Endoscopic submucosal dissection (ESD), a minimally invasive procedure, is constrained by the rapid dissipation of conventional submucosal cushions and the absence of sustained postoperative therapeutic support. Herein, we develop a multifunctional hydrogel integrating mucosal elevating, sustained local analgesia, wound protection, antibacterial and antioxidant capabilities for perioperative management of ESD. The hydrogel is formed via Schiff base reactions between carboxymethyl chitosan and oxidized dextran, incorporates methylene blue for intraoperative visualization, and integrates mesoporous silica nanoparticles loaded with Bupivacaine (BUP@MSNs) to enable local sustained-release analgesia. Targeting critical limitations of conventional mucosal elevating agents, the hydrogel rapidly forms a stable submucosal cushion through in situ gelation, enhancing ESD procedural efficiency and safety while enabling immediate wound sealing and protection. Under physiological conditions, the hydrogel exhibits excellent injectability, self-healing capacity, and tissue adhesion at the lesion site. It also exerts potent antioxidant and antibacterial activities, fulfilling requirements for perioperative microenvironment modulation. Furthermore, the hydrogel delivers early postoperative analgesia that persists for over 48 h, effectively covering the peak pain phase post-ESD. Comprehensive in vitro and in vivo safety evaluations confirm its favorable biocompatibility and tolerability. Collectively, the hydrogel represents an integrated, clinically operable, and cost-effective candidate for gastrointestinal endoscopic minimally invasive therapies.

Volumetric muscle loss (VML) is a severe condition in which the loss of skeletal muscle surpasses the body's intrinsic repair capabilities, leading to irreversible functional deficits and potential disability, with persistent inflammation and impaired myogenic differentiation. To address these challenges, a novel zinc-dihydromyricetin (Zn-DHM) nanozyme with superoxide dismutase (SOD)-like activity is developed, designed to neutralize excessive reactive oxygen species (ROS) and restore oxidative balance. Zn-DHM mitigates oxidative stress and promotes polarization of macrophages from the proinflammatory M1 phenotype to the anti-inflammatory M2 phenotype, thereby reducing chronic inflammation and creating a conducive environment for muscle repair. Further, Zn-DHM significantly enhances the myogenic differentiation of C 2 C 12 cells, accelerating wound healing processes. These studies confirm the biosafety and low toxicity of Zn-DHM. As per a murine tibialis anterior VML model, Zn-DHM effectively suppresses inflammation and markedly improves skeletal muscle repair outcomes. Single-cell RNA sequencing reveals that Zn-DHM treatment increases the expression of M2 macrophage markers and enhances the proliferation and differentiation capacity of muscle stem cells (MuSCs). In addition, intercellular communication analysis reveals interactions between MuSCs and macrophages in the Zn-DHM treatment group, suggesting that these interactions may drive tissue regeneration through the activation of the GAS and Notch signaling pathways.

Parkinson's disease (PD), a progressive neurodegenerative disorder, is marked by the degeneration of dopaminergic neurons. Emerging evidence highlights the critical involvement of neuronal ferroptosis and microglial pyroptosis in PD pathogenesis. In this study, we first characterized systemic inflammatory alterations in the peripheral blood of PD patients compared to healthy controls, revealing distinct pro-inflammatory signatures. Based on these findings, we engineered BV2 microglial membrane-camouflaged biomimetic multienzyme gallic acid-selenium nanoparticles (BM@GA-Se nanosomes) and evaluated their neuroprotective potential. In vitro and in vivo experiments demonstrated that BM@GA-Se nanosomes enhanced neuronal survival by simultaneously suppressing ferroptosis of neurons (via GPX4-mediated lipid peroxide reduction) and pyroptosis of microglia (through inhibition of the HSP90/NLRP3/Caspase-1 signaling pathway). These results position BM@GA-Se nanosome as a new dual-pathway targeting therapeutic strategy, offering a promising translational approach to mitigate PD progression by addressing interdependent inflammatory and oxidative mechanisms.

