A rapid solid-phase extraction coupled with HPLC method for sensitive determination of NSAIDs in water samples using hierarchical porous carbon flowers

Hao Wang,Jianbin Ju,Yuji Ruan,Guihua Ruan,Yipeng Huang

Pre-oxidation‑carbonization regulated PAN-derived porous carbon flowers, preventing collapse and enabling N/O co-doping. • PCF-400's 400 °C carbonization retains active sites, enhancing adsorption efficiency of NSAIDs. • PCF-400 exhibits superior NSAIDs selectivity, high NSAIDs adsorption (Qe ≥ 12.69 mg g −1 ) and fast kinetics (5 min equilibrium). Flower-shaped porous carbons are attractive in separation science. Herein, the controlled synthesis of porous carbon flowers (PCF) was realized by pre-oxidation and carbonization treatments of flower-shaped polyacrylonitrile (PAN) particles. The pre-oxidation treatment at 230 °C transferred the linear polymer chain of PAN to six-membered ring structure, enhancing the thermal stability to avoid the collapse of flower-shape in carbonization process. Then, the subsequent carbonization promoted the crosslinking and formation of alicyclic and graphitic structures. The carbonization at moderate temperature (400 °C) enabled the remaining of some oxygen-containing groups and the formation of nitrogen-doped carbon structure, which was responsible for the improved adsorption efficiency for non-steroidal anti-inflammatory drugs (NSAIDs). In addition, higher adsorption efficiency of PCF than that of porous carbons in other morphologies for NSAIDs was demonstrated. The PCF carbonized at 400 °C (PCF-400) showed sufficient adsorption capacity (12.69–18.59 mg g −1 ) and selectivity for NSAIDs. An SPE-HPLC method based on PCF-400 was proposed for extraction and analysis of trace NSAIDs in environmental water samples. The SPE-HPLC method showed a wide linear range (1–400 ng mL −1 ) and low detection limits (0.38–0.56 ng mL −1 ) for three kinds of NSAIDs. The intra-day and inter-day relative standard deviations (RSDs) of this method were 1.85%–2.77% and 3.17%–3.63%, respectively. The recoveries of NSAIDs in spiked samples ranged from 89% to 103% by the SPE-HPLC method. This study not only provides a simple synthetic method for preparing carbon materials with controllable morphology and adjustable heteroatom content, but also offers an analytical method for detection of trace NSAIDs in complex environmental matrices. Rational design of ibuprofen-based redox-responsive anti-cancer polymeric drug delivery systems

Hao Dule,Zhang Zheng,Cheng Yu,Cheng Long,Ji Yuanhui

An increasing number of studies have demonstrated that inflammation has a close correlation with multiple stages of tumor development, including tumor initiation, promotion, and migration. Cyclooxygenase-2 (COX-2), which is overexpressed in inflammation, has also been found to be overexpressed in several types of cancer. Some anti-inflammatory drugs have been shown to have potential effects on the inhibition of tumor pathogenesis and development. Among them, non-steroidal anti-inflammatory drugs (NSAIDs) play a role by inhibiting the overexpression of COX-2 with low price and few side effects. In this work, ibuprofen was linked to an amphiphilic polymer mPEG-PLA via disulfide bond to form a polymeric prodrug molecule with reductive sensitivity. The amphiphilic molecule can self-assemble into nanoparticles in an aqueous solution and encapsulate hydrophobic drug, doxorubicin, into the core. Electrostatic potential analysis and weak interaction analysis showed different weak interactions, including hydrogen bonds and π-π stackings, are the driving force of the formation of the complex system between doxorubicin and polymeric prodrug molecules. These quantitative calculations were helpful to understand the formation mechanism of prodrug polymeric carriers. Characterization methods such as TEM and DLS proved that mPEG-PLA- s -IHE/DOX (PDC/DOX) micelles had an excellent uniform nanostructure, and in vitro drug release experiments showed superior sustained-release and reductive environment-dependent release of this drug. More importantly, in vitro cytotoxicity and cell uptake studies showed that the PDC/DOX micelles induced apoptosis of A549 lung cancer cell lines. This study demonstrates the advantage of combining NSAIDs with chemotherapy agents in the treatment of lung cancer, which provides a feasible and novel polymer delivery system, and a reasonable explanation of formation mechanism. Constructing Zr/Fe-MOF-derived carbon intercalated MXene/Zr-Fe@C heterostructure for NSAIDs removal: Synergistic adsorption performance and mechanism studies in single and binary systems

Huixiao Duo,Yan Hao,Jiayuan Yue,Qianxing Lu,Yuetan Dong,Ke Chen,Yanyan Kong,Shushu Ding,Qing Zhu,Jiejia Li

