Functional analysis of Ornithine decarboxylase in manipulating the wing dimorphism in Nilaparvata lugens (Stål) (Hemiptera: Delphacidae)

Wan-Xue Li,Jing-Xiang Chen,Chuan-Chuan Zhang,Min-Shi Luo,Wen-Qing Zhang

The brown planthopper (BPH, Nilaparvata lugens ), a major insect pest of rice, can make a shift in wing dimorphism to adapt to complex external environments. Our previous study showed that NlODC ( Ornithine decarboxylase in N. lugens ) was involved in wing dimorphism of the brown planthopper. Here, further experiments were conducted to reveal possible molecular mechanism of NlODC in manipulating the wing dimorphism. We found that the long-winged rate (LWR) of BPH was significantly reduced after RNAi of NlODC or injection of DFMO (D, L-α-Difluoromethylornithine), and LWR of males and females significantly decreased by 21.7% and 34.6%, respectively. Meanwhile, we also examined the contents of three polyamines under DFMO treatment and found that the contents of putrescine and spermidine were significantly lower compared to the control. After 3rd instar nymphs were injected with putrescine and spermidine, LWR was increased significantly in both cases, and putrescine was a little bit more effective, with 5.6% increase in males and 11.4% in females. Three days after injection of ds NlODC , injection of putrescine and spermidine rescued LWR to the normal levels. In the regulation of wing differentiation in BPH, NlODC mutually antagonistic to NlAkt may act through other signaling pathways rather than the classical insulin signaling pathway. This study illuminated a physiological function of an ODC gene involved in wing differentiation in insects, which could be a potential target for pest control.

Various functions of detoxification enzymes against insecticides in Nilaparvata lugens selected by toxicity assays and RNAi methods

Jun Yang,Li-Yuan Sun,Ruiyan Ma,Rui Tang,Juan-Ying Zhao,Qing-Nian Cai

The brown planthopper (BPH), Nilaparvata lugens is a devastating agricultural pest of rice, and they have developed resistance to many pesticides. In this study, we assessed the response of BPH nymphs to nitenpyram, imidacloprid , and etofenprox using contact and dietary bioassays, and investigated the underlying functional diversities of BPH glutathione- S -transferase (GST), carboxylesterase (CarE) and cytochrome P450 monooxygenase (P450) against these insecticides. Both contact and ingestion toxicity of nitenpyram to BPH were significantly higher than either imidacloprid or etofenprox. Under the LC 50 concentration of each insecticide, they triggered a distinct response for GST, CarE, and P450 activities, and each insecticide induced at least one detoxification enzyme activity . These insecticides almost inhibited the expression of all tested GST , CarE , and P450 genes in contact bioassays but induced the transcriptional levels of these genes in dietary bioassays. Silencing of NlGSTD2 expression had the greatest effect on BPH sensitivity to nitenpyram in contact test and imidacloprid in dietary test. The sensitivities of BPH to insecticide increased the most in the contact test was etofenprox after silencing of NlCE , while the dietary test was nitenpyram. Knockdown of NlCYP408A1 resulted in BPH sensitivities to insecticide increasing the most in the contact test was nitenpyram, while the dietary test was imidacloprid. Taken together, these findings reveal that NlGSTD2 , NlCE , and NlCYP408A1 play an indispensable role in the detoxification of the contact and ingestion toxicities of different types of insecticides to BPH, which is of great significance for the development of new strategies for the sucking pest control.

Transcriptional Response of ATP-Binding Cassette (ABC) Transporters to Insecticide in the Brown Planthopper, Nilaparvata lugens (Stål)

Zhao Li,Tingwei Cai,Yao Qin,Yunhua Zhang,Ruoheng Jin,Kaikai Mao,Xun Liao,Hu Wan,Jianhong Li

The ATP-binding cassette (ABC) transporter superfamily is one of the largest groups of proteins and plays a non-negligible role in phase III of the detoxification process, which is highly involved in the response of insects to environmental stress (plant secondary metabolites and insecticides). In the present study, inNilaparvata lugens, we identified 32 ABC transporters, which are grouped into eight subfamilies (ABCA–H) based on phylogenetic analysis. The temporal and spatial expression profiles suggested that the nymphal stages (1st–5th) and adult males showed similarity, which was different from eggs and adult females, andNlABCA1,NlABCA2,NlABCB6,NlABCD2,NlABCG4,NlABCG12,NlABCG15, andNlABCH1were highly expressed in the midgut and Malpighian tubules. In addition,ABCG12, which belongs to the ABC transporter G subfamily, was significantly upregulated after exposure to sulfoxaflor, nitenpyram, clothianidin, etofenprox, chlorpyrifos, and isoprocarb. Moreover, verapamil significantly increased the sensitivity ofN. lugensto nitenpyram, clothianidin, etofenprox, chlorpyrifos, and isoprocarb. These results provide a basis for further research on ABC transporters involved in detoxification inN. lugens, and for a more comprehensive understanding of the response ofN. lugensto environmental stress. Keywords:ABC transporters;detoxification;Nilaparvata lugens;insecticides

