Dynamic culturing of large cell-loaded PCL/gelatin methacryloyl scaffolds for bone critical size defect repair

Guangxin Shen,Botao Gao,Jiayi Guo,Weikang Xu,Guangfu Chen,Shuai Huang,Zhiwen Zeng,Xiaodong Zhao

Due to the limited ability to self-repair, the regeneration of bone critical-sized defects (CSD) is a significant challenge. Bone tissue engineering scaffolds are considered promising candidates for CSD repair, but low cell infiltration efficiency and a lack of nutrients greatly restrict bone regeneration abilities. Herein, we developed a dynamic culturing of large biomimetic bone scaffolds, PCL/GelMA@cells that combining 3D printed polycaprolactone (PCL) multi-channel cylinder with gelatin methacryloyl (GelMA) encapsulated with bone marrow mesenchymal stem cells (BMSCs) and rat aortic endothelial cells (RAECs). A cell dynamic culture system was fabricated to simulate the dynamic microenvironment. Compared to static culturing, dynamic culturing proved to enhance the nutrient exchange within the large scaffold to promote the cells infiltration, growth, proliferation and induce osteogenic and angiogenic differentiation. Furthermore, a rat cranial CSD (D = 10 mm) repair model verified the accelerated vascular ingrowth and new bone formation with the implantation of dynamic culturing of PCL/GelMA@cells scaffold (∼10 times higher than Blank group), indicating the great potential of dynamical culturing of scaffolds for bone repair. In summary, the results highlight the significant advantages of the dynamical culturing of cell-loaded scaffolds for bone regeneration, offering a promising strategy for addressing critical size bone defects.

Recent progress of mechanism of mineralization process induced by Ta2O5/PCL scaffolds

Chen Gao,Junbo Jiang,Juan Wang,Yutong Wang,Yangyang Zhang,Caihua Rao,Yue Cheng,Zhongrong Chen,Runhuai Yang,Gang Zhao

Mineralization of hydroxyapatite mineralized by acidic metal oxides has attracted substantial research interest in the research and development of orthopedic substitutes. In this study, Polycaprolactone (PCL) nanofibers with nano-tantalum oxide (Ta 2 O 5 ) were successfully fabricated through electrospinning. The scaffold simulates the mineralization process of hydroxyapatite in simulated body fluid. However, in the process of mineralization, there is a chemical reaction between hydrophobic scaffolds and acid oxides, which is believed to have a significant impact on the biological effects of the scaffolds. The results of SEM, EDS, XRD, Mapping, FTIR, GPC, 13 CNMR, and 1 HNMR explain the principle of hydroxyapatite mineralization by Ta 2 O 5 and the reason why the composite scaffold is blurred, that is, the carbonyl carbon of PCL attacked by the surface hydroxyl groups of Ta 2 O 5 to catalyze its ring-opening polymerization to synthesize PCL/Ta 2 O 5 nanocomposites in one step. In order to accurately explore the effect of this reaction on the metabolism of biological macromolecules in the future, we provide a detailed route of Ag Nanoparticles -Dimercaptosuccinic acid@Transcription activator (AgNPs-DMSA@TAT) probe synthesis. The results provide a reference for the sedimentation of hydroxyapatite induced by acid oxides and its effect on traditional electrospun fiber scaffolds.

Improved surface adhesion and wound healing effect of madecassoside liposomes modified by temperature-responsive PEG-PCL-PEG copolymers

Meifeng Liu,Weichi Chen,Xingyu Zhang,Pengwen Su,Feng Yue,Shaoqun Zeng,Song Du

Madecassoside (MA) exhibits excellent therapeutic effects in wound healing and scar management. However, its high hydrophilic nature and low permeability through skin tissue limits its topical application. Liposomes are widely used to deliver drugs due to their high structural similarity and biocompatibility with cell membranes. However, normal liposome formulations are too fluid to maintain sufficient adhesion to the wound surface. In this study, in order to make an MA formulation conducive to topical administration, poly(ethylene glycol)-poly( ε -caprolactone)-poly(ethylene glycol) (PEG-PCL-PEG, PECE), a biodegradable and temperature-responsive copolymer material, was synthesized and applied to improve the adhesion properties of MA liposomes. The mean particle size of the PECE-modified MA liposomes was 213.43±4.68 nm, and the zeta potential was −23.80±15.37 mV under the optimal conditions of EPC (egg yolk lecithin) to PECE at a mass ratio of 1:1. Additionally, PECE-modified MA liposomes maintained a hydrogel state for better adhesion until the temperature reached 43°C. Furthermore, the PECE-modified MA liposomes showed superior wound contraction effects relative to the MA liposomes in second-degree burn experiments using a rat model. These results indicated that PECE-modified MA liposomes have better surface adhesion performance and healing effects than unmodified MA liposomes.

