Metabolomics analysis based on UHPLC-Q-TOF-MS/MS reveals effects of genistein on reducing mycotoxin citrinin production by Monascus aurantiacus Li AS3.4384

Shanshan He,Xin Liu,Yanling Wang,Jianhua Xie,Heng Gao,Xiujiang Li,Zhibing Huang

Monascus mold produces useful bioactive compounds such as Monascus pigments, monacolin K, and ergosterol . In this study, metabolomics analysis was used to investigate the metabolic profile changes in Monascus aurantiacus Li AS3.4384 (MALA) induced by genistein at different fermentation times using UHPLC-Q-TOF-MS/MS. The addition of genistein was found to not only reduce citrinin but also suppress significant differential metabolites. A total of 21 significant differential metabolites were involved in the most significant metabolic pathways (impact >0.1). These metabolites affected the aspartic acid , glutamic acid pathway, tricarboxylic acid (TCA) cycle, pyruvate synthesis pathway, and lysine degradation pathway. Thus, the addition of genistein may change the precursor for citrinin synthesis of Monascus . These findings provide significant insight into the Monascus metabolic pathways affected by genistein and will help to improve the quality control and safety of Monascus in food and supplements.

Analysis and Comparison of Lipids in Monascus-Fermented Cheese from Different Ripening Periods Based on UHPLC-QTRAP MS Quantitative Lipidomics

Yadong Wang,Qian Zhang,Junaid Yousaf,Fanyu Meng,Yanbo Wang,Bei Wang

Monascus-fermented cheese (MC) is a new cheese product that undergoes a multistrain fermentation process, which results in unique flavor qualities. Lipid metabolism plays an important role in the flavor formation of MC. To clarify the lipid composition of MC and the changes in lipid composition at four ripening periods, this study resolved the lipid profiles of MC by using UHPLC-QTRAP MS. The results identified 21 lipid classes and 748 lipid species, 72 of which were significantly different (VIP > 1, p < 0.01, |log 2FC| > 1). Additionally, 83 relevant metabolic pathways were enriched. Based on the enriched metabolic pathways and lipid changes, the pathways of differential lipid formation in MC were mapped. The results of this study comprehensively characterize the changes in lipids during the ripening process of MC, which can be used in the future to deeply analyze the mechanism of flavor formation in MC.

Development of soy protein isolate/κ-carrageenan composite hydrogels as a delivery system for hydrophilic compounds: Monascus yellow

Qian Zhang,Luping Gu,Yujie Su,Cuihua Chang,Yanjun Yang,Junhua Li

The aim of the present study was to develop soy protein isolate (SPI) and κ-carrageenan (KC) composite hydrogels as a delivery system for hydrophilic compounds. The pigment of monascus yellow was used as a model. A systematic study was performed to characterize the rheological, textural, microstructural properties and in vitro digestion release profile of monascus yellow of the composite gels. The results of power law modeling, electrophoresis patterns and fourier transform infrared spectroscopy (FTIR) confirmed that non-covalent interactions were involved in the formation of SPI/KC composite hydrogels. Compared to pure κ-carrageenan hydrogels, the incorporation of SPI could promote the formation of tougher, more uniform and compact composite gels with sustained-release property. In addition, the release behaviors of monascus yellow entrapped in the hydrogel network can be well described by the Ritger-Peppas mathematical model. Overall, our study provided a promising strategy to enhance the sustained release performance of hydrogels in digestive conditions.

Merging of a chemical reaction with microbial metabolism via inverse phase transfer catalysis for efficient production of red Monascus pigments

Bin Gu,Haisheng Xie,Xuehong Zhang,Zhilong Wang

Both chemical catalysis and microbial fermentation are essential ways to produce small molecules related to our everyday lives. The apparent mismatch between the approaches used in synthetic organic chemistry and the culture conditions required to support living micro-organisms remains one of the largest challenges in combining chemical catalysis with microbial fermentation in a one-pot procedure. In the present work, microbial fermentation of red Monascus pigments, which involves biosynthesis of orange Monascus pigments and non-enzymatic chemical conversion of hydrophobic orange Monascus pigments into red ones, was set up as a model. Inverse phase transfer catalysis (IPTC) consisting of a water–tetradecane two-phase system with hydroxypropyl-β-cyclodextrin as an inverse phase transfer catalyst was exploited to accelerate the chemical modification of hydrophobic orange Monascus pigments. At the same time, Monascus aurantiacus exhibited excellent biocompatibility in this system. Thus, microbial fermentation and further chemical modification of orange Monascus pigments had been merged successfully in one-pot by IPTC, where the shift of the microbial metabolic flux to orange Monascus pigments (almost undetectable yellow Monascus pigments) and efficient production of red Monascus pigments (corresponding to nearly 2 g l−1) were achieved. This is the first report about exploitation of IPTC as a biocompatible medium for merging of a chemical reaction with microbial metabolism.

