Nanoscale exopolymer reassembly-trap mechanism determines contrasting PFOS exposure patterns in aquatic animals with different feeding habitats: A nano-visualization study

Shuyan Xu,Pengfeng Zhu,Caiqin Wang,Daoyong Zhang,Ming Zhang,Xiangliang Pan

The behavior and fate of PFOS (perfluorooctanesulfonate) in the aquatic environment have received great attention due to its high toxicity and persistence. The nanoscale supramolecular mechanisms of interaction between PFOS and ubiquitous EPS (exopolymers) remain unclear though EPS have been widely-known to influence the bioavailability of PFOS. Typically, the exposure patterns of PFOS in aquatic animals changed with the EPS-PFOS interaction are not fully understood. This study hypothesized that PFOS exposure and accumulation pathways depended on the PFOS-EPS interactive assembly behavior and animal species. Two model animals, zebrafish and chironomid larvae, with different feeding habitats were chosen for the exposure and accumulation tests at the environmental concentrations of PFOS in the absence and presence of EPS. It was found that PFOS triggered the self-assembly of EPS to form large aggregates which significantly trapped PFOS. PFOS accumulation was significantly promoted in zebrafish but drastically reduced in chironomid larvae because of the nanoscale interactive assembly between EPS and PFOS. The decreased dermal uptake but increased oral uptake of PFOS by zebrafish with large mouthpart size could be ascribed to the increased ingestion of PFOS-enriched EPS aggregates as food. For the chironomid larvae with small mouthpart size, the PFOS-EPS assemblies reduced the dermal, oral and intestinal uptake of PFOS. The nano-visualization evidences confirmed that the PFOS-enriched EPS-PFOS assemblies blocked PFOS penetration through skin of both animals. These findings provide novel knowledge about the ecological risk of PFOS in aquatic environments.

Fabrication of tailored imprinted layer and cladded layer on magnetic nanomaterials with enhanced specificity for recognition of PFOS

Yi Hao,Xiaoyi Chen,Kaili Wang,Xingyan Wang,Long Zhang,Haiyan Jing,Ruixia Gao,Sicen Wang

Background Perfluorooctane sulfonate (PFOS) is a persistent organic pollutant with significant risks to ecosystems and human health. Magnetic molecularly imprinted polymers (MIPs) provide a promising solution for selectively extracting PFOS from contaminated water. However, while bifunctional monomer imprinting improves the imprinting effect by introducing diverse functional groups, it can also increase non-specific adsorption. To address this problem, a surface cladding strategy was applied after polymerization, forming an inert cladding layer to reduce non-specific binding and improve specificity. Results The magnetic MIPs with a cladding layer (Fe 3 O 4 @PFOS-cMIPs) exhibited enhanced adsorption performance, achieving a high adsorption capacity of 135.1 mg g -1 , an imprinting factor of 3.19, and a selectivity factor greater than 1.3, surpassing most reported PFOS-MIPs. The interactions between Fe 3 O 4 @PFOS-cMIPs and PFOS were confirmed to be a synergistic combination of electrostatic and hydrophobic interactions, as evidenced by FTIR analysis, zeta potential measurements, and pH studies. Additionally, Fe 3 O 4 @PFOS-cMIPs demonstrated excellent reusability, with stable performance across six adsorption-desorption cycles using a regeneration solution of acetone and NaCl. Furthermore, when coupled with LC-MS, Fe 3 O 4 @PFOS-cMIPs successfully detected trace levels of PFOS in complex environmental water samples. The method demonstrated high precision (RSD, 6.0%), excellent recoveries (90.9% - 103.3%), a low limit of detection (LOD, 0.06 ng L -1 ), and an enrichment factor of 299. Significance This study presents a practical and efficient strategy for developing magnetic MIPs with enhanced molecular recognition. The enhanced specificity and adsorption capacity highlight the strong potential of these MIPs for the targeted extraction of PFOS from various contaminated water sources, offering significant contributions to environmental analysis and remediation.

