo,Liao Jing,Ding Chun-yong,Zhang Ao,Tang Wei-yang,Zhang Ling-xi,Xu Lei-yu,Ning Fang-min,Liang Guang,Wei Xiao-hong,Wang Yi

Inflammatory bowel diseases (IBDs) are chronic inflammatory conditions primarily affecting the gastrointestinal tract. Previous studies established the role of the NF-κB signaling pathway in the development of IBDs, suggesting that anti-inflammatory therapies might offer a viable treatment strategy. Tanshinone IIA and salviadione, both derived from Salviae Miltiorrhizae Radix et Rhizoma , possess anti-inflammatory and anti-oxidative activities. A series of new compounds were synthesized by hybridizing salviadione with tanshinone. Among these compounds, 15a showed beneficial effects in LPS-induced acute lung injury and diabetes-induced renal injury mouse models. The current study explored the therapeutic efficacy of 15a using both acute and chronic colitis models and elucidated the underlying mechanisms. DSS-induced colitis models were established in mice, where acute colitis was treated with compound 15a (5 or 10 mg·kg −1 ·d −1 ) for 8 days, while chronic colitis mice received compound 15a (5 or 10 mg·kg −1 ·d −1 , i.g.) during 2.5% DSS administration. The 15a treatment significantly alleviated DSS-induced pathological and inflammatory damages in both acute and chronic colitis mouse models. In mouse intestinal epithelial cell line MODE-K, pretreatment with compound 15a (5 or 10 μM) significantly suppressed LPS + L18-MDP-induced inflammatory responses. The receptor-interacting serine/threonine kinase 2 (RIPK2) was identified as a direct binding target of compound 15a using microarrays and recombinant human proteins. Moreover, 15a could directly bind to and inhibit the phosphorylation of RIPK2, leading to the suppression of the NF-κB and MAPK signaling pathways. Furthermore, LEU153 and VAL32 were identified within the KD domain of RIPK2 as critical amino residues for the binding of 15a. Briefly, the current findings demonstrate that compound 15a holds promise as a therapeutic agent for managing acute and chronic colitis.

Promotion of Biomimetic Mineralization via Preinfiltration of Mineral Precursors

Shengjie Liang,Xin Gao,Xinyang Li,Chenmin Yao,Cui Huang

Intrafibrillar mineralization plays an important role in dentin repair. Current research on intrafibrillar mineralization primarily focuses on the precise positioning of mineral precursors within collagen and the reduction of mineralization time. Inspired by the multifunctionality of noncollagenous proteins (NCPs), we developed a dual-analogue system, 10-methacryloyloxydecyl dihydrogen phosphate (10-MDP) followed by 3-(3,4-dihydroxyphenyl)-l-alanine (l-DOPA)-stabilized amorphous calcium phosphate (M + LA), which integrated a nucleation inhibitor l-DOPA and inducer 10-MDP. In this study, we investigated the impact of a preinfiltration strategy using the M + LA system on intrafibrillar mineralization. l-DOPA with a concentration of 250 μg mL–1, acting as a mussel-inspired nucleation inhibitor, was demonstrated to stabilize amorphous calcium phosphate (ACP) precursors within 2 h at 37 °C, with the formed precursors termed “LACP” (l-DOPA-stabilized amorphous calcium phosphate). Numerous LACP precursors were immobilized within collagen via the M + LA system, referred to as “preinfiltration.” The M + LA system exhibited no significant cytotoxicity, as evidenced by the biocompatibility evaluation through a modified trans-dentin disk model. Notably, the M + LA preinfiltration strategy significantly reduced intrafibrillar mineralization time to 3 h, superior to that of bare collagen and MDP-modified collagen. Furthermore, the resulting mineralized M + LA-modified collagen achieved functional mechanical properties. Moreover, 10-MDP-Ca nanolayering was generated via a bottom-up approach during the preinfiltration process. This study introduces a preinfiltration strategy for promoting intrafibrillar mineralization, which provides a reference for understanding the mechanism of intrafibrillar mineralization and shows promising clinical application prospects for dentin repair.

