MBG/ PGA-PCL composite scaffolds provide highly tunable degradation and osteogenic features

Jiangfeng Li,Chunyi Wang,Guoxing Gao,Xing Yin,Ximing Pu,Bing Shi,Yang Liu,Zhongbing Huang,Juan Wang,Jingtao Li,Guangfu Yin

It remains a challenge to achieve satisfactory balance between biodegradability and osteogenic capacity in biosynthetic bone grafts. In this study, we aimed to address this challenge by incorporating mesoporous bioactive glass (MBG) into poly(caprolactone-co-glycolide) (PGA-PCL) at gradient ratios. MBG/PGA-PCL (PGC/M) scaffolds with MBG incorporation ratio at 0, 10%, 25% and 40% (PGC/M0-40) were synthesized using a modified solvent casting-particulate leaching method, and their physiochemical and biological properties were comprehensively evaluated. PGC/M scaffolds exhibited highly perforated porous structure with a large-pore size of 300–450 μm, with ordered MBGs of around 6.0 nm mesopores size uniformly dispersed. The increase in MBG incorporation ratio significantly improved the scaffold surface hydrophilicity, apatite-formation ability and pH stability, increased the weight loss rate while insignificantly influenced the molecular chains degradation of PGA-PCL component, and facilitated the attachment, spreading, viability and proliferation of rat bone marrow stromal cells (rBMSCs) on scaffolds. Moreover, rBMSCs cultured on PGC/M10-40 scaffolds demonstrated enhanced ALP activity and osteogenesis-related gene expression in a MBG dose-dependent manner as compared with those cultured on PGC/M0 scaffolds. When implanted to the rat cranial bone defect, PGC/M25 and PGC/M40 scaffolds induced significantly better bone repair as compared to PGC/M0 and PGC/M10 scaffolds. Besides, the biodegradability of PGC/M scaffolds correlated with the MBG incorporation ratio. These data suggested this novel PGC/M scaffolds as promising bone repair biomaterial with highly tunable hydrophilicity, bioactivity, cytocompatibility, osteogenic activity as well as biodegradability.

A colorimetric glucose sensor with gradient porous polyglycolic acid (PGA) microneedle array patch

Zhang Yunfan,Xue Zifan,Qin Boyu,Wu Jiaze,Park Jongho,Xu Chensong,Shen Shengnan,Li Hui,Kim Beomjoon

Rapid glucose detection holds significance to diabetes. Here, a novel colorimetric glucose sensor for interstitial fluid (ISF) glucose detection is proposed, comprising a gradient porous polyglycolic acid (PGA) microneedle array patch (MAP) for ISF extraction and a porous 3,3’,5,5’-tetramethylbenzidine (TMB)-functionalized PGA film for glucose sensing. Instead of using pore-forming agents, porous PGA structure is fabricated by solvent volatility method, which brings the benefit of eliminating residual agents and enabling the formation of uniform, tunable micropores. PGA MAP was designed and fabricated with a gradient porosity from tip (7.2 ± 2.3%, N  = 10) to the substrate (48.5 ± 1.1%, N  = 10), providing high mechanical strength at the tip and hygroscopicity across the microneedle. With a compression failure force of 0.51 ± 0.13 N ( N  = 10), it successfully punctured artificial skin model and absorbed 1.40 ± 0.06 µL of liquid in 1 min ( N  = 10). By using the co-precipitation method, TMB was uniformly distributed inside the microstructure of porous PGA film. It effectively prevents the color errors caused by coffee ring effect. Furthermore, enzymes were well immobilized onto TMB-functionalized PGA film due to its hydrophilicity and porosity. Finally, the sensor exhibits uniform, distinct, and linear color changes in response to glucose with a detection limit of 0.13 mM in the range of 2 to 10 mM. Biodegradable core-shell electrospun nanofibers based on PLA and γ-PGA for wound healing

Yue Fang,Xianqi Zhu,Na Wang,Xuan Zhang,Dongzhi Yang,Jun Nie,Guiping Ma

The repair process in wound healing is vital and complicated, which brings great challenge to researchers. Tissue engineering scaffolds provide a realizable approach for wound repair. In this study, nanofibers with Poly(γ-glutamic acid) (γ-PGA) as the core and Polylactic acid (PLA) as the shell materials were prepared via coaxial electrospinning. The effect of flow rate on the structure and diameter of the core-shell nanofibers was investigated. The microstructure and morphology of the core-shell nanofibers were observed by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). In laser scanning confocal microscopy experiments (LSCM), core-shell nanofibers loaded with rhodamine and coumarin-6 were excited with red and green light respectively to further investigate the structure of core-shell nanofibers. The hydrophobicity and degradation of core-shell nanofibers were studied by water contact angle test and degradation performance test experiments. The in vitro cell culture study showed favorable biocompatibility of nanofibers. The in vivo animal experiment showed that PLA/γ-PGA core-shell nanofiber membrane was conducive to wound repair, where more than 90% re-epithelialization was observed. It suggested a promising and dependable material for tissue engineering and wound healing.