Fluorinated LDHs for selective sorption of PFOS: Unveiling the roles of increased hydrophobicity and F-F interactions

Ao Xian,Changlong Wei,Zhiwen Tang,Yao Zhang,Qing Wang,Zhiyong Han,Xin Song

Layered double hydroxides have been proved to be effective adsorbents for the removal of per- and polyfluoroalkyl substances (PFASs) from aqueous solutions. In this study, a novel layered double hydroxides modified by perfluorooctyltriethoxysilane (LDHs-PFOTES) was synthesized at room temperature, demonstrating rapid and selective sorption of perfluorooctane sulfonic acid (PFOS). The covalent grafting of fluorosilane groups onto the LDHs surface enhanced hydrophobicity and introduced fluorine functional groups, while preserving the positive charge essential for electrostatic interactions. LDHs-PFOTES removed 97.21 % of PFOS from water within 5 min, with a sorption capacity of 1530.33 mg/g, nearly doubling the capacity of unmodified LDHs (831.79 mg/g). The thermodynamic analysis of PFOS sorption on LDHs-PFOTES revealed that the process is spontaneous and endothermic. LDHs-PFOTES maintained over 90 % PFOS removal efficiency under varying pH levels, ionic strengths, and the presence of competing compounds. Furthermore, the C-F structure on the LDHs-PFOTES enabled fluorine-fluorine (F-F) interactions with PFOS, further improving selective sorption. It was concluded that the combined effects of electrostatic interactions, hydrophobicity, F-F interactions, and ion exchange resulted in the efficient and selective removal of PFOS from water. LDHs-PFOTES demonstrated 95.37 % sorptive efficiency in removing PFOS when treating a mixture of 11 PFAS compounds. Additionally, LDHs-PFOTES effectively desorbed PFOS using a mixed solution of NaCl and methanol, maintaining approximately 95.08 % removal efficiency over three regeneration cycles. This fluorination strategy to modify LDHs offers a novel approach for selectively and efficiently removing PFASs from water.

Comparative Cytotoxicity and Toxicological Mechanisms of 6:2 Cl-PFAES and PFOS on Pancreatic β Cells: Implications for Glucose Metabolism Disruption

Xiaomin Ren,Jianying Wang,Fenqing Zhao,Pingping Zhang,Huan He,Zhixiang Xu,Bin Huang,Xuejun Pan

Previous studies have indicated that 6:2 chlorinated polyfluoroalkyl ether sulfonate (6:2 Cl-PFAES), a substitute for perfluorooctane sulfonate (PFOS), caused disruptions in glucose metabolism, but its toxicological mechanisms remain unclear. Pancreatic β cells are responsible for regulating glucose metabolism. This study used mouse insulinoma β cells (β-TC-6) to study the toxic effects of 6:2 Cl-PFAES and elucidate its potential mechanisms. Through cell counting kit-8, trypan blue staining, and apoptosis assays, we found that 6:2 Cl-PFAES showed toxicity to β-TC-6 cells, even surpassing that of PFOS. Mechanistic studies demonstrated that 6:2 Cl-PFAES induced oxidative stress by ascendant levels of reactive oxygen species (ROS) and malondialdehyde (MDA), alongside increased superoxide dismutase (SOD) activity. The oxidative stress-inducing capability of 6:2 Cl-PFAES was stronger than PFOS. Co-exposure with N-acetyl-L-cysteine (NAC), as the ROS scavenger, which obviously reduced the degree of cell apoptosis and the production of ROS, highlighting oxidative stress as a crucial mechanism of 6:2 Cl-PFAES-induced cell death. Transcriptomic analysis further revealed that 6:2 Cl-PFAES affected genes association with the oxidative phosphorylation pathway, confirming the oxidative stress effect, and altered diabetes-related genes expression, suggesting a heightened risk of diabetes. This study provides insights into the toxicity of 6:2 Cl-PFAES on β cells and its potential risks on glucose metabolism disruption.

Lysosomal iron accumulation and subsequent lysosomes-mitochondria iron transmission mediate PFOS-induced hepatocyte ferroptosis

Jixun Li,Ruzhen Feng,Wei Yang,Peiyao Liang,Tianming Qiu,Jingyuan Zhang,Xiance Sun,Qiujuan Li,Guang Yang,Xiaofeng Yao

