Di-(2-ethylhexyl) phthalate (DEHP) is currently the most frequently detected phthalic acid esters (PAEs) compounds and can induce diverse toxicities on aquatic organisms. To understand the molecular responses of fish to DEHP, we performed transcriptomic profiles in liver of tilapia ( Oreochromis niloticus ) which were exposed to environmental concentration of DEHP. A total of 30.10 Mb and 30.16 Mb clean reads were retrieved from the control and DEHP treated libraries, respectively. De-novo assembly of all the clean reads obtained 58,585 unigenes. After comparing the two libraries, 2814 and 1790 genes were identified as significantly increased and depressed, respectively. Gene ontology (GO) classification system and Kyoto Encyclopedia of Genes and Genomes (KEGG) database analysis demonstrated that DEHP significantly disturbed the expression level of genes associated with immunity, endocrine and reproductive system, lipid metabolism and so on. Quantitative real-time PCR was performed to validate the results of RNA-sequencing (RNA-seq) analysis. The resulting data provide new insights for exploring the molecular basis of tilapia response to DEHP exposure.

Flesh quality is influenced by diet components, but the underlying mechanism remains unclear. This study aimed to investigate the effect of replacing soybean meal (SBM) protein with cottonseed protein concentrate (CPC) at different levels (0%, CK; 15%, CPC15; 30%, CPC30 and 45%, CPC45) on the flesh quality of Nile tilapia. The results indicated that different protein sources influenced muscle amino acid composition instead of fatty acid composition. Lower muscle lipid content was found in CPC45, which in turn significantly altered the muscle texture. The hepatic lipid metabolism-related genes were detected and we found that CPC45 significantly suppressed the lipogenesis and promoted lipolysis. Higher content of microbiota-derived butyrate was found in the intestinal content of CPC45 and butyrate could decrease the lipid accumulation in vitro . Replacing SBM with CPC increased the intestinal butyrate to suppress the lipogenesis in the liver which may account for the increased muscle hardness.

Hepcidin (Hepc), as an antimicrobial peptide , plays a vital role in regulating the iron homeostasis and participating in innate immunity. In this study, the open reading frame of Nile tilapia ( Oreochromis niloticus ) hepcidin mRNA ( OnHepc ) was amplified, and its tissue distribution and expression pattern in response to bacterial infection and iron overload were characterized. The results showed that the ORF of OnHepc is 273 bp in length and encodes a peptide with 90 amino acids. Hepcidin has a certain degree of conservation among different species. OnHepc is widely expressed in ten tissues of natural Nile tilapia and is most abundant in the liver. The expression of OnHepc was apparently upregulated in liver, spleen, intestine and gills after challenged by S. agalactiae and A. hydrophila . In addition, the OnHepc expression in MO/Mø and hepatocytes was also upregulated after challenged by the above bacteria. On the other hand, the OnHepc transcripts were upregulated in the liver, spleen, intestine and gills after challenged by 1 μM and 10 μM FeCl 3 solutions, and the same trends were observed in the MO/Mø and hepatocytes after challenged by ferric ions. Moreover, the OnHepc expression was significantly upregulated after stimulated by (r)OnIL-6 protein, suggesting that inflammatory responses regulate the expression of OnHepc . In summary, the present study shows that OnHepc probably gets involved in against bacterial infection and regulating the iron homeostasis in O. niloticus .

Fava bean is currently utilized as a specialized feed ingredient to enhance the muscle texture of fish. When incorporated at appropriate levels, fava beans can improve fish muscle crispy texture. However, high dietary levels of fava beans may lead to liver damage in fish, although the specific mechanisms behind this effect remain unclear. In this study, 600 genetic improvement of farmed tilapia (GIFT; Oreochromis niloticus ) of uniform size (initial body weight 552.3 ± 10.12 g) and healthy appearance were randomly divided into four groups, each with three replicates of 50 fishes. The fish diets contained different levels of fava beans (0, 50%, 60%, and 70%), designated as D0, D50, D60, and D70, respectively, over a ten-week feeding experiment. A comprehensive analysis of the impact of fava beans on muscle texture and liver health in GIFT was performed. The results showed that 50%, 60%, and 70% dietary fava beans had no significant effect on the growth performance of GIFT ( P > 0.05), but 70% dietary fava beans increased feed conversion ratio (FCR) ( P = 0.022). Consumption of fava beans at levels of 50%, 60%, and 70% in the diet altered GIFT muscle texture ( P < 0.05), and adversely effected serum biochemistry and liver health ( P < 0.05). Non-targeted metabolome analysis of the liver revealed that dietary fava beans may cause irreversible damage to liver cells by disrupting the DNA methylation process ( P < 0.05). This finding enhances the understanding of the mechanisms behind the negative impact of fava bean consumption on liver function. It is crucial to investigate potential strategies to mitigate these adverse effects while maintaining the desired muscle quality when fava beans are used in aquatic feed.

