Regulation of Mechanical Properties and Degradation Behavior of Poly(L-Lactide-Co-ε-Caprolactone) (PLCL) by Magnesium Oxide Nanoparticle

Xiaofeng Wang,Hongyan Jin,Shuya Zhang,Zifan Zou,Tao Zhang,Yingzheng Lv,Zichen Yuan,Wenjuan Han,Qian Li

In tissue engineering and regenerative medicine, biodegradable PLCL polymers are highly valued. This is because their mechanical properties can be finely adjusted, and they are compatible with biological tissues. However, precise control over their degradation rate remains a challenge for biomedical applications. In this study, magnesium oxide (MgO) nanoparticles were incorporated into PLCL matrices to systematically modulate both mechanical properties and degradation behavior. To determine the microstructure and thermal robustness of the PLCL/MgO composites, a battery of tests was conducted. These tests included scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), differential scanning calorimetry (DSC), rheometry, and thermogravimetric analysis (TGA). The results revealed that MgO nanoparticles significantly enhanced the ultimate tensile strength and modulus of elasticity of PLCL while simultaneously accelerating its hydrolytic degradation. This was attributed to MgO's dual role: (1) stabilizing the local pH by neutralizing acidic degradation byproducts, thereby mitigating autocatalytic degradation of the PLCL matrix, and (2) acting as an alkaline catalyst to facilitate ester bond hydrolysis. Notably, the degradation rate increased in a MgO concentration-dependent manner, with minimal impact on the polymer's biocompatibility. This study demonstrates that MgO can effectively modulate the degradation process, thereby enabling the creation of bioresorbable PLCL-based materials with tailored lifespans. This advancement holds great promise for advanced biomedical applications.

Bioactive microgel coated electrospun membrane with cell-instructive interfaces and topology for abdominal wall defect repair

Renquan Xing,Rui Gao,Yini Huangfu,Yufeng Zhang,Shuangyang Li,Chuangnian Zhang,Pingsheng Huang,Weiwei Wang,Anjie Dong,Zujian Feng

Current mesh materials used for the clinical treatment of abdominal defects struggle to balance mechanical properties and bioactivity to support tissue remodeling. Therefore, a bioactive microgel coated electrospinning membrane was designed with the superiority of cell-instructive and topology in guiding cell behavior and function for abdominal wall defect reconstruction. The electrostatic spinning technique was employed to prepare a bioabsorbable PLCL fiber membrane with effective mechanical support. And decellularized matrix (dECM) derived bioactive microgels were further coated on the fiber membrane through co-precipitation with dopamine, which was expected to endow with cell-instructive hydrophilic interfaces, and also topological morphologies for cell adhesion. Moreover, the introduction of dECM into the microgel was able to promote myogenic proliferation and differentiation of C2C12 cells. Subsequently, in vivo experiments using a rat abdominal wall defect model demonstrated the bioactive microgel coating significantly contributed to the reconstruction of intact abdominal wall structures, highlighting its potential for clinical application in promoting the repair of soft tissue defects associated with abdominal wall damage. This study presented an effective mesh material for facilitating the reconstruction of abdominal wall defects, and also contributed novel design concepts for surface modification of scaffolds with cell-instructive interfaces and topology.

Development of Nanofiber Sponges-Containing Nerve Guidance Conduit for Peripheral Nerve Regeneration in Vivo

Binbin Sun,Zifei Zhou,Tong Wu,Weiming Chen,Dawei Li,Hao Zheng,Hany El-Hamshary,Salem S. Al-Deyab,Xiumei Mo,Yinxian Yu

In the study of hollow nerve guidance conduit (NGC), the dispersion of regenerated axons always confused researchers. To address this problem, filler-containing NGC was prepared, which showed better effect in the application of nerve tissue engineering. In this study, nanofiber sponges with abundant macropores, high porosity, and superior compressive strength were fabricated by electrospinning and freeze-drying. Poly(l-lactic acid-co-ε-caprolactone)/silk fibroin (PLCL/SF) nanofiber sponges were used as filler to prepare three-dimensional nanofiber sponges-containing (NS-containing) NGC. In order to study the effect of fillers for nerve regeneration, hollow NGC was set as control. In vitro cell viability studies indicated that the NS-containing NGC could enhance the proliferation of Schwann cells (SCs) due to the macroporous structure. The results of hematoxylin–eosin (HE) and immunofluorescence staining confirmed that SCs infiltrated into the nanofiber sponges. Subsequently, the NS-containing NGC was implanted in a rat sciatic nerve defect model to evaluate the effect in vivo. NS-containing NGC group performed better in nerve function recovery than hollow NGC group. In consideration of the walking track and triceps weight analysis, NS-containing NGC was close to the autograft group. In addition, histological and morphological analyses with HE and toluidine blue (TB) staining, and transmission electron microscope (TEM) were conducted. Better nerve regeneration was observed on NS-containing NGC group both quantitatively and qualitatively. Furthermore, the results of three indexes’ immuno-histochemistry and two indexes’ immunofluorescence all indicated good nerve regeneration of NS-containing NGC as well, compared with hollow NGC. The results demonstrated NS-containing NGC had great potential in the application of peripheral nerve repair.