Perfluorooctane sulfonate (PFOS) is known as a persistent organic pollutant. A significant correlation between PFOS and liver ferroptosis has been unveiled, but the precise mechanism needs to be elucidated. In prior research, we found that PFOS treatment provoked mitochondrial iron overload. In this study, we observed a gradual increase in lysosomal iron in L-O2 cells after exposure to PFOS for 0.5–24 h. In PFOS-exposed L-O2 cells, suppressing autophagy relieved the lysosomal iron overload. Inhibiting transient receptor potential mucolipin 1 (TRPML1), a calcium efflux channel on the lysosomal membrane, led to a further rise in lysosomal iron levels and decreased mitochondrial iron overload during PFOS treatment. Suppressing VDAC1, a subtype of voltage-dependent anion-selective channels (VDACs) on the outer mitochondrial membrane, had no impact on PFOS-triggered mitochondrial iron overload, whereas restraining VDAC2/3 relieved this condition. Although silencing VDAC2 relieved PFOS-induced mitochondrial iron overload, it had no effect on PFOS-triggered lysosomal iron overload. Silencing VDAC3 alleviated PFOS-mediated mitochondrial iron overload and led to an additional increase in lysosomal iron. Therefore, we regarded VDAC3 as the specific VDACs subtype that mediated the lysosomes-mitochondria iron transfer. Additionally, in the presence of PFOS, an enhanced association between TRPML1 and VDAC3 was found in mice liver tissue and L-O2 cells. Our research unveils a novel regulatory mechanism of autophagy on the iron homeostasis and the effect of TRPML1-VDAC3 interaction on lysosomes-mitochondria iron transfer, giving an explanation of PFOS-induced ferroptosis and shedding some light on the role of classic calcium channels in iron transmission.

Recovery of perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA) from water using foam fractionation with whey soy protein

Nan Hu,Xiaodan Sun,Nan Yao,Mei Yang,Yaoxi Chen,Zhijun Zhang

Perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA) are persistent anthropogenic chemicals that are widely distributed in the environment and pose significant risks to human health. Foam fractionation has emerged as a promising method to recover PFOS/PFOA from water. However, PFOS/PFOA concentrations in wastewater are often inadequate to generate stable foams due to their high critical micelle concentrations and the addition of a cosurfactant is necessary. In this study, we developed whey soy protein (WSP) as a green frother and collector derived from soybean meal (SBM), which is an abundant and cost-effective agro-industrial residue. WSP exhibited excellent foaming properties across a wide pH range and demonstrated strong collection capabilities that enhanced the recovery of PFOS/PFOA. The mechanism underlying this collection ability was elucidated through various methods, revealing the involvement of electrostatic attraction, hydrophobic interaction, and hydrogen bonding. Furthermore, we designed a double plate internal to improve the enrichment of PFOS/PFOA by approximately 2.3 times while reducing water recovery. Under suitable conditions (WSP concentration: 300 mg/L, pH: 6.0, air flowrate: 300 mL/min), we achieved high recovery percentages of 94–98% and enrichment ratios of 7.5–12.8 for PFOS/PFOA concentrations ranging from 5 to 20 mg/L. This foam fractionation process holds great promise for the treatment of PFOS/PFOA and other per- and polyfluoroalkyl substances.

Aberrant hepatic lipid metabolism associated with gut microbiota dysbiosis triggers hepatotoxicity of novel PFOS alternatives in adult zebrafish

Qiyu Wang,Jing Huang,Shuai Liu,Caiyun Wang,Yuanxiang Jin,Hong Lai,Wenqing Tu

Perfluorooctane sulfonate (PFOS) has been reported to induce hepatotoxicity in wildlife and humans. Novel PFOS alternatives have been widely used following restrictions on PFOS, but little is known about their potential toxicity. Here, the first comprehensive investigation on the chronic hepatotoxicity and underlying molecular mechanisms of PFOS, 6:2Cl-PFESA (F-53B), and sodium p -perfluorous nonenoxybenzene sulfonate (OBS) was carried out on adult zebrafish through a histopathological examination, biochemical measurement, and multi-omics analysis. PFOS and its alternatives caused changes in liver histopathology and liver function indices in the order of F-53B > PFOS > OBS, which was consistent with their concentration in the liver. In silico modeling and transcriptional profiles suggested that the aberrant hepatic lipid metabolism induced by F-53B and PFOS was initiated by the action on peroxisome proliferator-activated receptor γ (PPARγ), which triggered changes in downstream genes transcription and led to an imbalance between lipid synthesis and expenditure. Gut microbiome analysis provided another novel mechanistic perspective that changes in the abundance of Legionella , Ralstonia , Brevundimonas , Alphaproteobacteria , Plesiomonas , and Hyphomicrobium might link to alterations in the PPAR pathway based on their significant correlation. This study provides insight into the molecular mechanisms of hepatotoxicity induced by PFOS and its novel alternatives and highlights the need for concern about their environmental exposure risks.

