Colorectal cancer (CRC) poses a significant therapeutic challenge due to its immunosuppressive tumor microenvironmentl (TME). In this environment, triggering receptors expressed on myeloid cells 2 (TREM2+) tumor-associated macrophages (TAMs) drive immune evasion and therapy resistance. In this study, CSF1R-IN-3@Mn@MPDA-antiTREM2 (CMMT) is presented, a manganese-doped mesoporous polydopamine nanoplatform conjugated with anti-TREM2 antibodies, which integrates TAM reprogramming and magnetic resonance imaging (MRI)-guided immune monitoring. CMMT has three key characteristics: 1) pH-responsive release of the Colony Stimulating Factor 1 Receptor (CSF1R) inhibitor CSF1R-IN-3 within acidic TME; 2) selective targeting of TREM2+ M2 macrophages, achieving 4.2-fold higher uptake compared to M1 subsets ( P < 0.001); and 3) dual modulation of TREM2 and CSF1R pathways, polarizing M2-TAMs toward pro-inflammatory M1 phenotypes. In orthotopic CRC models, CMMT synergizes with anti-PD-L1 therapy, reducing tumor volume by 72.5% compared to 42.1% for monotherapy ( P < 0.001). It also extends median survival from 28 to 52 days. Importantly, CMMT acts as an MRI contrast agent, with T1 signal intensity correlating with intratumoral TREM2+ macrophage density (r- 0.71, P < 0.05), enabling non-invasive assessment of TME remodeling. This theranostic platform bridges immune modulation and real-time monitoring, providing a promising strategy to overcome immunotherapy resistance in CRC.

Chronic wounds, particularly those complicated by infection, present significant challenges in clinical management. The microenvironment of these wounds is typically characterized by the accumulation of reactive oxygen species (ROS) and abnormal local pH levels, both of which impede the healing process. Baicalin (BA), a natural flavonoid, exhibits anti-inflammatory activity, ROS-scavenging capability, and pro-healing effects. In this study, hydrogels were synthesized through photoinitiated radical polymerization of methacrylic anhydride (MAA) and dopamine (DA)-modified chondroitin sulfate (ChSMA-DA), grafting degrees of MA and DA were 58%, 23%, MPDA@MnO2nanoparticles (NPs), and methacrylated gelatin (GelMA). The gelation time, microtopography, swelling behavior, and water retention of the hydrogels were investigated, along with their degradation, rheological properties, and photothermal effects. The results indicate that swelling ratio (SR) and water retention (WR) of optimal HG-MPDA@MnO2-M sample were 5.7, 82.42%, exhibited responsive behavior upon weakly acidic environment with pH 6.5 and elevated ROS levels, and exhibited a stable photothermal effect (photothermal conversion efficiency was 22.7%) under 808 nm near-infrared (NIR) light. Following the incorporation of the drug model BA, the cumulative release percentage over 24 h under the combined stimulation of pH 6.5, 1 mmol·L−1H2O2, and 808 nm NIR was 81.1%, significantly higher than either factor alone. These hydrogels show promise as an injectable dressing for chronic wounds, effectively integrating the internal microenvironment of the wound tissue with external NIR to modulate drug release.

Despite the outstanding lightweight potential of Mg alloys, their poor corrosion resistance significantly limits their practical applications in engineering. As a more sustainable alternative to solvent-based resins, waterborne polyurethane (WPU) coatings offer notable environmental and safety benefits, including low VOC emissions, non-flammability, and ease of application. However, the inherent microporous defects formed during the curing process, coupled with the absence of self-healing capabilities, notably undermine their protective performance. In this study, a smart hybrid coating was developed to improve the corrosion protection of WPU on Mg alloys. The coating combines benzotriazole (BTA)-loaded mesoporous polydopamine (mPDA) microspheres and hydroxylated boron nitride (OH-BN) within the WPU matrix, achieving active-passive synergistic protection through dual pH/light responsiveness. The mPDA enhances OH-BN dispersion within the matrix and simultaneously acts as an intelligent carrier for the controlled release of BTA under acidic or light-irradiated conditions. Electrochemical impedance spectroscopy (EIS) showed that, after 6 h of immersion in NaCl solution (pH = 3), the damaged composite coating retained an impedance modulus of 2.37 × 10 5 Ω·cm 2 at 0.1 Hz, and maintained a value of 1.97 × 10 5 Ω·cm 2 under light irradiation-28 and 2.8 times higher, respectively, than pure WPU. These results confirm the coating's outstanding corrosion resistance and stimulus-responsive self-healing capability, which arise from the synergistic effects of the OH-BN barrier and BTA release under pH and light stimuli. This work presents a sustainable and intelligent corrosion protection strategy for Mg alloys in harsh environments.