Reinforcement of dental resin composite via zirconium hydroxide coating and phosphate ester monomer conditioning of nano-zirconia fillers

Xinyi Wu,Shiqi Dai,Ying Chen,Feng He,Haifeng Xie,Chen Chen

Objectives The present study aimed to evaluate effects of conditioning with the phosphate ester monomer 10-methacryloyloxydecyl dihydrogen phosphate (MDP), with and without precoating with zirconium hydroxide for nano-size zirconia fillers, on mechanical properties of dental resin composites . Materials and methods Nano-zirconia fillers coated with or without zirconium hydroxide [Zr(OH) 4 ] were prepared. Transmission electron microscopy (TEM), Fourier infrared spectroscopy (FTIR), and X-ray photoelectron spectroscopy (XPS) were used to observe the coating and to characterize Zr(OH) 4 coating on the zirconia filler surface . Zirconia fillers with or without Zr(OH) 4 coating were conditioned with MDP and were subsequently used to prepare experimental resin composites. XPS was used to analyze the Zr-O-P bonds on the filler surface after MDP conditioning. Moreover, three-point bending strength and elastic modulus of prepared resin composites were measured, and Weibull analysis was performed. Resin composites without addition of zirconia fillers and the ones with addition of untreated or silane conditioned-zirconia fillers were set as controls. Cell counting kit (CCK)-8 was used to test cell cytotoxicity of these zirconia fillers-containing experimental resin composites. Results Nano-zirconia fillers were coated with Zr(OH) 4 through chemical deposition . FTIR and XPS analysis confirmed the increase of hydroxyl groups after Zr(OH) 4 coating. XPS detected the highest contents of Zr-O-P bonds on MDP-conditioned zirconia fillers with pre-Zr(OH) 4 coating, followed by MDP-conditioned zirconia fillers. Resin composite with added MDP-conditioned zirconia fillers with and without Zr(OH) 4 coating exhibited greater three-point bending strength, elastic modulus values, and Weibull moduli . According to the cytotoxicity classification, resin composites containing experimental zirconia fillers were considered to have no significant cell cytotoxicity. Conclusion Nano-zirconia fillers conditioned with MDP, with or without precoating with Zr(OH) 4 , improve the mechanical properties of resin composites, and are potentially safe for clinical use.

Interactions of two phosphate ester monomers with hydroxyapatite and collagen fibers and their contributions to dentine bond performance

Fei Han,Xin Jin,Xiaojun Yuan,Zehua Bai,Qi Wang,Haifeng Xie

Objectives To evaluate the interactions of two phosphate ester monomers [10-methacryloyloxydecyl dihydrogen phosphate (10-MDP) and dipentaerythritol penta-acrylate phosphate (PENTA)] with hydroxyapatite and collagen and understand their influence on dentine bonding. Methods Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, X-ray diffraction, nuclear magnetic resonance, ultraviolet-visible, and molecular docking were applied for separately evaluating the interactions of two monomers with hydroxyapatite and collagen. Hydrophilicity tests and morphological observation were employed to characterize pretreated dentine. Microtensile bond strength (μTBS) and nanoleakage were investigated to evaluate the bonding performance. Hydroxyproline assay, in situ zymography, and matrix metalloproteinase-9 (MMP-9) activity assay were used to confirm the MMP inhibition. Results Chemoanalytic characterization confirmed the interactions of 10-MDP and PENTA with hydroxyapatite and collagen. The interactions of PENTA were weaker than 10-MDP. PENTA possessed better dentine tubule sealing after etching than 10-MDP. Dentine treated with PENTA was more hydrophilic than 10-MDP. 10-MDP and PENTA treating significantly increased the initial μTBS than the control group without primer conditioning. μTBS decreased significantly during aging, and the decrease was more severe in the PENTA group than 10-MDP. The 10-MDP and PENTA groups exhibited relatively less fluorescence than the control. The relative inhibition percentages of MMP-9 decreased in the order of 10-MDP–Ca salt, 10-MDP and PENTA. The 10-MDP, PENTA, and 10-MDP–Ca salt groups showed significantly lower hydroxyproline contents than the control. Conclusions Although PENTA adsorbed on hydroxyapatite, it did not form a stable calcium salt. The interactions of 10-MDP with hydroxyapatite and collagen are different than those of PENTA. Clinical significance The sealing of dentinal tubules by PENTA and the inhibition of MMP by 10-MDP and its calcium salts contribute to improving the dentine bonding durability.
