The co-occurrence of heavy metal contamination and biodegradable microplastic (polylactic acid, PLA) pollution poses increasing risks to terrestrial plant communities and soil functioning, yet species-specific responses to combined stress remain poorly understood. Cd and microplastics frequently co-occur in agricultural soils, where microplastics can alter cadmium mobility, bioavailability, and transport pathways, potentially modifying metal toxicity and plant stress responses compared with single-pollutant exposure. We investigated the responses of the invasive Bidens pilosa and the native Solanum nigrum grown in monoculture and mixed culture under combined cadmium (Cd) and biodegradable microplastic (PLA) stress by integrating plant growth, photosynthetic performance, oxidative physiology, and rhizosphere biochemical processes. Combined Cd–MP exposure markedly reduced plant growth, chlorophyll content (SPAD), photosystem II efficiency (Fv/Fm), nitrogen accumulation, biomass production, and rhizosphere enzyme activities associated with carbon, nitrogen, and phosphorus cycling. However, B. pilosa maintained greater physiological stability under stress, characterized by higher antioxidant enzyme activities (SOD, CAT, POD), lower reactive oxygen species (H 2 O 2 , O 2 ˙ − ) accumulation, and reduced lipid peroxidation (MDA), whereas S. nigrum exhibited stronger oxidative damage and functional impairment. Multivariate analyses further revealed that root antioxidant capacity was closely associated with rhizosphere microbial enzyme activity, suggesting a root-centered regulatory mechanism linking plant stress tolerance to soil functioning. Overall, the invasive species showed greater tolerance to combined contamination and maintained relatively higher rhizosphere functional activity than the native species, indicating that multi-pollutant stress may alter competitive interactions between invasive and native plants in contaminated environments.

Cutin and suberin in the rice seed coat serve as a lipid barrier that controls water entry into the seed. However, the molecular mechanisms underlying the permeability properties of seed coats remain largely unknown. In this study, we revealed that the rice EXTENSIN-like protein OsPEX1 influences seed germination by modulating water permeability in the seed coat. The germination rate of the gain-of-function pex1/pex1 mutant was lower than that of the wild-type seeds, although their seed vitality was comparable to that of the wild type. Morphological observations revealed that embryo growth in the pex1/pex1 mutant was inhibited. Further investigation indicated that water absorption in the seed coat around the micropyle was impaired during the early stage of germination. Integrated transcriptional and metabolic analyses suggested that the increased deposition of cutin and suberin may be associated with the overexpression of OsClo5 , a transcriptional repressor, in the mutant. It is proposed that abnormal deposition of cutin and suberin in the seed coat may be the major influencing factors contributing to the reduced impermeability of pex1/pex1 seeds. These results indicate that OsPEX1 plays a critical role in modulating water absorption in the seed coat during seed germination.

Nitrate serves not only as an essential nitrogen source but also as a signalling molecule to regulate plant growth and development. However, the mechanisms by which plants respond to nitrate to regulate root growth in rice remain largely unclear. In this study, we demonstrated that the overexpression of OsMADS27 significantly promoted root growth and nitrate absorption in response to nitrate supply, whereas RNA interference (RNAi) lines exhibited the opposite phenotype. OsMADS27 is predominantly expressed in the root, and the mutation of OsNAR2.1 impaired both the response rate and intensity of OsMADS27 under nitrate treatment. Notably, OsMADS27 is co-localised with OsNAR2.1, and the mutation of OsNAR2.1 affected the nuclear localisation of OsMADS27. Yeast two-hybrid (Y2H), bimolecular fluorescence complementation (BiFC) and co-immunoprecipitation (Co-IP) assays confirmed a direct protein interaction between OsMADS27 and OsNAR2.1. More importantly, chromatin immunoprecipitation followed by qPCR (ChIP-qPCR), dual-luciferase (Dual-LUC) and yeast one-hybrid (Y1H) assays demonstrated that OsMADS27 could directly regulate the expression of OsMADS57 to control root growth. Furthermore, OsMADS27 could also promote the expression of several nitrate transporter genes, including OsNRT1.2 , OsNRT2.1 , OsNRT2.3a and OsNRT2.4 , to regulate nitrate absorption in rice. Collectively, this study uncovers a novel pathway in which OsMADS27 promotes root growth in response to nitrate by directly regulating OsMADS57 expression in an OsNAR2.1-dependent manner. These findings will broaden our understanding and provide a novel regulatory network of OsMADS genes mediating root development through nitrate signalling in rice, which also has an important application value for molecular breeding to improve nitrogen use efficiency.