Piezoelectric conduit combined with multi-channel conductive scaffold for peripheral nerve regeneration

Ying Ma,Hao Wang,Qiqi Wang,Xiaodong Cao,Huichang Gao

Due to the complex structural hierarchy and regenerative microenvironment of peripheral nerves, developing a nerve guidance conduit (NGC) that can replace autografts for repairing long-distance nerve defects remains a challenge. To overcome this limitation, a multi-channel NGC with dual electrical activity of piezoelectricity and conductivity was constructed to enhance peripheral nerve regeneration. Silk fibroin (SF) was adopted to prepare a multi-channel cryogel scaffold by gradient freezing method, and then poly(3,4-ethylenedioxythiophene) (PEDOT) in situ polymerization based on the porous structure of the cryogel was carried out to achieve good conductivity. Meanwhile, a polyvinylidene fluoride (PVDF)/poly ( l -lactic acid- co -caprolactone) (PLCL) film with spontaneous electrical properties and high mechanical strength was constructed and wrapped in the outer layer of SF/PEDOT cryogel to construct an NGC with spontaneous electrical stimulation properties and surgical suture resistance, providing a biomimetic electrophysiological microenvironment for peripheral nerve regeneration. In vitro cell experiments indicated that the multi-channel electroactive NGC not only promoted the proliferation, maturation and myelination of Schwann cells (SCs), but also effectively promoted neuronal differentiation and axonal regeneration of Pheochromocytoma 12 (PC12) cells. Furthermore, a rat sciatic nerve defect model was constructed to evaluate the effect of multi-channel electroactive NGC on peripheral nerve repair and the results showed that electroactive NGC could significantly promote peripheral nerve regeneration and functional recovery after 12 weeks, achieving the repair effect of autografts through the positive synergistic effect of piezoelectric and conductive properties of NGC. These results demonstrated the great potential of multi-channel electroactive NGCs in repairing peripheral nerve injury (PNI).

Nanofibrous vascular scaffold prepared from miscible polymer blend with heparin/stromal cell-derived factor-1 alpha for enhancing anticoagulation and endothelialization

Weizhong Wang,Dinghua Liu,Dejian Li,Haibo Du,Jingtian Zhang,Zhengwei You,Maoquan Li,Chuanglong He

There is an urgent clinical demand to develop a functional small diameter vascular graft with long-term patency, which mainly relies on the anticoagulation and endothelialization of the vascular graft. In addition, improved degradation of vascular graft provides more space for cell infiltration and facilitates remodeling of blood vessel. In this study, an elastic and biomimetic nanofibrous vascular scaffold with improved degradability was prepared by adding poly(lactic-co-glycolic acid) (PLGA) into the poly(L-lactic acid) (PLLA)/poly(L-lactide-co-ε-caprolactone) (PLCL) blend using thermally induced phase separation (TIPS) technique. The incorporation of PLGA also improved the hydrophilicity of the composite scaffold. Then the vascular scaffold was surface modified by combination of heparin and stromal cell-derived factor-1 alpha (SDF-1α). The results of whole blood clotting kinetics and plasma recalcification profiles indicated that heparinized modification significantly enhanced the anticoagulation of vascular scaffold. In vitro cell culture assays demonstrated the immobilized SDF-1α facilitated recruitment of endothelial progenitor cells (EPCs), migration and proliferation of mature endothelial cells, human umbilical vein endothelial cells (HUVECs), and expression of VE-cadherin/CD144 and endothelial nitric oxide synthase (eNOS) genes in HUVECs, thereby accelerating the endothelialization of the modified vascular scaffold. Besides, the nanofibrous vascular scaffold modified with heparin and SDF-1α inhibited the proliferation of human vascular smooth muscle cells (HVSMCs). Therefore, the phase separated nanofibrous vascular scaffold modified with heparin and SDF-1α shows the promising in vitro performance as a functional small diameter vascular graft.