Diabetic wound healing poses a significant challenge in the biomedical domain. Inadequate angiogenesis and fibroblast dysfunction are two primary characteristics of diabetic microenvironment. Fibroblast dysfunction is characterized by excessive accumulation of reactive oxygen species (ROS) and fragmented mitochondria. Herein, we developed a microenvironment responsive zinc–gallic acid nanosheets (ZnGA NSs) for diabetic wound healing, which slowly release Zn 2+ and gallic acid (GA) in response to low pH and excessive ROS. The released Zn 2+ enhanced cell proliferation, migration, and endothelial tube formation. And the released GA activated antioxidant enzyme system via the KEAP1/NRF2 pathway and promoted mitochondrial fusion by upregulating MFN1/2 and OPA1, which ultimately reprograms dysfunctional fibroblast. The application of ZnGA NSs effectively facilitated wound healing in a rat diabetic model with full-thickness cutaneous defects. More importantly, skin wounds treated with ZnGA NSs exhibited enhanced angiogenesis, alleviated inflammation, and improved tissue regeneration. These findings highlight the potential of ZnGA NSs as a promising therapeutic modality for diabetic wound treatment.

The excess production of reactive oxygen species (ROS) will delay tooth extraction socket (TES) healing. In this study, we developed an injectable thermosensitive hydrogel (NBP@BP@CS) used to treat TES healing. The hydrogel formulation incorporated black phosphorus (BP) nanoflakes, recognized for their accelerated alveolar bone regeneration and ROS-scavenging properties, and dl-3-n-butylphthalide (NBP), a vasodilator aimed at enhancing angiogenesis. In vivo investigations strongly demonstrated that NBP@BP@CS improved TES healing due to antioxidation and promotion of alveolar bone regeneration by BP nanoflakes. The sustained release of NBP from the hydrogel promoted neovascularization and vascular remodeling. Our results demonstrated that the designed thermosensitive hydrogel provided great opportunity not only for ROS elimination but also for the promotion of osteogenesis and angiogenesis, reflecting the “three birds with one stone” concept, and has tremendous potential for rapid TES healing.

To obtain high-performance tissue-adhesive hydrogel embodying excellent mechanical integrity, a supramolecular hydrogel patch is fabricated through in situ copolymerization of a liquid-liquid phase separation precursor composed of self-complementary 2-2-ureido-4-pyrimidone-based monomer and acrylic acid coupled with subsequent corporation of bioactive epigallocatechin gallate. Remarkably, the prepared supramolecular hydrogel leverages hierarchical multi-strength hydrogen-bonds hinged strategy assisted by alkyl-based hydrophobic pockets, broadening the distribution of binding strength of physical junctions, striking a canonical balance between superb mechanical performance and robust adhesive capacity. Ultimately, the fabricated supramolecular hydrogel patch stands out as a high stretchability (1500 %), an excellent tensile strength (2.6 MPa), a superhigh toughness (12.6 MJ m −3 ), an instant and robust tissue adhesion strength (263.2 kPa for porcine skin), the considerable endurance under cyclic loading and reversible adhesion, a superior burst pressure tolerance (108 kPa) to those of commercially-available tissue sealants, and outstanding anti-swelling behavior. The resultant supramolecular hydrogel patch demonstrates the rapid hemorrhage control within 60 s in liver injury and efficient wound closure and healing effects with alleviated inflammation and reduced scarring in full-thickness skin incision, confirming its medical translation as a promising self-rescue tissue-adhesive patch for hemorrhage prevention and sutureless wound closure.

