Zr-MOF Nanoparticles Crafted from Conjugated Imidazolyl Carboxylic Acid Ligands for Sensitive Fluorescence Detection of 2,4-DNP and PNA

Fangfang Chen,Xiudu Zhang,Wensen Du,Chengcheng Ye,Meng Liu,Jun Du,Yonghong Ni

2,4-Dinitrophenol (2,4-DNP) and p-nitroaniline (PNA), as hazardous nitroaromatic explosives and persistent organic pollutants, exhibit high toxicity, nonbiodegradability, and carcinogenicity. Exposure to these compounds can cause severe health issues, including cardiovascular, neurological, and renal damage. In this study, a novel zirconium-based metal–organic framework (Zr-MOF, denoted as Zr-BPBI) was synthesized via a solvothermal reaction using the ligand 4,4′,4″,4‴-([1,1′-biphenyl]-4,4′-diyldi(1H-imidazole-2,4,5-triyl))tetrabenzoic acid (H4BPBI) and ZrCl4. The investigations showed that the original synthetic parameters, including surfactant concentration, acid type, reaction time, and temperature, could strongly affect the morphology, particle size, and fluorescence performance of the as-prepared Zr-BPBI. Interestingly, under excitation at 422 nm, the as-prepared Zr-BPBI nanoparticles (Zr-BPBI-NP) emitted blue luminescence centered at 502 nm, which demonstrated exceptional sensitivity toward PNA and 2,4-DNP with rapid fluorescence quenching responses (within 30 s for 2,4-DNP and 15 s for PNA). Simultaneously, common interfering substances (e.g., small molecules and ions) did not affect the above detection, suggesting high selectivity of the as-prepared Zr-BPBI-NP fluorescent probe. The corresponding limits of detection were 0.36 μM for 2,4-DNP and 0.4 μM for PNA. Obviously, the present Zr-BPBI-NP fluorescent probe is a new option for the highly sensitive and selective detection of PNA and 2,4-DNP in actual aqueous environments.

ACE2-PNA conjugates exploit viral endocytosis for targeted intracellular delivery and exhibit dual antiviral efficacy against SARS-CoV-2

Wang Yifei,Xu Jinghan,Chen Yiquan,Xiao Xin,Zhu Yiru,Yu Liyang,Shi Wen,Li Jingchao,Li Jianhua,Zhu Chenggang

The COVID-19 pandemic and its protracted consequences underscore the urgent need for more effective antiviral strategies. Current antiviral strategies face a persistent challenge achieving sufficient viral suppression while minimizing off-target toxicity, particularly against such highly mutable viruses. Here, we describe a Receptor-Drug Conjugate (RDC) strategy, in which a therapeutic payload is covalently linked to a decoy receptor, enabling virus-triggered targeted intracellular delivery. Angiotensin-converting enzyme 2 (ACE2), as the essential receptor for SARS-CoV-2 entry, has been widely exploited for virus-neutralizing strategies. Targeting SARS-CoV-2 as a proof-of-concept, we conjugated peptide nucleic acids (PNAs) designed to target the viral ORF1ab region (±30 bp) to ACE2-Fc. The ACE2-PNA conjugate demonstrated superior inhibitory efficacy against multiple SARS-CoV-2 variants relative to ACE2-Fc alone. We confirmed that ACE2-PNA retains the extracellular neutralization activity of soluble ACE2, while being selectively internalized into virus-infected cells via virus-mediated endocytosis. Owing to its inherent protease resistance, the PNA component remains intact upon cytoplasmic entry and subsequently exerts antisense inhibitory activity against viral RNA. The mechanistic feasibility of ACE2-PNA was further validated in a mouse model. Collectively, RDC represents a novel virus-triggered targeted delivery platform that confers dual antiviral efficacy through extracellular virion neutralization and intracellular inhibition of viral replication. It has significant implications for reducing off-target toxicity and enhancing antiviral potency, and is furthermore readily adaptable to diverse viral pathogens and therapeutic payloads.

