Cadmium, a bluish-white metallic, possesses a prolonged half-life in organisms, causing damage to multiple tissues and organs. Naringenin, a natural flavonoid antioxidant, mitigates cadmium-induced damage in organisms. However, the mechanism by which naringenin attenuates Cd-associated hepatocellular damage has not been fully elucidated. The present investigation consequently aimed to elucidate the mechanistic involvement of autophagy in cadmium hepatotoxicity using the rat model, with parallel assessment of naringenin's hepatoprotective efficacy. In this experiment, 24 SD rats underwent randomization to four experimental groups: (1) control (intraperitoneal saline injection), (2) Cd (1 mg/kg b.w. CdCl₂ intraperitoneal injection), (3) Cd + Nar (1 mg/kg b.w. CdCl₂ injection + 50 mg/kg b.w. naringenin oral gavage), and (4) Nar (50 mg/kg b.w. naringenin oral gavage) groups. After the 14-day interventions, serum and liver tissues were collected post-euthanasia. Hepatic injury markers (AST and ALT) and antioxidant enzymes (CAT and SOD) were quantified. Histopathology utilized HE and TUNEL staining. RT-qPCR and western blot analyses determined mRNA and protein expression levels of autophagy-related factors (p62 and LC3) and apoptosis-related factor (Caspase-3). The results demonstrated that cadmium exposure significantly reduced body weight, increased relative liver weight, elevated serum AST/ALT levels, and diminished hepatic CAT/SOD activity. Cadmium significantly upregulated both mRNA and protein expression levels of p62 and Caspase-3 while suppressing LC3 expression. Naringenin co-administration attenuated cadmium-induced hepatic injury, oxidative stress, impaired autophagy, and enhanced apoptosis. These findings collectively demonstrate that cadmium exposure induces hepatic injury and oxidative stress in rats through autophagy inhibition and apoptotic activation, while naringenin exerts protective effects by modulating these pathological processes.

Naringenin (NRG), a flavanone compound present in citrus fruits, has a variety of beneficial physiological active functions such as antioxidant, anti-inflammatory, and hypoglycaemic. In this study, an sensor was constructed by electrodeposition and used for the electrochemical study of NRG. Reduced graphene oxide (rGO) and glutathione (GSH) showed the ability to synergistically amplify NRG signals and possessed good linearity in the concentration range of 10.00–1200.00 μmol l−1. The linear equation is Ip = 0.0776logc + 0.9353 (R2 = 0.9901), and the limit of detection is 3.33 μmol l−1. The sensor performed well in terms of reproducibility, stability, and selectivity, which in turn enabled the detection of NRG in tomatoes. The average recovery of the sensor is 95.68% to 111.92%, with RSD less than 11.89%. The results were also verified by Ultraviolet–visible spectroscopy(UV-vis). Furthermore, density-functional theory was employed to analyse the front track of the NRG, speculating that the NRG underwent a transfer of two electrons and two protons.

Naringenin (NAR) exhibits many biological and pharmacological activities. However, its poor water-solubility and dissolution rate restrict its oral-intestinal absorption rate and bioavailability. In order to improve its dissolution rate, micronized NAR was successfully prepared using a supercritical anti-solvent process. A mixed acetone/dichloromethane solution and CO 2 were used as the solvent and anti-solvent, respectively. The optimal temperature (35 °C), pressure (12 MPa), drug concentration (15 mg/mL), solution flow rate (1 mL/min), and volume percentage of dichloromethane (45%) were determined using a single-factor method. Under optimal conditions, a mean particle size of 611 ± 32 nm was obtained. XRD and FTIR analyses indicated that the unprocessed NAR and micronized NAR using solution-enhanced dispersion by supercritical CO 2 (SEDS) processes exhibited the same crystal structure and chemical structure, and the samples showed similar DSC thermograms. An in vitro dissolution test showed that micronization significantly enhanced the dissolution of NAR.

Triple-negative breast cancer (TNBC) represents a highly aggressive subtype with limited treatment options, primarily relying on chemotherapy. Emerging therapies, including photothermal therapy (PTT) and related combination therapies, are considered highly promising. Naringenin is a flavonoid glycoside with diverse biological activities can enhance immune responses as well as inhibit cancer cell proliferation. We developed photothermal bovine serum albumin (BSA)-biomineralized MnO 2 nanoparticles (MnO 2 @BSA) by directly reducing KMnO 4 with BSA, which were then combined with naringenin to achieve immuno-enhanced PTT for TNBC. The results show that MnO 2 @BSA exhibits strong light absorption and tumor-targeting capabilities. Naringenin and PTT together significantly suppress TNBC cell proliferation and migration while enhancing antitumor immune responses. In vivo experiments confirm that the combined therapy reduces tumor growth and metastatic risk compared to monotherapy. Additionally, the treatment exhibits a favorable safety profile, offering potential for improved clinical outcomes in breast cancer treatment.

