Nanocellulose materials have undergone rapid development in recent years as promising biomedical materials due to their excellent physical and biological properties, in particular their biocompatibility, biodegradability, and low cytotoxicity. In this study, we prepared 2,3-dialdehyde nanofibrillated cellulose (DANFC) by sodium periodate oxidation, which is a mild oxidation process. With increasing oxidation time, the antimicrobial activity of DANFC against both Staphylococcus aureus (S. aureus) and methicillin-resistant Staphylococcus aureus (MRSA) improved. DANFC also displays good biocompatibility with mammalian cells, and shows good blood compatibility. In addition, animal studies and histology results reveal that DANFC can accelerate wound healing and enhance the formation of blood vessels and epithelium.

Methicillin-resistant Staphylococcus aureus (MRSA) is a ubiquitous foodborne pathogen capable of causing food poisoning and producing enterotoxins in foodstuffs, which ultimately results in various illnesses, such as vomiting, diarrhea, and pneumonia. In this study, we present a MoS 2 NF@AuNPs enhanced electrochemiluminescence (ECL) biosensor for MRSA detection based on a DNA branching tree structure formed via enzyme-free hybridization chain reaction (HCR) amplification. This approach integrates bacterial aptamers with multistage HCR amplification to achieve highly sensitive and specific target recognition. Furthermore, MoS 2 NF@AuNPs act as a synergistic catalyst for Ru(bpy) 3 2+ molecules, which in turn substantially enhances the signal readout of the biosensor. The biosensor exhibits a robust linear correlation for MRSA detection over a concentration range of 10 0 to 10 7 cfu/mL, with a detection limit of 1 cfu/mL. In addition, the biosensor demonstrates excellent selectivity and high sensitivity when applied to real samples such as water, juice, and milk.

The rising prevalence of multidrug-resistant pathogens poses a significant threat to human health, with methicillin-resistant Staphylococcus aureus (MRSA) being particularly concerning. Although vancomycin has long been the standard treatment for MRSA, its repeated use can lead to antibiotic tolerance and resistance. The accumulation of bacterial metabolites contributes to this tolerance, suggesting that modulation of bacterial metabolic pathways may provide a novel solution. Herein, we developed Ga/Cu-MOF nanomyze, characterized by a high specific surface area, which could be capable of loading antibiotics and serve as a dual-metal ion reservoir for the sustained release of gallium ions (Ga 3 + ) and copper ions (Cu 2+ ). These ions independently inhibited nitric oxide (NO) production and promoted the decomposition of S-nitrosothiols (RSNO), significantly reducing MRSA's tolerance to vancomycin. Moreover, Ga/Cu-MOF induced programmed cell death in MRSA, specifically targeting ferroptosis and cuproptosis. Antibiotic-loaded nanomyze exhibited robust synergistic antibacterial properties, including the inhibition of biofilm formation and the clearance of intracellular bacteria in macrophages. In preclinical models of skin defect infections in mice and osteomyelitis in rats, Ga/Cu-MOF showed the ability to regulate the host inflammatory response, facilitating faster recovery. Our goal is to reduce bacterial tolerance by modulating metabolic pathways and enhance antibacterial efficacy through bacterial cell death, thereby presenting a potential novel therapeutic strategy for the clinical management of multidrug-resistant bacteria.