Erucic acid (EA) in rapeseed oil has adverse effects on terrestrial animal and fish health. However, its antinutritional role in fish remains unclear due to the limited information on EA. Therefore, this study was conducted to assess the impact of EA on growth performance, antioxidative capacity, fatty acid profile, and lipid metabolism in tilapia. Six diets containing different amounts of EA (0, 3, 6, 12, 20, and 27 g/kg diet) were fed to tilapia (initial weight: 3.01 ± 0.01 g) for 8 weeks. The results exhibited that dietary EA did not affect growth performance but remarkedly increased the crude lipid contents (in the whole body, liver, and muscle). It also markedly increased the levels of low-density lipoprotein cholesterol, total cholesterol, nonesterified fatty acids, and triglyceride in the liver and serum in a dose-dependent manner. The EA groups had lower values of total superoxide dismutase, total antioxidant capacity, catalase, and higher activities of aspartate aminotransferase and alanine aminotransferase, as dietary EA levels increased. Feeding fish with diets containing EA (20 and 27 g/kg diet) significantly increased the malondialdehyde content. Moreover, dietary EA greatly altered the fatty acid profile in the liver and muscle. It especially elevated the percentages of C18 : 2n-6, C20 : 1n-9, and C22 : 1n-9 while decreasing the C18 : 0 and C16 : 0 levels. When the levels of EA in diets were 12, 20, and 27 g/kg, genes correlated with lipophagy, lipolysis, and β -oxidation were significantly reduced. Meanwhile, genes concerned in triglyceride synthesis were largely increased in the liver and muscle. In summary, high-dose EA (20 g/kg diet) in the diets significantly induced fat accumulation, hepatic oxidative damage, and abnormal lipid metabolism in tilapia. The current findings expand our understanding on the antinutritional role of EA in lipid homeostasis and fish health.

Hemopexin, a high affinity heme-binding protein is widely involved in variety physiological and pathological processes. It is an important acute phase response protein, and is important in regulating the inflammatory response. In this study, the open reading frame of Nile tilapia hemopexin ( OnHpx ) gene was amplified. The expression pattern of OnHpx in natural and bacterial challenged tilapia tissues were analyzed through RT-qPCR. The results indicated the OnHpx was most abundant in liver, and increased significantly in liver, spleen, head kidney and peripheral blood after bacterial challenge. Furthermore, the OnHpx mRNA was also significantly up-regulated in monocytes/macrophages and hepatocytes under the stimulation of S. agalactiae or A. hydrophila . In addition, the recombinant OnHpx protein could effectively reduce the bacteria proliferation and alleviate the inflammatory reaction caused by bacteria. Moreover, the (r)OnHpx also regulated the respiratory burst of monocytes/macrophages and played an important role in the antioxidant process. To our knowledge, these results provide the first evidence on the antibacterial and anti-inflammatory response mechanism of Hpx in early vertebrates. This brings new insights about the understanding of the evolutionary origins and ancient roles of the Hpx in the innate immune defense.