Comparison of toxicokinetics and toxic effects of PFOS and its novel alternative OBS in zebrafish larvae

Yilong Zou,Yongming Wu,Qiyu Wang,Jinbao Wan,Mi Deng,Wenqing Tu

Sodium p -perfluorous nonenoxybenzene sulfonate (OBS), a novel alternative to perfluorooctanesulfonate (PFOS), is widely used in industry as a surfactant, firefighting foam and photographic material. The occurrence of OBS in the aquatic environment has been recently reported, but little information is available on its accumulation and toxic effects in aquatic organisms. In this study, zebrafish larvae (3 d post-fertilization) were subjected to OBS (10, 100 μg/L) and PFOS (10 μg/L) for a period of 48 h, followed by a 24 h of depuration period. The bioconcentration and depuration kinetics, oxidative stress and possible molecular mechanisms of OBS and PFOS were investigated in zebrafish larvae. Our results showed that the uptake and depuration of both OBS and PFOS fitted well with a first-order kinetic model. The uptake rate constant of OBS was similar to that of PFOS, but the depuration rate constant was much higher than PFOS with a half-life of 69.7–85 h for OBS and 222.2 h for PFOS. The calculated BCFs of OBS and PFOS were 238.0–242.5 and 644.2, respectively. In our acute toxicity assay, the enhanced expression of Nrf2 protein accompanied by the upregulation of CAT and SOD protein expressions indicated OBS and PFOS induced oxidative stress in zebrafish larvae, and the Nrf2-ARE signaling pathway was involved in this process. Collectively, OBS has a lower bioconcentration potential than PFOS, but its toxic effect on oxidative stress was comparable to PFOS in zebrafish larvae.

A bifunctional UIO-66-NH2@ RhB-based fluorescence and colorimetric dual-mode ratiometric sensor and hydrogel-based smart phone–assisted naked eye detection of perfluorooctanesulfonate (PFOS)

Aham Emmanuel Chigozie,Ravikumar A.,Feng Weiwei,Arunjegan A.,Hu Zhang,Zhang Zhen,Zhao Hongjun

A bifunctional fluorescent and colorimetric sensor based on UiO-66-NH₂@RhB was developed for the highly sensitive detection of perfluorooctanesulfonate (PFOS), a persistent environmental pollutant. The sensing mechanism relies on the inner filter effect (IFE), where Rhodamine B (RhB) modulates the fluorescence of UiO-66-NH₂, enabling effective PFOS recognition. In the presence of PFOS, UiO-66-NH 2 @RhB exhibits dual emissions in blue fluorescence at 448 nm and red fluorescence at 565 nm at a single excitation wavelength. Upon the addition of increasing concentrations of PFOS, the blue fluorescence intensity increased and a small change was observed in the red fluorescence intensity. Besides, UiO-66-NH 2 @RhB solutions display an excellent colorimetric response for PFOS and produce an obvious color change from red to colorless. The method demonstrated high sensitivity, achieving linear detection ranges of 1–10 nM with detection limits of 0.84 nM (fluorescence) and 0.91 nM (colorimetry). Furthermore, the dual-sensing platform successfully detected PFOS in real water samples with satisfactory outcomes. Its integration with a portable smartphone further enables intelligent, real-time, on-site monitoring and is successfully applied to monitoring PFOS in environmental samples. This study not only broadens the application of MOFs but also provides valuable insights for designing multifunctional sensors for diverse analytical applications. Graphical abstract

Distinct Effects of PFOS and OBS on Neurotoxicity via PMK-1 Mediated Pathway in Caenorhabditis elegans

