Point of care testing (POCT) of cholesterol in blood serum via a moving reaction boundary electrophoresis titration chip

Muhammad Idrees Khan,Qiang Zhang,Youli Tian,Shah Saud,Yiren Cao,Jicun Ren,Weiwen Liu,Chengxi Cao

Cholesterol (CHO) in human blood is one of the most frequently and crucially quantified substances in diagnostic laboratories. However, visual and portable point of care testing (POCT) methods have been rarely developed for the bioassay of CHO in blood samples. Here, we developed an electrophoresis titration (ET) model, a chip device of ∼60 grams, and a quantification method for the POCT of CHO in blood serum based on a moving reaction boundary (MRB). In this model, the selective enzymatic reaction is integrated with an ET chip for visual and portable quantification. At first, CHO reacted with cholesterol oxidase (CHOx) in the anode well, producing H2O2 and cholest-4-en-3-one in the solution. H2O2 further oxidized the colorless and chargeless leucocrystal violet (LCV) dye into violet colored positively charged crystal violet (CV+) and, under the influence of the electric field, the CV+ migrates in the ET channels and is titrated by the alkali of sodium hydroxide immobilized in the ET channels. The length covered by the MRB was measured as a function of the CHO content. The relevant experiments validated the feasibility of the model and method. Furthermore, the experiments revealed the high selectivity, portability, and visuality of the ET-MRB model, device, and method. Finally, the experiments showed a fair sensitivity of LOD of 5 μM, good linearity of 10–1000 μM (r2 = 0.9919), fair stability (intra-day RSD of less than 5.09% and an inter-day RSD of less than 6.36%), and high recovery (99.4–105%). All the data and results indicate the potential of the ET-MRB model, chip device, and method for POCT of CHO in human blood samples.

One-Step, On-Site Chemical Printing of a 3D Plasmon-Coupled Silver Nanocoral Substrate toward SERS-Based POCT

Xuefei Zhao,Yueliang Wang,Yuanyuan Yao,Lifen Chen,Bingyong Lin,Wenhan Zheng,Yanbo Zeng,Lei Li,Yuanbin She,Longhua Guo

Surface-enhanced Raman scattering (SERS) with the advantages of high sensitivity, nondestructive analysis, and a unique fingerprint effect shows great potential in point-of-care testing (POCT). However, SERS faces challenges in rapidly constructing a substrate with high repeatability, homogeneity, and sensitivity, which are the key factors that restrict its practical applications. In this study, we propose a one-step chemical printing strategy for synthesizing a three-dimensional (3D) plasmon-coupled silver nanocoral (AgNC) substrate (only need about 5 min) without any pretreatments and complex instruments. The galvanic replacement between AgNO3 and Cu sheets will provide both Ag0 for the formation of silver nanostructures and Cu2+ for the polymerization of fish sperm DNA (FSDNA). The protection of AgNCs is facilitated by the crosslinked FSDNA, which can improve the stability of the substrate and promote the control of its coral-like morphology. The obtained substrate displays excellent capacity of signal enhancement due to the 3D plasmon coupling both between nanocoral tentacles and between nanocorals and Cu sheets as well. Therefore, the AgNC substrates display high activity (enhancement factor = 1.96 × 108) and uniformity (RSD < 6%). Food colorants have been widely used in various foods to improve their color, but the inevitable toxicity of colorants seriously threatens food safety. Therefore, the proposed AgNC substrates were used to directly quantify three kinds of weak-affinity food colorant molecules including Brilliant Blue, Allura Red, and Sunset Yellow assisted by the capture by cysteamine hydrochloride (CA), showing the detection limits (S/N = 3) of 0.053, 0.087, and 0.089 ppm, respectively. The SERS method has been further applied in the detection of the three kinds of food colorants in both complex food samples and urine with recoveries of 91–119%. The satisfactory detection results suggest that the facile preparation strategy of AgNC substrates will be widely used in SERS-based POCT to promote the development of food safety and on-site healthcare.

A 3D homogeneous microreactor with high mixing intensity at wide Re range for MOF preparation and POCT application

