In this study, a pH/reduction dual responsive carrier containing 42nt-nucleic acid HApt based on mesoporous silica nanoparticles (MSNs) was designed. Two kinds of low molecular weight oligomeric hyaluronic acid (HA) were used to graft onto MSN for better drug encapsulation. Crosslinked MSN-HA3000gel and MSN-HA11000gel were prepared by crosslinking the HA chain through the sulfhydrylization of the carboxyl group on the HA side chain. An appropriate amount of sulfhydryl nucleic acid (HApt-SH) was added during the crosslinking reaction, which realized the targeting ability and apoptosis function to cancer cells overexpressing the HER2 receptor. Crosslinked HA had a good effect on decreasing the side effect of DOX that the drug leakage was less than 20% under a normal body environment. However, it could realize rapid and efficient drug release in a tumor environment. As to the release of HApt, it exhibited a good response to GSH. The cytotoxicity test showed that HApt contained in HAgel had a great targeting effect and significant cytotoxicity to SKBR3 cells. As a whole, this MSN-HAgel enabled the combination of gene therapy and chemotherapy, showing the synergistic effect of “1 + 1 > 2”, providing a novel idea for cancer treatments.

Targeted drug delivery to colon cancer cells can significantly improve the efficiency of treatment. We firstly synthesized carboxyl-modified mesoporous silica nanoparticles (MSN–COOH) via two-step synthesis, and then developed calcium leucovorin (LV)-loaded carboxyl-modified mesoporous silica nanoparticles based on galactosylated chitosan (GC), which are galectin receptor-mediated materials for colon-specific drug delivery systems. Both unmodified and functionalized nanoparticles were characterized by scanning electron microscopy (SEM), transmission electron microscope (TEM), X-ray diffraction (XRD), Fourier transform infrared (FT-IR), nitrogen sorption, and dynamic light scattering (DLS). Drug release properties and drug loading capacity were determined by ultraviolet spectrophotometry (UV).[email protected]–COOH/GC had a high LV loading and a drug loading of 18.07%. In vitro, its release, mainly by diffusion, was sustained release. Cell experiments showed that in SW620 cells with the galectin receptor, the[email protected]–COOH/GC metabolized into methyl tetrahydrofolic acid (MTHF) and 5-fluorouracil (5-FU)@MSN–NH2/GC metabolized into FdUMP in vivo. MTHF and 5-fluoro-2′-deoxyuridine 5′-monophosphate (FdUMP) had combined inhibition and significantly downregulated the expression of thymidylate synthase (TS). Fluorescence microscopy and flow cytometry experiments show that MSN–COOH/GC has tumor cell targeting, which specifically recognizes and binds to the galectin receptor in tumor cells. The results show that the nano-dosing system based on GC can increase the concentrations of LV and 5-FU tumor cells and enhance their combined effect against colon cancer. Keywords:calcium leucovorin;MSN–COOH;galactosylated chitosan (GC);[email protected]–COOH/GC;galectin receptor;SW620 cells

Rod-shape nanoplatform have received tremendous attention owing to their enhanced ability for cell internalization and high capacity for drug loading. MoS 2 , widely used in electronic devices, electrocatalysis, sensor and energy-storage, has been studied as photothermal agents over the years. However, the efficacy of rod-shape MoS 2 based photothermal agents for photothermal therapy has not been studied before. Here, a near-infrared (NIR) light-absorbing MoS 2 nanosheets coated mesoporous silica nanorods with human serum albumin (HSA) modifying and Ce6 loading (MSNR@MoS 2 -HSA/Ce6) were constructed for combined photothermal and photodynamic therapy. Methods : The near-infrared (NIR) light was used to trigger the synergistic anti-tumor therapy. In addition, breast cancer cell line was applied to evaluate the in vitro anti-tumor activity. The multi-modal imaging capacity and tumor-killing efficiency of the designed nanocomposites in vivo was also demonstrated with the 4T1 tumor-bearing mouse model. Results : These nanocomposites could not only perform NIR light triggered photodynamic therapy (PDT) and photothermal therapy (PTT), but also achieve in vivo fluorescence (FL) /multispectral optical tomography (MSOT)/X-ray computed tomography (CT) triple-model bioimaging. What's more, the rod-shape nanoplatform could be endowed with better anti-tumor ability based on the EPR effect and HSA-mediated active tumor targeting. At the same time, the hyperthermia generated by MoS 2 could synergistically improve the PDT effect with the acceleration of the blood flow, leading to the increase of the oxygen level in tumor tissue. Conclusion : MSNR@MoS 2 -HSA/Ce6 proves to be a promising multi-functional nanoplatform for effective treatment of tumor.