Synergistic and Additive Interactions of Zhongshengmycin to the Chemical Insecticide Pymetrozine for Controlling Nilaparvata lugens (Hemiptera: Delphacidae)

Zhao Ruoying,Li Danting,Wang Xinlong,Li Zhong,Yu Xiaoping,Shentu Xuping

Management of the rice brown planthopper Nilaparvata lugens Stål is challenging because it can rapidly adapt to new pesticides within several generations. Combined use of chemical insecticides and antimicrobials was proposed as an alternative strategy to control N. lugens. Our previous experiments identified two effective agents (chemical insecticide: pymetrozine and antimicrobial: zhongshengmycin) that act on different targets in N. lugens. However, conditions and effectiveness of combinations of antimicrobials and insecticides against N. lugens are still unknown. Here, we evaluated separate and combined effects of pymetrozine and zhongshengmycin on third instar nymphs of N. lugens under laboratory and greenhouse conditions. Results showed that zhongshengmycin exerts significant inhibitory effects on the three endosymbionts Pichia guilliermondii, Cryptococcus peneaus, and Pichia anomala cultured in vitro of N. lugens. Combinations of pymetrozine and zhongshengmycin under laboratory conditions produced additive or synergistic effects on N. lugens and caused higher mortality in third instar nymphs than either of them used alone. Experiments under greenhouse conditions further demonstrated that effective component quality ratio of pymetrozine to zhongshengmycin of 1:10 and 1:40 with co-toxicity coefficients of 221.63 and 672.87, respectively, also produced significant synergistic effects against N. lugens. Our results indicated that chemical insecticides combined with antimicrobials may provide a potential novel strategy for controlling N. lugens by inhibiting its endosymbionts.

Molecular and in vitro biochemical assessment of chemosensory protein 10 from brown planthopper Nilaparvata lugens at acidic pH

Muhammad Irfan WARIS,Aneela YOUNAS,Rana Muhammad Kaleem ULLAH,Fatima RASOOL,Muhammad Muzammal ADEEL,Man-qun WANG

Chemosensory proteins (CSPs) are important molecular components of the insect olfactory system, which are involved in capturing, binding, and transporting hydrophobic odour molecules across the sensillum in sensillar lymph in regulating insect behavior. This protein family (CSPs) is also involved in many other systems that are not linked to olfactory receptors in olfactory sensilla. The brown planthopper (BPH) is a monophagous pest of rice that causes damage by sucking phloem sap and transmitting a number of diseases caused by viruses. In this study, fluorescence competitive binding assay and fluorescence quenching assay at acidic pH were performed as well as homology modelling to describe the binding affinity of NlugCSP10. Fluorescence competitive binding assay (FCBA) demonstrated that NlugCSP10 bound strongly to nonadecane, farnesene, and 2-tridecanone at acidic pH. The results of FCBA indicated that NlugCSP10 bound different ligands at the physiological pH (5.0) of the bulk sensillum lymph. Fluorescence quenching assay demonstrated that NlugCSP10 generated a stable complex with 2-tridecanone, while two ligands nonadecane and farnesene collided due to molecular collisions. The interaction of selected ligands with the modelled structure of NlugCSP10 was also analyzed, which found the key amino acids (Gln23, Gln24, Gln25, Asn27, Met33, Ser34, Ile35, Tyr36, Asn42, Met43, Val45, Asn46, Asn93, Arg96, Ala97, Lys99, and Ala100) in NlugCSP10 that were involved in binding of volatile compounds. The present study contributes to the binding profile of NlugCSP10 that promotes the development of behaviorally active ligands based on BPH olfactory system.