Wearable biosensor with high specific surface area PGA-CNTs electrode for sweat glucose detection

Yuting Zheng,Yunfan Zhang,Zifan Xue,Hui Li,Shengnan Shen

Sweat glucose sensors have a high potential to detect blood glucose concentration conveniently and quickly, making them a focus of research on glucose detection methods. We prepare a polyglycolic acid-carbon nanotubes (PGA-CNTs) film with a high specific surface area, loaded with glucose oxidase (GOx) to develop a PGA-CNTs sweat glucose sensor. PGA films with a porous three-dimensional (3D) network structure were prepared by solvent evaporation, CNTs were deposited to obtain PGA-CNTs electrodes, and GOx was adsorbed through the high specific surface area structure of the 3D network. The results confirmed that the mesh structure formed by carbon nanotubes attached to PGA could substantially improve the charge transfer ability, and a considerable amount of GOx could be stably immobilized in the three-dimensional structure of the electrode. The PGA-CNTs sweat glucose sensor has excellent conductivity stability under bending and accurately detects glucose in the presence of other interference. The linear range of glucose response is 20–300 μM, and the sensitivity is 78.45 μA mM −1 cm −2 . Compared to existing products, the PGA-CNTs biosensors are more competitive in detection performance and cost.

Vaccine adjuvant platform and fluorescence imaging of amphiphilic γ-PGA-IMQ-LA-FL conjugates

Wenzhu Yin,Dechun Xuan,Bihua Deng,Mingxu Zhou,Fang Ma,Jinqiu Zhang,Yu Lu

Adjuvants have proven to be integral components in most vaccines for promoting appropriate immune responses at both innate and adaptive levels. Imiquimod (IMQ, R837), an agonist for Toll-like receptors 7 (TLR7), is a potent molecular activator of the innate immune system and has been developed as an adjuvant for immunotherapy. However, due to its pharmacokinetic profile, IMQ may induce strong local and systemic inflammatory reactions and is poorly tolerated. To overcome these hurdles, herein, we conjugate IMQ and lauryl alcohol (LA) to the backbone of biodegradable poly (γ-glutamic acid) (γ-PGA) to prepare an amphiphilic polymer (shortened to γ-PGA-IMQ-LA) with improved stability and biodistribution of IMQ. In order to investigate the distribution of the amphiphilic polymer in vitro and in vivo, fluorescein was grafted to the lateral chain to synthesize fluorescent-labeled γ-PGA-IMQ-LA-FL (shortened to FIP). The novel amphiphilic FIP polymer was characterized via1H NMR, FI-IR, UV-vis, fluorescence spectra and zeta potential. Using ovalbumin (OVA) as a model antigen and FIP as an adjuvant for immunization, the formulations of FIP + OVA in different dosages could significantly promote the production of OVA-specific IgG in mice and prolong immune responses with acceptable safety.

Preparation and characterization of CS/γ-PGA/PC complex nanoparticles for insulin oral delivery

Liu Kedong,Chen Yun,Yang Zhaoqi,Jin Jian

In recent years, natural polyelectrolyte complex chitosan/poly-γ-glutamic acid (CS/γ-PGA) has emerged as a promising nanocarrier delivery system for drug delivery due to its non-toxic and easily degradable properties. However, tightly bound CS/γ-PGA complexes often lead to lower encapsulation efficiency of peptide drugs without coordination of metal ions. In this investigation, we find that procyanidin (PC), as a cross-linking agent, is able to combine with CS and insulin through hydrogen bonding, which significantly improve encapsulation efficiency of insulin up to 90% in CS/γ-PGA/PC complex nanoparticles (CNPs). The results of this study demonstrate that CNPs release insulin in a pH-dependent approach and greatly reduce its enzymatic degradation in vitro. The transepithelial permeability of the CNPs is 8.1-fold higher than that of native insulin via both endocytosis and paracellular passways. Further pharmacokinetic studies reveal an oral pharmacological availability of 9.81 ± 1.32% in type I diabetic rats after intragastric administration of freeze-dried CNPs which loaded in enteric-coated capsules. In consequence, CNPs not only improve oral bioavailability of insulin but also are a promising delivery platform for insulin or other peptide/protein drugs. Graphical Abstract

