Oil-water separation membranes with excellent performance are crucial for addressing the issue of marine oil pollution and treating industrial wastewater. To overcome the limitations of traditional polymeric membranes, such as large pore sizes and limited emulsion selectivity, an asymmetric Janus nanofibrous membrane with switchable separation capabilities for oil–water emulsions was developed through a synergistic combination of layer-by-layer electrospinning and electrospraying techniques. The Janus membrane consists of a hydrophilic layer with a fine pore structure (0.46 μ m) and a hydrophobic layer composed of nano-nets connected by nanofibers (56 nm) and microspheres. The distinctive structural configuration endows the membrane with dual functionality: the hydrophilic surface demonstrates superhydrophilicity (0°) and underwater oleophobicity (140°), facilitating efficient oil-in-water emulsions separation with a flux of 4233 L m -2 h −1 and a separation efficiency of 99.36 % at 5 kPa. Conversely, the hydrophobic surface exhibits superoleophilicity (0°) and underwater superhydrophobicity (>150°), achieving permeation flux of 1887 L m -2 h −1 with 99.32 % separation efficiency for water-in-oil emulsions under gravity. Furthermore, the membrane demonstrates unidirectional moisture transport capability, superior mechanical strength, and recyclability.

Background CD276 (B7-H3), a pivotal immune checkpoint, facilitates tumorigenicity, invasiveness, and metastasis by escaping immune surveillance in a variety of tumors; however, the underlying mechanisms facilitating immune escape in esophageal squamous cell carcinoma (ESCC) remain enigmatic. Methods We investigated the expression of CD276 in ESCC tissues from patients by using immunohistochemistry (IHC) assays. In vivo, we established a 4-nitroquinoline 1-oxide (4NQO)-induced CD276 knockout (CD276 wKO ) and K14cre; CD276 conditional knockout (CD276 cKO ) mouse model of ESCC to study the functional role of CD276 in ESCC. Furthermore, we used the 4NQO-induced mouse model to evaluate the effects of anti-CXCL1 antibodies, anti-Ly6G antibodies, anti-NK1.1 antibodies, and GSK484 inhibitors on tumor growth. Moreover, IHC, flow cytometry, and immunofluorescence techniques were employed to measure immune cell proportions in ESCC. In addition, we conducted single-cell RNA sequencing analysis to examine the alterations in tumor microenvironment following CD276 depletion. Results In this study, we elucidate that CD276 is markedly upregulated in ESCC, correlating with poor prognosis. In vivo, our results indicate that depletion of CD276 inhibits tumorigenesis and progression of ESCC. Furthermore, conditional knockout of CD276 in epithelial cells engenders a significant downregulation of CXCL1, consequently reducing the formation of neutrophil extracellular trap networks (NETs) via the CXCL1–CXCR2 signaling axis, while simultaneously augmenting natural killer (NK) cells. In addition, overexpression of CD276 promotes tumorigenesis via increasing NETs’ formation and reducing NK cells in vivo. Conclusions This study successfully elucidates the functional role of CD276 in ESCC. Our comprehensive analysis uncovers the significant role of CD276 in modulating immune surveillance mechanisms in ESCC, thereby suggesting that targeting CD276 might serve as a potential therapeutic approach for ESCC treatment.

For the first time, two-dimensional (2D) gelatin nano-nets are fabricated by regulating the solution properties and several process parameters during electrospinning/electro-netting. The spider-web-like nano-nets that comprise interlinked one-dimensional (1D) ultrathin nanowires (10–35 nm) are stacked layer-by-layer and widely distributed in the three-dimensional (3D) porous membranes. The final morphology of the gelatin nano-nets, including nanowire diameter, area density and pore-width of the nano-nets, is highly dependent on the solution concentration, salt concentration, kinds of solvents, applied voltage, ambient temperature and relative humidity (RH). The occurrence of rapid phase separation on the splitting-film and the formation of hydrogen bond among gelatin molecules during electro-netting are proposed as the possible mechanisms for the formation of these spider-web-like nano-nets.