Collectins, a subfamily of the C-type lectins, are able to bind non-self glycoconjugates on the surface of microorganisms and inhibit infection by direct neutralization, agglutination and/or opsonization , which play important roles in innate immunity . In this study, a CL11X1-like collectin (OnCL11X1) was identified from Nile tilapia ( Oreochromis niloticus ) and characterized at expression and agglutination functional levels. The open reading frame of OnCL11X1 is 840 bp of nucleotide sequence encoding polypeptides of 279 amino acids . The deduced amino acid sequence is highly homology to teleost and similar to mammalian CL11X1, containing a canonical collagen-like region, a carbohydrate recognition domain and a neck region. Expression analysis revealed that the OnCL11X1 was highly expressed in the liver, and widely exhibited in other tissues including kidney, intestines and spleen. In addition, the OnCL11X1 expression was significantly up-regulated in spleen and anterior kidney following challenges with a Gram-positive bacterial pathogen ( Streptococcus agalactiae ) and a Gram-negative bacterial pathogen ( Aeromonas hydrophila ). The up-regulation of OnCL11X1 expression was also demonstrated in hepatocytes and macrophages in vitro stimulation with S. agalactiae and A. hydrophila . Recombinant OnCL11X1 protein was able to agglutinate both S. agalactiae and A. hydrophila in vitro and promote the phagocytosis by macrophages. Taken together, the results of this study indicated that OnCL11X1, possessing apparent agglutination and opsonization ability to bacterial pathogens, might be involved in host defense against bacterial infection in Nile tilapia.

A high‐level fava beans diet has been proven to enhance the texture characteristics of tilapia muscle, yet it can also induce hepatointestinal injury. Rutin, as a nutritional additive, has antioxidant and immune‐boosting effects. This study explored the regulatory effect of 150 and 300 mg/kg rutin on the muscle characteristics and hepatointestinal health of tilapia after being fed with a 60% fava beans diet. Two hundred and forty tilapias (average weight: 371.50 ± 9.29 g) were evenly divided into four groups and fed with the diet of groups C0 (0% fava bean), R0 (60% fava bean), R1 (60% fava bean + 150 mg/kg rutin), and R2 (60% fava bean + 300 mg/kg rutin) for 10 weeks, respectively. The results indicated that the growth performance in R1 and R2 showed no significant changes compared to R0, while muscle hardness, gumminess, and resilience increased, along with an elevation in muscle crude protein deposition. The addition of 150 and 300 mg/kg rutin enhanced the antioxidant capacity (AOC) of the muscles, liver, and intestines, alleviated liver damage by regulating liver lipid metabolism compared with R0. Dietary supplementation with 150 mg/kg rutin improved the composition of intestinal microbiota, significantly upregulating the abundance of beneficial bacteria Muribaculaceae unclassified and reduced the abundance of harmful bacteria Rhodobacteraceae , Chloroplast_unclassified , and Rothia . In summary, rutin can effectively function as a nutritional supplement to alleviate liver and intestinal damage caused by 60% fava bean feed, while also improving muscle texture simultaneously. This study provides a crucial theoretical basis for optimizing crispy tilapia aquaculture through rutin supplementation, promoting innovation in aquatic feed formulations.

Microfibril-associated glycoprotein 4 (MFAP4), a pattern recognition-like molecule with a fibrinogen-like domain (FBG), has the ability to combine and agglutinate pathogens, playing an essential role in the first line of innate immune defense. In this study, the sequence of Nile tilapia (Oreochromis niloticus) microfibril-associated glycoprotein 4 (OnMFAP4) open reading frame (ORF) was amplified and identified. The ORF of OnMFAP4 is 720 bp of nucleotides and codes for 239 amino acids. Spatial mRNA encoding analysis indicated that OnMFAP4 was highly produced in liver, intestine and head kidney in healthy tilapia, and with the lowest expression in muscle. After challenges with Streptococcus agalactiae ( S. agalactiae ) and Aeromonas hydrophila ( A. hydrophila ), the expression of OnMFAP4 mRNA was prominently produced in the liver, spleen and head kidney. The up-regulation of OnMFAP4 expression was also presented in head kidney monocytes/macrophages (MO/MΦ) and hepatocytes. Recombinant OnMFAP4 ((r)OnMFAP4) could bind and agglutinate both bacterial pathogens. Moreover, (r)OnMFAP4 could take part in the modulation of inflammation and phagocytosis . In conclusion, this study revealed that OnMFAP4 might take effect in host defense against bacterial infection in Nile tilapia , with agglutination and opsonization capability to bacterial pathogens.