Polyamino acid antibacterial hydrogel with photothermal effect for wound healing

Nian Liu,Sen Liang,Jingtao Li,Qi Li,Shuo Bai,Jing Hu

Poly (amino acid)s-based hydrogels with high biological safety and natural antibacterial capabilities have significant clinical value in wound repair dressings. Poly ( γ -glutamic acid) (PGA) and ε -poly-lysine (EPL) are among the few poly (amino acid)s that are widely available and can be biosynthesized on a large scale. In this study, PGA and EPL were selected as backbone molecules, and the complex of protocatechuic aldehyde and iron (PA@Fe) with photothermal effects was introduced to prepare the poly (amino acid)s-based antibacterial wound repair gel PGA-EPL-PA@Fe (PEPF). The PEPF gel dressing exhibited excellent adhesive, antibacterial, antioxidant, and other multiple properties, significantly promoting wound healing in rats, indicating the high clinical application prospects of poly (amino acid)s-based dressings.

A Safe and Multifunctional γ-PGA Hydrogel Platform: Endotoxin-Controlled Injectable Fillers and Antimicrobial Wound Dressings

Bingbing Wang,Zejing Chu,Jingyu Wei,Ruixiang Mai,Yuan Wang,Xiaocui Wang,Yi Hou,Na Zheng,Jiao Sun,Biao Dong

In response to the limitations of hyaluronic acid (HA)—such as its high cost, short durability, and instability—in anti-aging aesthetic applications, this study developed a novel injectable micelle system, with a triple network structure. It is the particle size of approximately 400 nm and the elevated potential that enhance the crosslinking density and mechanical strength of the hydrogel. Importantly, following ultrafiltration and purification processes, the material’s hemolysis rate measured by spectrophotometry was only 3.25%, and endotoxin levels measured by the LAL assay were less than 0.5 EU/mL (test conditions: 37 °C, pH = 7, detection limit: 0.125 EU/mL). Building on this safe and stable material platform, we further designed an antibacterial wound dressing by functionalizing γ-PGA with penicillin or benzalkonium chloride. It reduced the cellular activity of Staphylococcus aureus by 78.9% and 84.2%, respectively. The outstanding safety profile, combined with customizable functionality, positions this γ-PGA-based platform as a promising multifunctional biomaterial meeting practical standards for both aesthetic medicine and wound care applications.

Double-network PVA/γ-PGA/fucoidan hydrogels with favourable biocompatibility and anti-inflammatory activity for wound healing

Aoyu Li,Jun Yang,Yingying He,Changfeng Qu,Jinlai Miao

Acute wound healing still faces numerous challenges, such as excessive inflammatory response, prolonged hemostasis time, and insufficient tissue regeneration, which often lead to delayed wound healing and scar formation. To address these issues, this study developed a multifunctional “physical-ionic” dual-network hydrogel (PPF hydrogel). This hydrogel combines an elastic polyvinyl alcohol (PVA) scaffold with a rigid ionic network composed of γ-polyglutamic acid (γ-PGA) and fucoidan, cross-linked via Ca 2+ . The resulting hydrogel possesses a highly interconnected porous structure, exhibiting excellent mechanical strength and moderate tissue adhesion. In vitro and in vivo evaluations showed that the PPF hydrogel has good blood and cell compatibility, rapidly achieving hemostasis and promoting the proliferation and migration of L929 fibroblasts. In a mouse full-thickness skin resection model, the PPF significantly accelerated wound closure while reducing epidermal hyperplasia and promoting orderly collagen deposition. Mechanistically, the PPF effectively modulates the immune microenvironment by downregulating pro-inflammatory cytokines (TNF-α, IL-6) and upregulating anti-inflammatory factors (IL-10) and VEGF. This regulation promotes M2 macrophage polarization and angiogenesis, thereby accelerating the transition from the inflammatory phase to the remodeling phase. This dual-network hydrogel offers a promising therapeutic strategy for high-quality acute wound repair in regenerative medicine.