Plant cuticle as hydrophobic barrier covers almost all aerial plant organs. Herein the cuticular chemical components and the transpiration of various organs of Chinese flowering cabbage (CFC) and Chinese kale (CK) were comprehensively characterized. Numerous species- and organ-specific differences in morphological, chemical, and physiological levels were found. The various organs were relatively smooth in surface for CFC but glaucous with hollow tube- and plate-type crystals for CK. The chemical composition of cuticular waxes were very-long chain n -alkanes, ketones, secondary alcohols with a prominent carbon chain of C 29 in CK, primary alcohols dominated by C 26 , and aldehydes prominently C 30 in CFC. Cutin monomers accumulated with similar levels as waxes and were dominated by α , ω -dicarboxylic acids and fatty acids without added groups. The minimum water conductance differed considerably among species and various organs ranging between 8.9 × 10 −5 (CK leaf) and 3.7 × 10 −4 m s −1 (CFC leaf petiole). These differences in transpiration properties were proposed to be largely related to the cuticular chemicals in various organs and species. The presented results provide further insights to link the transpiration barrier functions with surface characteristics and cuticular chemicals.

The plant parasitic nematode Aphelenchoides besseyi is a major pest that poses serious threats to different vegetables and crop plants. In the present study, volatiles isolated from Bacillus spp. were utilized as green biocontrol agents to overcome nematodes. In in vitro experiment, Bacillus spp. GBSC56, SYST2, and FZB42 showed the strongest nematicidal activity with killing rates of 80.78%, 75.69%, and 60.45%, respectively, as compared with control. The selected synthetic volatile organic compounds (VOCs), namely albuterol, benzaldehyde (BDH), 1,2-benzisothiazol-3(2H)-one (1,2-HIT), dimethyl disulfide (DMDS), 2-undecanone (2-UD), and 1,3-propanediole (1,3-PD), exhibited strong nematicidal activity, with A. besseyi killing rate of 85.58%, 82.65%, 81.75%, 80.36%, 84.45%, and 82.36%, respectively, at 400 μg/mL. Microscopic analysis proved that the rapid mortality was due to the production of reactive oxygen species (ROS). Molecular docking attributed this ROS production to the nematicidal effect of synthetic VOCs on NADH DEHYDROGENASE SUBUNIT 2, which is known to play a critical role in the suppression of ROS in nematode models. In a greenhouse experiment, the Bacillus strains GBSC56, SYST2, and FZB42 and their synthetic VOCs significantly improved the physiological parameters in terms of growth promotion traits. In addition, selected genes related to growth promotion and defense genes showed a significant upregulation of their expression in rice seedlings treated with those synthetic VOCs. Overall, these findings revealed that the selected Bacillus strains and their synthetic VOCs possess high potential against A. besseyi . Moreover, this study also sheds new light on the mechanisms by which specific Bacillus nematicidal VOCs influence important genes involved in rice plant growth promotion and could effectively be used to suppress plant parasitic nematodes.

