Polybrominated diphenyl ethers (PBDEs) are a typical group of persistent organic pollutants (POPs) widely present in the environmental media and aquatic products. However, knowledge about the content and source of PBDEs in aquatic products from freshwater farms is lacking. Herein, a sensitive and robust method for the detection of 25 PBDE congeners was established. The concentrations of PBDEs were analyzed in aquatic products (fish, shrimp, and crab), along with their feed, water, and sediment collected from the lower reaches of the Yangtze River. Results showed the mean concentration of ∑PBDEs in crab (210.2 pg g −1 ww) was significantly higher than in shrimp (33.3 pg g −1 ww), crucian carp (33.2 pg g −1 ww), and black carp (24.3 pg g −1 ww), indicating a higher bioaccumulation rate of PBDEs in crab than the others. The highest concentrations of PBDEs were found in crab hepatopancreas, accounting for over 77.6% of the relative body burdens (RBBs) of these contaminants. The content of PBDE congeners in different aquatic products exhibits interspecies variation, whereas similar patterns are observed within the same aquatic products across different bases. No obvious dietary risk was found for all collected aquatic products under the two assessment strategies. Commercial feed was the main pollution route for all the farming bases and, in farming bases with snails and frozen fish, the contribution of these animal baits is also non-negligible. This study comprehensively elucidates the occurrence, risk and source of PBDEs contamination in farmed freshwater aquatic products, offering significant insights into exposure risks and the development of prevention strategies, which are crucial for ensuring the safety of aquatic products.

This study investigated the expediated transformation of halophenols in the presence of nitrite (NO 2 − ) under slightly acidic conditions in ice, whereas such transformation was negligible in liquid water at 4 °C. We proposed that this phenomenon was attributed to the freeze-concentration effect, incurring a pH drop and the aggregation of NO 2 − and halophenols within the liquid-like grain boundary layer amid ice crystals. Within this micro-environment, NO 2 − underwent protonation to generate reactive nitrous acid (HNO 2 ) and nitrosonium ions (NO + ) that facilitate the nitration and oxidation of halophenols. When 10 μМ halophenol was treated by freezing in the presence of 5 μМ NO 2 − , the total yields of nitrated products reached 2.4 μМ and 1.4 μМ within 12 h for 2-chlorophenol (2CP) and 2-bromophenol (2BP), respectively. NO + drove oxidative coupling reactions, generating hydroxyl polyhalogenated diphenyl ethers (OH-PBDEs) and hydroxyl polyhalogenated diphenyls via C-O or C-C coupling. These two pathways were intricately intertwined. The presence of natural organic matter (NOM) mitigated the formation of nitrated products and completely suppressed the coupling products. This study offers valuable insights into the fate of halophenols in ice and suggests potential pathways for the formation of nitrophenolic compounds and OH-PBDEs in natural cold environments. These findings also open up a new avenue in environmental chemistry research.

Polybrominated diphenyl ethers (PBDEs) and their metabolite 4-bromodiphenyl ether (BDE3) are pervasive environmental contaminants suspected of endocrine-disrupting effects. This study investigates their inhibitory potential and mechanism of action against human (h3β-HSD1) and rat (r3β-HSD4) placental 3β-hydroxysteroid dehydrogenase, a pivotal enzyme in progesterone biosynthesis. Comprehensive enzymatic analysis revealed that among eight tested PBDE congeners and the structural analog 4-bromobiphenyl (BDP), only BDE3 and BDP exhibited significant inhibition. BDE3 was a potent, mixed/competitive inhibitor with IC 50 values of 5.86 μM (h3β-HSD1) and 12.35 μM (r3β-HSD4), significantly more potent than BDP (IC 50 ∼ 100 μM). Mechanistic studies using pregnenolone as substrate confirmed mixed/competitive inhibition for both compounds, indicating binding at the steroid substrate site, while analysis with NAD⁺ suggested mixed/noncompetitive inhibition. Molecular docking and dynamics simulations corroborated these findings, indicating BDE3 and BDP binding at the steroid/NAD⁺ interface with favorable binding energies, stabilizing the active-site conformation. Network toxicology analysis identified HSD3B1 (encoding h3β-HSD1) as a central hub gene linking PBDE exposure to preeclampsia, enriched in steroidogenesis and estrogen signaling pathways. Single-cell RNA-seq analysis revealed predominant HSD3B1 expression in extravillous and syncytiotrophoblasts, with reduced expression in preeclampsia. In silico virtual knockout of HSD3B1 predicted downstream disruption of cell cycle regulation and steroidogenic pathways. Pharmacokinetic predictions indicated both compounds have high intestinal absorption but potential hepatotoxicity. These results suggest that BDE3 may act as a potent inhibitor of placental 3β-HSD, potentially linking its disruption of progesterone synthesis to adverse pregnancy outcomes.

Purpose Remediation of polybrominated diphenyl ethers (PBDEs)/heavy metals/antibiotics mixed contaminated soil with high abundance of antibiotic resistance genes (ARGs) has being greatly challenging. Materials and methods An elevated soil washing procedure was developed to investigate the potential of applying DNA solution to extract PBDEs, Cu, Pb, tetracycline, and sulfadiazine from a high abundance of ARG-affected soil, followed by microbial augmentation to restore the microbiological functioning of washed soil. Results and discussion Two successive washings with 15 g L −1 DNA solution plus ultrasonication (25 kHz for 15 min) were optimal for extracting 80.6 % BDE28, 85.2 % BDE47, 64.7 % Cu, 60.3 % Pb, 100 % tetracycline, and 100 % sulfadiazine. Simultaneously, the ARG abundance decreased to the level of 10 −7 –10 −8 (ARGs copies/16S copies). Moreover, combined treatment with PBDE-degrading bacteria inoculation and nutrient application for 3 months to the second washed soil resulted in the further degradation of residual BDE28 and BDE47. Furthermore, the bacterial community composition was restored and the transfer risk of residual pollutants was limited, as indicated by high-throughput sequencing technology analysis and heavy metals stability model evaluation. Conclusions The combined cleanup technology is an environmentally friendly and effective technology to remediate mixed contaminated soils.

Here is reported effervescent reaction-enhanced switchable hydrophilic-hydrophobic solvent-based salting-out microextraction (ERSSM) with high-performance liquid chromatography with photodiode array detection (HPLC-PDA) for the preconcentration, extraction, and trace-level determination of polybrominated diphenyl ethers (PBDEs) in milk and water. The important variables were rigorously optimized to be 150 µL of hexanoic acid as the extraction solvent, 400 µL of Na2CO3 as the alkaline source, 350 µL of H2SO4 solution as the acid source, and NaCl as the salting-out agent. The optimized conditions provided a linear range from 0.2 to 50 µg L−1 with limits of detection and quantitation from 0.026 to 0.085 µg L−1 and 0.087 to 0.28 µg L−1 for six PBDEs. The intra-day and inter-day precision values were from 2.48% to 6.75%, and the fortified recoveries for PBDEs were between 78.06% and 103.07% in water and milk samples. The developed procedure avoids the use of a traditional organic disperser and mechanical mixing allowing the determination of PBDEs outdoors by synchronous extraction method and high efficiency for the analytes. These advantages suggest a high potential for the routine monitoring of PBDEs in liquid food and environmental samples.
