d-tyrosine enhances disoctyl dimethyl ammonium chloride on alleviating SRB corrosion

Jingyi Zhou,Hongyi Li,Shichu Gong,Shuguang Wang,Xianzheng Yuan,Chao Song

Microbiologically influenced corrosion (MIC) caused by sulfate reducing bacteria (SRB) is a serious challenge in many industries, but biofilm greatly decreases the toxicity of bactericides to cell inside. d-amino acids are potential enhancers for bactericides due to their excellent performance on biofilm inhibition. However, the mechanism of d-amino acid cooperating with bactericides for MIC inhibition is still unknown. In this study, d-tyrosine(D-Tyr)and disoctyl dimethyl ammonium chloride (DDAC) were selected as the typical d-amino acid and bactericide, respectively, to evaluate their synergetic inhibition on the corrosion caused by Desulfovibrio vulgaris . D-Tyr obviously enhanced the role of DDAC in inhibiting corrosion with high corrosion inhibition efficiency at 77.23 %. The attachment of EPS and live cells on the coupon surface decreased in the presence of D-Try, leading to more cells directly exposed to DDAC. Besides, D-Try decreased the amount of live cells on the surface and thus reduced the utilization of Fe by SRB and corrosion current. Moreover, dead cells settling to the coupon surface may form a protective lay to retard the contact between live SRB and Fe, leading to slow cathode reaction and less corrosion. Therefore, D-Tyr can reduce the coverage of biofilm, thereby reducing its protective effect on SRB and achieving better corrosion inhibition effect. This work provides a new strategy for improving bactericides and inhibiting MIC.

Rosin modified aminated mesoporous silica adsorbed tea tree oil sustained-release system for improve synergistic antibacterial and long-term antibacterial effects

Songfa Qiu,Fan Gao,Zhijun Liang,Ximing Zhong,Li Hao,Huayao Chen,Xinhua Zhou,Hongjun Zhou

Tea tree oil, a natural antibacterial compound, cannot be used effectively because of its volatile nature. In this work, a biocompatible carrier was prepared and loaded with tea tree essential oil. The carrier was prepared via the electrostatic or chemical action of aminated mesoporous silica and sodium rosin for achieving a low volatilization rate of tea tree essential oil. A synergistic antibacterial effect was observed between sodium rosin and tea tree essential oil. This method utilized the positive charge of the amino group and the condensation reaction with the carboxyl group to achieve physical and chemical interactions with sodium rosin. Fourier Transform Infrared, Brunauer–Emmet–Teller, Zeta potential, SEM, TEM, and TG were performed to characterize the structure and properties of the samples. Compared to the electrostatic effect, the chemically modified system exhibited a longer sustained release, and the sustained release curve followed the Korsmeyer–Peppas release model. Also, the antibacterial properties of the chemically modified system exhibited better minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) respectively, the MIC and MBC for E. coli were 0.3 mg ml−1 and 0.6 mg ml−1 respectively, for S. aureus were 0.15 mg ml−1 and 0.3 mg ml−1 respectively. More strikingly, the sample also demonstrated long-term antibacterial performance. Therefore, this work provides a new way for the delivery of volatile antibacterial drugs to achieve sustained-release and long-lasting antibacterial effects.

Antibacterial mechanisms of diacetyl on Listeria monocytogenes and its application in Inner Mongolian cheese preservation via gelatin-based edible films

Haiying Cui,Yangyang Chen,Tariq Aziz,Fahad Al-Asmari,Maher S. Alwethaynani,Ce Shi,Lin Lin

Cheese spoilage caused by Listeria monocytogenes ( L. monocytogenes ) contamination results in food waste, economic losses, and even threatens human health. Diacetyl, a natural microbial metabolite derived from lactic acid bacteria with efficient antimicrobial activity, is commonly found in fermented dairy products, but its inhibitory effect on L. monocytogenes was few reported. This study aimed to explore the antibacterial effects and mechanisms of diacetyl on L. monocytogenes , and its application in Inner Mongolian cheese preservation was conducted by fabricating it into gelatin-based edible films. Diacetyl showed excellent antioxidant and antibacterial activities. The minimal inhibitory concentration (MIC) of diacetyl was 1 mg/mL. Upon exposure to 2MIC diacetyl, the bacterial membrane integrity was significantly impaired, leading to intracellular materials leakage, and the cytoplasmic reactive oxygen species (ROS) level was markedly elevated. To overcome the disadvantages of diacetyl such as unstable and volatile, gelatin-based edible films incorporated with diacetyl were fabricated, and the characterization tests showed the composite edible films exhibited good mechanical, barrier, and antibacterial and antioxidant properties. In the challenge test, the composite edible films could extend the shelf life of cheese samples by maintaining the stability of moisture and pH, stabilizing the color and texture changes, and slowing down the bacterial growth and lipid oxidation. These findings offer new insights into the antibacterial effects of diacetyl on L. monocytogenes, and the fabricated edible films were anticipated to serve as a promising packaging material for preserving Inner Mongolian cheese.

