Pen-2 Negatively Regulates the Differentiation of Oligodendrocyte Precursor Cells into Astrocytes in the Central Nervous System

Jinxing Hou,Huiru Bi,Zhuoyang Ye,Wenhui Huang,Gang Zou,Xiaochuan Zou,Yun Stone Shi,Ying Shen,Quanhong Ma,Frank Kirchhoff,Yimin Hu,Guiquan Chen

Mutations on γ-secretase subunits are associated with neurologic diseases. Whereas the role of γ-secretase in neurogenesis has been intensively studied, little is known about its role in astrogliogenesis. Recent evidence has demonstrated that astrocytes can be generated from oligodendrocyte precursor cells (OPCs). However, it is not well understood what mechanism may control OPCs to differentiate into astrocytes. To address the above questions, we generated two independent lines of oligodendrocyte lineage-specific presenilin enhancer 2 (Pen-2) conditional KO mice. Both male and female mice were used. Here we demonstrate that conditional inactivation of Pen-2 mediated by Olig1-Cre or NG2-CreERT2 causes enhanced generation of astrocytes. Lineage-tracing experiments indicate that abnormally generated astrocytes are derived from Cre-expressing OPCs in the CNS in Pen-2 conditional KO mice. Mechanistic analysis reveals that deletion of Pen-2 inhibits the Notch signaling to upregulate signal transducer and activator of transcription 3, which triggers activation of GFAP to promote astrocyte differentiation. Together, these novel findings indicate that Pen-2 regulates the specification of astrocytes from OPCs through the signal transducer and activator of transcription 3 signaling.SIGNIFICANCE STATEMENT Astrocytes and oligodendrocyte (OLs) play critical roles in the brain. Recent evidence has demonstrated that astrocytes can be generated from OL precursor cells (OPCs). However, it remains poorly understood what mechanism governs the differentiation of OPCs into astrocytes. In this study, we took advantage of OL lineage cells specific presenilin enhancer 2 (Pen-2) conditional KO mice. We show that deletion of Pen-2 leads to dramatically enhanced astrocyte differentiation from OPCs in the CNS. Mechanistic analysis reveals that deletion of Pen-2 inhibits Hes1 and activates signal transducer and activator of transcription 3 to trigger GFAP activation which promotes astrocyte differentiation. Overall, this study identifies a novel function of Pen-2 in astrogliogenesis from OPCs.

Effect of oligonucleotide MT01 delivered by N-isopropylacrylamide modified polyethyleneimine for bone regeneration

Qian Zhang,Xingyuan Qu,Chen Liang,Hongyan Li,Siyu Du,Chang Wang,Yuandong Xie,Yi Zheng,Lei Wang

Objective: This study aimed to investigate the regulatory effect of N-isopropylacrylamide-modified polyethyleneimine (PEN)-delivered oligodeoxynucleotide (ODN) MT01 on bone regeneration in vitro and in vivo . Methods: A polyethylenimine (PEI) derivative, PEN, was constructed through Michael addition and employed as a carrier for ODN MT01 transfection. PEN/MT01 nanocomposites were characterized using agarose gel retardation assay, size distribution, zeta potential and transmission electron microscopy. The Cell Counting Kit-8 (CCK-8) assay was used to detect the effect of PEN on cell viability. Alkaline phosphatase (ALP) staining was used to detect the osteogenic differentiation ability of PEN/MT01 nanocomposite. Real-time quantitative PCR (q RT-PCR) and enzyme-linked immunosorbent assay (ELISA) were used to detect the regulatory effects of PEN/MT01 nanocomposite on osteogenic differentiation gene expression. Rat model was observed using the skull defect method and verified using micro-computed tomography (CT), serum biochemical indices, hematoxylin and eosin (H&E) staining and Immunohistochemistry (IHC). Results: PEN had good biological properties and could deliver MT01 well to achieve efficient transmission of MT01. PEN/MT01 nanocomposites were effectively transfected into MC3T3-E1 cells at a ratio of 6.0. CCK-8 assay displayed that PEN had no cytotoxicity to MC3T3-E1 cells. Additionally, PEN/MT01 nanocomposites could promote the expression of osteogenic genes. In vivo results revealed that PEN/MT01 nanocomposites could promote bone regeneration more effectively than the other groups. Conclusion: PEN has good biocompatibility and low toxicity, which is a good carrier for ODN MT01. PEN-delivered MT01 can be potentially employed as a useful approach to achieving bone regeneration.