The discharge of large volumes of textile dyeing wastewater, characterized by poor biodegradability and high toxicity, poses severe threats to the environment. In this study, polyvinylidene difluoride (PVDF) membranes were prepared using the nonsolvent-induced phase separation (NIPS) method, with porous amino-functionalized mesoporous silica nanoparticles (Am-MSNs) mixed into the casting solution to fabricate the Am-MSN/PVDF mixed matrix membranes. By varying the amount of Am-MSNs added, the microstructure and overall performance of the membranes were comprehensively analyzed. The results demonstrated that the addition of Am-MSNs significantly enhanced the hydrophilicity of the membranes. The high specific surface area and amino groups of Am-MSNs facilitated interactions with dye molecules, such as Reactive Black 5 (RB5), through hydrogen bonding, electrostatic attraction, and physical adsorption, resulting in a marked improvement in RB5 rejection rates. Static adsorption tests further validated the superior adsorption capacity of the Am-MSN/PVDF mixed matrix membranes for RB5. Additionally, the nanoscale mesoporous structure of Am-MSNs enhanced the mechanical strength of the membranes. The synergistic effects of the mesoporous structure and amino groups significantly increased the efficiency and stability of the Am-MSN/PVDF mixed matrix membranes in dye removal applications, providing an effective and sustainable solution for the treatment of dye-contaminated wastewater.

The validity and biocompatibility of irrigating agents are imperative for the success of root canal therapy. The imperfections in the currently available irrigants highlight the fact that more advanced technologies and strategies are required for complete disinfection in endodontic treatments. In the present study, a Fenton reaction-enhanced antimicrobial sonodynamic therapy (SDT) platform was fabricated for root canal disinfection. Firstly, mesoporous silica nanoparticles (MSNs) were synthesized, grafted with an amino group and then conjugated with sonosensitizer protoporphyrin IX (PpIX). Iron ions were then anchored (M@P-Fe) to initiate a Fenton reaction. Nanoparticle characterization by size and zeta potential measurements, scanning electron microscopy, transmission electron microscopy and thermogravimetric analysis confirmed that the platform was successfully developed. Reactive oxygen species (ROS) generation assessment, methylene blue degradation and electron spin resonance assays illustrated upon ultrasound (US) irradiation, that augmented ROS, can be produced by US activated PpIX and iron mediated Fenton reactions from low concentration H2O2 (0.01%). In vitro anti-Enterococcus faecalis efficacy was demonstrated by growth curve and colony forming unit measurements. Confocal laser scanning microscopy and scanning electron microscopy observations illustrated the effectiveness of the platform on in situ biofilm eradication in root canal. Owing to the stronger oxidizing capability and short lifetime of ROS, the Fenton reaction-enhanced SDT can induce detrimental oxidative damage to bacteria upon activation of US while avoiding nonspecific toxicity to cells, which was verified by cytotoxicity evaluations using CCK-8 assay and morphology observation of MC3T3-E1 cells. Compared to commonly used NaClO, this nanoplatform displayed desirable anti-bacterial, anti-biofilm abilities and better biocompatibility. These results highlight that the integrated M@P-Fe + US + H2O2 platform is a promising candidate for US enhanced root canal irrigation and disinfection.