Role of Broad-Complex (Br) and Krüppel homolog 1 (Kr-h1) in the Ovary Development of Nilaparvata lugens

Jiang Jianru,Xu Yili,Lin Xinda

Ovarian development plays an important role in the life history of insects and is crucial for control of the insect population. The metamorphosis of an insect is precisely regulated by the interaction of the juvenile hormone and ecdysone. To understand the role of NlBr and NlKr-h1 in ovary development, we used RNA interference (RNAi) to down-regulate the expression of Broad-Complex (Br) and Krüppel homolog 1 (Kr-h1), two important down-stream transcription factors of juvenile hormone and ecdysone signaling. We further investigated their effects on metamorphosis and ovary development. The results showed that both NlBr and NlKr-h1 are induced by ecdysone. The down-regulation of NlBr and NlKr-h1 alone or together by RNAi is more effective than the topical application of ecdysone on the number of ovarioles, suggesting the necessity of NlBr and NlKr-h1 in determining the number of ovarioles. The ovarian grade was significantly increased/decreased by the topical application of ecdysone and down-regulation of NlBr and NlKr-h1. The pre-oviposition period was also increased. When NlBr and NlKr-h1 were down-regulated together, the ovary grade was not significantly different compared to the control (dsGFP), indicating that the development of the ovary is under the control of both NlBr and NlKr-h1. The interaction between the NlBr and NlKr-h1 on the number of ovarioles and the development of the ovary indicates cross-talk between both juvenile hormone and ecdysone signaling at the transcription level in the brown planthopper. Both genes are nuclear transcription factors and may regulate signaling via down-stream genes. These results would help to both enhance the current understanding of the regulatory mechanism of the interaction between juvenile hormone and ecdysone signaling pathways during ovarian development and to design chemicals to control pests.

Mechanism of metabolic resistance to pymetrozine in Nilaparvata lugens: over-expression of cytochrome P450 CYP6CS1 confers pymetrozine resistance

Li-Xiang Wang,Sha Tao,Yan Zhang,Ya-Long Jia,Shun-Fan Wu,Cong-Fen Gao

BACKGROUND Pymetrozine is commonly used for the control of Nilaparvata lugens , and resistance to pymetrozine has been frequently reported in the field populations in recent years. However, the mechanism of brown planthopper resistance to pymetrozine is still unknown. RESULTS In this study, a pymetrozine-resistant strain (PMR) was established, and the potential biochemical resistance mechanism of N. lugens to pymetrozine was investigated. Pymetrozine was synergized by the inhibitor piperonyl butoxide (PBO) in the PMR with 2.83-fold relative synergistic ratios compared with the susceptible strain (Sus). Compared with the Sus, the cytochrome P450 monooxygenase activity of PMR was increased by 1.7 times, and two P450 genes ( NlCYP6CS1 and NlCYP301B1 ) were found to be significantly overexpressed more than 6.0-fold in the PMR. Pymetrozine exposure induced upregulation of NlCYP6CS1 expression in the Sus, but the expression of NlCYP301B1 did not change significantly. In addition, RNA interference (RNAi)-mediated suppression of NlCYP6CS1 gene expression dramatically increased the toxicity of pymetrozine against N. lugens . Moreover, transgenic lines of Drosophila melanogaster expressing NlCYP6CS1 were less susceptible to pymetrozine, and had a stronger ability to metabolize pymetrozine. CONCLUSIONS Taken together, our findings indicate that the overexpression of NlCYP6CS1 is one of the key factors contributing to pymetrozine resistance in N. lugens . And this result is helpful in proposing a management strategy for pymetrozine resistance.

Silencing a Simple Extracellular Leucine-Rich Repeat Gene OsI-BAK1 Enhances the Resistance of Rice to Brown Planthopper Nilaparvata lugens

Miaofen Ye,Peng Kuai,Shuting Chen,Na Lin,Meng Ye,Lingfei Hu,Yonggen Lou

Many plant proteins with extracellular leucine-rich repeat (eLRR) domains play an important role in plant immunity. However, the role of one class of eLRR plant proteins—the simple eLRR proteins—in plant defenses against herbivores remains largely unknown. Here, we found that a simple eLRR protein OsI-BAK1 in rice localizes to the plasma membrane. Its expression was induced by mechanical wounding, the infestation of gravid females of brown planthopper (BPH)Nilaparvata lugensor white-backed planthopperSogatella furciferaand treatment with methyl jasmonate or abscisic acid. SilencingOsI-BAK1(ir-ibak1) in rice enhanced the BPH-induced transcript levels of three defense-related WRKY genes (OsWRKY24,OsWRKY53andOsWRKY70) but decreased the induced levels of ethylene. Bioassays revealed that the hatching rate was significantly lower in BPH eggs laid on ir-ibak1plants than wild-type (WT) plants; moreover, gravid BPH females preferred to oviposit on WT plants over ir-ibak1plants. The exogenous application of ethephon on ir-ibak1plants eliminated the BPH oviposition preference between WT and ir-ibak1plants but had no effect on the hatching rate of BPH eggs. These findings suggest that OsI-BAK1 acts as a negative modulator of defense responses in rice to BPH and that BPH might exploit this modulator for its own benefit.Keywords:rice;OsI-BAK1;Nilaparvata lugens;ethylene;plant defense responses;simple extracellular leucine-rich repeat protein