Redox-sensitive micelles assembled from amphiphilic mPEG-PCL-SS-DTX conjugates for the delivery of docetaxel

Huiyuan Zhang,Kaiming Wang,Pei Zhang,Wenxiu He,Aixin Song,Yuxia Luan

Docetaxel (DTX) can produce anti-tumor effects by inhibiting cell growth and inducing apoptosis. However, the poor solubility of DTX restricts its application and its clinical formulation has caused serious adverse reaction due to the use of Tween-80. In the present study, DTX was conjugated to an amphiphilic di-block polymer to solve these problems. Methoxy poly(ethylene glycol)-poly(ε-caprolactone) (mPEG-PCL) was selected as the polymer skeleton and a redox sensitive disulfide bond was used as the linker between DTX and mPEG-PCL. The synthesized mPEG-PCL-SS-DTX conjugates were characterized by 1 H-nuclear magnetic resonance ( 1 H NMR) and Fourier transform infrared spectroscopy (FTIR). Interestingly, the mPEG-PCL-SS-DTX conjugates could self-assemble into micelles in aqueous solution. The critical micelle concentration (CMC) of mPEG-PCL-SS-DTX micelles was about 2.3   mg   L −1 determined using pyrene molecule fluorescent probe method while the size of mPEG-PCL-SS-DTX micelles was determined to be ca . 17.6   nm and 116.0   nm with a bimodal distribution by dynamic light scattering (DLS). The in vitro release results indicated that the as-prepared micelles exhibited a sustained release profile with good redox sensitive properties. In particular, the hemolytic toxicity test indicated the as-prepared mPEG-PCL-SS-DTX micelles had negligible hemolytic activity, demonstrating their safety in drug delivery system. Cytotoxicity assay of the mPEG-PCL-SS-DTX micelles verified their highly enhanced cytotoxicity to MCF-7/A and A549 cells. These results thus demonstrated that the present redox-sensitive mPEG-PCL-SS-DTX micelle was an efficient and safe sustained drug delivery system in the biomedical area.

Electrospun PLGA/PCL Nanofiber Film Loaded with LPA Promotes Full-Layer Wound Healing by Regulating the Keratinocyte Pyroptosis

Xinqi Huang,Jianghuiwen Lu,Yumei An,Mingyuan Xu,Xueshi Chen,Chao Liu,Xuefeng Zhou,Haiyan Shan,Yunzhu Qian,Mingyang Zhang

Electrospun nanofibers have a number of qualities that make them a suitable choice for skin wound healing. Lysophosphatidic acid (LPA) stimulates the keratinocytes and fibroblasts to proliferate, differentiate, and migrate and enhances skin wound healing. Here, we developed the electrospun scaffolds contained in polycaprolactone (PCL) and polylactic-co-glycolic acid (PLGA). The scaffolds loaded with LPA nanoparticles retained a porous nanofiber structure and showed better physicochemical properties and biocompatibility. The scaffold continuously releases LPA to quickly initiate cell signaling and maintain long-term anti-inflammatory activity. In this study, we found that PP scaffold with LPA reduces the disordered collagen deposition and the thickness of the newborn epidermis, improves skin healing, and reduces scar formation. Explaining the mechanism of LPA mineralized tissue regeneration in skin wound healing, LPA inhibited the pyroptosis of keratinocyte, a cell death process that induces inflammation and scar formation by inhibiting the expression of TNF-α and β-catenin proteins. Thus, the electrospun PP scaffold with LPA can be potentially developed as a therapeutic avenue to target skin wound healing.

Fabrication and characterization of electrospun PCL/Antheraea pernyi silk fibroin nanofibrous scaffolds

Xiufang Li,Qiang Zhang,Dezhan Ye,Jie Zhang,Yuhang Guo,Renchuan You,Shuqin Yan,Mingzhong Li,Jing Qu

To engineer tissue restoration, it is necessary to provide a bioactive, mechanically robust scaffold. Electrospun poly(ε-caprolactone) (PCL) nanofiber is a promising biomaterial candidate with excellent mechanical properties, but PCL scaffolds are inert and lack natural cell recognition sites. To overcome this problem we investigated the incorporation of Antheraea pernyi silk fibroin (ASF) containing inherent RGD tripeptides with PCL in electrospinning process. The mixing ratios showed remarkable impact on the properties of hybrid nanofibers. Increasing PCL content significantly enhanced the mechanical properties of nanofibers. In particular, the mechanical properties were remarkably enhanced when PCL content increased from 50 wt% to 70 wt%. Moreover, the biological assays based on endothelial cells showed promoted cell viability when ASF content reached to 30 wt%. The data demonstrated that the nanofiber containing 70% of PCL and 30% of ASF achieved the most balanced performances for integrating the mechanical properties of PCL and the bioactivity of ASF. Furthermore, biomimetic alignment of 70PCL/30ASF nanofibers was achieved, which could support PC12 neuron-like cell growth and guide neurite outgrowth, providing a potentially useful option for the engineering of oriented tissues. The results show that the PCL/ASF hybrid nanofibers can be considered as a promising candidate for tissue engineering scaffolds. POLYM. ENG. D@MSNs-P/PCL antibacterial nanofibers combined with DBD cold plasma for fresh pork preservation