The extensive use of non-steroidal anti-inflammatory drugs (NSAIDs), particularly diclofenac (DIC) and indomethacin (IND), poses escalating ecological and health risks due to incomplete degradation in water systems. Conventional remediation methods face challenges including low efficiency and toxic intermediate generation. Herein, we developed a novel MXene/Zr-Fe-MOF-derived carbon heterostructure (MXene/Zr-Fe@C) featuring bimetallic Zr/Fe active sites anchored on MXene nanosheets. The heterostructure design of MXene/Zr-Fe@C enables exceptional synergistic adsorption of NSAIDs from aqueous environments. Systematic adsorption experiments in single-component (DIC or IND) and binary (DIC+IND) systems revealed competitive adsorption behaviors and synergistic effects. MXene/Zr-Fe@C demonstrated satisfactory adsorption capacities (277.7 mg/g for DIC and 238.09 mg/g for IND, surpassing conventional adsorbents. These exceptional performances stem from the bimetallic site selectivity and synergistic domain interactions of MXene/Zr-Fe@C. The Zr/Fe@C provide selective coordination site, while MXene and carbon matrix promote the non-polar interaction with aromatic rings of NSAIDs. Remarkably, the composite exhibited excellent regenerability, retaining 83% of adsorption capacity after 54 regeneration cycles. Overall, the synergistic interfacial effects between MXene's conductive surfaces and ordered porosity of MOF-derived carbon enhance structural stability, efficient mass transfer and site accessibility. These findings establish MXene/Zr-Fe@C heterostructures as versatile platforms for NSAIDs remediation, providing foundational insights for sustainable water purification technologies addressing emerging pharmaceutical contamination challenges.

Development and application of metal-organic framework@GA based on solid-phase extraction coupling with UPLC-MS/MS for the determination of five NSAIDs in water

Yiyuan Zhou,Jiabi Xu,Nan Lu,Xinyu Wu,Yu Zhang,Xiaohong Hou

MIL-101(Cr) and graphene aerogel (GA) were hybridized as a multifunctional adsorbent for the preconcentration of trace analytes. The novel MIL-101(Cr)@GA was prepared and characterized by FTIR, XRD and SEM, where GA is serving as a carrier for MIL-101(Cr). The synthesized composite as a solid phase extraction (SPE) sorbent combined with UPLC-MS/MS was applied for the determination and quantification of five NSAIDs (phenacetin, meloxicam , naproxen , diclofenac sodium and carprofen) in different environmental water samples. The parameters influencing the whole extraction process were systematically optimized. Under the most favorable. conditions, good sensitivity was achieved with a limit of detection between 0.006 and 0.012 ng mL −1 , the linear range of 0.02–2 ng mL −1 for phenacetin , meloxicam and 0.05–5 ng mL −1 for naproxen , diclofenac sodium , carprofen ( r 2  ≥ 0.9940). The satisfactory recoveries of the target analytes were in the range from 77.2 to 103.3% with relative standard deviation (RSD) from 0.6% to 8.4%. The established method was proved to be simple, highly sensitive and accurate. The adsorbent could be reusable up to nine cycles without any decrease in performance, which provides economic strategy and little waste generation. Adsorption behaviors were explored by choosing two typical types of NSAIDs as models to measure the adsorption capacity of MIL-101(Cr)@GA. The maximum adsorption capacities for PHE and NAP were 232.5 and 333.3 mg g −1 , respectively.

Differential impacts of nonsteroidal anti-inflammatory drugs on lifespan and healthspan in aged Caenorhabditis elegans

Jia-Jun Tu,Zhen-Zhen Yu,Mei-Ling Ou,Jin-Xiong Cen,Kun Xue,Jing Zhou,Shao-Jun Li,Guo-Dong Lu

Aging and age-related diseases are intricately associated with oxidative stress and inflammation. Nonsteroidal anti-inflammatory drugs (NSAIDs) have shown their promise in mitigating age-related conditions and potentially extending lifespan in various model organisms. However, the efficacy of NSAIDs in older individuals may be influenced by age-related changes in drug metabolism and tolerance, which could result in age-dependent toxicities. This study aimed to evaluate the potential risks of toxicities associated with commonly used NSAIDs (aspirin, ibuprofen, acetaminophen, and indomethacin) on lifespan, healthspan, and oxidative stress levels in both young and old Caenorhabditis elegans . The results revealed that aspirin and ibuprofen were able to extend lifespan in both young and old worms by suppressing ROS generation and enhancing the expression of antioxidant SOD genes. In contrast, acetaminophen and indomeacin accelerated aging process in old worms, leading to oxidative stress damage and reduced resistance to heat stress through the pmk-1/skn- 1 pathway. Notably, the harmful effects of acetaminophen and indomeacin were mitigated when pmk-1 was knocked out in the pmk-1 (km25) strain. These results underscore the potential lack of benefit from acetaminophen and indomeacin in elderly individuals due to their increased susceptibility to toxicity. Further research is essential to elucidate the underlying mechanisms driving these age-dependent responses and to evaluate the potential risks associated with NSAID use in the elderly population.