Jiahong Jiang,Qi Liu,Boxiang Zhang,Lei Zhao,Dan Xu

Sodium p-perfluorous nonenoxybenzenesulfonate (OBS) has been proposed as a substitute for perfluorooctanesulfonic acid (PFOS), yet it has garnered increasing attention due to its environmental persistence and potential toxicity. Despite these concerns, the neurotoxic mechanisms of OBS remain unclear. This study investigates and compares the neurotoxic effects and mechanisms of OBS and PFOS inCaenorhabditis elegans. L4-stage worms were exposed to OBS (0.1–100 μM) or PFOS (100 μM) for 24 h. Neurobehavioral analysis showed that OBS exposure induced concentration-dependent neurobehavioral deficits, with 100 μM OBS significantly reducing pharyngeal pumping rate (29.8%), head swing frequency (23.4%), and body bending frequency (46.6%), surpassing the effects of PFOS. Both compounds decreased the fluorescence intensity of dopaminergic, glutamatergic, and γ-aminobutyric acid neurons and downregulated neurotransmitter-associated genes. They also increased ROS generation and inhibited antioxidant gene expression. Molecular docking revealed that OBS had a stronger binding affinity to p38 MAPK key protein (PMK-1) than PFOS. OBS and PFOS upregulatedpmk-1andskn-1, modulating oxidative stress and neuronal function.pmk-1mutation differentially affected OBS-induced neurobehavioral changes and gene expression alterations. Our findings indicate that OBS exhibits stronger neurotoxicity than PFOS inCaenorhabditis elegans, mediated through the PMK-1 pathway. These results highlight the need for further investigation into the safety of OBS as a PFOS alternative.

The VDAC1 oligomerization regulated by ATP5B leads to the NLRP3 inflammasome activation in the liver cells under PFOS exposure

Yu Ma,Wei Yang,Peiyao Liang,Ruzhen Feng,Tianming Qiu,Jingyuan Zhang,Xiance Sun,Qiujuan Li,Guang Yang,Xiaofeng Yao

As a persistent organic pollutant, perfluorooctane sulfonate (PFOS) has a serious detrimental impact on human health. It has been suggested that PFOS is associated with liver inflammation. However, the underlying mechanisms are still unclear. Here, PFOS was found to elevate the oligomerization tendency of voltage-dependent anion channel 1 (VDAC1) in the mice liver and human normal liver cells L-02. Inhibition of VDAC1 oligomerization alleviated PFOS-induced nucleotide-binding domain and leucine-rich repeat protein-3 (NLRP3) inflammasome activation. Cytoplasmic membrane VDAC1 translocated to mitochondria was also observed in response to PFOS. Therefore, the oligomerization of VDAC1 occurred mainly in the mitochondria. VDAC1 was found to interact with the ATP synthase beta subunit (ATP5B) under PFOS treatment. Knockdown of ATP5B or immobilization of ATP5B to the cytoplasmic membrane alleviated the increased VDAC1 oligomerization and NLRP3 inflammasome activation. Therefore, our results suggested that PFOS induced NLRP3 inflammasome activation through VDAC1 oligomerization, a process dependent on ATP5B to transfer VDAC1 from the plasma membrane to the mitochondria. The findings offer novel perspectives on the activation of the NLRP3 inflammasome, the regulatory mode on VDAC1 oligomerization, and the mechanism of PFOS toxicity.

Effect Mechanism of Low-level PFOS Isomers to A. thaliana Leaves Using High-throughput Untargeted Metabolomics

Wang Hao,Zhang Yue,Zhao Liuqing,He Zeying,Zhang Jingran,Liu Bingjie,Liu Xiaowei,Zhang Yanwei

The ecotoxicity of perfluorooctane sulfonate(PFOS) to plants has been reported, but the lack of study on the effect of PFOS isomers is of concern. In this paper, the effect mechanism of Arabidopsis thaliana ( A. thaliana ) against two different concentrations of L -PFOS or perfluoro-3-methylheptanesulfonate (P3MHpS) was investigated via metabolomics. The metabolism of Arabidopsis thaliana leaves was investigated via the metabolomics strategy and statistical analysis. And 41 biomarkers for L -PFOS and 32 biomarkers for P3MHpS were found and identified. There was no significant difference of saccharides between L -PFOS and P3MHpS. More amino acids were affected by P3MHpS than by L -PFOS in high-level exposure group, while the opposite phenomenon was observed in low-level exposure group. More phytohormones were affected by L -PFOS than by P3MHpS in the high-level exposure group, however, the elevated level of indole phytohormones was significantly higher by P3MHpS than by L -PFOS. The level of up-regulation of (poly)phenols in L -PFOS group was significantly higher than that in P3MHpS group. The affected aliphatic glucosinolate metabolism was firstly observed with higher up-regulation induced by L -PFOS than by P3MHpS. New insight of phytotoxicity of PFOS isomers to plants was provided.