Discovery of acetohydroxyacid synthase inhibitors as anti-tuberculosis lead compounds from natural products

Yanhong Niu,Zhili Wu,Qianfang Hu,Yuchen Wu,Qihua Jiang,Xiaolan Yang

Acetohydroxy acid synthase (AHAS) is a key enzyme that catalyzes the synthesis of branched-chain amino acids, which is indispensable for the survival and growth of Mycobacterium tuberculosis ( Mtb ). Aim to discover new AHAS inhibitors from natural products, here we performed computer assistant target-based screening for Mtb- AHAS inhibitors using Discovery Studio on TCMSP and SELLECK libraries. Mtb -AHAS structure was first simulated and verified for docking, and 80 compounds with top LIBDOCK and CDDOCK scores were obtained. By experimental verification, four compounds namely Salvianolic acid A, Embelin, Celastrol and Wushanicaritin showed inhibition potency against Mtb- AHAS with IC50 ranging from 805.5 nM–32.36 μM. The most potential inhibitor Celastrol exhibited bacteriostatic activity for both Mycobacterium smegmatis and Mycobacterium tuberculosis with MIC of 62.5 μM and 80 μM, respectively. This study revealed that Celastrol is the potential Mtb- AHAS inhibitor as an anti-tuberculosis lead compound.

Inhibitory effects of lactobionic acid on Vibrio parahaemolyticus planktonic cells and biofilms

Qiuxia Fan,Yahong Yuan,Ting Zhang,Wei Song,Qinglin Sheng,Tianli Yue

Vibrio parahaemolyticus is the primary pathogenic bacteria associated with seafood-borne illnesses . Lactobionic acid (LBA) is an organic acid with multiple biological activities that has recently been a focus of interest as an antibacterial agent. The aim of this study was to investigate the inhibitory effects of LBA on Vibrio parahaemolyticus planktonic cells and biofilms. The minimum inhibitory concentrations (MICs) and minimal bactericidal concentrations (MBCs) of LBA against Vibrio parahaemolyticus were both identified as 4 mg/mL. LBA exerted antimicrobial effects against planktonic Vibrio parahaemolyticus cells by damaging their membranes, as revealed by reduced intracellular ATP concentrations, increased protein leakage, abnormal cell morphology and diminished membrane integrity after treatment. At 1/16 × MIC and 1/8 × MIC, LBA inhibited biofilm formation and downregulated the expression of some biofilm-related genes, which was confirmed by crystal violet staining, field-emission scanning electron microscopy (FESEM) observations, and real-time quantitative PCR analysis. Moreover, LBA inactivated Vibrio parahaemolyticus cells within biofilms (on polystyrene and stainless steel surfaces), destroyed biofilm structures on stainless steel surfaces, and also reduced the levels of polysaccharides and proteins in the biofilms. Therefore, LBA is a potential low-cost agent that can be used to control planktonic Vibrio parahaemolyticus and its biofilms.

A combination of cuminaldehyde with citral- / trans-2-hexen-1-al alleviates green mold disease in citrus fruit

Reymick Okwong Oketch,Duan Bin,Feng Zhao,Zhang Yong Hua,Ouyang Qiuli,Kaaya Archileo N.,Tao Nengguo

Management of Penicillium digitatum requires use of highly effective fungicides that are not prone to drug resistance and are safe to humans and the environment. This study examined the fungicidal potential of combinations of cuminaldehyde (CA) + trans-2-hexen-1-al (TH) and CA + Citral. Both CA + TH and CA + Citral exhibited enhanced fungitoxicity against P. digitatum than CA, citral or TH alone. The minimum inhibitory concentration (MIC) of CA + Citral consisted of 0.13 ml/L CA and 0.3 ml/L citral, representing a synergistic interaction. MIC of CA + TH consisted of 0.12 ml/L CA and 0.15 ml/L TH, representing an additive interaction. The minimum fungicidal concentration (MFC) of CA + Citral and of CA + TH consisted of 0.26 ml/L CA and 0.6 ml/L citral, and 0.18 ml/L CA and 0.24 ml/L TH, respectively. 10×MFC CA + Citral in Tween-80 (WCI) delayed onset of hyphal growth in fruits inoculated with P. digitatum to day 5 compared to day 4 in the 10×MFC CA + TH in Tween-80 (WCT) treatment, and day 3 for the control samples. Daily green mold disease incidence and disease severity were much lower in WCI and WCT treatments than in the 10×MFC CA in Tween-80 (WCA) treatment and in control samples. Our results show that WCI was much fungitoxic against P. digitatum than WCT and WCA, its effect being comparable to that of imazalil. Additionally, at 25 ± 2 °C and 85–90% relative humidity, both WCI and WCT markedly preserved fruit quality by day 6 of treatment. WCI and WCT are thus, safe, cheap and highly effective alternatives to chemical fungicides being used to control postharvest green mold in citrus fruits.