A PNA-DNA2 Triple-Helix Molecular Switch-Based Colorimetric Sensor for Sensitive and Specific Detection of microRNAs from Cancer Cells

Mengjia Xu,Pan Fu,Shu Xing,Yang Zhao,Chao Zhao

Graphical Point-of-care cancer catching ? A novel PNA-DNA 2 triplex consisting of a homopurine PNA strand and two homopyrimidine DNA strands is reported for the first time. On the basis of this phenomenon, a simple, inexpensive colorimetric PNA-DNA 2 triple-helix molecular switch-based sensing platform for sensitive and selective detection of miRNAs from cancer cells is demonstrated. Peptide nucleic acids (PNAs), the synthetic DNA mimics that can bind to oligonucleotides to form duplexes, triplexes, and quadruplexes, could be advantageous as probes for nucleic acid sequences owing to their unique physicochemical and biochemical properties. We have found that a homopurine PNA strand could bind to two homopyrimidine DNA strands to form a PNA-DNA 2 triplex. Moreover, the cyanine dye DiSC 2 (5) could bind with high affinity to this triplex and cause a noticeable color change. On the basis of this phenomenon, we have designed a label-free colorimetric sensing platform for miRNAs from cancer cells by using a PNA-DNA 2 triple-helix molecular switch (THMS) and DiSC 2 (5). This sensing platform can detect miRNA-21 specifically with a detection limit of 0.18 nM, which is comparable to that of the THMS-mediated fluorescence sensing platform. Moreover, this colorimetric platform does not involve any chemical modification or enzymatic signal amplification, which boosts its applicability and availability at the point of care in resource-limited settings. The universality of this approach can be simply achieved by altering the sequences of the probe DNA for specific targets.

Highly sensitive electrochemical biosensor for MMP-2 detection using multi-stage amplification with PNA T7 RNA polymerase and split CRISPR/Cas12a

Yuanxun Gong,Jiayi Zhang,Huiying Liang,Yanling Wang,Zichun Song,Jihua Wei,Qianli Tang,Kai Zhang,Xianjiu Liao

Matrix metalloproteinases (MMPs) are critical for the degradation of extracellular matrix components, which is essential for processes such as tissue remodeling, wound healing, and angiogenesis. MMP-2, in particular, degrades type IV collagen and is associated with diseases such as cancer and cardiovascular conditions, making it a significant biomarker. This study presents a novel electrochemical biosensor for the detection of MMP-2. The biosensor combines the specificity of a programmable RNA detection system with the amplification capabilities of T7 RNA polymerase and CRISPR/Cas12a. In the absence of MMP-2, Peptide Nucleic Acid (PNA) adsorbs on the DNA template, preventing RNA amplification and resulting in a low electrochemical signal. When MMP-2 is present, it cleaves a specific peptide, releasing the DNA template and enabling RNA synthesis. This activation leads to the cleavage of single-stranded DNA probes, increasing the electrochemical signal. The biosensor demonstrates high sensitivity with a detection limit of 0.496 fM for MMP-2 and excellent specificity, effectively distinguishing MMP-2 from other proteins. It successfully detected MMP-2 in biological samples, such as liver cancer cell supernatants, indicating its potential for clinical diagnostics. This innovative biosensor offers a powerful tool for the early detection and monitoring of diseases associated with MMP-2 activity.

Electrochemical detection of DNA hybridization using a water-soluble branched polyethyleneimine–cobalt(III)–phenanthroline indicator and PNA probe on Au electrodes

Xianggang Liu,Xiangjin Qu,Hai Fan,Shiyun Ai,Ruixia Han

An electrochemical method for the detection of DNA hybridization using a novel electroactive, cationic, and water-soluble branched polyethyleneimine (BPEI)–cobalt(III)–phenanthroline(phen) polymeric indicator and single-stranded neutral peptide nucleic acid (PNA) probe on the Au electrode was developed. The indicator possesses some free amine groups, as well as cationic cobalt complexes in the polymer chain. It does not bind to neutral PNA capture probe alone. However, the indicator strongly interacts with the negatively charged backbone of the complementary oligonucleotide bound to the PNA probe through electrostatic interactions . The coordination spherical moieties also interact with the probe by embedding into the double-helix structure of PNA–DNA. These two interactions enable transduction of hybridization, producing a clear electrical signal in differential pulse voltammetry (DPV). The correlation against non-complementary DNA, three-base and one-base mismatch DNA was sharp, and the signal of indicator for the target DNA demonstrated a linear relationship within the concentration range of 5.0   ×   10 −9 to 2.5   ×   10 −7   M ( R   =   0.9940) with a detection limit of 5.6   ×   10 −10   M. These studies showed that the novel polymeric indicator and single-stranded PNA probe could be used to fabricate an electrochemical biosensor for DNA detection. This technique can provide an alternative route for expanding the range of detection methods available for DNA hybridization.