An Injectable CMCS/γ-PGA/PRP Bioadhesive With Antibacterial, Adhesive, and Regenerative Properties for Infected Wound Healing

Wenqing Xu,Jinglve Zhang,Yutian Xiao,Tsz Lam Yau,Yuwei Luo,Yujun Shao,Ze Cheng,Chenchen Zuo,Junlong Tian,Shaorong Lei,Dingyu Wu

Infected traumatic wounds remain a formidable clinical challenge because existing bioadhesives provide only tissue adhesion and lack concurrent antimicrobial and regenerative capabilities. Here, we developed an injectable bioadhesive (CγR) that integrates an “antibacterial–adhesive–regenerative” (AAR) triad by precisely tuning the ratio of carboxymethyl chitosan (CMCS), autologous platelet-rich plasma (PRP), and γ-polyglutamic acid (γ-PGA). CγR exhibits > 99.9% bactericidal efficacy against Escherichia coli and Staphylococcus aureus , delivers instantaneous wet-tissue adhesion (> 3 kPa), and sustains a gradient release of PDGF, TGF-β, and VEGF. In a rat model of infected full-thickness wounds, a single application achieved sterile sealing and complete epithelialization within 6 days, increased collagen deposition by 1.5-fold, and markedly drove macrophage polarization toward the prohealing M2 phenotype. By condensing the conventional “suture–antibiotic–dressing” regimen into a one-step procedure, CγR offers a biosafe and readily scalable platform for managing contaminated complex wounds and holds promise for clinical translation. Graphical The injectable bioadhesive CγR integrates carboxymethyl chitosan (CMCS), platelet-rich plasma (PRP), and γ-polyglutamic acid (γ-PGA) to provide antimicrobial, adhesive, and regenerative effects. CγR bioadhesive exhibits sustained release of growth factors, promotes rapid wound closure, and accelerates tissue regeneration in infected wound models.

PGA-TMC/PTMC/nHA composite membrane with synergistic barrier and osteogenic functions for enhanced bone defect regeneration

Wang Jing,Wang Pei,Wang Baojin,Liu Jian,Huang Min,Han Wei,Li Xinzhe,Gao Yun,Xiong Wei

Balancing mechanical strength, controllability of degradation and bioactivities, remains a significant challenge for absorbable barrier membranes in guided bone regeneration (GBR) for the treatment of bone defects. To overcome these limitations, a novel absorbable synthetic PGA-TMC/PTMC/nHA (PGTTH) barrier membrane was developed using electrospinning technology to prolong barrier duration and promote rapid bone repair. The PGTTH membrane integrates three functional components in a one-step fabrication process: a poly (glycolic acid-co-trimethylene cabonate) (PGA-TMC) copolymer matrix providing mechanical integrity, polytrimethylene carbonate (PTMC) imparting flexibility and a slow, linear degradation profile to ensure long-term structural stability, nano-hydroxyapatite (nHA) delivering bioactive ions to enhance osteogenesis and biomineralization. To systematically evaluate the performance of the PGTTH membrane, mechanical characterization and in vitro degradation tests confirmed that the membrane exhibits appropriate mechanical properties and a slow degradation profile, thereby demonstrating its ability to maintain structural space over an extended period. Furthermore, in vitro and in vivo studies demonstrated that the PGTTH membrane significantly promotes cell adhesion, proliferation, and osteogenic differentiation. In conclusion, this study not only presents a promising GBR barrier membrane material with potential for clinical application, but also offers a new design strategy and experimental basis for developing absorbable membranes that combine excellent barrier functionality with osteoinductive capacity.

HRP-catalyzed grafting of MXene@PGA to silk fibers for visualization of dual-driven heating smart textile

Biaobiao Yan,Shuaibing Huang,Yiwen Ren,Man Zhou,Yuanyuan Yu,Bo Xu,Li Cui,Qiang Wang,Ping Wang

MXene-based functional textiles have been widely studied and applied in many fields. However, the service stability of MXene combined with textile substrates in the environment is far from ideal, which makes its practical application a great challenge. Here we introduced gallic acid (GA), as natural reactive polyphenol compound to silk fibers through enzymatic polymerization, which significantly improved the durability of its conductivity. The small molecules of GA can covalently bind to the titanium atoms on the MXene nanosheets, and the tyrosine residues from silk fibroins can be enzymatically oxidized by horseradish peroxidase (HRP) and further coupled with GA simultaneously, thus forming a covalent cross-linked network on the fiber surfaces. Furthermore, the durable MXene-based textile was used to manufacture smart dual-driven thermal devices with temperature monitoring, which can judge the real-time temperature during heating by changes in its apparent color. More importantly, the textile with smart temperature visualization also offers good EMI shielding and superior UV resistance, while retaining its inherent moisture-wicking, breathable and softness. The present work provides a new insight for the preparation of MXene-based multifunctional textile, and the smart visualization of dual-driven heating shows promising applications in practical personal thermal management.
