Bilayered HA/CS/PEGDA hydrogel with good biocompatibility and self-healing property for potential application in osteochondral defect repair

Baihao You,Qingtao Li,Hua Dong,Tao Huang,Xiaodong Cao,Hua Liao

The fabrication of osteochondral tissue engineering scaffolds comprised of different layers is a big challenge. Herein, bilayers comprised of double network hydrogels with or without nano hydroxyapatite (HAp) were developed by exploiting the radical reaction of poly(ethylene glycol) diacrylate (PEGDA) and the Schiff-base reaction of N-carboxyethyl chitosan (CEC) and oxidized hyaluronic acid sodium (OHA) for osteochondral tissue engineering. The bilayered osteochondral scaffold was successfully fabricated based on the superior self-healing property of both hydrogels and evaluated by scanning electron microscopy, macroscopic observation and mechanical measurements. In addition, the hydrogels exhibited good biocompatibility as demonstrated by the in vitro cytotoxicity and in vivo implantation tests. The results indicated that the bilayered hydrogel had great potential for application in osteochondral tissue engineering.

Construct functional PEGDA/CSMA/CuII photosensitive ink for SLA 3D printing of blood contact appliance

Jiang Shao,Yanqiu Liu,Yang Li,Yuanfeng Wang,Ruolan Li,Li Yao,Yian Chen,Yunji Fei,Jingyue Wang,Ansha Zhao

Stereolithography (SLA) is an emerging technology in three-dimensional printing that has garnered significant attention in the biomedical field due to its high printing fidelity, good biocompatibility, and high spatial resolution. Polyethylene glycol diacrylate (PEGDA) is commonly used as a photosensitive printing ink in SLA, and it possesses excellent biocompatibility, photosensitivity, processability, water solubility, and biomechanical tunability. Additionally, PEGDA exhibits unique anti-protein, platelet, and cell adhesion possibilities, making it a promising material for blood contact applications. However, cross-linking PEGDA using chain growth photopolymerization results in a high concentration of reactive functional groups, leading to high print volume shrinkage, decreased precision of the finished product, warping, deformation, and cracking. This study introduces a chemically modified methacryloylated chitosan (CSMA) as a cross-linking agent into the PEGDA ink. This strategy effectively reduces the volume shrinkage of the ink and imparts antimicrobial properties to the final product. The dual anticoagulation properties of PEGDA were achieved by loading divalent copper ion (Cu II ) into the ink, which better addresses the antimicrobial and anticoagulant needs in the physiological environment of blood. The results show that SLA printable inks based on PEGDA/CSMA/Cu II have low swelling, high print fidelity, high stability, and good cell compatibility. Furthermore, by verifying the functionality of PEGDA/CSMA/Cu II , the research shows that the bioprinting ink exhibits excellent antibacterial and anticoagulant properties, making it a promising material candidate for SLA 3D printing of blood purification, central venous catheter, and other semi-implanted blood materials. This study provides new ideas for developing multifunctional photosensitive inks in biomedical engineering.

Chiral, pH responsive hydrogels constructed by N-Acryloyl-alanine and PEGDA/α-CD inclusion complex: preparation and chiral release ability

Jie Liu,Ru Cheng,Jianping Deng,Youping Wu

Chiral, pH-responsive hydrogels are constructed by poly(ethylene glycol) diacrylate/ α -cyclodextrin (PEGDA/ α -CD) inclusion complex and L- N -acryloyl-alanine or D- N -acryloyl-alanine (L-NAA or D-NAA) by an effective free radical polymerization approach. PEGDA containing two C=C end groups was used simultaneously to introduce α -CD units in the resulting hydrogels and to serve as a cross-linking agent, by which forming the designed hydrogels in quantitative yield. Hydrophilic α -CD moieties acted as pore-forming agent, while the L(D)-NAA-based polymer chains bearing –COOH groups enabled the hydrogels to display remarkable swelling–deswelling behavior in response to pH variation. The chiral NAA monomer-derived polymer chains rendered the hydrogels with intriguing optical activity, according to circular dichroism spectra. Scanning electron microscopy revealed the uniformly porous microstructures of hydrogels. More remarkably, the L-NAA-based hydrogels preferentially adsorbed trans -4-hydroxy- d -proline and preferentially released trans -4-hydroxy- l -proline, while D-NAA-based hydrogels provided opposite results. The hydrogels also demonstrated remarkable enantioselective release ability towards chiral drug ibuprofen. Copyright © 2015 John Wiley & Sons, Ltd.