A label-free electrochemical biosensor with magnetically induced self-assembly for the detection of CYP2C9*3 gene

Ruijiang Liu,Shuai Pan,Min Liu,Wei Huang,Zhixiang Lv,Aolin He

A label-free electrochemical biosensor with magnetically induced self-assembly based on peptide nucleic acids (PNA) and Fe 3 O 4 /Fe 2 O 3 heteroplasmon nanorods was successfully fabricated to detect the single nucleotide polymorphism (SNP) of CYP2C9*3. The sensitivity of the electrochemical biosensor was improved by introducing PNA and Fe 3 O 4 /Fe 2 O 3 @Au nanocomposites. The Fe 3 O 4 /Fe 2 O 3 @Au nanocomposites were assembled on the surface of the magnetic glassy carbon electrode (MGCE) by magnetic field, and the thiolated PNA were connected to the nanocomposites through Au-S bonds. The biosensor exhibited a good linear correlation between 1 pM and 1 µM (R 2  = 0.9941), and the limit of detection (LOD) and the limit of quantitation (LOQ) were 0.95 pM and 3.18 pM, respectively. Besides, the electrochemical biosensor could recognize single-base mismatch DNA, and it could be stored stably for 15 days at 4 °C.

Ultrasensitive label-free detection for lung cancer CYFRA 21-1 DNA based on ring-opening polymerization

Xiaofei Li,Yawen Zhang,Lulu Hao,Yanju Liu,Xia Wang,Huaixia Yang,Jinming Kong

Cytokeratin fragment antigen 21-1 (CYFRA 21-1) DNA is perceived as sensitive tumor marker for the diagnosis of non-small cell lung cancer and other tumor. Herein, linear chain poly(ε-caprolactone) (PCL) synthesized by ring-opening polymerization is applied to ultrasensitive label-free electrochemical impedance detection system for CYFRA 21-1 DNA. First, thiolated peptide nucleic acid (PNA) is self-assembled into the Au electrode surface through the formation of Au–S bonds, allowing the PNA to act as biomolecular probe and form PNA/DNA heteroduplex with the target DNA via specific hybridization. Then, PCL is conjugated to the immobilized DNA on the electrode via “carboxylate–Zr 4+ –phosphate” bridges. Finally, the electrochemical response of modified PNA/DNA/Zr 4+ /PCL electrode is determined by electrochemical impedance method to quantify of CYFRA 21-1 DNA. Under optimal conditions, this method exhibits highly sensitivity with a broad linear range (0.1 fM – 1 nM) ( R 2  = 0.995) and the limit of detection (LOD) is as low as 10.73 aM, which is equivalent to just 64 molecules in a 10 μL sample. What's more, the high selectivity , good anti-interference, label-free operation, and real-time monitoring in complex samples of the proposed strategy demonstrate its broad application for the early diagnosis and clinical monitoring.

Coenzyme-mediated electro-grafting for ultrasensitive electrochemical DNA biosensing

Qiong Hu,Luofeng Su,Yanyu Huang,Zhuohua Chen,Xiaojing Cao,Yilin Luo,Dongdong Qin,Li Niu

Signal amplification through the grafting of polymers has attracted increasing attention in biodetection at ultralow concentrations. Herein, an ultrasensitive peptide nucleic acid (PNA)-based electrochemical DNA biosensor is reported by using the coenzyme-mediated electro-grafting of polymers as a novel amplification method. For the capture of DNA target, PNA is used as the probe. After hybridization, reversible addition-fragmentation chain-transfer (RAFT) agents are site-specifically tethered to the deoxyribose phosphate backbone of the captured DNA targets via the carboxylate-Zr(IV)-phosphate cross-linking chemistry. Subsequently, electro-grafting of polymers through the nicotinamide adenine dinucleotide (NAD + ) coenzyme-mediated electro-RAFT (NAD + -eRAFT) polymerization of ferrocenylmethyl methacrylate (FcMMA) recruits numerous ferrocene redox tags for voltammetric measurement. As the electro-grafting of polymers involves simply i) the tethering of RAFT agents and ii) the NAD + -eRAFT polymerization, it is highly efficient, easy to use, and low-cost. Under optimal conditions, the voltammetric signal correlates linearly with the logarithm of DNA concentration over the range from 0.1 fM to 0.1 nM ( R 2  = 0.996), with a detection limit of 0.067 fM. The PNA-based electrochemical DNA biosensor can also differentiate even single base mismatch and is applicable to DNA detection in the presence of complex serum matrices, thus showing great promise in the sequence-specific detection of DNA targets at ultralow concentrations.