Mannose receptor (MR) as a member of the pattern recognition receptors (PRRs) plays an important role in the immune response. In mammals, the role of MR in the regulation of phagocytosis is clarified; however, its contribution to opsonize phagocytosis remains unclear in bony fish. In this study, the expression pattern of Nile tilapia mannose receptor gene ( OnMR ) was investigated and its regulation of the phagocytosis of monocytes/macrophages to pathogenic bacteria was identified. The full-length of OnMR open reading frame is 4314 bp, encoding a peptide containing 1437 amino acid residues. The deduced amino acid sequence revealed that OnMR contained a cysteine-rich domain, a fibronectin type II domain, multiple C-type lectin-like domains, a transmembrane domain and a short cytoplasmic domain. Tissue distribution analysis showed the OnMR transcripts was widely distribute in the ten detected tissues, and highly expressed in head kidney, hind kidney, intestine and spleen . After S. agalactiae and A. hydrophila infection, the expression of OnMR in head kidney and spleen increased significantly. Moreover, the expression of OnMR in MO/Mø were also upregulated post the infection of bacteria and mannose solutions in vitro. This suggested that MR, as a mannose receptor on macrophage surface, could respond strongly to the stimulation of pathogenic bacteria. In addition, the (r)OnMR protein could effectively bind and agglutinate S. agalactiae and A. hydrophila , and regulate the phagocytic ability of monocytes/macrophages to pathogenic bacteria. These results suggest that OnMR is involved in response against bacterial infection in Nile tilapia, and this study will help us better understand the function of MR in teleost fish.

B cell linker protein (BLNK), an adapter of SLP-76 family, plays an extremely important role in B cell receptor (BCR) signaling pathway that regulates proliferation, differentiation, and apoptosis of B cell. BLNK participates in the signal transduction of BCR activation through its typical structure and interaction network with SYK, BTK etc. In this study, Nile tilapia BLNK (OnBLNK) open reading frame is 1470 bp in length for 489 amino acids. OnBLNK composes of a SAM domain at the N-terminal, a central proline-rich region and a highly conserved SH2 domain containing four conserved YXXP tyrosine phosphorylation sites at the C-terminal. Alignment of BLNK sequence and phylogenetic analysis indicated that OnBLNK was highly homologous to that of other teleost fish and mammals. Tissue distribution analysis revealed that OnBLNK showed a wide distribution in all examined tissues and with the highest expression in head kidney . After challenge with Streptococcus agalactiae ( S. agalactiae ) in vivo , the OnBLNK expression was significantly up-regulated in spleen and head kidney. In addition, the significant up-regulation of OnBLNK mRNA expression was also observed in head kidney leukocytes (HKLs) stimulated with S. agalactiae and lipopolysaccharide (LPS) in vitro . Further, it was also demonstrated that expression of OnBLNK mRNA was obviously up-regulated when HKLs were stimulated by mouse anti-tilapia IgM monoclonal antibody in vitro , and declined to normal after treatment with LYN, SYK or BTK inhibitor, respectively. Overall, the results suggest that OnBLNK might take part in host defense during bacterial infection, and get involved in the BCR signaling pathway. This study might aid to better understand the regulatory mechanism of BCR signaling pathway in teleost fish.

Dissolved organic matter (DOM) is likely to coexist with microplastics (MPs) in aquatic environments. However, little is known about the effects of different DOM fractions on the stability and aquatic toxicity of MPs. In this study, we separated humic acid (HA) into three molecular weight (MW) fractions (> 30, 3–30, and < 3 k Da) and examined their effects on the toxicity of polyamide 66 (PA66)-MPs to red tilapia (Oreochromis niloticus). Generally, addition of HA enhanced the stability and thus the exposure of tilapia to MPs, leading to the increased accumulation of MPs in the gut, liver, and brain. However, compared with MPs alone, the accumulation of MPs in the gills of tilapia was reduced. Compared with the presence of high-MW HA, the accumulation of MPs in the fish liver was significantly enhanced by 1.1 times in the presence of medium-MW HA (m-HA) after a 10-day exposure. The protein peroxidation on the liver induced by MPs was alleviated by adding all HA fractions. However, compared with MPs alone, the presence of m-HA resulted in a more severe lipid peroxidation, while the presence of low-MW HA alleviate the lipid peroxidation caused by MPs. In addition, the Integrated Biological Responses version 2 (IBRv2) results suggested that the oxidative stress in the liver tilapia caused by MPs could be alleviated by the presence of all HA fractions, which was largely related to the reduced damage caused by lipid peroxidation and/or protein peroxidation. Collectively, our results suggest that the presence of different MW HA fractions could induce complex changes in the MP toxicity on aquatic organisms.