Simultaneous degradation of NSAIDs in aqueous and sludge stages by an electron-Fenton system derived from sediment microbial fuel cell based on a novel Fe@Mn biochar GDC

Shuai Zhao,Hanyan Li,Jia Zhou,Tawatchai Sumpradit,El-Sayed Salama,Xiangkai Li,Jianhang Qu

Electro-Fenton (e-Fenton) reactions boosted using microbial fuel cells (MFCs) can degrade contaminants with high efficiency, the treatment is generally performed in the aqueous phase and the sludge pollution is ignored in such MFCs. In this study, a sediment MFC e-Fenton (SMFC-e-Fenton) system was proposed for the degradation of nonsteroidal anti-inflammatory drugs (NSAIDs) in both aqueous and solid phases. A Fe@Mn-codoped biochar gas diffusion cathode was fabricated to produce hydroxyl radicals without any additional power input. The studied NSAIDs of ibuprofen (IBU), naproxen (NPX), and diclofenac (DCF) exhibited total degradation efficiencies (including aqueous and sludge phase) of 99.1%, 98.5%, and 80.6%, respectively, in the SMFC-e-Fenton system. Compared with sludge anaerobic digestion, the residual amounts of IBU, NPX, and DCF reduced from 10.2%, 24.6%, and 32.2% to 2.7%, 4.4%, and 8.5%, respectively, in the sludge phase. Moreover, the electricity generation and sludge reduction improved owing to the e-Fenton process. Using the SMFC-e-Fenton system, the effects of NSAIDs were reduced and the richness and diversity of the microbial community are maintained, improving the overall system performance. These results demonstrate that the SMFC-e-Fenton method is a high-efficiency, economic and environmental friendly technology for simultaneously treating NSAIDs in the aqueous and sludge phases.

Dummy-surface molecularly imprinted polymers as a sorbent of micro-solid-phase extraction combined with dispersive liquid–liquid microextraction for determination of five 2-phenylpropionic acid NSAIDs in aquatic environmental samples

Guo Ping,Yuan Xucan,Zhang Jingjing,Wang Binjie,Sun Xiaoyang,Chen Xiaohui,Zhao Longshan

A highly binding dummy template surface of molecularly imprinted polymers (MWNTs-MIPs) was synthesized on multi-walled carbon nanotubes surface using 2-phenylpropionic acid as dummy template, 4-vinylpyridine as the functional monomer, ethylene glycol dimethacrylate as the cross-linker, and DMF as porogen by precipitation polymerization method. MIPs were characterized by FT-IR spectroscopy, scanning electron microscope, thermo-gravimetric analysis, and nitrogen adsorption-desorption experiment. Adsorption and selectivity experiments of MIPs and non-imprinted polymers (NIPs) verified that the MIPs had a good selectivity and adsorption properties for five 2-phenylpropionic acid nonsteroidal anti-inflammatory drugs (NSAIDs). Imprinted polymer was used as a sorbent material for μSPE in current work and μSPE-DLLME method was selected for pretreatment of water samples. The μSPE-DLLME method was successfully used for the pre-concentration of five non-steroidal anti-inflammatory drugs in different environmental water samples prior to ultra-high performance liquid chromatography-tandem mass spectrometry. Efficiencies of μSPE and DLLME were thoroughly investigated and optimized in this study. The optimal results were obtained by using 3 mL of 1% formic acid-acetonitrile as elution solvent and dichloroethane and acetonitrile as extractant and disperser solvent, respectively. Limits of detection and quantification of five NSAIDs for different water matrices varied from 0.50 to 1.10 ng L −1 and 0.93 to 2.20 ng L −1 , respectively. Each target analyte had a good linearity in its corresponding concentration range. Enrichment factors of target analytes ranged from 91 to 215. Recoveries of the target analytes were between 72.43 and 113.99% at the concentration levels of 0.02, 0.1, and 0.5 μg L −1 . The developed method was successfully applied to extraction and analysis of NSAIDs in different water samples with satisfactory results which could help us better understand their environmental fate and risk to ecological health. Dispersive solid-phase extraction of non-steroidal anti-inflammatory drugs in water and urine samples using a magnetic ionic liquid hypercrosslinked polymer composite

Pengzhao Han,Kai Hu,Yuanpeng Wang,Lixin Li,Pan Wang,Weixia Zhu,Haiyan Gong,Zhenqiang Zhang,Shusheng Zhang