Lignin, as a natural polyphenol, displays anti-oxidant activity by trapping and binding free radicals through its free phenolic hydroxyl groups. However, the most accessible form, industrial lignins, generally has low phenolic hydroxyl content, which severely limits their application value and scenarios. Herein, we showed that potassium-glycerate deep eutectic solvent (PG-DES) treatment can be combined with laccase oxidation to afford prepared high antioxidant lignin nanoparticles (HA-LNPs) with notably improved anti-oxidant activities benefiting from both the enhanced phenolic hydroxyl content 170.8 % and reduced average particle size (59.0 nm). At concentrations as low as 60 μg/mL, HA-LNPs showed favorable effects in promoting collagen formation. When HA-LNPs were used as an active ingredient in the anti-aging mask formulation, the reactive oxygen species (ROS) scavenging activity of mask samples containing 0.4 % HA-LNPs reached 37.2 %. The data suggest great promise of HA-LNPs as a natural antioxidant for formulating in anti-aging skin care products.

The method of adding additives at once not only increases costs, but also poses a risk of excessive consumption. Therefore, the slow-release technology was employed for improving the utilization efficiency and edible safety of additives in this work. The poly (N-isopropylacrylamide) (PNIPAM) and polylactic acid (PLA) composite hydrogels with sheath-core structure were fabricated by in-situ spinning method. The mechanical bearing capacity of PNIPAM hydrogel was improved by controlling the structure of PLA electrospun fiber membrane. The results showed that the compression resistance of the samples coated with PLA was increased 931 % at 50 % deformation. In addition, the surface wettability of samples was studied to ensure the stable water permeability. The composite hydrogel was used for the sustained release of tea polyphenol (TP) antioxidant. The release effect of TP at different temperatures, and radical scavenging effect of 2, 2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2-Phenyl-4,4,5,5-tetraMethyliMidazoline-3-oxide-1-oxyl) (PTIO) were investigated. It was found that the release rate for TP could reach to 77.8 % at 30 °C. The radical clearance rates of ABTS and PTIO were 99.2 % and 46.6 %, respectively. This work provides effective ideas for the use of antioxidants in food preservation processes.

Microemulsions are extensively utilized in food, pharmaceuticals, and cosmetics due to their superior solubilizing capacity and protective capabilities. However, a longstanding critical issue with microemulsions is the quantitative relationship between the HLB value and the physicochemical properties. Herein, to quantify the effect of the HLB value on microemulsions, Juniperus chinensis Roxb. essential oil (CEO) microemulsions were fabricated. It is certified that HLB values between 8.5 and 11.5 correspond to the smallest biphasic regions, while values between 9.5 and 13.5 correspond to the largest monophasic regions. Besides, the phase evolution from lamellar structures to microemulsions was monitored by the Small-Angle X-ray Scattering (SAXS) patterns. The physicochemical properties of the resulting emulsions, e.g., polydispersity indices (PDI), zeta potential, thermal stability, viscosity, and conductivity, were also investigated. The quantification of the effect of the HLB value on the formation of microemulsions will provide significant quantitative and theoretical guidance to researchers and industry professionals.

This study explored the extraction of polyphenols from Juniperus chinensis Roxb. using natural deep eutectic solvents (NADES), emphasizing the role of hydrogen bonding strength in enhancing extraction efficiency. Choline chloride-fructose emerged as the most effective NADES after evaluating the correlation between hydroxyl stretching vibrations and polyphenol yield. Using the response surface methodology, the optimal extraction process (liquid to solid ratio 50.6 mL/g, extraction temperature 41.1 °C, extraction time 29.8 min, ultrasonic time 14.9 min) was determined to have a yield of 27.90 ± 0.14 mg/g, more than double that of water extraction. Choline chloride-fructose showed a more pronounced disruptive effect on plant powders. Additionally, compared to water extracts, the extracts using choline chloride-fructose exhibited higher antioxidant potential and quercitrin content. Overall, this study explores the correlation between hydrogen bond strength and extraction yield, while optimizing a green and efficient alternative process for polyphenol extraction from plants. These findings align with green chemistry principles, contributing to the development of sustainable and environmentally friendly extraction methods.