Incorporating redox-sensitive nanogels into bioabsorbable nanofibrous membrane to acquire ROS-balance capacity for skin regeneration

Shihao Zhang,Yamin Li,Xiaofeng Qiu,Anqi Jiao,Wei Luo,Xiajie Lin,Xiaohui Zhang,Zeren Zhang,Jiachan Hong,Peihao Cai,Yuhong Zhang,Yan Wu,Jie Gao,Changsheng Liu,Yulin Li

Facing the high incidence of skin diseases, it is urgent to develop functional materials with high bioactivity for wound healing, where reactive oxygen species (ROS) play an important role in the wound healing process mainly via adjustment of immune response and neovasculation. In this study, we developed a kind of bioabsorbable materials with ROS-mediation capacity for skin disease therapy. Firstly, redox-sensitive poly(N-isopropylacrylamide-acrylic acid) (PNA) nanogels were synthesized by radical emulsion polymerization method using a disulfide molecule as crosslinker. The resulting nanogels were then incorporated into the nanofibrous membrane of poly( l -lactic acid) (PLLA) via airbrushing approach to offer bioabsorbable membrane with redox-sensitive ROS-balance capacity. In vitro biological evaluation indicated that the PNA-contained bioabsorbable membrane improved cell adhesion and proliferation compared to the native PLLA membrane. In vivo study using mouse wound skin model demonstrated that PNA-doped nanofibrous membranes could promote the wound healing process, where the disulfide bonds in them were able to adjust the ROS level in the wound skin for mediation of redox potential to achieve higher wound healing efficacy.

A Fluorescent Sensor Based on Reversible Addition-Fragmentation Chain Transfer Polymerization for the Early Diagnosis of Non-small Cell Lung Cancer

Liu Wenwen,Ma Ligang,Guo Zhuangzhuang,Liu Tao,Liu Yanju,Wang Dazhong,Kong Jinming

We propose a novel, ultrasensitive and low-cost sensor using reversible addition-fragmentation chain transfer (RAFT) polymerization as a signal amplification strategy for the detection of CYFRA 21-1 DNA fragment, a tumor marker of non-small cell lung carcinoma. The peptide nucleic acid (PNA) probes were firstly immobilized on magnetic beads (MBs) to capture the CYFRA 21-1 DNA specifically. After hybridization, CPAD was tethered to the hetero duplexes through carboxylate-Zr 4+ -phosphate chemistry. Subsequently, a number of fluorescent tags were introduced to the heteroduplexes through RAFT polymerization, leading to an amplification of the fluorescence signal. The sensor demonstrates a low limit of detection (LOD) of 0.02 fM. It has great selectivity with respect to base mismatch DNA, and high anti-interference ability in normal human serum. Overall findings of the study suggest that proposed sensor holds enormous potential to be used as a tool for the early-stage diagnosis of lung cancers.

Uniform and Easy-To-Prepare Glycopolymer-Brush Interface for Rapid Protein (Anti-)Adhesion Sensing

Junji Li,Xiao-Yang Tian,Li-Ping Zong,Qiang Zhang,Xue-Ji Zhang,Robert Marks,Serge Cosnier,Dan Shan

Glycopolymers have emerged as powerful and versatile glycan analogues for the investigation of cellular signal transduction. In this study, a layer of the glycopolymer-brush (GlyB) interface was functionalized on the surface of gold substrates. In order to enhance the capability and accessibility of this transducer interface, a combined protocol of copper(0)-mediated living radical polymerization (Cu(0)-LRP) with subsequent “CuAAC” click reaction was utilized to synthesize a set of novel glycopolymer precursors with a tunable scaffold structure and pyranose ligands. The resulting glycopolymer exhibited a fine-tuned molecular weight with a minor dispersity of 1.27. Through surface plasmon resonance (SPR) analysis, various GlyB interfaces exhibiting different saccharide moieties (glucose, mannose, and galactose) were examined to study their adhesion or antiadhesion potential toward three types of proteins, concanavalin A, bovine serum albumin, and peanut agglutinin (PNA). The strong affinity between poly(galactose) and PNA was further employed to construct a proof-of-concept aggregation-mediated sensing system. This minimal naked-eye sensor that consisted of only two substances, namely, gold nanoparticles and glycopolymers, was characterized and tested for its potential in protein quantification.