Zhumiao Ye,Haidu Yuan,Jianhao Zhang,Wenjing Yan,Changrui Xing

Fresh pork is easily contaminated by microorganisms, leading to spoilage. Dielectric barrier discharge (DBD) cold plasma technology has shown significant advantages in the preservation of meat. However, severe lipid oxidation of pork resulting from DBD treatment greatly limits its development. Herein, the antibacterial D@MSNs-P/PCL nanofiber membranes were prepared by modification polycaprolactone (PCL) with pre-reported nanomaterials (D@MSNs-P) through electrostatic spinning. Compared with the PCL membranes, the D@MSNs-P/PCL membranes showed better water stability, lower water vapor transmission rate, stronger hydrophobicity, and potent antibacterial activity. The D@MSNs-P/PCL membranes can be used for wrapping fresh pork before treatment with DBD plasma, which can not only improve the antibacterial efficiency but effectively inhibit the lipid oxidation and color change of fresh pork, extending the shelf life for 2 days. The developed D@MSNs-P/PCL membranes can also be used alone to preserve other fresh meat or oily food, with a wide range of application prospects.

Biomimetic glycopeptide hydrogel coated PCL/nHA scaffold for enhanced cranial bone regeneration via macrophage M2 polarization-induced osteo-immunomodulation

Yaping Wang,Jingrong Wang,Rui Gao,Xiang Liu,Zujian Feng,Chuangnian Zhang,Pingsheng Huang,Anjie Dong,Deling Kong,Weiwei Wang

The reconstruction of large cranial bone defects by bioactive materials without exogenous cells or growth factors remains a substantial clinical challenge. Here, synthetic fibrous glycopeptide hydrogel (GR gel ) self-assembled by β-sheet RADA16-grafted glucomannan was designed to mimic the glycoprotein composition and the fibrillar architecture of natural extracellular matrix (ECM), which was non-covalently composited with 3D-printed polycaprolactone/nano hydroxyapatite (PCL/nHA) scaffold for cranial bone regeneration . The glycopeptide hydrogel significantly promoted the proliferation, osteogenic differentiation of bone mesenchymal stem cells (BMSCs), which was further augmented by GR gel -induced macrophage M2-phonotype polarization and the effective M2 macrophage-BMSC crosstalk . The repair of critical-size skull bone defect in rat indicated a superior efficacy of PCL/ [email protected] gel implant on bone regeneration and osseointegration , with an average bone area of 83.3% throughout the defect location at 12 weeks post treatment. Furthermore, the osteo-immunomodulatory GR gel induced a reparative microenvironment similar with that in normal cranium, as characterized by an increased percentage of anti-inflammatory M2 macrophages and osteoblasts, and high-level vascularization . Collectively, the composite scaffold developed here with macrophage polarization-mediated osteo-immunomodulation may represent a promising implant for expediting in situ bone regeneration by providing biochemical and osteoinductive cues at the injured tissue.

pH/redox dual-sensitive nanoparticles based on the PCL/PEG triblock copolymer for enhanced intracellular doxorubicin release

Yan Cao,Junqiang Zhao,Yumin Zhang,Jianfeng Liu,Jinjian Liu,Anjie Dong,Liandong Deng

pH/redox dual-sensitive nanoparticles (NPs) based on a poly(ε-caprolactone) (PCL) and polyethylene glycol (PEG) triblock copolymer were developed and investigated aiming at improving the drug release property of PCL in cancer chemotherapy. A redox-sensitive disulfide bond and pH-sensitive benzoic-imine linkers were introduced to the backbone of the amphiphilic block copolymer termed SCHE, which could self-assemble into stable spherical NPs in aqueous solutions with an average size of about 100 nm. Doxorubicin (DOX) as a hydrophobic anticancer drug was loaded into the NPs by a dialysis method. The in vitro drug release results showed that the accumulative release amount of DOX at pH 5.0 with 10 mM glutathione (GSH) was accelerated apparently by more than twice compared to that at pH 7.4 without GSH. After a pre-treatment with GSH, the intracellular fluorescence intensity of Hela cells was enhanced compared to those without the pretreatment, which indicated faster DOX release in cells with higher GSH concentration. In an MTT assay, no obvious toxicity of blank NPs was found. In conclusion, SCHE NPs could serve as a novel colloidal drug delivery system in cancer chemotherapy and the introduced unstable linkers could enhance the drug release rate at tumor sites.