Monascus pilosus YX-1125: An efficient digester for directly treating ultra-high-strength liquor wastewater and producing short-chain fatty acids under multiple-stress conditions

Yuxi Zheng,Tianyuan Zhang,Yun Lu,Li'ao Wang

Ultra-high-strength liquor wastewater (UHS-LWW) is rich in organic matter, and the required treatment is expensive. Here, an extremophilic strain Monascus pilosus YX-1125 was isolated for the direct conversion of UHS-LWW to short-chain fatty acids (SCFAs). Strain YX-1125 is an efficient SCFA producer with carbohydrate metabolic flexibility under multiple-stress conditions. Moreover, strain YX-1125 could tolerate up to 75 g/L, 100 g/L, and 50 g/L of ethanol, organic acids, and salt, respectively, without inhibition. In repeated-cycle fermentations, 17.8 g/L of butyric acid and 2.0 g/L of propionic acid were produced from UHS-LWW at the fifth cycle, which are the highest concentrations of wastewater-derived SCFAs reported to date. After SCFA recovery, a 98.9% COD reduction was achieved, which is estimated to reduce treatment costs by 91.7%. Results indicate that M. pilosus YX-1125 is a promising strain for the direct treatment of UHS-LWW, and for converting it into valuable biochemicals without any pre-treatment.

Monascus Yellow Pigments Ameliorate Hyperuricemia via Dual Mechanisms: Xanthine Oxidase Inhibition and Uric Acid Transporter Modulation (ABCG2, URAT1, and GLUT9)

Furong Xue,Renqin Zhu,Jiaxing Li,Zheng Liu,Lidan Niu,Wei Chen,Chengtao Wang,Jie Zheng

The increasing global prevalence of hyperuricemia (HUA), particularly among younger populations, underscores the urgent need for safe and effective dietary interventions.Monascusfungi, long utilized in East Asian food culture, ferment rice to produce red yeast rice (RYR), a functional food rich in monacolin K and Monascus pigments. Among these,Monascusyellow pigments (MYPs)—natural azaphilone compounds used as food additives and colorants—have shown antioxidant, anti-inflammatory, and metabolic regulatory activities. However, their potential to alleviate hyperuricemia remains unexplored. This study investigates the urate-lowering and organ-protective effects of MYPs through a combination of in vitro, in vivo, and gut microbiota analyses. MYPs exhibited significant xanthine oxidase (XOD) inhibitory activity, and molecular docking confirmed that monascin (MS) and ankaflavin (AK) competitively bind to the XOD active site. In a murine HUA model, MYPs significantly reduced serum uric acid (SUA) levels without causing hepatic or renal toxicity. Mechanistically, MYPs downregulated renal UA reabsorption transporters (URAT1, GLUT9) and upregulated the excretory transporter ABCG2, enhancing uric acid (UA) excretion. These findings highlight MYPs as promising food-derived bioactives with dual XOD inhibition and uricosuric effects, offering a novel nutraceutical strategy for hyperuricemia prevention and management.

Evaluating Antitumor and Antioxidant Activities of Yellow Monascus Pigments from Monascus ruber Fermentation