The overexpression of neutrophil extracellular traps (NETs) serves as a critical biomarker and mediator of tumorigenesis and metastasis. By inhibiting their formation or degrading existing structures, therapeutic strategies targeting NETs have shown promising potential in cancer treatment and metastasis prevention. Deoxyribonuclease I (DNase), while effective in NETs degradation, suffers from rapid plasma clearance, necessitating delivery vehicles to prolong its therapeutic activity. In this study, we developed superparamagnetic Fe 3 O 4 nanoclusters via a one-step solvothermal method for co-delivery of doxorubicin (DOX) and DNase. The resulting nanoplatform demonstrated: (1) enhanced magnetic retention at tumor sites for improved local drug concentration and targeted delivery, and (2) synergistic therapeutic effects under alternating magnetic field (AMF) exposure, combining magnetic hyperthermia with chemotherapy while simultaneously remodeling the tumor microenvironment through NETs degradation. Comprehensive evaluations revealed that this combined approach significantly enhanced antitumor efficacy compared to monotherapies, with the additional benefit of metastasis prevention through tumor microenvironment modulation. This multifunctional nanoplatform thus represents a promising strategy for synergistic cancer therapy, addressing both primary tumor control and metastatic suppression.

Summary The pathogenesis of hyperalgesia is complex and can lead to poor clinical treatment. Our study revealed that epididymal white adipose tissue (eWAT) from spared nerve injury (SNI) mice is involved in the occurrence of hyperalgesia after adipose tissue transplantation. We also showed that lysophosphatidylcholine (LPC) is enriched in the eWAT of SNI mice using non-targeted metabolomic analysis and verified that the levels of LPC in plasma and the anterior cingulate cortex (ACC) region increased following eWAT transplantation. Based on the immunohistochemistry results, we observed that LPC in the ACC region activated microglia via the TRPV1/CamkⅡ pathway. Meanwhile, the disruption of perineuronal nets (PNNs) around PV + neurons in ACC promoted hyperalgesia, and the loss of PNNs and PV + interneurons might be due to microglial phagocytosis. These findings elucidate the mechanism underlying hyperalgesia from the perspective of lipid metabolite LPC and PNNs and provide potential strategies for the treatment of hyperalgesia.

Neutrophils can capture and kill pathogens by releasing neutrophils extracellular traps (NETs), which play critical roles in anti-microbial infection in mammals; however, the mechanisms involved in NETs formation and its role in anti-bacterial infection in teleost fish remains largely unknown. In this study, to explore the function of NETs in turbot , we established an in vitro bacterial infection model in head kidney derived neutrophils, and found that the haemolysin over-expressed Edwardsiella piscicida ( ethA + ) could induce a robust phenotype of NETs, compared with that in wild type or ethA mutant ( ethA + - ΔethA ) strains. Besides, the NETosis was mediated by ethA + -induced pyroptosis , and arms the ability of bacterial killing in neutrophils of turbot. Moreover, we found that neutrophils elastase (NE) might involves in this pyroptotic signaling, rather than inflammatory Sm caspase . Taken together, this study reveals the important role of pyroptosis in NETs formation in turbot neutrophils, suggesting that NETs formation is a critical immune response during bacterial infection in teleost fish.

Effective promotion of air filtration applications proposed for fibers requires their real nanoscale diameter, optimized pore structure, and high service strength; however, creating such filter medium has proved to be a tremendous challenge. This study first establishes a strategy to design and fabricate novel poly( m -phenylene isophthalamide) nanofiber/nets (PMIA NF/N) air filter via electrospinning/netting. Our strategy results in generation of a bimodal structure including a scaffold of nanofibers and abundant two-dimensional ultrathin (~20 nm) nanonets to synchronously construct PMIA filters by combining solution optimization, humidity regulation, and additive inspiration. Benefiting from the structural features including the true nanoscale diameter, small pore size, high porosity, and nets bonding contributed by the widely distributed nanonets, our PMIA NF/N filter exhibits the integrated properties of superlight weight (0.365 g m −2 ), ultrathin thickness (~0.5 μm), and high tensile strength (72.8 MPa) for effective air filtration, achieving the ultra-low penetration air filter level of 99.999% and low pressure drop of 92 Pa for 300–500 nm particles by sieving mechanism. The successful synthesis of PMIA NF/N would not only provide a promising medium for particle filtration, but also develop a versatile platform for exploring the application of nanonets in structural enhancement, separation and purification.