Anti-Neoplastic Cytotoxicity of SN-38-Loaded PCL/Gelatin Electrospun Composite Nanofiber Scaffolds against Human Glioblastoma Cells In Vitro

Xiaodong Zhu,Shilei Ni,Tongliang Xia,Qingyu Yao,Haoyuan Li,Benlin Wang,Jiangang Wang,Xingang Li,Wandong Su

ABSTRACT: Electrospun poly(ε-caprolactone) (PCL)/gelatin (GT) scaffolds were developed to provide controlled release of 7-ethyl-10-hydroxy camptothecin (SN-38). Acetic acid was introduced to improve the miscibility of PCL and GT to produce a homogeneous nanofiber membrane mixture. The effect of SN-38 content in binary mixtures on processability, fiber morphology, water sorption, swelling, and drug release was investigated. Electrospun PCL/GT blend nonwoven fibers showed fiber surface roughness, decreased PCL crystallinity, and increased swelling with increasing drug content of 1, 2, and 4 wt %. Additionally, increasing the SN-38 concentration reduced the degradation rate of the GT. Furthermore, we hypothesize the existence of a drug content saturation point in the monoaxial fiber to explain the different drug release patterns of PG2 compared with those of PG1 and PG4. The matrix also showed good biodegradation and anti-tumor function. Our results demonstrate that SN-38-loaded PCL/GT fibers can be obtained by electrospinning. The SN-38-loaded fibers merit further evaluation as a means to potentially prevent locoregional recurrence following surgical tumor resection.

Construction of a novel GAP/PCL energetic self-healing blend adhesive system for propellants based on the synergistic effect of hydrogen bond reorganization and disulfide bond exchange reactions

Mianji Qiu,Zhi Geng,Wangjian Cheng,Chengyuan Hua,Xing Yan,Baoyun Ye,Chongwei An,Jingyu Wang

Solid propellants are essential for rocket engine efficiency and safety. The introduction of energetic self-healing adhesives can improve its energy density, mechanical properties, and safety, while extending its service life. Therefore, this paper proposes a novel energetic self-healing adhesive that utilizes asymmetric alicyclic isophorone diisocyanate (IPDI) and 2-(aminophenyl) disulfide (2-AFD), which has a bent biphenyl ring structure, as hard segments, combined with semicrystalline polymer polycaprolactone (PCL) and energetic adhesive glycidyl azide polymer (GAP) as soft segments to synthesize a series of self-healing adhesives. By adjusting the hard segment content, a balance between mechanical properties and self-healing performance is achieved. Results show that increasing the hard segment content enhances toughness (from 26.60 MJ·m −3 to 58.54 MJ·m −3 ), but decreases self-healing efficiency (from 90 % to 58 %). GPPU-2 exhibits 38.95 MJ·m −3 toughness and recovers 86 % of its toughness within 90 min at 80 °C via dynamic disulfide and hydrogen bond interactions. The GPPU-2-based propellant, GPPU80, recovers 88.22 % of tensile strength after 24 h at 80 °C, demonstrating effective crack propagation inhibition. Compared to inert adhesive-based propellants, GPPU80 shows superior combustion performance and energy release. This study offers insights for designing high-performance self-healing adhesives to enhance propellant safety and energy density.

3D-printed CoSi/PCL composite scaffold with NIR-II photothermal ability and enhanced adipogenic activity for breast reconstruction after mastectomy

Jupei Zhang,Hangbin Xia,Xuerui Zhou,Zhaoxu Meng,Qishu Jin,Dongmin Chen,Xiaojuan Xia,Yiren Jiao,Jiang Chang,Zhihong Dong,Zhen Zeng,Hongshi Ma,Chen Yang

Breast reconstruction after mastectomy aims to restore breast appearance and function. Current scaffolds often fail to meet the complex clinical demands of post-mastectomy breast reconstruction, which include personalized shaping, prevention of tumor recurrence and metastasis, and promotion of adipose tissue regeneration. This study aims to address these challenges by combining a readily processable biopolymer, polycaprolactone (PCL), with a multifunctional bioactive ceramic, cobalt orthosilicate (Co 2 SiO 4 , CoSi) to form a multi-functional CoSi/PCL composite scaffold through 3D printing technology. The scaffold exhibits a controllable shape with designed macroporous architecture. Its photothermal performances are well presented in both near-infrared (NIR) I and NIR-II regions, enabling effective tumor ablation with minimal side effects. Additionally, the release of bioactive silicate and cobalt ions promotes adipogenesis and angiogenesis, thereby enhancing the integration of the scaffold with surrounding tissues. This approach offers a promising strategy for combining tumor therapy with breast reconstruction, providing a clinically relevant solution for post-mastectomy patients.