Hailing Tan,Ziyi Xing,Gong Chen,Xiaofei Tian,Zhenqiang Wu

YellowMonascuspigments can be of two kinds: Natural and reduced, in which natural yellowMonascuspigments (NYMPs) attract widespread attention for their bioactivities. In this study, the antioxidative and antibreast cancer effects of the water-soluble NYMPs fermented byMonascus ruberCGMCC 10910 were evaluated. Results showed that water-soluble NYMPs had a significantly improved antioxidative activities compared to the reduced yellowMonascuspigments (RYMPs) that were chemically derived from orange or redMonascuspigments. Furthermore, NYMPs exhibited a concentration-dependent inhibition activity on MCF-7 cell growth (p< 0.001). After a 48-h incubation, a 26.52% inhibition yield was determined with 32 μg/mL of NYMPs. NYMPs also significantly inhibited the migration and invasion of MCF-7 cells. Mechanisms of the activities were associated with a down-regulation of the expression of matrix metalloproteinases and vascular endothelial growth factor. Rather than being alternatively used as natural colorants or antioxidants, this work suggested that NYMPs could be selected as potential functional additives in further test of breast cancer prevention and adjuvant therapy. Keywords:natural yellowMonascuspigments;water-soluble;antioxidation;migration;invasion;MCF-7 cells

Analysis of metabolites of coix seed fermented by Monascus purpureus

Qi Mei,Zhaona Xu,Quanye Wu,Likang Qin,Haiying Zeng,Yi Zhu

In this study, an untargeted metabolomics method based on LC-MS was used to investigate the differential metabolites of coix seed after fermentation by Monascus purpureus . The compounds with highly correlated variable importance in projection (VIP) values of orthogonal partial least squares discriminant analysis (OPLS-DA) were selected as the important metabolites of coix seed samples. Coix seed was fermented by M. purpureus for 9 d. The color value units (CVU) of pigment in fermented coix seed reached 157.47 CVU/g, and the contents of total phenols and total flavonoids were 0.32 mg/g and 1.2 mg/g, respectively. A total of 270 differential metabolites were detected, including amino acids, peptides, fatty acids, phenols, steroids, and their derivatives, of which the relative contents of 187 metabolites showed a significant upward trend (p < 0.05). Amino acids, peptides and analogues accounted for the largest proportion at 26%, among which most metabolites were enriched. For example, N-alpha-acetyllysine, l -tryptophan and l -arginine exhibited a fold change (FC) of 6200.87, 330.29, and 35.59, respectively. In particular, the detected steroid metabolites accounted for 3% of the total metabolites, and they all participated in the same pathway, in which androsterone, 2-methoxyestrone and etiocholanolone were upregulated. The synthesis of these steroids was related to the precursors and enzymes in the pathway, such as dehydroepiandrosterone, androstenedione, and 3beta-hydroxy-delta5-steroid dehydrogenase (HSD3B). The results of this study reveal M. purpureus fermentation can successfully transform coix seed, improving the functional activities of metabolites. Thus, this process has high development value with important applications in the food industry.

Monascus pigment-protected bone marrow-derived stem cells for heart failure treatment

Tian Yue,Wentai Zhang,Haifeng Pei,Dunzhu Danzeng,Jian He,Jiali Yang,Yong Luo,Zhen Zhang,Shiqiang Xiong,Xiangbo Yang,Qisen Ji,Zhilu Yang,Jun Hou

Mesenchymal stem cells (MSCs) have demonstrated significant therapeutic potential in heart failure (HF) treatment. However, their clinical application is impeded by low retention rate and low cellular activity of MSCs caused by high inflammatory and reactive oxygen species (ROS) microenvironment. In this study, monascus pigment (MP) nanoparticle (PPM) was proposed for improving adverse microenvironment and assisting in transplantation of bone marrow-derived MSCs (BMSCs). Meanwhile, in order to load PPM and reduce the mechanical damage of BMSCs, injectable hydrogels based on Schiff base cross-linking were prepared. The PPM displays ROS-scavenging and macrophage phenotype-regulating capabilities, significantly enhancing BMSCs survival and activity in HF microenvironment. This hydrogel demonstrates superior biocompatibility, injectability, and tissue adhesion. With the synergistic effects of injectable, adhesive hydrogel and the microenvironment-modulating properties of MP, cardiac function was effectively improved in the pericardial sac of rats. Our results offer insights into advancing BMSCs-based HF therapies and their clinical applications.