Resistance monitoring and cross-resistance patterns of three rice planthoppers, Nilaparvata lugens, Sogatella furcifera and Laodelphax striatellus to dinotefuran in China

Xi-Chao Mu,Wei Zhang,Li-Xiang Wang,Shuai Zhang,Kai Zhang,Cong-Fen Gao,Shun-Fan Wu

Three rice planthoppers , brown planthopper , Nilaparvata lugens , white-backed planthopper, Sogatella furcifera and small brown planthopper, Laodelphax striatellus , are important pests of cultivated rice in tropical and temperate Asia. They have caused severe economic loss and developed resistance to insecticides from most chemical classes. Dinotefuran is the third neonicotinoid which possesses a broad spectrum and systemic insecticidal activity. We determined the susceptibility of dinotefuran to field populations from major rice production areas in China from 2013 to 2015. All the populations of S. furcifera and L. striatellus were kept susceptible to dinotefuran (0.7 to 1.4-fold of S. furcifera and 1.1-to 3.4-fold of L. striatellus ) However, most strains of N. lugens (except FQ15) collected in 2015 had developed moderate resistance to dinotefuran, with resistance ratios (RR) ranging from 23.1 to 100.0 folds. Cross-resistance studies showed that chlorpyrifos-resistant and buprofezin-resistant Sogatella furcifera , chlorpyrifos-resistant and fipronil-resistant L. striatellus , imidacloprid-resistant and buprofezin-resistant Nilaparvata lugens exhibited negligible or no cross-resistance to dinotefuran. Synergism tests showed that piperonyl butoxide (PBO) produced a high synergism of dinotefuran effects in the DY15 and JS15 populations (2.14 and 2.52-fold, respectively). The obvious increase in resistance to dinotefuran in N. lugens indicates that insecticide resistance management strategies are urgently needed to prevent or delay further increase of insecticide resistance in N. lugens .

Physicochemical Properties and Bioactivity of a New Guar Gum-Based Film Incorporated with Citral to Brown Planthopper, Nilaparvata lugens (Stål) (Hemiptera: Delphacidae)

Xiubing Gao,Can Guo,Ming Li,Rongyu Li,Xiaomao Wu,Anlong Hu,Xianfeng Hu,Feixu Mo,Shuai Wu

The brown planthopper (BPH), Nilaparvata lugens (Stål), is the most notorious rice insect pest. In order to repel BPH effectively while being environmentally friendly, a new film based on guar gum incorporated with citral (GC film) was formulated. A toxicity bioassay of citral and guar gum at different proportions (ratios of 3:1, 2:1, 1:1, 1:2, and 1:3 inw/w) of GC film-forming emulsion to BPH was performed with the rice stem dipping method. Results showed that the most effective ratio of citral to guar gum was 1:1 with the median lethal concentration (LC50) of 4.30 mg/mL, far below the LC50of guar gum (GG)/citral individual (141.51 and 44.38 mg/mL, respectively). The mortality of BPH adults and nymphs in the third instar treated with different dilution multiples of GC film-forming emulsion ranged from 46.67% to 82.22% and from 37.78% to 71.11%, respectively. These indicated that GC film-forming emulsion had a direct toxicity on BPH, and the mixture of citral and GG had synergistic interactions. Subsequently, Fourier-transform infrared spectroscopy showed that the incorporation of guar gum with citral was successful and did not result in the formation of new chemical bonds. The GC film exhibited a darker color and rougher surface topography with larger apertures and deeper gullies (Ra = 1.42 nm, Rq = 2.05 nm, and Rmax = 25.40 nm) compared to the guar gum film (GG film) (Ra = 1.00 nm, Rq = 1.33 nm, and Rmax = 16.40 nm), as determined by transmission electron microscopy and atomic force microscopy. The GC film exhibited a 50.4% lower solubility in water (30.30% vs. 15.00%) and 71.3% oxygen permeability (8.26 × 10−9vs. 2.37 × 10−9cm3/m2·d·Pa) (p <0.05) but did not demonstrate any significant difference in mechanical properties, such as thickness (39.10 vs. 41.70 mm), tensile strength (41.89 vs. 38.30 N/mm2), and elongation at break (1.82% vs. 2.03%) (p <0.05) compared to the GG film. Our findings established a link between physicochemical properties and bioactivity, which can provide useful information on developing and improving GC films and may offer an alternative approach for the control of BPH in the near future.