The emergence of multidrug-resistant (MDR) bacteria represented by MRSA poses a great challenge to current anti-infection treatment. The traditional antibacterial agents containing Ag + have been receiving intense attention due to their broad-spectrum activity. Compared with the Ag + , the high-valent silver ions including Ag 2+ and Ag 3+ have superior antibacterial ability. However, their extreme instability has severely limited their development as antimicrobials, and so far, no high-valent silver antimicrobials have been reported. Herein an unusual and water-soluble high-valent silver (II + III)-porphyrin complex (AgTMPyP) which can stably exist in an aqueous medium for at least 200 days, its corresponding composite AgTMPyP@GO was also successfully prepared. AgTMPyP@GO shows an excellent photothermal conversion rate (32.17 %) under excitation by an 808 nm near-infrared laser along with dual enzyme-like (peroxidase-like and oxidase-like) activities. When used for photothermal therapy, AgTMPyP@GO can effectively destroy biofilms and kill methicillin-resistant Staphylococcus aureus (MRSA) in vitro . The composite efficiently eradicates MRSA from wounds and rapidly promote wound healing in vivo . Moreover, hemolysis, blood routine, and blood biochemistry tests all demonstrate that AgTMPyP@GO has good biocompatibility. This work reveals that AgTMPyP@GO provides a synergistic antibacterial platform with effective antibacterial performance both in vivo and in vitro .

Persistent microbial infections and excessive reactive oxygen species (ROS) accumulation severely impede diabetic wound healing. Herein, we developed an ultrasound-switchable BiVO 4 /fullerene piezoelectric heterostructure via a one-pot solvothermal method, enabling on-demand transition between bactericidal action and ROS scavenging for treating infected diabetic wounds. Under 8-min ultrasound (US) irradiation, the heterojunction sonosensitizer leveraged piezoelectric polarization to generate substantial ROS in real-time through a narrowed energy band gap and enhanced charge carrier separation and migration efficiency, resulting in the disruption of bacterial membrane integrity and 99.9 % eradication of methicillin-resistant Staphylococcus aureus (MRSA). Upon US withdrawal, the sonosensitizer spontaneously transitioned to an antioxidative state through fullerene-mediated ROS scavenging, effectively neutralizing excess ROS and restoring cellular redox balance. In an MRSA-infected diabetic wound model, this ultrasound-responsive duality effectively suppressed bacterial proliferation, reduced inflammation, enhanced angiogenesis, and ultimately accelerated wound healing within 14 days. This ultrasound-switchable therapeutic strategy offers promising insights for managing drug-resistant infections and other ROS-mediated biomedical challenges.

Owing to the poor penetration depth of light, phototherapy, including photothermal and photodynamic therapies, remains severely ineffective in treating deep tissue infections such as methicillin-resistant Staphylococcus aureus (MRSA)-infected osteomyelitis. Here, we report a microwave-excited antibacterial nanocapturer system for treating deep tissue infections that consists of microwave-responsive Fe 3 O 4 /CNT and the chemotherapy agent gentamicin (Gent). This system, Fe 3 O 4 /CNT/Gent, is proven to efficiently target and eradicate MRSA-infected rabbit tibia osteomyelitis. Its robust antibacterial effectiveness is attributed to the precise bacteria-capturing ability and magnetic targeting of the nanocapturer, as well as the subsequent synergistic effects of precise microwaveocaloric therapy from Fe 3 O 4 /CNT and chemotherapy from the effective release of antibiotics in infection sites. The advanced target-nanocapturer of microwave-excited microwaveocaloric-chemotherapy with effective targeting developed in this study makes a major step forward in microwave therapy for deep tissue infections.

Bacterial infection, especially multidrug-resistant bacteria-induced infection, threatens human health seriously, which has posed great challenges for clinical therapy. The overuse of conventional antibiotics has given rise to bacterial resistance that severely restricts the clinical treatment options of conventional antibiotics. The development of highly effective antibacterial materials and therapeutic strategies to inhibit the multidrug-resistant bacteria-induced infections is of great urgency. Although silver nanoparticles (AgNPs) have exhibited certain effectiveness in killing multidrug-resistant bacteria, their antibacterial efficacy and biosafety are still unsatisfactory. In this work, we prepared graphene quantum dots (GQDs) by a green synthesis method with the natural polymer starch as a precursor for uniformly decorating AgNPs to form GQDs coated AgNPs (GQDs@Ag). The nanocomplex was comprehensively characterized, and its antibacterial activity and biosafety were systematically investigated. The characterization results revealed that the successfully constructed GQDs@Ag hybrids with improved dispersion and stability are composed of AgNPs closely and uniformly surrounded by the GQDs. Furthermore, in vitro and in vivo results demonstrated that GQDs@Ag hybrids with superior biosafety showed a markedly enhanced effect in killing MRSA and accelerating MRSA-infected wound healing as compared to AgNPs alone. Collectively, these results suggest that the biocompatible nanosystem of GQDs@Ag exhibits great potential in clinical application for MRSA infection.