Rhizoctonia solani and Xanthomonas oryzae pv. oryzae ( Xoo ) are the two major diseases affecting the quality and quantity of rice production. In the current study, volatile organic compounds (VOCs) of Bacillus spp. were used as green biocontrol agents for plant diseases. In in vitro experiments, Bacillus spp. FZB42, NMTD17, and LLTC93-VOCs displayed strong antimicrobial volatile activity with inhibition rates of 76, 66, and 78% for R. solani and 78, 81, and 76% for Xoo , respectively, compared to control. The synthetic VOCs, namely Pentadecane (PDC), Benzaldehyde (BDH), 1,2-Benz isothiazol-3(2H)-one (1,2-BIT), and mixture (MIX) of VOCs showed high volatile activity with inhibition rates of 86, 86, 89, and 92% against R. solani and 81, 81, 82, and 86%, respectively, against Xoo as compared to control. In addition, the scanning and transmission electron microscopes (SEM and TEM) analyses were performed to examine the effect of Bacillus and synthetic VOC treatments on R. solani and Xoo morphology. The analysis revealed the deformed and irregularized morphology of R. solani mycelia and Xoo cells after VOC treatments. The microscopic analysis showed that the rapid inhibition was due to severe oxidative productions inside the R. solani mycelia and Xoo cells. By using molecular docking, it was determined that the synthetic VOCs entered the active binding site of trehalase and NADH dehydrogenase proteins, causing R. solani and Xoo cells to die prematurely and an accumulation of ROS. In the greenhouse experiment, FZB42, NMTD17, and LLTC93-VOCs significantly reduced the lesions of R. solani 8, 7, and 6 cm, and Xoo 7, 6, and 6 cm, respectively, then control. The synthetic VOCs demonstrated that the PDC, BDH, 1,2-BIT, and MIX-VOCs significantly reduced R. solani lesions on leaves 6, 6, 6, and 5 cm and Xoo 6, 5, 5, and 4 cm, respectively, as compared to control. Furthermore, plant defence-related genes and antioxidant enzymes were upregulated in rice plants. These findings provide novel mechanisms by which Bacillus antimicrobial VOCs control plant diseases.

In terms of ligand fishing, the amount and the relative activity recovery of enzymes immobilized on magnetic particles and nanoparticles are not preeminent. Therefore, the metal-organic framework (MOF) UiO-66-NH 2 was synthesized to immobilize the porcine pancreatic lipase (PPL) via precipitation-cross-linking, and the resulting novel biological matrices named PPL@MOF manifested high PPL loading capacity (98.31 mg/g) and relative activity recovery (104.4%). Moreover, the novel enzyme-MOF composite was applied to screen lipase inhibitors from Prunella vulgaris L. to enrich and improve the techniques of ligand fishing. As a result, 13 lipase ligands were obtained, and 12 compounds were determined by HPLC-Q-TOF-MS/MS. All of these ligands were further confirmed to be potential inhibitors through the verification of the activity assay and molecular docking . The proposed approach based on PPL@MOF was superior in terms of abundant protein loading capacity, high enzyme catalytic activity and easy preparation process. Taken together, our newly developed method provided a new platform for efficient discovering bioactive molecules from natural herbs.

This study developed two novel magnetic nanocomposites with immobilized porcine pancreatic lipase (PPL) for simultaneously removing patulin (PAT) and ochratoxin A (OTA)—two hazardous environmental contaminants—from fruit juices. All detoxification experiments were conducted in real fruit juice matrices (apple juice for PAT, grape juice for OTA) under ambient temperature (25 °C) with realistic contamination ranges. The results demonstrated MNP@UiO-66-(SH)₂@PPL achieved removal rates of 77.03% for PAT in apple juice and 45.03% for OTA in grape juice, while MNP@UiO-66-NH₂@PPL showed removal rates of 85.74% for OTA and 48.44% for PAT. LC-MS analysis indicated that PAT undergoes ring-opening to ascladiol, while OTA is hydrolyzed to phenylalanine and OTα. Molecular docking revealed that PPL spontaneously binds to both toxins via hydrogen bonding and hydrophobic interactions. CCK-8 assays confirmed low cytotoxicity of the materials and their degradation products, with no adverse impact on juice quality. The composites also exhibited magnetic recovery rates > 93%, enabling facile separation and recyclability. This work provides a safe, efficient, and reusable strategy for multi-mycotoxin decontamination with demonstrated environmental relevance.