A novel airborne particulate filtration medium, consisting of a two-tier composite structure, i.e., a nano-fiber/net (NFN) top layer and a conventional nonwoven microfibrous support, was demonstrated for highly efficient and low pressure drop filtration for the first time. The polyamide-66 (PA-66) NFN structured top layer, which is composed of traditional electrospun nanofibers and two-dimensional (2D) spider-web-like nano-nets, was electro-spinning/netting (ESN) deposited on the nonwoven polypropylene (PP) scaffold for constructing this new concept of filter. The morphology of NFN architecture, including fiber diameter, coverage rate, pore-width and layer-by-layer packing structure of the nano-nets, can be finely controlled by regulating the solution properties and several ESN process parameters. Taking advantage of several fascinating features such as extremely small diameter, high porosity, controllable coverage rate, nano-nets bring to the NFN/nonwoven composite filtration medium several excellent filtration features such as high filtration efficiency (up to 99.9%), low pressure drop, facile filters cleaning, and more lightweight.

Periodontitis, a chronic inflammatory disease affecting over one billion people worldwide, is characterized by bacterial infections and hyperactive immune responses. Recent studies have revealed that the formation of neutrophil extracellular traps (NETs) contributes significantly to periodontal tissue destruction, and NETs degradation plays a critical role in periodontitis treatment. Current treatments, including mechanical debridement and systemic antibiotics, face limitations such as antibiotic resistance and insufficient local efficacy. To integrate antibacterial and NETs-elimination strategies, the authors propose a novel therapeutic approach using bifunctional core-shell microneedles (MNs) that deliver two types of nanozymes: a peroxidase (POD)-like palladium (Pd) nanozyme in the inner core layer and a DNase-like dendritic mesoporous silica nanoparticles (DMSN)-cerium (Ce) nanozyme in the outer layer. The Pd/Ce MNs are designed to facilitate the rapid release of Pd for bacterial eradication and the sustained release of DMSN-Ce for NETs degradation. This study details the synthesis and characterization of two nanozymes and core-shell structured MNs, followed by evaluations of their catalytic activities, in vitro biocompatibility, antibacterial efficacy and NETs-cleavage ability. In vivo testing using a rat model of periodontitis demonstrates significant improvements in bacterial clearance, inflammation reduction, and alveolar bone preservation. In conclusion, these findings suggest that Pd/Ce MNs with superior antibacterial and NETs-hydrolyzing properties represent a promising therapeutic strategy for the management of periodontitis.

A facile, ultrasensitive, reproductive and selective sensor strip utilizing electrospun polyamide-6/nitrocellulose (PA-6/NC) nano-fibers/nets (NFN) membranes assemble bovine serum albumin decorated Au nanoparticles (BAu probe) for naked-eye colorimetric assay of Pb 2+ has successfully prepared through dual-component alternate distribution multifluidic electrospinning technique . Benefiting from the extremely large specific surface areas and high porosity of NFN membranes, the stability of BAu probe dramatically increased and the strips presented a significant absorbance decreasing band at 546 nm which induce the visual color changes from deep pink to white after incubated in Pb 2+ liquor with a low detection limit of 0.2 μM without any assistance of equipment. Upon exposure to a series of metal ions , only Pb 2+ could induce a pink-to-white color change, which clearly exhibited that BAu probe immobilized PA-6/NC membranes could act as highly selective strips to detect Pb 2+ with little interference from other metal ions. Additionally, the colorimetric responses are represented in visualized quantitative by calculated color difference from L ⁎ a ⁎ b ⁎ coordinates which are presented with lightness and chrome values. Furthermore, the sensitivity of NFN membrane-based strips is much higher than that of film-based ones. The results indicate that this promising cost-effective sensing system could potentially allow for assaying of Pb 2+ in human urine or blood as preliminary screening of lead poisoning.