Tandem catalysts represent a promising paradigm for synergistic electrochemical CO 2 reduction (CO 2 RR) to high-value multi-carbon (C 2 ) products, yet coordinate regulation of key intermediates to boost C 2 selectivity remains unclear. Herein, a dual-phase templating coupled with gas-phase reduction strategy is exploited to fabricate a series of CuAg@MSN tandem catalysts with tunable Cu/Ag mass ratios. A volcano-type relationship between catalyst composition and C 2 product selectivity is observed, with CuAg@MSN-3 (15 wt.% Ag, 40 wt.% Cu) delivering the maximum C 2 Faradaic efficiency (FE) of 75.4% at −1.4 V vs RHE. And a high FE of 63.5% and a large partial current density of 330 mA/cm −2 toward C 2 products on CuAg@MSN-3 at a cell voltage of 4.5 V in a membrane-electrode assembly (MEA). In situ infrared spectroscopy and density functional theory (DFT) calculations jointly corroborate that CO generated on Ag sites spontaneously spills over and becomes confined within the mesopores of the MSN framework, markedly promoting CO re-adsorption on adjacent Cu sites and elevating the * CO surface coverage. This intermediate regulation effectively enhances the reaction rate of C─C coupling and consequently boosts C 2 product selectivity. The present work exemplifies a rational, confinement-guided design of tandem catalysts for steering reaction intermediates toward desired multicarbon products.

Purpose The purpose of this paper is to prepare smart coatings with self-healing and self-reporting dual functionalities. Design/methodology/approach Benzotriazole (BTA) and 1,10-phenanthroline-5-amine (APhen) were encapsulated within mesoporous silica nanocontainers (MSN) via one-step method. Then, BTA+APhen@MSN were mixed with epoxy for preparing BTA+APhen@MSN/EP coatings. The microstructure, structure and sustained-release properties of BTA+APhen@MSN nanocontainers were investigated by transmission electron microscopy, Fourier-transform infrared spectroscopy, Brunauer–Emmett–Teller analysis and UV-Vis spectroscopy. The warning performance and corrosion resistance of BTA+APhen@MSN/EP coatings were characterized by optical microscopy and electrochemical impedance spectroscopy. Findings BTA+APhen@MSN exhibit higher corrosion inhibitors loading capacity (31.2 wt.%) and higher corrosion inhibition efficiency (53.07%) than BTA@MSN and APhen@MSN. The BTA+APhen@MSN/EP coatings show excellent self-healing performance. After soaking in 3.5 wt.% NaCl solution for five days, the fitted resistance of scratched coatings increased by 18%. The BTA+APhen@MSN/EP coatings also show superior self-reporting capability, which can provide a warning of coating damage within 30 min by a visible color change. Originality/value The corrosion inhibitors and corrosion warning agent are encapsulated in a nanocontainer to prepare multifunctional nanocomposite. This multifunctional nanocomposite presents a new approach for designing smart coatings with self-healing and self-reporting dual functionalities.

A temperature, glutathione (GSH), and H2O2multi-responsive composite nanocarrier (MSN-SS-Fc@β-CD-PNIPAM) based on β-cyclodextrin-poly(N-isopropylacrylamide) (β-CD-PNIPAM) star polymer capped ferrocene modified mesoporous silica nanoparticles (MSN-SS-Fc) was successfully prepared. The surface of the mesoporous silica was first modified by ferrocene (Fc) via a disulfide bond (–SS–) to form an oxidizing and reducing site and then complexed with a β-CD-PNIPAM star shaped polymer through host–guest interactions as a nano-valve to provide temperature responsive characteristics. The structure and properties of the complex nanoparticles were studied by FTIR, TGA, EDS, Zeta potential, and elemental analysis. Doxorubicin (DOX) and Naproxen (NAP), as model drugs, were loaded into nanocarriers to assess drug loading and release behaviour. The release of drugs from nanocarriers was enhanced with an increase of the GSH, H2O2concentration, or temperatures of the solution. The kinetics of the release process were studied using different models. This nanocarrier presents successful multi-stimuli responsive drug delivery in optimal stimuli and provides potential applications for clinical treatment. Keywords:disulfide bond;ferrocene;multi-responsive;nanocarrier;MSNs