Finger temperature-driven thermogalvainc gel-based smart pen: Utilized for identity recognition, stroke analysis, and grip posture assessment

Shengbo Sang,Chenhui Bai,Wenxu Wang,Saeed Ahmed Khan,Zhaosu Wang,Xinru Yang,Zhiyi Zhang,Hulin Zhang

Inappropriate pen-holding postures can result in increased muscle strain and reduced writing speed, and may even contribute to the onset of conditions such as myopia, arthritis, and cervical spondylosis. However, challenges persist in current one-size-fits-all pens toward actively monitoring handwriting habits due to structural complexity and indispensability of power supply. In this study, a finger temperature-driven thermoelectric gel-based smart pen has been invented for identity recognition, stroke analysis, and grip posture assessment by coupling thermogalvanic and piezoresistive effects derived from finger temperature and pressure on a PVA/gelatin gel. This dual-network gel exhibits not only a high thermopower of − 2.05 mV K −1 and a rapid response of 130 ms, but also full flexibility, significant resistance to desiccation, and a high tensile capacity of up to 300%. By integrating thermogalvanic gel units within a pen shaft, our self-powered smart pen achieved a recognition accuracy of 98.1% for eight basic Chinese character strokes with the aid of deep learning. Furthermore, with a four-channel expansion, the recognition accuracy reaches 98.8% for eight distinct identities and 99.1% for nine different pen-holding postures, respectively, while offering real-time visualization of monitoring results on a user-friendly human-machine interface. This machine learning-assisted self-powered smart gel pen presents a new viable solution for wearable bioelectronics toward future intelligent era.

Using a Flexible Fountain Pen to Directly Write Organic Semiconductor Patterns with Crystallization Regulated by the Precursor Film

Bingyang Liu,Jialin Wang,Guoxin Zhang,Gengxin Du,Huihui Xia,Weiwei Deng,Xinyan Zhao

Organic semiconductor (OSC) films fabricated by meniscus-guided coating (MGC) methods are suitable for cost-effective and flexible electronics. However, achieving crystalline thin films by MGC methods is still challenging because the nucleation and crystal growth processes are influenced by the intertwined interactions among solvent evaporation, stochastic nucleation, and the fluid flow instabilities. Herein, a novel flexible fountain pen with active ink supply is designed and used to print OSCs. This direct-write method allows the flexible pen tip to contact the substrate, maintaining a robust meniscus by eliminating the gap found in conventional MGCs. An in situ optical microscopy observation system shows that the precursor film plays a critical role on the crystallization and the formation of coffee rings and dendrites. The computational fluid dynamics simulations demonstrate that the microstructure of the pen promotes extensional flows, facilitating mass transport and crystal alignment. Highly-aligned ribbon-shaped crystals of a small organic molecule (TIPS-pentacene), as well as a semiconducting polymer (N2200) with highly-ordered orientations, have been successfully printed by the flexible fountain pen. Organic field-effect transistors based on the flexible pen printed OSCs exhibit high performances and strong anisotropic mobility. In addition, the flexible fountain pen is expandable for printing multiple lines or large-area films.

A Highly Stretchable, Ultrastable, and Antifatigue Ionic Organogel as an Environment-Tolerant Flexible Sensor and Sensitive Capacitive Pen

Zhengxu Jin,Hongyan Liu,Debin Wang,Mengju Yuan,Huijuan Zhang

High-performance flexible sensors require excellent temperature and fatigue resistance without a significant sacrifice of high sensitivity. In this study, an environmentally resistant ionic organogel was fabricated via a two-step approach. An ultrastable binary solvent system was first synthesized using ethylene glycol and halogenated metal ionic liquid. Then, a loosely chemically cross-linked poly(acrylic acid) was incorporated as a backbone in the binary solvent, in which graphene oxide and halogenated metal ionic liquids synergistically formed an efficient conductive network. The distinct binary solvent and ample interactions endowed the gel with remarkable stretchability (elongation at break of 1105%), superior fatigue resistance (mechanical loss of only 4.05% after 500 compression cycles), and stable mechanical properties over a wide temperature range (from −80 to 150 °C). Additionally, its mechanical properties could be preserved to a great extent after 15 days of exposure to an open environment. Due to its outstanding conductivity and sensing capabilities, the gel is also suitable for transmitting Morse code signals or being integrated into capacitive pens for seamless digital screen interaction. This multifunctional ionic organogel holds great promise for applications in electronic skin, motion monitoring, and human machine interaction (HMI) in extreme environments.