Effects of Fungicide Propiconazole on the Yeast-Like Symbiotes in Brown Planthopper (BPH, Nilaparvata lugens Stål) and Its Role in Controlling BPH Infestation

Shentu Xuping,Wang Xiaolong,Xiao Yin,Yu Xiaoping

Yeast-like symbiotes (YLS), harbored in the abdomen fat-body cells of the rice brown planthopper (BPH), Nilaparvata lugens Stål (Hemiptera: Delphacidae), are vital to the growth and reproduction of their host. It is feasible to manipulate BPH infestation on rice by inhibiting YLS using fungicide. In this study, the fungicide propiconazole was injected into the hemolymph of BPH thorax via microinjection to investigate its effect on YLS, especially the dominant species, Hypomyces chrysospermus, and their host BPH. Propiconazole markedly reduced the total number of YLS and H. chrysospermus in BPH hemolymph and fat body, thereby leading to an obvious higher mortality and lower fecundity of BPH than the negative control (PBS, phosphate buffer solution). After microinjecting propiconazole, the survival rate of BPH nymphs at the 5th instar was significantly lower than that obtained after PBS treatment. Eight days after propiconazole microinjection, the BPH survival rate dropped to 40%, only half of BPH survival rate treated with PBS microinjection. For female adults (1-day-old), there were significant differences in the survival rates between BPHs treated with propiconazole and those treated with PBS at days 5–8. The fecundity of BPH decreased significantly by microinjecting propiconazole and averaged only 229 eggs per female, which was 20% less than that of the negative control. Furthermore, we reared BPH on the susceptible variety TN1 sprayed with propiconazole to prove the feasibility manipulating field occurrence of BPH by inhibiting YLS using fungicides. The number of YLS and H. chrysospermus in BPH obviously declined. Subsequently, the survival rate and fecundity of BPH significantly decreased after feeding on rice treated with propiconazole. Meanwhile, the propiconazole residue was detected in the hemolymph and gut of BPH by HPLC analysis within 1 day of feeding. Inhibiting YLS using fungicides was a novel and effective way to control BPH infestation.

Silver and copper-oxide nanoparticles prepared with GA3 induced defense in rice plants and caused mortalities to the brown planthopper, Nilaparvata lugens (Stål)

Amr S. Abou El-Ela,Eric Siaw Ntiri,Asim Munawar,Xiao-Xiao Shi,Chao Zhang,Joko Pilianto,Yadong Zhang,Ming Chen,Wenwu Zhou,Zeng-Rong Zhu

Background Nanoparticles have been employed as nanopesticides for pest control in agriculture. However, the harmful effects of their chemical synthesis on human and environmental health have resulted in increased use of green synthetic approaches, including the use of plant extracts. The brown planthopper, Nilaparvata lugens (Stål) (BPH), is a severe pest of rice plants ( Oryza sativa L.), especially in Asia. It is usually controlled chemically but has developed resistance against many insecticides. Results In this study, we synthesized metallic silver (Ag-NPs) and copper-oxide (CuO-NPs) nanoparticles using the exogenous phytohormone, gibberellic acid (GA 3 ), as a reducing agent. We then sprayed them separately on rice plants and BPH together and evaluated their effects on the plants and insects. SEM and TEM images showed that the synthesis was successful, indicated by the sizes (25–60 nm), uniform shape and spherical and cubical structures of Ag-NPs, as well as by the rugby sheet-like of CuO-NPs with lateral sizes of 150–340 nm and thickness of 30–70 nm. Independent applications of the nanoparticles and GA 3 on rice plants induced different volatile profiles, of which the highest number emitted was under Ag-NPs, including the highest emission of linalool. Transcriptome analysis showed that Ag-NPs-treated rice plants showed different transcriptome profiles compared to the control, 24 h after treatment, including the upregulation of the linalool synthase gene, genes of plants transcription factors such as WRKY, bHLH and NAC and other genes involved in plant defense responses. In all treatments, the mortality rate of BPH increased with an increase in NPs concentrations over time but was prominent under Ag-NPs treatment. The LC 50 values for Ag-NPs and CuO-NPs decreased with an increase in time. Also, the nanoparticles increased the activities of protective enzymes (POD, SOD and CAT), inhibited that of detoxification enzymes (A-CHE, ACP and AKP), and reduced total protein concentrations in the BPH. Conclusions These results show that synthesizing nanoparticles using phytohormones may be a safer and environmentally friendly option, which also holds promise for controlling the BPH in rice production.