A Controlled Biodegradable Triboelectric Nanogenerator Based on PEGDA/Laponite Hydrogels

Zhe Li,Cong Li,Wei Sun,Yuan Bai,Zhou Li,Yulin Deng

Implantable and wearable transient electronics based on nanogenerators have been applied in self-powered sensing, electrical-stimulation therapy, and other fields. However, the existing devices have a poor ability to match with the shapes of human tissues, and the degradation processes cannot meet individual needs. In this work, a PEGDA/Lap nanocomposite hydrogel was prepared that was based on biocompatible polyglycol diacrylate (PEGDA) and laponite, and a biodegradable single-electrode triboelectric nanogenerator (BS-TENG) was built. The PEGDA/Lap hydrogel has enhanced flexibility and mechanical and electrical performance. Its strain was 1001.8%, and the resistance was 10.8. The composite hydrogel had a good biocompatibility and could effectively promote the adhesion of cells. The BS-TENG could be used as a self-powered device to light an LED and serve as an active sensor for real-time monitoring of breath and various human movements. More importantly, the device could be degraded controllably without any harm. Therefore, BS-TENGs will be mainstream in diagnosis and treatment and play an important role in biomedical science.

In situ cross-linked fluorinated gel polymer electrolyte based on PEGDA-enabled lithium-ion batteries with a wide temperature operating range

Han He,Yue Wang,Meng Li,Jingyi Qiu,Yuehua Wen,Junhong Chen

Lithium-ion batteries (LIBs) require a wide operating temperature window to meet their actual demand. Herein, a fluorinated gel polymer electrolyte (F-GPE) was synthesised by in situ thermal polymerisation method using poly (ethylene glycol) diacrylate (PEGDA) as a cross-linking agent. The F-GPE showed excellent electrochemical stability in a wide voltage window in the range of 2–5.6 V, high ionic conductivity (above 1.2 mS cm −1 at –40 °C), superior lithium-ion transfer number (t Li + ) of 0.51 at room temperature, and good compatibility with lithium metal. Additionally, the LiCoO 2 ||F-GPE||graphite pouch battery, which the surface density of LiCoO 2 was 30 mg cm −2 , showed an outstanding discharge capacity in a wide temperature from –40 to 60 °C. The results displayed the retention rate of discharge capacities at –40 and 60 °C were 64.9% and 106.3% of that at room temperature, respectively. F-GPE retained the low-temperature performance of the electrolyte and improved the stability and electrochemical behavior at high temperature, providing a new strategy for the large-scale application of LIBs in a wide operating temperature.

Recombinant collagen coating 3D printed PEGDA hydrogel tube loading with differentiable BMSCs to repair bile duct injury

Yang Xiang,Yuanhui Gao,Qiuhua Cheng,Zhongwen Lei,Xiaoyu Zhang,Yijun Yang,Jianquan Zhang

Bile duct injury presents a significant clinical challenge following hepatobiliary surgery, necessitating advancements in the repair of damaged bile ducts is a persistent issue in biliary surgery. 3D printed tubular scaffolds have emerged as a promising approach for the repair of ductal tissues, yet the development of scaffolds that balance exceptional mechanical properties with biocompatibility remains an ongoing challenge. This study introduces a novel, bio-fabricated bilayer bile duct scaffold using a 3D printing technique. The scaffold comprises an inner layer of polyethylene glycol diacrylate (PEGDA) to provide high mechanical strength, and an outer layer of biocompatible, methacryloylated recombinant collagen type III (rColMA) loaded with basic fibroblast growth factor (bFGF)-encapsulated liposomes (bFGF@Lip). This design enables the controlled release of bFGF, creating an optimal environment for the growth and differentiation of bone marrow mesenchymal stem cells (BMSCs) into cholangiocyte-like cells. These cells are instrumental in the regeneration of bile duct tissues, evidenced by the pronounced expression of cholangiocyte differentiation markers CK19 and CFTR. The PEGDA//rColMA/bFGF@Lip bilayer bile duct scaffold can well simulate the bile duct structure, and the outer rColMA/bFGF@Lip hydrogel can well promote the growth and differentiation of BMSCs into bile duct epithelial cells. In vivo experiments showed that the scaffold did not cause cholestasis in the body. This new in vitro pre-differentiated active 3D printed scaffold provides new ideas for the study of bile duct tissue replacement.