Utilization of waste polyethylene terephthalate (PET) depolymerization products as binders for ballpoint pen ink: preparation, optimization, and performance evaluation

Kim Ri Myong,Ryang Il Song,Oh Jun Hyok,Kim Jong Nam,Kim Yong Il,Kim Guk Chan,Ho Kuang Min,Hong Son Il,Son Ryong Chol

This study presents a sustainable approach for upcycling waste polyethylene terephthalate (PET) into a functional binder for ballpoint pen ink. PET was depolymerized via glycerolysis under systematically optimized conditions: PET-to-glycerol mass ratio of 1:1.1, catalyst loading of 0.6 wt%, temperature of 230 °C, and reaction time of 4 h. Comprehensive characterization by FT-IR, HPLC, and GPC elucidated the structural features and molecular composition of the depolymerization products. FT-IR confirmed the presence of hydroxyl end groups and preservation of ester linkages, providing evidence for a transesterification-based depolymerization mechanism. GPC analysis revealed a number-average molecular weight (M n ) of approximately 1,800 g/mol, confirming successful cleavage of PET chains into oligomeric fragments suitable for binder applications. HPLC analysis of the reaction effluent further corroborated the proposed reaction pathway, offering insights into the by-products generated during depolymerization. The formulated ink exhibited excellent performance characteristics: appropriate viscosity, rapid drying (≤ 19 s), thermal stability across − 20 to 40 °C, UV resistance exceeding 72 h, and storage stability over 340 days. These properties meet critical industry standards, demonstrating the practical viability of the upcycled product. This study successfully establishes a practical route to converting PET waste into commercially valuable materials, providing a cost-effective and sustainable alternative to conventional petrochemical binders in the stationery industry while contributing to plastic pollution reduction. Double-layered hydrogels based on phase change material and pen ink for continuous and efficient solar-driven seawater desalination

Zhiheng Zheng,Huan Liu,Xiaodong Wang

A novel double-layered hydrogel based on the polyacrylamide matrix, pen ink, and phase-change microcapsules was developed to construct a hydrogel-based evaporator for efficient and continuous seawater desalination . The n -eicosane (core)@crystalline TiO 2 (shell) phase-change microcapsules were fabricated through interfacial polycondensation , and the resulting phase-change microcapsules obtained a considerably high latent-heat capacity of 187 J·g −1 . The upper layer of the hydrogel exhibits a high solar absorptivity of over 90 % due to the introduction of pen ink as a solar absorber, which helps the hydrogel-based evaporator to gain an excellent photothermal energy conversion capability. A high evaporation efficiency of 93.89 % was obtained for the developed hydrogel-based evaporator along with a high evaporation rate of 1.85 kg·m −2 ·h −1 under one-sun illumination. The phase-change microcapsules dispersed in the lower layer of the hydrogel can act as a latent heat-storage unit to store a great quantity of photothermal energy through melting phase change by the phase change material core. This imparts a consecutive evaporation capability to the developed hydrogel-based evaporator. As a result, its water production was increased by 11 % under 50-min one-sun illumination and 50-min non-illumination.

A fluorescent detection pen for sensitive, specific, and real-time detection of phosgene based on a novel rhodamine probe

Yingying Li,Jie Zhang,Zengqiang Liang,Ran Yang,Lingbo Qu,Zhaohui Li,Yuanqiang Sun

It is of great significance for real-time monitoring of phosgene to protect public safety. In this paper, a new rhodamine derivate with highly rapid, special and sensitive response towards phosgene was synthesized and used as a phosgene probe based on its spirolactam ring-opening property and nucleophilic reaction with phosgene. To realize in situ visualization of phosgene, a fluorescent detection pen with gas-liquid reaction model was proposed for the first time using the mixture of the probe and polystyrene as ink. The detection film could be formed in situ by simply writing on desired surface and respond toward phosgene gas within 30 s with an ultra-low detection limit (2 ppm) with the naked eye. Such a fluorescence pen detection mode supplied a new strategy for the fabrication of an excellent platform for real-time detection of phosgene and other gas analytes.

PEN-coated superparamagnetic iron-mediated delivery of siSnail2 to inhibit metastasis and promote ferroptosis in the treatment of cancer

Yue Hu,Qing Nie,Xianling Cong,Wen Wu,Qiong Wu,Qihui Liu,Yuanyuan Li,Haiyan Liu,Jingyan Ge,Fangfang Chen

Cancer represents a significant global public health challenge, and conventional cancer therapies such as surgery and chemoradiotherapy are not enough due to the increased complexity of cancer. Nanotechnology has the potential to revolutionize tumor treatments by integrating gene therapy, tumor targeting, and drug delivery. In this study, we demonstrated that Snail2 plays a crucial role in the migration and invasion of lung and liver carcinoma. We proposed a novel approach to synergize the aminated crosslinking dextran coat of superparamagnetic iron oxide nano worms (CLIO-NH 2 , CN) with small interfering Snail2 RNA (siSnail2). The efficiency of siSnail2 delivery was significantly improved by coating CN with N-Isopropylacrylamide-modified polyethylenimine (CNP). In vitro, experiments revealed that CNP@siSnail2 effectively inhibited cancer cell EMT, migration, and invasion. Moreover, CNP@ siSnail2 promoted cancer cell death through various mechanisms, including apoptosis and ferroptosis . The combination of CNP@ siSnail2 and cisplatin significantly improved the anti-tumor effect of the treatment. Animal models demonstrated that the combined treatment of CNP@ siSnail2 and cisplatin resulted in excellent tumor inhibition effects. Our findings provide a potential combined treatment strategy for cancer therapy.