In this paper, Fe 3 O 4 nanoparticles coated with mesoporous silica were prepared successfully, noted as Fe 3 O 4 at the mobile composition of matter No. 41 (MCM-41). Also, Fe 3 O 4 at MCM-41 was grafted by both 3-aminopropyltriethoxysilane (APTS) and 3-chloropropyltriethoxysilane (CPS), noted as Fe 3 O 4 at MCM-41/APTS and Fe 3 O 4 at MCM-41/CPS. The compounds were characterized by X-ray powder diffraction, scanning electron microscopy, transmission electron microscopy, Fourier transform infrared spectroscopy, vibrating sample magnetometry, thermogravimetry and N 2 adsorption/desorption. Then, the enzyme, porcine pancreas lipase (PPL), was immobilized onto these modified nanoparticles by covalent attachment, physical adsorption and cross-linking, noted as Fe 3 O 4 at MCM-41/CPS-PPL, Fe 3 O 4 at MCM-41-PPL and Fe 3 O 4 at MCM-41/APTS-PPL, respectively. The results showed that Fe 3 O 4 at MCM-41/CPS was the best nanomaterial for PPL immobilization, exhibiting enhanced immobilization efficiency (maximum 96%), maximum relative activity (up to 96%), high stability and reusability (83% 56 days and 86.7% ten cycles). Additionally, it offered some other advantages, such as easy recycling and reuse, complying with the trend of green chemistry. Therefore, Fe 3 O 4 at MCM-41/CPS in combination with a relevant method can be proposed for commercial applications.

Objectives . Preparation of a multifunctional drug-loaded phase-change nanoparticle (NP), pirfenidone perfluoropentane liposome NPs (PPL NPs), and combined with low-intensity focused ultrasound (LIFU) to influence epithelial mesenchymal transition (EMT) for hepatocellular carcinoma (HCC) by inhibiting the activity of activated Hepatic Stellate Cells (a-HSCs ). Methods . PPL NPs were prepared by the thin film dispersion method. The appearance, particle size, zeta potential, encapsulation efficiency, drug loading rate, drug release in vitro, and stability of PPL NPs were tested. The role of a-HSCs in HCC metastasis was studied by CCK-8, colony formation assay, apoptosis, cellular uptake assay, wound healing assay, and Transwell assay. Western blot was used to detect the related protein expression levels. In vitro and vivo, the acoustic droplet vaporization (ADV) of PPL NPs was tested at different times and LIFU intensities. Biosafety of the PPL NPs was assessed by measuring nude mouse body weight and hematoxylin and eosin (H&E) staining . Results. The results showed that the PPL NPs had good biosafety, with an average particle size of 346.6 ± 62.21 nm and an average zeta potential of -15.23 mV. When the LIFU power is 2.4 W/cm 2 , it can improve the permeability of cells, further promote the uptake of drugs by cells, and improve the toxicity of drugs. In vitro experiments showed that PPL NPs could inhibit the proliferation of a-HSCs cells, thereby affecting the metastasis of HCC, and were related to the TGF β -Smad2/3-Snail signaling pathway. Both in vivo and in vitro PPL NPs enhanced ultrasound imaging by LIFU-triggered ADV . Conclusion. The PPL NPs designed and prepared in this study combined with LIFU irradiation could significantly alter the EMT of HCC by inhibiting LX2. Clinically, PPL NPs will also be considered a promising contrast agent due to their ultrasound imaging capabilities.

Dioscoreae nipponica Makino ( D. nipponica ) as the rhizome of dioscoreaceae rich in steroidal saponins, has been reported to have the hypolipidemic effects etc. However, it is still unclear which exact active components are primary responsible for the beneficial effects. This study was conducted to fish out the lipase inhibitors from D. nipponica , and evaluate the inhibitory activity on porcine pancreatic lipase (PPL) through in vitro kinetic assay using p -nitrophenyl palmitate as substrate. Accordingly, the ethanolic extract was subjected to D101 macroporous resin purification for spectrophotometric screening, high performance liquid chromatography (HPLC) separation and structural characterization by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry. Through orlistat validation, the PPL inhibitory activity and IC 50 value of the extract were respectively 68.34 ± 1.47 % and 107.05 μg/mL under the optimized inhibition conditions. From 6 steroidal saponins identified, the inhibitory components named the protodioscin, protogracillin, dioscin and gracillin were fished out by grouping separation and HPLC analysis. Furthermore, dioscin and gracillin with the parent structure of diogenin were confirmed as the major inhibitors by virtue of stability tests based on transformation of protodioscin and protogracillin. Finally, the inhibitory mechanism of the major inhibitors toward PPL was further clarified by kinetic analysis and molecular docking analysis. The proposed method not only revealed the PPL inhibitory components in D. nipponica , but also provided an effective approach to hierarchical screening of PPL inhibitors from natural plants.