In this work, Friedel-Crafts alkylation was successfully applied to prepare a magnetic ionic liquid hypercrosslinked polymer composite (Fe 3 O 4 @IL-HCP), which was subsequently employed as magnetic solid-phase extraction (MSPE) adsorbent for the isolation and enrichment of trace non-steroidal anti-inflammatory drugs (NSAIDs). The developed composite was comprehensively characterized using various techniques, with the results indicating that it possessed high saturation magnetization (39.44 em g   −   1 ), large specific surface area (175 m 2 g   −   1 ), and high adsorption capacity for NSAIDs. The adsorption behavior and mechanisms were also investigated in detail. NSAIDs were adsorbed onto the Fe 3 O 4 @IL-HCP sorbent via a heterogeneous multilayer process consisting of hydrogen bonding and π-π and electrostatic interactions. Additionally, the composite's large surface area and multiple active sites enabled extraction equilibrium within 6 min. By coupling with high performance liquid chromatography (HPLC), the developed MSPE/HPLC method was applied for the determination of selected NSAIDs in water and urine samples. The developed method displayed wide linear ranges, low limits of detection (0.12–0.30 ng mL −1 and 0.15–1.5 ng mL −1 in water and urine samples, respectively), sufficient recoveries (92.8–109%), and good precision (relative standard deviations ≤ 4.6%). Thus, the findings of this work provide an appealing alternative for the extraction and determination of trace NSAIDs in environmental water and biological samples.

Fabrication of sea urchin shaped polyaniline-modified magnetic microporous organic network for efficient extraction of non-steroidal anti-inflammatory drugs from animal-derived food samples

Yan-Hong Li,Xu-Hui Li,Yuan-Yuan Cui,Abdukader Abdukayum,Cheng-Xiong Yang

In this work, a novel polyaniline-modified magnetic microporous organic network (MMON-PANI) composite was fabricated for effective magnetic solid phase extraction (MSPE) of five typical nonsteroidal anti-inflammatory drugs (NSAIDs) from animal-derived food samples before high performance liquid chromatography (HPLC) detection. The core-shell sea urchin shaped MMON-PANI integrates the merits of Fe 3 O 4 , MON, and PANI, exhibiting large specific surface area, rapid magnetic responsiveness, good stability, and multiple binding sites to NSAIDs. Convenient and effective extraction of trace NSAIDs from chicken, beef and pork samples is realized on MMON-PANI via the synergetic π-π, hydrogen bonding, hydrophobic, and electrostatic interactions. Under optimal conditions, the MMON-PANI-MSPE-HPLC-UV method exhibits wide linear ranges (0.2–1000 μg L −1 ), low limits of detection (0.07–1.7 μg L −1 ), good precisions (intraday and inter-day RSDs < 5.4 %, n = 3), large enrichment factors (98.6–99.9), and less adsorbent consumption (3 mg). The extraction mechanism and selectivity of MMON-PANI are also evaluated in detail. This work proves the incorporation of PANI onto MMON is an efficient way to promote NSAIDs enrichment and provides a new strategy to synthesize multifunctional MON-based composites in sample pretreatment.

Mechanisms of action and adaptive responses to diclofenac and meloxicam during the early life stages of Oryzias melastigma

Chuan Liang,Yang Chen,Yunzhe Ling,Peirun Li,Jiahao Liu,Xuantian Li,Yanhua Xu,Zhiying Liu

Non-steroidal anti-inflammatory drugs (NSAIDs), though designed to target specific molecular pathways, pose significant environmental risks to non-target organisms, particularly marine fish. This study investigated the toxicity mechanisms and adaptive responses to diclofenac (DCF) and meloxicam (MEX) during the early life stages (ELS) of Oryzias melastigma at environmentally relevant concentrations over a 31-day period. Mechanistic investigations of sub-lethal effects were conducted using Enzyme-Linked Immunosorbent Assay (ELISA), RNA sequencing (RNA-Seq) and quantitative PCR (qPCR). The results revealed that cyclooxygenase (COX) inhibition disrupted the renin-angiotensin system, leading to an accumulation of angiotensin II and cardiovascular developmental defects. Additionally, downregulation of the pla2 gene reduced substrates essential for COX enzyme activity, exacerbating the effects. Although NSAIDs are known to affect the digestive system, no significant effects on developmental factors were observed. RNA-Seq and qPCR analyses revealed an adaptive upregulation of key genes, including ace2 and cyp7a1 , involved in cardiovascular and metabolic regulation. Furthermore, 16S rRNA sequencing identified shifts in the microbial community, particularly in g_Rubritalea and g_Sphingomonas genera. Both the upregulated genes and the altered microbial taxa likely played a role in mitigating toxic effects and promoting homeostasis. Moreover, molecular docking suggested that MEX exhibited stronger sub-lethal effects than DCF, likely due to its higher binding affinity to COX. These findings provide valuable insights into NSAID toxicity mechanisms in marine fish, highlighting the importance of adaptive responses in countering environmental stress and underscoring the long-term ecological risks of chronic NSAID exposure.
