Electroactive and antibacterial wound dressings based on Ti3C2Tx MXene/poly(ε-caprolactone)/gelatin coaxial electrospun nanofibrous membranes

Xu Shixin,Du Chen,Zhang Miaomiao,Wang Ruoying,Feng Wei,Wang Chengwei,Liu Qinsong,Zhao Wen

Endogenous electric fields (EFs) are capable of regulating the behaviors of skin cells in wound healing. However, majority of current dressings are primarily engaged in the passive repair of defective tissue, as they lack the ability to actively respond to physiological electrical signals. In this work, a series of nanofibrous membranes (NFMs) were fabricated by coaxial electrospinning, combining the good mechanical properties of poly(ε-caprolactone) (PCL), the bioactivity of gelatin and the electroactivity of Ti 3 C 2 T x MXene, as electroactive and antibacterial dressings for cutaneous wound healing. The obtained NFMs exhibited suitable mechanical properties and hydrophilicity, excellent electroactivity, antibacterial activity, and biocompatibility. Especially, Ti 3 C 2 T x MXene/PCL/gelatin-6 (MPG-6, 6 wt.% of Ti 3 C 2 T x MXene in sheath spinning liquids) showed the optimal conductivity and antibacterial activity. Excitingly, this scaffold significantly promoted the adhesion, proliferation, and migration of NIH 3T3 cells under the electrical stimulation (ES). The in vivo evaluation in a full-thickness wounds defect model demonstrated that the MPG-6 films significantly accelerated wound closure, increased granulation tissue formation, increased collagen deposition, and promoted wound vascularization. In summary, the versatile scaffold is expected to be an ideal candidate as wound dressings due to its ability to promote the transmission of physiological electrical signals and thus improved the therapeutic outcomes of wound regeneration.

Dual functional NaYF4:Yb3+, Er3+@NaYF4:Yb3+, Nd3+ core–shell nanoparticles for cell temperature sensing and imaging

Zengliang Shi,Yue Duan,Xingjun Zhu,Qiwei Wang,DongDong Li,Ke Hu,Wei Feng,Fuyou Li,Chunxiang Xu

Lanthanide-doped up-conversion nanoparticles (UCNPs) provide a remote temperature sensing approach to monitoring biological microenvironments. In this research, the UCNPs of NaYF4:Yb3+, Er3+@NaYF4:Yb3+, Nd3+ with hexagonal (β)-phase were synthesized and applied in cell temperature sensing as well as imaging after surface modification with meso-2, 3-dimercaptosuccinic acid. In the core–shell UCNPs, Yb3+ ions were introduced as energy transfer media between sensitizers of Nd3+ and activators of Er3+ to improve Er3+emission and prevent their quenching behavior due to multiple energy levels of Nd3+. Under the excitations of 808 nm and 980 nm lasers, the NaYF4:Yb3+, Er3+@NaYF4:Yb3+, Nd3+ nanoparticles exhibited an efficient green band with two emission peaks at 525 nm and 545 nm, respectively, which originated from the transitions of 2H11/2 → 4I15/2 and 4S3/2 → 4I15/2 for Er3+ ions. We demonstrate that an occurrence of good logarithmic linearity exists between the intensity ratio of these two emission peaks and the reciprocal of the inside or outside temperature of NIH-3T3 cells. A better thermal stability is proved through temperature-dependent spectra with a heating–cooling cycle. The obtained viability of NIH-3T3 cells is greater than 90% after incubations of about 12 and 24 (h), and they possess a lower cytotoxicity of UCNPs. This work provides a method for monitoring the cell temperature and its living state from multiple dimensions including temperature response, cell images and visual up-conversion fluorescent color.

Fabrication of copper ions-substituted hydroxyapatite/polydopamine nanocomposites with high antibacterial and angiogenesis effects for promoting infected wound healing

Bailong Tao,Chuanchuan Lin,Ai Guo,Yonglin Yu,Xian Qin,Kai Li,Hongchuan Tian,Weiwei Yi,Dengliang Lei,Lixue Chen

Infected wound healing remains a critical threat, which frequently delays the healing process and even leads to severe life-threatening complications. Herein, we reported an effective anti-infection approach, which was based on copper ions-releasing hydroxyapatite/polydopamine (HA-Cu/PDA) nanocomposites with photothermal effect. The HA-Cu/PDA nanocomposites was fabricated through a co-precipitation reaction between polydopamine (PDA)-coated hydroxyapatite nanoparticles (HA)-loaded Cu 2+ (HA-Cu). Through a synergistic effect of released Cu 2+ and photothermal efficiency of PDA coating, and the HA-Cu/PDA nanocomposites exhibited extraordinary antibacterial capacities against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). The nanocomposites presented good biocompatibility for mouse embryonic fibroblast (NIH-3T3) cells and promoted NIH3T3 cells to migrate toward wound sites. Additionally, this nanocomposite could stimulate the tissue remodeling-related gene expression to induce the blood vessels formation, granulation tissues and collagen deposition, and eventually enhance wound healing. In vivo study further verified that HA-Cu/PDA nanocomposites with NIR irradiation could significantly improve bacterial infected wound healing through the prominent antibacterial property, reduced inflammatory response, the formation of granulation tissue, collagen deposition, and angiogenesis ability. Thus, this study develops a versatile strategy for a broad range of wound healing and skin reconstruction caused by bacterial infection.

pH-Sensitive Nanoparticles Composed Solely of Membrane-Disruptive Macromolecules for Treating Pancreatic Cancer

Feng Fan,Lijun Jin,Lihua Yang

Pancreatic tumor is extremely lethal because its cancerous structures are sheltered by dense stromal barriers that hinder the infiltration of therapeutics. To facilitate the infiltration of therapeutics through the stromal barrier, remodeling the stroma with an adjuvant prior to or together with gemcitabine—the current chemotherapeutic standard for pancreatic cancer—is a widely studied strategy; nevertheless, the intrinsic nonuniformity in distribution (spatial and/or temporal) of the adjuvant and gemcitabine has raised the increased risk of tumor metastasis as a major concern. In this work, we propose long-circulating, pH-sensitive nanoparticles composed solely of cellular membrane-disruptive molecules as a new approach for treating pancreatic cancer. Using a micelle of a polymeric mimetic of host defense peptides as the model for such nanoparticles, we showed that this nanoparticle exhibited acid-activated cytotoxicity indiscriminately to both cancerous and fibroblast cells, and the underlying activity mode was acid-activatable disruption of cellular membrane integrity. As a result, our acid-activatable nanoparticle effectively permeabilized the stromal barrier and eradicated the otherwise sheltered pancreatic cancer cells, as demonstrated with a three-dimensional spheroid in which a shell of fibroblast NIH-3T3 cells was cultured over a core of pancreatic BxPC-3 cells. When administered intravenously into mouse models bearing xenograft pancreatic BxPC-3 tumors, our acid-activatable nanoparticle efficiently inhibited tumor growth without causing noticeable off-target adverse effects or promoting tumor metastasis. Notably, this nanoparticle permeabilized the otherwise dense pancreatic tumor tissue while significantly suppressing the expression of extracellular matrix components and activated cancer-associated fibroblasts. Although the feasibility of our approach was demonstrated with a micelle of a polymeric molecule, we trust that future research efforts in this pathway may eventually offer translational formulations for improving the therapeutic efficacy of pancreatic cancer.