Zhongjian Tan,Huanhuan Shi,Yun Zheng,Yu Cao

Most microreactors can only work under high Re or extremely low Re conditions to obtain enough mixing efficiency, the inefficient reaction intensity undoubtedly limits its application scenarios. In this study, a novel 3D homogeneous microreactor (3D-HM) was reported that demonstrate a strong mixing intensity over a wide range of Re conditions. The 3D geometries of shunt and lateral secondary flow structure were optimized in details, and then were added to the Zigzag mixing channel to promote mixing index (MI) (over 94 % from Re  = 0–10, and more than 98 % for Re  > 10). Compared with the traditional T-shaped, Split and Recombine micromixer, a shorter mixing distance ( L mix ) and mixing time ( τ mix ) were needed to realize fully mixing. The 3D-HM was used for the preparation of Zeolitic imidazolate framework (ZIF-8), the particle size, yield and drug loading capacity of ZIF-8 prepared using the 3D-HM under a milder Re conditions were better than those prepared by traditional and existing microfluidic methods under a harsh Re conditions. The promotion to diffusion mixing with 3D-HM channel was then verified using the enzymatic catalytic reaction driven by syringe pump at low Re and capillary pump, respectively. Enzyme and substrates in the laminar flow in the 3D-HM are more likely to bind each other than that in the T-shaped diffusion mixing. In addition, the laminar diffusion mixing process was scaled up by microfluidic channel design, so that the results of enzymic catalytic reaction was visualized for Point-of-care Testing (POCT) application. The MI, L mix and τ mix directly determine the homogeneous microreaction intensity. The proposal of the 3D-HM may provide more possibilities for the application of micromixing-based reactors.

Methylene-Blue-Encapsulated Metal-Organic-Framework-Based Electrochemical POCT Platform for Multiple Detection of Heavy Metal Ions in Milk

Huiwen Xiong,Pintao Li,Fei Cun,Hui Chen,Jilie Kong

Considering the high risk of heavy metal ions (HMIs) transferring through the food chain and accumulating in milk, a flexible and facile point-of-care testing (POCT) platform is urgently needed for the accurate, sensitive, and highly selective on-site quantification of multiple HMIs in milk. In this work, a cost-effective disk with six screen-printed electrodes (SPEs) was designed for hand-held electrochemical detection. Metal organic frameworks (MOFs) were adopted to amplify and enhance the electrochemical signals of methylene blue (MB). Using differential pulse voltammetry (DPV) methods, low limits of detection for four HMIs (Cd2+, 0.039 ppb; Hg2+, 0.039 ppb; Pb2+, 0.073 ppb; and As3+, 0.022 ppb) were achieved within four minutes. Moreover, the quantitative POCT system was applied to milk samples. The advantages of low cost, ease of on-site implementation, fast response, and accuracy allow for the POCT platform to be used in practical monitoring applications for the quantitation of multiple HMIs in milk samples. Keywords:heavy metal ions;point-of-care testing;electrochemical detection;metal organic frameworks;on-site

One-Pot Rlock-Mediated CRISPR/Cas12a-Driven RCA Cycle for Rapid and High-Sensitive APE1 Detection

Junzhuo Shan,Yixiao Sheng,Lun Luo,Wenhai Wang,Xinjiang Liu,Yi Ma,Jufang Wang

Apurinic/apyrimidinic endonuclease 1 (APE1) is a key enzyme involved in DNA repair and cellular redox regulation, and is frequently overexpressed in tumor cells. This highlights the urgent need for a rapid and high-sensitive point-of-care testing (POCT) strategy for APE1 to facilitate early cancer diagnosis. Rolling circle amplification (RCA) is a widely used isothermal DNA amplification method; however, its application in APE1 detection remains rare. Here, we introduce a versatile RCA-Lock (Rlock) conversion platform that enables the transformation of RCA-based nucleic acid detection technologies into APE1-responsive assays. Building upon this platform, we further developed a novel POCT method for APE1 detection─Rlock-mediated, CRISPR/Cas12a-driven RCA cycle (RCRE)─which, for the first time, integrates CRISPR/Cas12a with RCA into a one-pot APE1 detection system. The RCRE assay achieves a limit of detection of 8.86 × 10–4 U/mL within 30 min, while requiring minimal equipment, low cost, and no complex handling procedures. This Rlock-based conversion strategy represents a transformative advance in the field of APE1 diagnostics and offers conceptual inspiration for the design of programmable nucleic acid–based biosensors. The resulting RCRE assay significantly broadens the technological landscape for early cancer detection and paves the way for future clinical translation.

Sodium alginate hydrogelation mediated paper-based POCT sensor for visual distance reading and smartphone-assisted colorimetric dual-signal determination of L-lactate

Wenjuan Wang,Danrong Chen,Yujiao Cai,Zijing Liu,Hongfen Yang,Hongbin Xie,Jinquan Liu,Shengyuan Yang

Herein, we presented a paper-based POCT sensor based on lactate dehydrogenase-mediated alginate gelation combined with visual distance reading and smartphone-assisted colorimetric analysis dual-signal to determine the concentrations of L-lactate in yogurt samples. In this research, L-lactate was transformed to pyruvate by lactate dehydrogenase. The pyruvate then triggered the gelation of the sol mixture, increasing the viscosity (ηs) of the mixture, which showed a decrease in diffusion diameter on the paper-based sensor. Also, protons from pyruvate accelerated the degradation of Rhodamine B, causing color fading of the mixture, which could be analyzed by a RGB analysis application software. Under the optimal experimental conditions, the linear ranges of the visual distance reading and smartphone-assisted colorimetric analysis were 0.1-15 μM for the former and 0.3-15 μM for the latter, and the detection limits were 0.03 μM and 0.07 μM, respectively. As a proof-of-concept application, we then exploited the paper-based sensor to determine the concentrations of L-lactate in yogurt samples. The results from the dual-signal paper-based sensor are consistent with the ones from HPLC analysis. In short, this study developed a simple, convenient, cost-effective and feasible method for the quantitative detection of L-lactate in real samples.