Total flavonoids from Potentilla kleiniana Wight et Arn inhibits biofilm formation and virulence factors production in methicillin-resistant Staphylococcus aureus (MRSA)

Junyu Tao,Shilun Yan,Chuyue Zhou,Qiong Liu,Hui Zhu,Zhen Wen

Ethanopharmacological relevance Potentilla kleiniana Wight et Arn is a wide-spread wild plant in the mountainous areas in southern China. The whole herb has been used as a traditional herbal medicine to treat fever, arthritis , malaria, insect and snake bites , hepatitis, and traumatic injury. In vitro studies have reported the antibacterial activity use of the plant in traditional medicinal systems. Aim of the study The aim of this study was to investigate the inhibitory activity of total flavonoid from Potentilla kleiniana Wight et Arn (TFP) on methicillin-resistant Staphylococcus aureus (MRSA) in planktonic state and biofilm state. Materials and methods Antibacterial activities of TFP on planktonic MRSA were determined by agar diffusion method, microtiter plate assay and time-kill curve assay. Electrical conductivity, membrane permeability, membrane potential and autoaggregation were analyzed to study TFP effects on planktonic MRSA growth. Crystal violet (CV) staining and confocal laser scanning microscopy (CLSM) were analyzed to study TFP effects on aggregation and maturation of MRSA biofilm. After TFP treatment, extracellular polymeric substances (EPS) production were examined. Morphological changes in planktonic and MRSA biofilm following TFP treatment were determined with scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Moreover, α-Toxin protein expression and adhesion-related gene expression were also determined. Results The minimum inhibitory concentration (MIC) of TFP against MRSA was 20 μg/mL. The agar diffusion method and time-kill curve assay results indicated that TFP inhibited planktonic MRSA growth. TFP treatment significantly inhibited planktonic MRSA growth by inhibiting autoaggregation, α-hemolysin activity, α-Toxin protein expression, but increasing electrolyte leakage, membrane permeability and membrane potential and impacting cell structure. Moreover, TFP treatment significantly inhibited aggregation and maturation on MRSA biofilm by decreasing surface hydrophobicity , EPS production and adhesion-related gene expression. Conclusion The results of this trial provide scientific experimental data on the traditional use of Potentilla Kleiniana Wight et Arn for traumatic injury treatment and further demonstrate the potential of TFP to be developed as a novel anti-biofilm drug.

AI-guided precise design of antimicrobial polymers through high-throughput screening technology on an automated platform

Tianyi Zhang,Yuhui Wu,Ye Tian,Youxiang Wang,Peng Zhang,Qiannuan Shi,Qun Fang,Jianzhang Pan,Qiao Jin,Jian Ji

Antimicrobial peptides (AMPs)-mimicking antimicrobial polymers show great potential as therapeutic alternatives to antibiotics in the looming “post-antibiotic era”. However, the discovery of new AMP-mimicking antimicrobial polymers is challenging due to the vast chemical space of side-chain combinations. The advancement of AI-guided high-throughput screening enables more efficient, precise, and intelligent material design. Herein, we integrate combinatorial chemistry, machine learning, and automated high-throughput synthesis and characterization platforms to establish a new paradigm for the design of antimicrobial polymers with excellent biocompatibility. Starting with a library of 13,728 combinations, a seed dataset of 400 structures is generated, followed by four Design-Build-Test-Learn iterations using a new machine learning model. 7 top-performing candidates are screened with a minimum inhibitory concentration (MIC) ≤ 8 μg/mL and an inhibitory concentration causing 20 % cell death (IC 20 ) ≥ 64 μg/mL. The highest-performing polymer (MIC 2 μg/mL, IC 20 256 μg/mL) shows similar in vivo therapeutic efficacy with ceftazidime. Overall, the integration of AI-guided high-throughput screening and combinatorial chemistry accelerates the discovery of new antimicrobial polymers, which provides a scalable strategy for developing novel antimicrobial agents.