On-demand fabrication of surface-enhanced Raman scattering arrays by pen writing, and their application to the determination of melamine in milk

Han Caiqin,Li Yuqi,Jia Qin,Bradley Layne Hyer,Gan Yun,Yao Yue,Qu Lulu,Li Haitao,Zhao Yiping

An on-demand, disposable and portable surface-enhanced Raman scattering (SERS) array has been prepared by modifying the surface of cellulose paper and patterning it with silver nanoparticles (AgNPs) via pen-writing technique. The paper is initially modified with hexadecenyl succinic anhydride to generate a hydrophobic top layer that is capable of concentrating melamine (and other hydrophobic analytes) by preventing the mainly aqueous sample from spreading on the paper. The AgNPs are subsequently written onto the hydrophobic paper with a high degree of control to form large-scale SERS sensing arrays. The resulting arrays demonstrate outstanding Raman scattering enhancement as demonstrated for the fluorescent dye Rhodamine 6G which can be detected by SERS with an 80 pM limit of detection (LOD) without interference by fluorescence. The reproducibility in the preparation of spots has a relative standard deviation of <15% (for n  = 128 spectra). The arrays were applied to the determination of the illicit milk additive melamine. The plot of SERS intensity at Δν = 681 cm −1 versus the logarithm of melamine concentration is linear in the concentration range from 0.3 to 20 mg·L −1 , and the LOD is 0.27 mg·L −1 . In our perception, the disposable arrays presented here integrate the advantages of easy production and reliable reproducibility, and therefore are an ideal choice for use in SERS detection. An on-demand, disposable and portable surface-enhanced Raman scattering (SERS) array has been developed by pen writing. Such a SERS array can be applied for the highly sensitive determination of the illicit milk additive melamine, which holds great promise for field-deployable applications.

Highly Efficient Photothermal Conversion of Ti3C2Tx/Ionic Liquid Gel Pen Ink for Smoothly Writing Ultrasensitive, Wide-Range Detecting, and Flexible Thermal Sensors

Haoge Cheng,Qiongxia Liu,Shengpeng Han,Shuai Zhang,Xiao Ouyang,Xun Wang,Zhilong Duan,Hao Wei,Xinyue Zhang,Ning Ma,Mianqi Xue

Photothermal conversion behavior has a vital application to disease therapy, water purification, or uncontacted heaters. The fabrication of high-performance photothermal conversion materials especially for near-infrared (NIR) light and microstructures has attracted a great deal of attention. Among numerous substances, MXene as a new type of 2D material with semi-metallic and unique electromagnetic properties presents a broader absorption of light and even a typical plasmonic absorption near the NIR-I area (808 nm), which has made it suitable for photothermal conversion. Here, we propose a facile approach for preparing a Ti3C2Tx/ionic liquid ink with a high photothermal conversion efficiency. The as-prepared ink has showed good wettability of various substrates as well as the high sensitivity of 808 nm NIR light irradiation and a wide range of thermal variation. After packing the ink into a gel pen refill, the flexible thermal chips could be easily obtained just by pen writing on the soft surface with the designed size, which also have become an optimal candidate for the thermal alarm system.

Temporal evolution stages classification and aging time prediction of gel-pen ink using GC-IMS combined with machine learning for forensic science applications

Wenhui Lu,Jing Chen,Liangliang Zhang,Zixuan Nie

Determining the temporal evolution of inks remains a critical challenge in forensic document analysis. The temporal evolution stages classification and aging time prediction of gel-pen ink were investigated by integrating gas chromatography-ion mobility spectrometry (GC-IMS) with machine learning algorithms. Ink-specific volatile markers were correlated with aging mechanisms through kinetic modeling and heatmap analysis. Three distinct temporal evolution stages were categorized into the rapid evaporation stage, slow-release stage, and chemical stabilization stage through multivariate analysis of volatiles. Furthermore, six tree-based machine learning algorithms were systematically evaluated, with the Categorical Boosting (CatBoost) model achieving superior performance (accuracy=100 %) in classifying five detailed aging stages of gel-pen ink. The decision tree regression model demonstrated high temporal prediction accuracy (test R²=0.954) through interpretable feature engineering. A stepwise strategy combining classification and regression models was proposed, enabling simultaneous ink characterization and age estimation. This methodology is expected to provide a validated approach for classifying temporal evolution stages and predicting aging time, significantly improving the efficiency of forensic analysis in judicial investigations.