A digitalized isothermal nucleic acid testing platform based on a pump-free open droplet array microfluidic chip

Ping Mao,Lei Cao,Zedong Li,Minli You,Bin Gao,Xianghong Xie,Zhenrui Xue,Ping Peng,Chunyan Yao,Feng Xu

Digital PCR has shown great potential for quantitative nucleic acid testing (NAT), but most existing platforms are dependent on large auxiliary equipment (e.g., vacuum pump, amplification instrument, fluorescence microscope) to achieve target dispersion, amplification, signal capture and result analysis. Such complex, expensive and bulky NAT platforms have limited their applications in resource-limited areas, especially for point-of-care testing (POCT). In this work, we designed a digital isothermal NAT platform based on a pump-free open droplet array microfluidic chip. A pump-free microfluidic chip was developed based on an open microdroplet array in the form of thousands of independent microdroplets for spontaneous sample dispersion, without the need for external power. Combined with a handheld fluorescent signal reader based on a smartphone, this digital NAT platform can accurately quantify as low as 1 copy per μL of λDNA. Therefore, our integrated NAT platform, as a potable, robust and low-cost tool for highly accurate NA quantitative analysis, holds great potential for POCT applications.

Integrated hand-held electrochemical sensor for multicomponent detection in urine

Jiang Liu,Wei Lu,Lu Zhang,Jiao Yang,Zhong-Ping Yao,Yongcheng He,Yingchun Li

Electrochemical sensors have shown great advantage and application potential in point-of-care testing (POCT) related scenarios. However, some fatal problems plague its widespread utilization, which include the susceptibility of sensors to interference in real samples (e.g. pH), the contradiction between the limited objects detectable for most sensors and the requirement of multi-target analysis in most cases, and the complicated procedures in sensor preparation as well as in routine use. This paper contributed a tip-like electrochemical sensor prototype. By integrated with a commercial pipettor, it fulfilled semi-automatic assay procedure of sampling, detection and rinsing, thus saving operational time and manual work. The tip sensor owns the property of simple fabrication and is free from any modification of extra bio/chem materials. Moreover, built on multiple electrochemical signal outputs including open circuit potential , peak current and potential of specific electrochemical reaction , this work established a novel multi-component sensing strategy, wherein detection of uric acid (UA), urea and pH in urine samples was realized by using one single working electrode . The detection range for the above targets is 5.0~600 μM for UA, 4.0~8.0 for pH and 0.5~7.0 mM for urea with the detection limits ( S / N  = 3) of 0.05 μM for UA and 5.4 μM for urea, and the sensitivity of pH assay is 73 mV/pH. Notably, as variation of sample pH has impact on electrochemical analysis , the pH-related parameter was introduced for calibration to diminish such interference. The developed tip sensor and the novel sensing strategy may open a new window for electrochemical technology and broaden its application in POCT.

A smartphone-assisted photoelectrochemical POCT method via Z-scheme CuCo2S4/Fe3O4 for simultaneously detecting co-contamination with microplastics in food and the environment

Jing Zeng,Wenqin Wu,Xiao Chen,Shenling Wang,Huimin Wu,Ahmed A. El-Kady,Amnart Poapolathep,Alejandro Cifuentes,Elena Ibañez,Peiwu Li,Zhaowei Zhang

As emerging contaminants, microplastics threaten food and environmental safety. Dibutyl phthalate (DBP, released from microplastics) and benzo[ a ]pyrene (BaP, adsorbed on microplastics) coexisted in food and the environment, harming human health, requesting a sensitive and simultaneous testing method to monitor. To address current sensitivity, simultaneousness, and on-site portability challenges during dual targets in complex matrixes, CuCo 2 S 4 /Fe 3 O 4 nanoflower was designed to develop a smartphone-assisted photoelectrochemical point-of-care test (PEC POCT). The carrier transfer mechanism in CuCo 2 S 4 /Fe 3 O 4 was proven via density functional theory calculation. Under optimal conditions, the PEC POCT showed low detection limits of 0.126, and 0.132 pg/mL, wide linearity of 0.001–500, and 0.0005–50 ng/mL for DBP and BaP, respectively. The smartphone-assisted PEC POCT demonstrated satisfied recoveries (80.00%–119.63%) in real samples. Coherent results were recorded by comparing the PEC POCT to GC–MS (DBP) and HPLC (BaP). This novel method provides a practical platform for simultaneous POCT for food safety and environment monitoring.