Background Ovarian cancer is one of the most prevalent gynecologic malignancies and ranks as a leading cause of death in female cancers. Chemotherapy remains the primary treatment modality, however, the development of chemoresistance poses a significant obstacle in ovarian cancer therapy, contributing to its high mortality rate. Results In this paper, we report a multifunctional nanoparticle that enables laser-controlled release of co-loaded paclitaxel (PTX) and STMN1 siRNA for synergistic combination therapy. This nanoparticle can improve the microtubule stability by interfering the expression of STMN1, thereby increasing the sensitivity to PTX, blocking cells in the G2/M phase, and ultimately leading to cancer cell death. Additionally, it exhibits enhanced bioavailability, reduced systemic toxicity, and photothermal properties suitable for in vivo imaging. Conclusions Our nanoparticle offers an innovative strategy for the concurrent therapy and monitoring of ovarian cancer, enhancing therapeutic efficacy and diagnostic precision. Graphical abstract

Obesity and its metabolic complications urgently require safe, effective, and minimally invasive interventions. Here, we engineered an injectable, self-healing hydrogel system (OCMU hydrogel) based on oxidized hyaluronic acid (OHA) and carboxymethyl chitosan (CMCS) for the localized delivery of plant-based ursolic acid (UA) loaded in mesoporous polydopamine (MPDA) nanoparticles. This system synergistically induces white adipose browning through combined photothermal therapy (PTT) and pharmacological action, effectively counteracting obesity. The OCMU hydrogel exhibited excellent self-healing properties, high UA loading capacity (81.04%), efficient photothermal conversion, and favorable biocompatibility. Mechanistically, the system promotes lipolysis, mitochondrial biogenesis, and adipose browning via activation of the AMPK/SIRT1/PGC-1α pathway, with a 439% increase in UCP1 expression in vitro. In obese mice, OCMU combined with PTT reduced body weight by 10.94% and inguinal fat by 56.32%, while significantly improving metabolic profiles and alleviating inflammation. Notably, the combination of mild PTT and naturally derived UA achieved superior anti-obesity efficacy compared to monotherapies, highlighting a favorable safety and efficacy profile. This work establishes a promising platform in which UA and mild PTT act in concert to reprogram adipocyte metabolism, offering a safe and synergistic approach to combat obesity. Graphical

Introduction Tirzepatide (TZP), a dual glucose-dependent insulinotropic polypeptide/glucagon-like peptide 1 receptor agonist, has been showing superior benefits in weight loss and glucose lowering in patients with obesity, while the tissue-specific mechanisms of TZP as well as its side effects remain to be investigated. Objectives We aimed to explore whether localized injection of TZP into inguinal white adipose tissue (iWAT) would be as effective as subcutaneous injection for weight loss, in order to understand the local effects and signaling pathways of TZP for precision medicine. Methods Mesoporous polydopamine was synthesized and mixed with TZP for encapsulation (MPDA@TZP), followed by characterization of its retention in iWAT after local injection. Both diet-induced obesity (DIO) mice and db/db mice received local injection of TZP, and transcriptomic analysis of iWATs was performed. iWATs were also dissected for ex vivo assays. Results MPDA@TZP successfully increased the retention time of TZP in the iWAT of mice and had a more dramatic effect on weight loss and improvement in plasma lipid profiles compared to TZP alone in DIO mice, while showing comparable glucose-lowering efficacy. Transcriptomic analysis indicated that iWAT injection of MPDA@TZP improved leptin resistance, beiging, lipid metabolism, mitochondrial activity and branched-chain amino acid (BCAA) catabolism in iWAT. Leptin receptor deficiency in db/db mice abolished the weight reduction effect of MPDA@TZP via iWAT local injection compared to that in DIO mice, while the glucose-lowering effects were comparable in both db/db and DIO mice. Conclusion These findings indicate that the retention of TZP in iWAT via MPDA encapsulation amplified its effect on weight loss in mice through leptin receptor signaling compared with TZP alone, which provides new insights into the tissue-specific mechanism and alternative delivery strategies of TZP for targeting specific WAT tissues.

Excessive oxidative stress, dysregulated immune homeostasis, and disruption of the intestinal epithelial barrier are crucial features of inflammatory bowel disease (IBD). Traditional treatments focusing solely on inflammation resolution remain unsatisfactory. Herein, a yeast-inspired orally administered nanocomposite was developed. First, the MD@MPDA core was fabricated by integrating manganese dioxide (MnO2) nanozymes onto diallyl trisulfide (H2S prodrug)-loaded mesoporous polydopamine nanoparticles (MPDA). Then, yeast cell wall (YCW) was chosen to encapsulate MD@MPDA, namely, YMD@MPDA. The β-glucan embedded in the YCW shell not only protected the nanocomposite from the harsh gastrointestinal environment but also allowed the targeting enrichment in the inflamed colon. Furthermore, M1 macrophages triggered the intracellular GSH-responsive H2S release in the pathological microenvironment. MD@MPDA effectively alleviated inflammatory responses by MnO2-mediated ROS-scavenging and H2S-participated immunomodulation. The synergistic action contributed to macrophage mitochondrial function restoration and M2 polarization by suppressing NOX4 signaling and p38 MAPK pro-inflammatory signaling. In the mice model of dextran sulfate sodium (DSS)-induced IBD, the multipronged manner of scavenging oxidative stress, remodeling innate and adaptive immune homeostasis, and reshaping gut microbiota caused by YMD@MPDA effectively ameliorated inflammation and restored intestinal barrier functions. Overall, the YMD@MPDA nanocomposite provides a promising codelivery strategy of antioxidative nanozymes and gas prodrugs for the comprehensive management of IBD.