Effects of acid, alkaline, cold, and heat environmental stresses on the antibiotic resistance of the Salmonella enterica serovar Typhimurium

Shang Wu,Yang Yang,Tingwei Wang,Jiadi Sun,Yinzhi Zhang,Jian Ji,Xiulan Sun

Antibiotic resistance in Salmonella enterica serovar Typhimurium ( S . ser. Typhimurium) has become a critical safety hazard in food. Sublethal environmental stresses can influence resistance in Salmonella during food processing. This study simulated environmental stresses in food processing. The antibiotic resistance of three strains of S . ser. Typhimurium (the ATCC 14028 strain and two wild-type isolates from chicken and pork product processing) was evaluated under different pH levels (5.0, 5.5, 6.0, 8.0, and 9.0). Also, dynamic changes in resistance with treatment duration under cold (4 °C, −20 °C) and heat (55 °C) treatment were studied. The results showed that acid and alkaline stresses reduced the resistance of S. ser. Typhimurium to eight antibiotics; meanwhile, the resistance of meropenem (MERO) increased. The minimal inhibitory concentration (MIC) of MERO was increased 16- to 64-fold. With acid or alkaline stress, the extracellular ATP content increased, and the scanning electron microscopy (SEM) result clearly revealed the appearance of wrinkles and holes on the outer membrane of Salmonella . These observations imply changes in membrane permeability, which may decrease the antibiotic resistance of Salmonella . Cold or heat stress increased the resistance of S . ser. Typhimurium to tetracycline, cefotaxime, ceftazidime, nalidixic acid, azithromycin, and ampicillin; the MIC increased 2- to 4-fold. The antibiotic resistance only changed when cold and heat stresses occurred over a certain period of time and remained unchanged when the stress persisted. This study reports on the ability of S. ser. Typhimurium to develop antibiotic resistance after environmental stresses. It can provide valuable information for meat processing to improve interventions and risk management.

The effect of alkyl chain length of (R)-3-Hydroxybutyric alkyl ester on antibacterial activity and its antibacterial mechanism

Zao Cheng,Junfei Gao,Qianqian Liu,Qun Gu

This work describes the relationship between the antibacterial activity and the ester chain length (C1-C8) of (R)-3-Hydroxybutyric ((R)-3-HB) alkyl esters that synthesized from (R)-3-HB acid ((R)-3-HBA) by esterification reaction. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) decrease as the length of the (R)-3-HB alkyl ester chain increases from 1 to 6, but (R)-3-HB-C7 and (R)-3-HB-C8 have their own rules for different microorganisms. Among them, the (R)-3HB-C6 has the relatively best antibacterial and antifungal properties, which MIC were 1.95 mg mL−1 against E. coli and S. aureus; 0.98 mg mL−1 against C. albicans and B. subtilis; 0.49 mg mL−1 against A. niger. Finally, the antimicrobial mechanisms of the (R)-3HB-C6 are revealed, and these include disruption of biofilm and the bacterial wall/membrane, leakage of the intracellular content, and change in the transmembrane potential. These results imply the potential application of (R)-3-HB alkyl ester as new antimicrobial agents; future research can use this as an antibacterial element to synthesize new polymer materials or agents with high-efficiency antibacterial activity.

Development of cationic peptide chimeric lysins based on phage lysin Lysqdvp001 and their antibacterial effects against Vibrio parahaemolyticus: A preliminary study

Houqi Ning,Yu Cong,Hong Lin,Jingxue Wang

Cationic peptide chimeric lysins, Lysqdvp001-5aa, Lysqdvp001-10aa and Lysqdvp001-15aa, were designed based on lysin Lysqdvp001 from Vibrio parahaemolyticus ( V. parahaemolyticus ) phage qdvp001. These chimeric lysins showed equivalent peptidoglycan hydrolysis activities with Lysqdvp001 and could lyse the bacteria from the outside. The antibacterial activity as well as outer and inner membrane permeabilization of Lysqdvp001 and chimeric lysins against V. parahaemolyticus were Lysqdvp001-15aa>Lysqdvp001-10aa>Lysqdvp001-5aa>Lysqdvp001. Lysqdvp001-15aa exhibited an excellent antibacterial activity with minimum inhibition and bactericidal concentrations (MIC and MBC) of 0.2 and 0.4 mg/mL, respectively, and its antibacterial spectrum was much broader than phage qdvp001. Membrane hyperpolarization and membrane phospholipid exposure of V. parahaemolyticus were observed after Lysqdvp001-15aa treatments. Transmission electron microscope (TEM) showed Lysqdvp001-15aa destroyed structure integrity of V. parahaemolyticus . Besides, MIC and MBC of Lysqdvp001-15aa decreased V. parahaemolyticus counts in oyster by 3.20 and 4.03 log 10 CFU/g. Lysqdvp001-15aa at MBC eradicated about 50% of V. parahaemolyticus biofilms and inhibited over 90% of the formation of the bacterial biofilms.