Novel field amplification for sensitive colorimetric detection of microalbuminuria on a paper-based analytical device

Yu Cai,Ji-Cheng Niu,Xin-Li Du,Fang Fang,Zhi-Yong Wu

Field-amplified stacking (FAS) is a commonly used method for enhancing the sensitivity of charged species from low conductive media in capillary electrophoresis . FAS also showed significant sensitivity enhancement effect on a uniform paper fluidic channel by proper design of the electrolyte. In this paper, a novel method of introducing electric field gradient is proposed by geometry design of a 2D paper fluidic channel, and field amplification effect was successfully demonstrated with reduced requirement on the sample's conductivity . Sensitive colorimetric detection of microalbuminuria (MAU) from urine samples was demonstrated by mobile phone camera. Experimental results showed that, with active electric field motivation, up to 93.5% of the loaded protein probe could be effectively transferred and stacked into a narrow band on the newly designed paper fluidic channel. A limit of detection (LOD) of 6.5 mg‧L −1 HSA was achieved with a dynamic range of 10–300 mg‧L −1 (linear in the range of 10–100 mg‧L −1 , R 2  = 0.991). Combined with selective staining of albumin with bromophenol blue (BPB), the established method was applied to the detection of MAU from clinical urine samples, and consistent results with that of the clinical method were obtained. With this paper-based analytical device (PAD), MAU from highly conductive urine samples can be directly loaded and detected without any pretreatment . This method provides a way to develop highly sensitive point-of-care test (POCT) for rapid screening of some diseases.

The fabrication of quantum dots microgel based POCT microfluidic chip for simultaneously detecting multiple pathogenic heavy metal ions and its applications in actual water samples

Lianlian Wang,Yongfeng Cui,Yiduo Mi,Luyao Wang,Bing Han,Yadan Guo

Heavy metal ions pollution is one of the most threatened issues for human health and ecosystem balance. Herein, a promising microfluidic chip with microgel arrays for realizing the simultaneously point of care test to multiple pathogenic heavy metal ions was designed, which consisted of a sample perfusion area in the center (pipe width 200 μm), and four detection chambers (width 8000 μm, length 20000 μm) with microcolumn arrays (radius 100 μm, width 550 μm), all the heights were 100 μm. With simulation of the fluid flow, the flow rate of 5 mL/min made the microgel successfully patterning the precursor in microcolumn. By utilizing different kinds of quantum dots microgels, this chip could achieve specific and on-site quick detection for Pb 2+ , Co 2+ , Cu 2+ and Fe 3+ . Moreover, the gel composition had almost no effect on the performance of quantum dots and owned enough space after losing water to absorb the sample solution for reacting with the analytes. Furthermore, this POCT microfluidic chip was applied in the detection of actual water samples, and the fluorescence quenching of the chip was visible to naked eyes. It possessed the potential to achieve the ions quantitative determination. Therefore, this method realized the simultaneous detection of multiple pathogenic heavy meatal ions rapidly and availably and could further applied in actual water pollution samples with enormous advantages.

Rapid photothermal detection of foodborne pathogens based on the aggregation of MPBA-AuNPs induced by MPBA using a thermometer as a readout

Laibao Zheng,Wenjia Dong,Chaochuan Zheng,Yunqiu Shen,Ruolan Zhou,Zhenxing Wei,Zhixuan Chen,Yongliang Lou

Rapid and portable detection of foodborne pathogens is of great significance for food safety and public health. The colorimetric methods based on naked-eye have been demonstrated to be a suitable qualitative method for point-of-care testing (POCT). However, analytical instruments like a microplate reader must be needed for the quantitative assay. To overcome its limitation, we herein report a novel photothermal method for foodborne pathogens based on the photothermal effect of aggregated mercaptophenylboronic acid-functionalized AuNPs (MPBA-AuNPs) induced by MPBA to translate the colorimetric detection into a simple temperature measurement using thermometers as the readout. The aggregated AuNPs show higher photothermal conversion efficiency than well-separated AuNPs under 660 nm laser irradiation. In the presence of bacteria, MPBA-AuNPs will attach to the surface of bacteria and keep separated from aggregation induced by excess MPBA, resulting in a lower temperature increase under 660 nm laser irradiation. Using E. coli O157:H7 as a model target, a good linear relationship is observed between temperature increase and bacteria concentration from 1.00 × 10 5 -1.00 × 10 9 cfu mL −1 (R 2 = 0.9877) with a detection limit of 1.97 × 10 4 cfu mL −1 , which is three orders of magnitude lower than of the MPBA-AuNPs-based colorimetric assays . The proposed photothermal method provided a universal platform for rapid and portable detection of broad-spectrum bacteria strains in real samples.