Transdermal delivery of therapeutics through dissolvable gelatin/sucrose films coated on PEGDA microneedle arrays with improved skin permeability

Ya Gao,Mengmeng Hou,Ruihao Yang,Lei Zhang,Zhigang Xu,Yuejun Kang,Peng Xue

Microneedles are primarily designed for enhancing transdermal drug delivery in a minimally invasive manner. In particular, coated microneedles have attracted increasing attention due to the resulting mode of rapid and highly efficient drug administration. In this study, we developed a novel gelatin/sucrose film-coated poly(ethylene glycol)diacrylate (PEGDA) microneedle patch, which can effectively improve the skin permeability of therapeutic drugs and realize convenient and efficient drug administration. A simple and reliable technique was proposed to produce uniform film coatings on microneedles at room temperature. After introducing a prepolymer solution into the mold cavity, a film-coated microneedle patch was fabricated via a photo-induced polymerization process. Four categories of molecular models, including rhodamine B (RhB), bovine serum albumin (BSA), doxorubicin (DOX) and indocyanine green (ICG), were encapsulated into the gelatin/sucrose film to evaluate their transdermal delivery and therapeutic effects both in vitro and in vivo. The presented methodology represents a facile technique for rapid formation of film-coated microneedles for efficient transdermal delivery of small molecular drugs and large proteins.

An initiating system with high efficiency for PEGDA photopolymerization at 532 nm

Junjie Li,Yuanyuan Peng,Jhair Peña,Jinfeng Xing

Nanogels based Polyethylene glycol diacrylate (PEGDA) attracted much attention due to their promising application in drug delivery and tissue engineering. Here, PEGDA photopolymerization at 532 nm with different initiating systems was investigated. Nanogels can be obtained using single 2′,4′,5′,7′-tetrabromofluorescein disodium salt (EY) as the photoinitiator. To increase the polymerization rate, EY, triethanolamine (TEOA), and diphenyl iodonium hexafluorophosphate (DPI) were investigated as a three-component initiating system. With that initiating system, the conversion of PEGDA double bond was higher than 90 % in 1 min at extremely low irradiation energy density (P= 0.95 mW/cm 2 ). However, the conversion of PEGDA double bond was only 51.5 % in 1 min at the same irradiation energy density when using EY and TEOA as two-component initiating system. Subsequently, the corresponding mechanism of the three-component (EY-TEOA-DPI) initiating system was illustrated. DPI might participate in two ways of photooxidation and reduction, which dramatically improved the polymerization rate. The EY-TEOA-DPI initiating system is promising to be applied in the preparation of multifunctional hydrogels.

Free-standing interpenetrating PVA-PEGDA films with excellent antifogging performance and mechanical robustness

Xiuxian Gu,Junhui He,Xiaochun Hu

Robust antifogging films have significant potential for a wide range of applications on the surfaces of transparent materials. Nevertheless, currently either their preparation methods are complex or their mechanical properties such as hardness, wear, scrub and water resistance are yet to be improved before they can be applied in practical applications. In this work, we have developed a facile room temperature approach to fabricating free-standing films of excellent antifogging properties and mechanical robustness. Glutaraldehyde (GA) was used to cross-link polyvinyl alcohol (PVA) and ultraviolet (UV) light was applied to initiate polymerization of poly (ethylene glycol) diacrylate (PEGDA), leading to robust interpenetrating networks (IPN). The films also had excellent mechanical properties with pencil hardness up to 8H, Taber wear-resistance up to 10,000 cycles, and sponge scrub resistance up to 300 cycles. The tensile strength and tearing strength of films reached as high as 100 MPa and 700 kN/m, respectively, much higher than those of free-standing films reported so far. This approach may hold great potential for broad application on various substrates, particularly those with low glass transition temperatures (e.g., transparent plastic films). It may expand the possibilities for future applications of free-standing antifogging films for screens, optical lenses and so on.

MXene-Enhanced PEGDA Crosslinked Quasi-Solid Electrolytes: A Flame-Retardant 3D Network for High-Performance Sodium-Ion Batteries

Lin Chen,Yirou Du,Yuhui Xie,Guowei Jia,Yuanzhi Zhu,Dong Feng,Yang Meng,Yi Mei,Delong Xie

Semi-solid polymer electrolytes (SPEs) play a pivotal role in the development of high-performance sodium-ion batteries (SIBs) by providing benefits such as enhanced safety, flexibility, and high ionic conductivity. However, traditional SPEs face challenges including poor mechanical strength, limited ionic conductivity, and insufficient electrochemical stability. Furthermore, safety concerns arising from flammability and dendrite growth significantly limit their practical applications. In this study, a novel quasi-solid electrolyte (QSE) is developed by incorporating MXene nanosheets into a PVDF-HFP matrix through UV-initiated in-situ polymerization of PEGDA monomers, forming a 3D interconnected MXene-P(PEGDA)-PVDF-HFP composite structure. The integration of MXene enhances both ionic conductivity and Na+ mobility, while simultaneously improving the electrolyte’s flame retardancy and thermal stability. The QSE-1.5M (with 1.5 wt% MXene) demonstrates an ionic conductivity of 1.01 × 10-3 S cm-1 at 30 °C, and the NVP||QSE-1.5M||Na half-cell exhibits a high initial discharge capacity of 105.5 mAh g⁻1 at 2C, with 90.1% capacity retention after 2500 cycles and an average Coulombic efficiency exceeding 99%. The full cell configuration, NVP||QSE-1.5M||HC, delivers stable cycling performance for over 3000 cycles at 3C, maintaining a capacity retention of 84.5%, with a reversible capacity of approximately 100 mAh g⁻1 at 10C. Moreover, QSE-1.5M exhibits superior thermal stability with a high decomposition temperature and a reduced heat release rate. The combustion test further highlights its self-extinguishing capability within 0.01 s, confirming excellent flame retardancy. These improvements, coupled with effective dendrite suppression, position QSE-1.5M as a promising candidate for high-safety, long-cycle-life SIBs. This work demonstrates the potential of MXene-enhanced PEGDA-based QSEs in advancing both the performance and safety of SIBs, paving the way for more efficient and secure energy storage systems.