Role of genistein on the yield, structure and immunomodulatory activity of Monascus exopolysaccharides

Liuming Xie,Zhibing Huang,Hui Meng,Zhibing Fan,Xiaoyi Shi,Jianhua Xie

Manipulating the structures, physicochemical properties, and monosaccharide compositions of exopolysaccharides (EPS) isolated from microorganisms has been reported to enhance their biological activities. Hence, the aim of this work was to examine the effects of genistein addition during fermentation on the amount, physicochemical properties, and immunomodulatory activity of EPS secreted by M. purpureus. Results showed that genistein addition significantly increased M. purpureus biomass and EPS yield to 2.42 g L−1 and 6.08 g L−1, respectively, and affected the physicochemical properties and structures of EPS. Furthermore, EPS produced by genistein-treated M. purpureus (G-EMP) improved the immunomodulatory activity of RAW264.7 macrophages by increasing the secretion of nitric oxide and cytokines. Moreover, phospho-Jun N-terminal kinase (p-JNK), phospho-extracellular regulated protein kinase (p-ERK), phospho-p38 (p-p38) mitogen-activated protein kinase (MAPK) and nuclear factor-kappa B (NF-κB) phospho-p65 (p65) proteins were remarkably upregulated by G-EMP stimulation, blocking Toll-like receptor 4 (TLR4) that dramatically reduced the pinocytic and phagocytic capacities. Overall, these findings provide potential rationales for the application of genistein in improving the EPS yield of M. purpureus.

Monascus fermentation enhances the lipid-lowering properties of ginseng in vivo and component assay

Qingsong Qu,Qing Zhou,Xinhui Peng,Zhixun Li,Mingyue Jia,Yingying Zhang,Cuiting Yang,Xingxing Dai,Mengke Sheng,Jichuan Kang,Xinyuan Shi

Hyperlipidemia is a global health concern characterized by elevated lipid levels and increased cardiovascular risk. Ginseng ( Panax ginseng ) has lipid-lowering properties, and Monascus fermentation can yield monacolin K, a natural statin. However, it remains unclear whether Monascus -fermented ginseng (MFG) offers synergistic benefits over ginseng or Monascus alone. This study aimed to compare the lipid-lowering efficacy of MFG versus unfermented ginseng, Monascus , and their non-fermented combination in hyperlipidemic mice. Multi-omics analyses (hepatic transcriptomics, serum/fecal metabolomics, and gut microbiota profiling) were integrated to elucidate underlying mechanisms, and fecal microbiota transplantation (FMT) assessed the causal role of MFG-altered microbiota. MFG supplementation markedly improved serum lipid profiles and attenuated hepatic steatosis, outperforming ginseng, Monascus , or their unfermented combination. Phytochemical analysis confirmed that fermentation introduced monacolin K into MFG and increased its rare ginsenoside content, distinguishing it from unfermented ginseng. Mechanistically, MFG increased hepatic p-AMPK and upregulated fatty acid oxidation while downregulating lipogenesis (SREBP-1c, FAS). Metabolomics revealed elevated short-chain fatty acid levels and enhanced bile acid biotransformation in MFG-treated mice. MFG reshaped gut microbiota composition and enriched beneficial taxa. Importantly, FMT from MFG-fed donors recapitulated the lipid-lowering effects in recipients, confirming a microbiota-mediated mechanism. In conclusion, MFG exerts superior lipid-lowering effects through complementary host metabolic and gut microbial modulation, highlighting fermented herbal products as promising interventions against hyperlipidemia.

Synthesis and characterization of functional Monascus yellow pigment-tannic acid complexes via Co-pigmentation

Kaize Zheng,Xueling Qin,Lingchun Meng,Yusheng Liang,Junwei Qin,Zhenqiang Wu,Zhenfeng Huang

Natural Monascus yellow pigments (MYPs) have potential in the food industry due to their safety, but their application is limited by low biological activity and stability. Novel MYPs complexes, Monascus yellow pigments-tannic acid (MYPs-TA), were synthesized by spontaneously incorporating TA through intermolecular co-pigmentation in this work. Structural analysis revealed that multiple interactions contributed to the formation of complexes, including hydrogen bonding, π-π stacking, and hydrophobic interactions. The MYPs-TA exhibited characteristic ultraviolet signals of both MYPs (380 nm) and TA (276 nm), and the crystalline structure of MYPs transitioned to an amorphous form. These phenomena confirmed the tight combination of MYPs and TA. Color tonality, physicochemical stability as well as antimicrobial and antioxidant activities of MYPs were significantly improved after interaction with TA. Besides, MYPs-TA exhibited optimal stability and functionality in high-temperature processed foods such as baked goods or pasteurized products. This study provides a novel approach for synthesizing functional MYPs complexes with superior stability.