Abstract Naringenin, an herbal ingredient possesses a diversity of pharmacological action and bioactive functions. In this study, surface molecularly imprinted polymers named as Zeolite imidazolate framework-8 were successfully synthesized and applied in the selective adsorption of naringenin. A series of adsorption experiments evaluated the adsorption properties of polymers. The results showed that the molecularly imprinted polymers had a higher adsorption capacity than non-imprinted polymers. In addition, they have good selectivity for Naringenin compared with other flavonoid compounds, and its imprinting factor for naringenin is as high as 3 in single solutions. The Scatchard plots showed two distinct linear portions, indicating that two kinds of binding sites exist in the molecularly imprinted polymers. After eight cycles, the adsorption capacity of polymers did not decrease significantly in the initial affinity. These results demonstrated that prepared molecularly imprinted polymers could be promising functional materials for the selective adsorption of naringenin.

Drug cocrystals can modulate the physical and chemical properties of drugs, even complement each other’s strengths, and have a synergistic enhancement effect. Here, we reported a drug cocrystal prepared by norfloxacin (NOR) and naringenin (NAR), comprising an antibacterial drug and an antitumor active pharmaceutical ingredient. The cocrystal hydrate obtained by crystal transformation methods enhanced the antibacterial activity of NOR against one Gram-positive bacterium (Staphylococcus aureus), three Gram-negative bacteria (Escherichia coli, Dysentery bacillus, Pseudomonas aeruginosa), and one fungus (Candida albicans), improved the solubility of NAR, reduced the hygroscopicity of NOR, and enhanced the anticancer activity of NAR against human breast cancer cells MDA-MB-231. This combination has potential therapeutic synergy in clinical terms.

The antimicrobial mechanism of naringenin, one of the citrus antibacterial flavonoids against food-borne Staphylococcus aureus ATCC 6538, was investigated in this study. Analysis of gas chromatography-mass spectrometry (GC-MS) and fluorescence showed that relatively low concentrations of naringenin caused perturbations in the membrane fatty acid composition and the conformation of membrane proteins through changing the microenvironment of phenylalanine, tyrosine, and tryptophan residues. Exposure of naringenin at higher levels significantly increased membrane permeability and changed the morphology of S. aureus cells. The genomic DNA-binding of naringenin was also quantitatively monitored using UV-vis spectra in combination with multivariate curve resolution-alternating least squares (MCR-ALS) analysis, and the concentration and pure spectra profiles for the three reaction species (DNA, naringenin, and DNA–naringenin) were obtained. Moreover, the thermal behavior of DNA and docking studies revealed that naringenin preferentially bound to the A–T base pair regions of genomic DNA via groove binding, and atomic force microscopy and circular dichroism showed that naringenin induced mild secondary structure and obvious morphological variations of this biomacromolecule. These results suggested that naringenin exerting its antibacterial effects might be connected with disruption of the cytoplasmic membrane and DNA targeting effects in Staphylococcus aureus.

Introduction: With the rapid development of nanotechnology, the research and development of nano-drugs have become one of the development directions of drug innovation. The encapsulation of the nanoparticles can change the biological distribution of the drug in vivo and improve
the bioavailability of the drug in vivo. Naringenin is poorly soluble in water and has a low bioavailability, thus limiting its clinical application. The main purpose of this study was to develop a nano-sized preparation that could improve the oral bioavailability of naringenin.
Methods: Chitosan oligosaccharide modified naringenin-loaded bovine serum albumin nanoparticles (BSA-COS@Nar NPs) were prepared by emulsification solvent evaporation and electrostatic interaction. The nanoparticles were characterized by HPLC, laser particle size analyzer, transmission electron
microscope and X-ray diffraction analysis. The release in vitro was investigated, and the behavior of nanoparticles in rats was also studied. The caco-2 cell model was established in vitro to investigate the cytotoxicity and cellular uptake of nanoparticles. Results: BSA-COS@Nar
NPs were successfully prepared, and the first-order release model was confirmed in vitro release. In vivo pharmacokinetic results indicated that the area under the drug concentration- time curve (AUC) of BSA-COS@Nar NPs was 2.37 times more than free naringenin. Cytotoxicity and
cellular uptake results showed that BSA-COS@Nar NPs had no significant cytotoxic effect on Caco- 2 cells and promoted cellular uptake of the drug. Conclusion: BSA-COS@Nar NPs could improve the in vivo bioavailability of naringenin.