Atherosclerosis is a chronic inflammatory disease characterized by lipid streaks, which are produced by aggregates of lipid-rich foam cells. Foam cells intensify atherosclerosis by secreting a range of inflammatory mediators. Neutrophil extracellular traps produced by activated neutrophils, which are abundantly present in lipid-accumulating plaques. However, the relationship between neutrophil extracellular traps and foam cells inflammation is still unclear. Paeonol is well known for its anti-inflammatory effects in atherosclerosis. Nevertheless, the exact pharmacological mechanisms by which paeonol affects atherosclerosis are not fully understood which require further investigation. The purpose of this study is to investigate the effects of paeonol on the neutrophil extracellular traps' formation and foam cell inflammation caused by neutrophil extracellular traps, and to explore the potential mechanisms. A high-fat diet was administered to ApoE −/− mice for a period of 12 weeks to induce an atherosclerosis model. Our findings demonstrated that paeonol notably suppressed the advancement of atherosclerosis in ApoE −/− mice, curtailed the formation of neutrophil extracellular traps, and lowered inflammatory factor levels within the plaque. In vitro studies have shown that neutrophil extracellular traps could enhance the inflammation in foam cells. CitH3 played a role in the cellular communication between neutrophil extracellular traps and foam cells. Concurrently, NLRP3 acted as a key receptor in the inflammation mediated by this interaction. Paeonol is capable of regulating NE, thereby affecting the formation of neutrophil extracellular traps. Most notably, the foam cell inflammation caused by neutrophil extracellular traps was significantly mitigated by the inclusion of paeonol. Our findings suggested that paeonol inhibited foam cell inflammation which induced by neutrophil extracellular traps through the CitH3/NLRP3/caspase-1 signaling pathway, shedding new lights on its anti-atherosclerotic pharmacological mechanism.

Neutrophil extracellular traps (NETs) are reticular structures formed by myeloperoxidase (MPO), histones, and neutrophil elastase (NE) that are released from neutrophils in response to pathogenic stimuli. Chlorpyrifos (CPF) is wildly used as an organophosphorus pesticide that causes a range of toxicological and environmental problems. Exposure to CPF can increase the production of neutrophils in carps, and this increase can be considered a biomarker of water pollution. To explore a relationship between NETs and CPF and its mechanism of influence, we treated neutrophils from the blood of carp with 1 μg/mL phorbol 12-myristate 13-acetate (PMA), 0.325 mg/L CPF, or 20 μM necrostatin-1 (Nec-1). The production of MPO and NETs was reduced in the CPF+PMA group compared with that in the PMA group. CPF can cause an increase in reactive oxygen species (ROS), while inhibiting respiratory burst caused by PMA stimulation. We found that the expression levels of protein-coupled receptor 84 (gpr84), dystroglycan (DAG), proto-oncogene serine/threonine kinase (RAF), protein kinase C (PKC), and mitogen-activated protein kinase 3 (MAPK3) in the CPF+PMA group were lower than those in the PMA group, indicating that the PKC-MAPK pathway was suppressed. The expression levels of cylindromatosis (CYLD), mixed lineage kinase domain-like pseudokinase (MLKL), receptor-interacting serine-threonine kinase 1 (RIP1), and receptor-interacting serine-threonine kinase 3 (RIP3) were increased, and the expression levels of caspase 8 were reduced by CPF, indicating that CPF may cause necroptosis. The addition of Nec-1 restored the number of NETs in the CPF+PMA group. The results indicate that CPF reduced the production of NETs by inhibiting respiratory burst and increasing necroptosis. The results contribute to the understanding of the immunotoxicological mechanism of CPF and provide a reference for comparative medical studies.