Bioinspired Design of PVDF/SiO2-g-PEGDA Membranes Via Vapor-Induced Phase Separation for Oil/Water Separation

Ran Yu,Liying Pu,Xiaofang Wang,Peng Liu,Xiang Shen

The eco-friendly nature and energy efficiency of polymeric membranes have established them as pivotal materials for oil-in-water emulsion separation. However, persistent membrane fouling caused by oil deposition remains a major challenge, significantly hindering their practical implementation in industrial settings. In this work, thiol–ene photochemistry was employed to synthesize PEGylated silica nanoparticles (SiO2-g-PEGDA) using poly(ethylene glycol) diacrylate (PEGDA) and hexanedithiol (HD) as monomers. Biomimetic PVDF membranes were engineered via vapor-induced phase separation (VIPS) by integrating SiO2-g-PEGDA nanoparticles, followed by secondary coagulation in a dopamine (DA) solution. The experimental results demonstrate that the biomimetic membranes exhibit a characteristic sponge-like cross-sectional morphology and well-defined rough surface microstructures, which are attributed to the synergistic roles of SiO2-g-PEGDA as nanofillers and DA as interfacial cross-linker during secondary phase separation. Energy-dispersive X-ray spectroscopy (EDS) and X-ray photoelectron spectroscopy (XPS) analyses confirm that the combined action of nanoparticles and PDA significantly increases the density of hydrophilic functional groups on membrane surfaces and within pore channels. The membranes exhibit excellent hydrophilicity and underwater superoleophobicity, achieving an outstanding emulsion flux of 3537 Lm–2 h–1 bar–1 and an oil removal efficiency of 98.5%. After 15 cycles of oil–water separation, the membranes maintain a high flux recovery rate of 85.6%. Moreover, the biomimetic membranes possess exceptional operational stability, as evidenced by consistent surface wettability under organic solvents, strong acid and high-salinity conditions.

Layered GelMA/PEGDA Hydrogel Microneedle Patch as an Intradermal Delivery System for Hypertrophic Scar Treatment

Zhijing Chen,Xiaole Hu,Zhengjie Lin,Haoran Mao,Zhen Qiu,Kerong Xiang,Tiancheng Ke,Lihua Li,Lu Lu,Liling Xiao

Hypertrophic scar (HS) is an unfavorable skin disorder that typically develops after trauma, burn injury, or surgical procedures and causes numerous physical and psychological issues in patients. Currently, intralesional multi-injection of corticosteroid, particularly compound betamethasone (CB), is one of the most prevalent treatments for HS. However, injection administration could result in severe pain and dose-related side effects. Additionally, the vacuum therapeutic efficacy of this treatment relies on the level of expertise of the healthcare professional. To overcome the limitations of conventional injections, a new method that is convenient, painless, and self-administrable is urgently required. In this study, we developed a methacrylate gelatin (GelMA)/polyethylene glycol diacrylate (PEGDA) double-network hydrogel microneedle patch loaded with CB (CB-HMNP) as an intradermal delivery system for HS treatment. The double-network structure conferred the CB-HMNP with sufficient mechanical properties to successfully penetrate scar tissue while also helping to regulate the drug’s sustained release rate. Subsequently, we confirmed that the CB-HMNP had a pronounced inhibitory effect on human HS fibroblasts (hHSFs), whereas drug-free HMNPs had no effect on hHSFs, indicating its high biocompatibility. In order to assess the therapeutic efficacy of CB-HMNPs, HS models of New Zealand rabbit ears were developed. The administration of CB-HMNP three times significantly decreased the scar elevation index (SEI), collagen I/III, and transforming growth factor-β1 (TGF-β1) protein. Therefore, the CB-HMNP may offer an administration pathway for the treatment of HS that is less painful, more convenient, less invasive, and sustain-released.