Atherosclerosis is the leading cause of global cardiovascular morbidity and mortality associated with inflammatory and immunological mechanisms. Immunotherapy has demonstrated promising efficacy in the management of atherosclerosis. Nevertheless, certain immunotherapeutic approaches are associated with limitations, including suboptimal efficacy and non-negligible adverse effects. Upon the pivotal role of macrophage phenotypes in atherosclerosis progression, naringenin-loaded manganese-doped mesoporous silica nanoparticles (MMSN@NAR) were designed and synthesized to reprogram M1 macrophages toward the M2 phenotype, thereby offering a potential therapeutic strategy for atherosclerosis treatment. High loading capacity of naringenin was achieved in MMSN carriers, with superior biocompatibility profiles compared to naringenin dissolved in dimethyl sulfoxide, while maintaining pH-dependent release behavior as demonstrated by dialysis assays. MMSN@NAR is preferentially phagocytosed by M1 macrophages, attenuates inflammatory responses, protects against oxidative stress, and promotes M2 polarization via the AMP-activated protein kinase (AMPK) pathway in vitro. In the ApoE −/− mouse unilateral carotid artery ligation model of atherosclerosis, MMSN@NAR demonstrated marked accumulation in plaques and excellent biocompatibility. Compared to using naringenin or MMSN alone, it could further reduce plaque area by approximately 40% or 60% by inducing macrophage phenotype transformation, which was confirmed by section staining and immunofluorescence. Collectively, this study highlights enhanced macrophage M2 polarization inhibiting atherosclerosis by MMSN@NAR as a promising nanoplatform, offering a novel therapeutic approach based on anti-inflammatory immune regulation.

Polyphenols have various functions to improve the nutritional value, stability and organoleptic properties of foods. The purpose of this study is to explore the effects of adding naringenin on physicochemical properties of potato starch. Nuclear magnetic resonance spectroscopy and X-ray photoelectron spectroscopy showed successful combination of naringenin and potato starch. Through thermogravimetric and DSC analysis, it was found that the thermal stability of potato starch was enhanced due to the reordering of molecules after gelatinization in the temperature range of 25–160 °C, while the thermal stability of the gelatinized potato starch decreased at around 300 °C. The experimental results showed that the pasteurisation process and the amount of naringenin added led to different changes in the swelling power and solubility of the samples. The elasticity, reversibility, hardness, chewability and gelling properties of the gel samples were lower than those of potato starch. The potato starch-naringenin composite granules were found to have a higher content of resistant starch (c = 8%–20%) by digestive characterisation. There was a significant impact of naringenin on the properties of potato starch, showing potential application prospects in food industry. This study provides a basis for the application of naringenin in potato starch-based food formula.

Naringenin (Nar), a dihydroflavone, is widely used in medicine, food industry, and other fields. In this study, naringenin magnetic surface molecularly imprinted polymers (Nar/SMIPs) were prepared by etching nanomagnetic carriers and using computer-aided material design for highly selective adsorption of naringenin. The Nar/SMIPs exhibited a high adsorption capacity of 37.64 mg·g–1 in 30 min. They also showed excellent selectivity for naringenin, with an imprinting factor of 2.48, and the selection coefficients (K) for structurally similar compounds, diosmetin, chrysin, and alizarin, were 3.27, 2.82, and 3.07, respectively. Furthermore, they demonstrated high recoveries (89.47–95.95%) and precisions (relative standard deviation: 1.38–1.90%) in grapefruit peel extract. Thus, Nar/SMIPs hold tremendous potential for the selective extraction of naringenin.

Polymethoxyflavones (PMFs) are a distinct class of plant flavonoids with significant therapeutic potential, particularly as anti-cancer agents. This potent bioactivity is largely attributable to O-methylation. However, a major bottleneck for the industrial-scale production of PMFs is the lack of suitable precursors and available O-methyltransferases (OMTs), which are essential for constructing a viable biosynthetic pathway. Here, we discover a caffeic acid O-methyltransferase (CsOMT5) whose transcriptional dynamics closely mirrored the PMF levels in the flavedo of sweet orange (Citrus sinensis). In vivo validation further verified that CsOMT5 indeed promoted PMF accumulation in citrus. And its recombinant protein possessed broad substrate promiscuity for various PMF intermediates. Notably, CsOMT5 exhibited specialized C5-regioselectivity for naringenin, a readily available precursor for de novo synthesis of PMFs. Furthermore, critical residues involved this methylation activity (N14, T18, I120, I256, and G305) were revealed through mutational analysis. This study not only provides deeper insights into the PMF biosynthetic pathway but also opens new avenues for the industrial biosynthesis of PMFs.