Wound infections are a primary cause of delayed wound healing, and film dressings have garnered significant attention in wound management. However, the development of biologically sourced and green-synthesized film dressings with rapid antimicrobial functionality remains an urgent technological breakthrough. In this study, we successfully developed gelatin sodium alginate/sea sheath molecularly modified berberine hydrochloride film (GS/CB). The incorporation of sea sheath powder significantly enhances the mechanical properties of the gelatin‑sodium alginate film. The calcium ions (Ca 2+ ) provided by the sea sheath create a cross-linking effect within the sodium alginate-gelatin network, resulting in tensile properties of the GS/CB films that are 2.6 times greater than those of the control films. This enhancement allows the films to meet the requirements for wound applications. In in vitro antimicrobial experiments, the films incorporated with sodium oleate-modified berberine hydrochloride showed good antimicrobial effects, with an inhibition rate of 97.79 % against Staphylococcus aureus ( S. aureus ) and 98.57 % against methicillin-resistant Staphylococcus aureus ( MRSA ). In in vivo wound healing experiments, the film effectively eliminated MRSA from the wound, achieving a bacterial clearance rate of 98.5 %. Compared to transparent film dressings, the GS/CB film demonstrated superior wound care efficacy and significantly accelerated wound healing. Furthermore, both in vivo and in vitro tests indicated that the film possesses excellent biocompatibility. Therefore, the GS/CB film holds great potential and application value as a new wound dressing for the treatment of infectious wounds.

MRSA -induced arthritis is a prevalent and highly debilitating orthopedic condition. The inflammatory response induced by bacterial infection hinders tissue repair and exacerbates bone loss. Traditional antibiotic therapies are limited by low bioavailability, substantial side effects, and narrow efficacy, rendering them inadequate for comprehensive treatment of arthritis. Nitric oxide (NO) has demonstrated considerable potential in overcoming bacterial resistance, modulating immune responses, and facilitating tissue repair. Therefore, a stage-specific NO release strategy, tailored to the distinct phases of bacterial arthritis, is essential for effective treatment. In this study, mesoporous polydopamine nanoparticles were utilized as NO donors (mPDA/NONOate) and encapsulated within a supramolecular hydrogel formed via the host-guest interaction between α-cyclodextrin (α-CD) and Pluronic F127. The injectable nature of the resulting NO/PDA-Gel hydrogel ensured uniform distribution within irregular bone joint infection sites, minimizing NO donor loss and enhancing local bioavailability. Notably, upon near-infrared (NIR) irradiation, the hydrogel induces a rapid increase in local temperature, facilitating rapid NO release. At the same time, the synergistic photothermal effect effectively kills bacteria and rapidly controls the infection. Without light irradiation, NO is sustainably and stably released from the NO/PDA-Gel, modulating the bone immune microenvironment, alleviating inflammation, promoting chondrocyte proliferation and differentiation, and accelerating bone tissue repair, thus significantly shortening the healing time of MRSA -induced arthritis. In conclusion, the injectable self-assembled NO/PDA-Gel offers a precise, stage-matched therapeutic approach for MRSA -induced arthritis and holds promise for the treatment of deep-seated infections caused by other multidrug-resistant pathogens.