Alfalfa is rich in flavonoid compounds, which are known for their antioxidative and anti-inflammatory properties, suggesting therapeutic potential for alfalfa flavonoids (AF) in inflammation-related diseases. This study investigated the effects of AF on Salmonella -induced colitis, a severe inflammatory bowel disorder characterized by oxidative damage and inflammatory response. In vitro, antioxidant assays revealed AF's concentration-dependent radical scavenging, significantly reducing electron paramagnetic resonance (EPR) signals for HO • and O 2 •− by 42% and 54%, respectively. In vivo, AF treatment significantly mitigated body weight (BW) loss by 6%, increased colon length by 11%, and reduced liver and spleen weights by 19% and 81%, respectively, compared to the colitis group. Mechanistically, AF suppressed inflammation by downregulating the Toll-like receptor 4 (TLR4)/IκB/nuclear factor-κB (NF-κB)/cyclooxygenase-2 (COX-2) pathway and inhibiting nucleotide-binding domain-like receptor protein 3 (NLRP3) inflammasome activation, thereby lowering levels of pro-inflammatory cytokines (tumor necrosis factor alpha [TNF-α], interleukin 6 [IL-6], interleukin 1 beta [IL-1β]). Concurrently, AF enhanced antioxidant defense via the Kelch-like ECH-associated protein 1 (Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2) pathway, reducing reactive oxygen species (ROS) and malondialdehyde (MDA) levels while increasing catalase (CAT) (69%), glutathione peroxidase (GPx) (83%), and superoxide dismutase (SOD) (21%) activities. Moreover, AF preserved epithelial and mucosal barriers by reducing apoptosis and upregulating tight junction proteins (Claudin1, ZO-1, E-cadherin) and goblet cell marker Ulex europaeus (Gorse) Agglutinin I (UEA-1). Microbiota analysis revealed that AF significantly enriched beneficial bacteria, including Akkermansia , Oscillibacter , and butyrate-producing taxa, thereby counteracting Salmonella -induced dysbiosis. Furthermore, AF restored the disrupted profile of short-chain fatty acids (SCFAs), strengthening the relationship between symbiotic microbiota and mucosal defense. Overall, AF exerted multifaced protection against Salmonella -induced colitis by alleviating oxidative stress, stabilizing intestinal homeostasis, and thus attenuating inflammation. These findings make AF a promising phytopharmaceutical for the prevention and treatment of inflammatory diseases.

Graphical The conjugation of a polyphenolic antioxidant with a small peptide generates a self-assembled nanodrug, which suppresses ocular inflammation by attenuating oxidative stress in mitochondria after escaping from lysosome. As an investigation of anti-inflammation by the mitochondrial localization of antioxidant, this work establishes a foundation by regulating cellular redox balance for developing drug delivery systems in the treatment of ocular disorders. External environments ( e. g ., pollutants, irritants, ultraviolet radiation, etc ) probably activate oxidative stress on the ocular surface, further leading to inflammatory responses and cellular apoptosis. For treating ophthalmic diseases, one of the strategies is to regulate oxidative stress through antioxidants. Here we conjugate a polyphenolic antioxidant drug ( i. e ., caffeic acid) with a small peptide of protein tag to generate a peptide-drug conjugate as a nanodrug. With a self-assembled ability to form nanoparticles, the nanodrug mainly enters human corneal epithelial cells (HCEC) by caveolin-mediated endocytosis, succeeds lysosomal escape, and achieves mitochondrial localization of caffeic acid. Revealed by free radical scavenging experiments, the nanodrug shows considerable antioxidant capacities. In mouse leukemia cells of monocyte macrophage (RAW 264.7) induced by lipopolysaccharide (LPS), the nanodrug inhibits various pro-inflammatory cytokines ( e. g ., NO, IL-6, and TNF-α) by up-regulating the expression of anti-apoptotic protein ( e. g ., Bcl-2). As an investigation of alleviating oxidative stress by the mitochondrial localization of antioxidant, this work may establish an experimental foundation for the regulation of cellular redox balance, as well as provide different insights for the clinical development of antioxidant drug delivery systems in the treatment of ocular disease.