Introduction RNA interference (RNAi) stands as a widely employed gene interference technology, with small interfering RNA (siRNA) emerging as a promising tool for cancer treatment. However, the inherent limitations of siRNA, such as easy degradation and low bioavailability, hamper its efficacy in cancer therapy. To address these challenges, this study focused on the development of a nanocarrier system (HLM-N@DOX/R) capable of delivering both siRNA and doxorubicin for the treatment of breast cancer. Methods The study involved a comprehensive investigation into various characteristics of the nanocarrier, including shape, diameter, Fourier transform infrared (FT-IR) spectroscopy, X-ray photoelectron spectroscopy (XPS), encapsulation efficiency, and drug loading. Subsequently, in vitro and in vivo studies were conducted on cytotoxicity, cellular uptake, cellular immunofluorescence, lysosome escape, and mouse tumor models to evaluate the efficacy of the nanocarrier in reversing tumor multidrug resistance and anti-tumor effects. Results The results showed that HLM-N@DOX/R had a high encapsulation efficiency and drug loading capacity, and exhibited pH/redox dual responsive drug release characteristics. In vitro and in vivo studies showed that HLM-N@DOX/R inhibited the expression of P-gp by 80%, inhibited MDR tumor growth by 71% and eliminated P protein mediated multidrug resistance. Conclusion In summary, HLM-N holds tremendous potential as an effective and targeted co-delivery system for DOX and P-gp siRNA, offering a promising strategy for overcoming MDR in breast cancer.

In order to achieve a controlled release drug delivery system (DDS) for cancer therapy, a pH and redox dual-responsive mesoporous silica nanoparticles (MSN)-sulfur (S)-S- chitosan (CS) DDS was prepared via an amide reaction of dithiodipropionic acid with amino groups on the surface of MSN and amino groups on the surface of CS. Using salicylic acid (SA) as a model drug, SA@MSN-S-S-CS was prepared by an impregnation method. Subsequently, the stability, swelling properties and drug release properties of the DDS were studied by x-ray diffraction, scanning electron microscopy, Fourier transform infrared microspectroscopy, size and zeta potential as well as Brunauer–Emmett–Teller surface area. Pore size and volume of the composites decreased after drug loading but maintained a stable structure. The calculated drug loading rate and encapsulation efficiency were 8.17% and 55.64%, respectively. The in vitro drug release rate was 21.54% in response to glutathione, and the release rate showed a marked increase as the pH decreased. Overall, double response functions of MSN-S-S-CS had unique advantages in controlled drug delivery, and may be a new clinical application of DDS in cancer therapy. Keywords:mesoporous silica nanoparticles;double response;drug delivery;drug release

The metabolite profiles and distributions of procyanidin B2 were qualitatively described using UPLC-DAD-ESI-IT-TOF-MSn without help of reference standards, and a possible metabolic pathway was proposed in the present study. Summarily, 53 metabolites (24 new metabolites) were detected as metabolites of procyanidin B2, and 45 of them were tentatively identified. Twenty seven metabolites were assigned as similar metabolites of (−)-epicatechin by scission of the flavanol interflavanic bond C4–C8, including 16 aromatic metabolites, 5 conjugated metabolites, 3 ring-cleavage metabolites, and 2 phenylvalerolactone metabolites. Additionally, 14 metabolites were conjugates of free procyanidin B2, comprising 9 methylation metabolites, 8 sulfation metabolites, 5 hydration metabolites, 2 hydroxylation metabolites, 1 hydrogenation metabolites, and 1 glucuronidation metabolites. The results of metabolite distributions in organs indicated that the conjugated reaction of free procyanidin B2 mainly occurred in liver and diversified metabolites forms were observed in small intestine. The metabolic components of procyanidin B2 identified in mice provided useful information for further study of the bioactivity and mechanism of its action.