Intracellular MRSA is extremely difficult to eradicate by traditional antibiotics, leading to infection dissemination and drug resistance. A general lack of facile and long-term strategies to effectively eliminate intracellular MRSA. In this study, glabridin (GLA)-loaded pH-responsive nanoparticles (NPs) were constructed using cinnamaldehyde (CA)-dextran conjugates as carriers. These NPs targeted infected macrophages/MRSA via dextran mediation and effectively accumulated at the MRSA infection site. The NPs were then destabilized in response to the low pH of the lysosomes, which triggered the release of CA and GLA. The released CA downregulated the expression of cytotoxic pore-forming toxins, thereby decreasing the damage of macrophage and risk of the intracellular bacterial dissemination. Meanwhile, GLA could rapidly kill intracellularly entrapped MRSA with a low possibility of developing resistance. Using a specific combination of the natural antibacterial agents CA and GLA, NPs effectively eradicated intracellular MRSA with low toxicity to normal tissues in a MRSA-induced peritonitis model. This strategy presents a potential alternative for enhancing intracellular MRSA therapy, particularly for repeated and long-term clinical applications. Statement of significance Intracellular MRSA infections are a growing threat to public health, and there is a general lack of a facile strategy for efficiently eliminating intracellular MRSA while reducing the ever-increasing drug resistance. In this study, pH-responsive and macrophage/MRSA-targeting nanoparticles were prepared by conjugating the phytochemical cinnamaldehyde to dextran to encapsulate the natural antibacterial agent glabridin. Using a combination of traditional Chinese medicine, the NPs significantly increased drug accumulation in MRSA and showed superior intracellular and extracellular bactericidal activity. Importantly, the NPs can inhibit potential intracellular bacteria dissemination and reduce the development of drug resistance, thus allowing for repeated treatment. Natural antibacterial agent-based drug delivery systems are an attractive alternative for facilitating the clinical treatment of intracellular MRSA infections.

This study investigated the anti-methicillin-resistant Staphylococcus aureus (anti-MRSA) activity and chemical compositions of ether extracts from Rhizoma Polygoni Cuspidati (ET-RPC). Significant anti-MRSA activities of ET-RPC against MRSA252 and MRSA clinical strains were tested in in vitro antibacterial experiments, such as inhibition zone diameter test, minimal inhibitory concentration test, and dynamic bacterial growth assay. Subsequently, 7 major compounds of ET-RPC were purified and identified as polydatin, resveratrol-4-O-d-(6′-galloyl)-glucopyranoside, resveratrol, torachryson-8-O-glucoside, emodin-8-O-glucoside, 6-hydroxy-emodin, and emodin using liquid chromatography – electrospray ionization – tandem mass spectrometry. After investigation of anti-MRSA activities of the 7 major compounds, only emodin had significant anti-MRSA activity. Further, transmission electron microscopy was used to observe morphological changes in the cell wall of MRSA252, and the result revealed that emodin could damage the integrity of cell wall, leading to loss of intracellular components. In summary, our results showed ET-RPC could significantly inhibit bacterial growth of MRSA strains. Emodin was identified as the major compound with anti-MRSA activity; this activity was related to destruction of the integrity of the cell wall and cell membrane.

The multi-drug resistance of methicillin-resistant Staphylococcus aureus (MRSA) and complex wound microenvironment challenge the repair of MRSA infected wound. Herein, in this study, α-tocopherol modified glycol chitosan (TG) nanoparticles encapsulated with phytochemical rhein (Rhein@TG NPs) were prepared for comprehensive anti-infection and promotion of MRSA infected wound healing. Rhein@TG NPs could not only specifically release rhein in the infection site in response to low pH and lipase of infectious microenvironment, but also up-regulated M1 macrophage polarization in the infection stage, thus achieving synergistically bacterial elimination with low possibility of developing resistance. Additionally, the NPs reduced the levels of pro-inflammatory factors in the post-infection stage, scavenged the ROS, promoted cell migration and angiogenesis, which significantly improved the microenvironment of infected wound healing. Therefore, this antibiotic-free NPs enabling anti-infection and promotion of wound healing provides a new and long-term strategy for the treatment of MRSA infected wound.
