Yongqiang Jiang,Meiting Zhang,Mingxuan Liu,Zhilian Su,Yuqian Pu,Wen Li,Hong Shao,Peng Shi,Rong Zhang,Ling Zhao,Yumeng Wei

Introduction FTA is a new FT derivative developed by our team by modifying FT with aspirin, and it has been shown to boost anti-breast cancer activity. Methods To significantly enhance the anti-breast cancer efficacy and biosafety of FTA, this study employed Poloxamer F127 and Poloxamer F68 to construct a composite carrier system, successfully prepared FTA-loaded liposomes (FTA-LN) and their thermosensitive gel formulation (FTA-LN-TSG), with systematic in vitro and in vivo assessments performed. Results Simultaneously, FTA-LN exhibited an average particle size of 131.03 ± 4.35 nm, polydispersity index (PDI) of 0.265 ± 0.003, zeta potential of −21.46 ± 2.49 mV, and encapsulation efficiency of 94.55 ± 1.10%. FTA-LN followed first-order kinetics, with a cumulative release rate of 68.48% within 4 hours. Conversely, FTA-LN-TSG displayed sustained release characteristics consistent with Higuchi kinetics, reaching a cumulative release of 52.56% at 8 hour. In vitro efficacy evaluation revealed that FTA-LN-TSG exhibited significantly superior inhibitory effects against breast cancer cells compared to FTA (24 h and 48 h inhibition rates: 2.72- and 5.02-fold of FTA; cell migration rate: 37% of FTA). Furthermore, FTA-LN-TSG demonstrated significant differences in inducing apoptosis and cell cycle arrest (Total apoptosis rate of FTA: 25.74, total apoptosis rate of FTA-LN-TSG: 48.84%). Pharmacokinetic results indicated that FTA-LN-TSG exhibited a 3.5-fold prolongation in half-life (t1/2), and a 6-fold extension in time to peak concentration (Tmax) compared with FTA group. In tumor model mice, The FTA-LN-TSG injected beside the tumor was achieved a tumor inhibition rate of 64.42 ± 5.60% (1.82-fold of the FTA group, 1.45-fold of the S-1 group) with favorable biosafety. Conclusion In summary, as a novel drug delivery system, FTA-LN-TSG may represent a promising option for breast cancer therapy.

Triethanolamine-modified poloxamer temperature-sensitive hair care gel for intelligent controlled release of sodium thioglycolat

Wenrui Zheng,Yue Lei,Yuheng Li,Haiyan Gao,Liangliang Lin,Hujun Xu

A triethanolamine-modified poloxamer temperature-sensitive hair care gel was formulated in order to reduce the adverse health effects associated with sodium thioglycolate (TGA(Na)), the active component in hair treatment agents, while maintaining excellent efficacy. It was found that TEA-3 gelled at 32 °C with a triethanolamine concentration of 3 %. This formulation exhibited a reversible phase transition, transitioning to a solid state upon contact with the scalp (>32 °C) and reverting to a liquid state when in contact with hair (<32 °C). The release behavior of TGA(Na) from the gel was assessed by both in vitro release studies and transdermal experiments. In addition, it was observed that the triethanolamine-modified poloxamer gel exhibited improved hair care efficiency compared to conventional hair care agents. These results suggest that the triethanolamine-modified poloxamer gel is a promising alternative to conventional hair care products, offering improved efficacy while minimizing potential adverse health effects.

Amino-functionalized poloxamer-hyaluronate in situ gels for sustained delivery of timolol maleate in glaucoma therapy

Xiaoxuan Ji,Ying Bao,Zhen Zhang,Yujing Huang,Haibing He,Guoqing Zhang,Hongfei Liu

Poloxamer-based in situ gels face limitations in ocular drug delivery due to weak mucoadhesion and low mechanical strength. Herein, we developed an amino-functionalized poloxamer-hyaluronate graft copolymer (Am-P407-HA) to overcome these challenges. Comprehensive characterizations confirmed successful synthesis of copolymer, which interacted with mucin to enhance the mucoadhesion of poloxamer. Using timolol maleate (TM) as a model drug, an ophthalmic in situ gel (TM@Am-P407-HA) was developed. The thermosensitive gel demonstrated sustained release properties (93.07 % cumulative release over 10 h) following Fickian diffusion kinetics. In vivo fluorescence tracking indicated that the TM@Am-P407-HA prolonged the TM residence time on the ocular surface. Pharmacokinetic study of TM@Am-P407-HA revealed 4.46-fold and 2.29-fold increases in AUC 0-t for tear fluid and aqueous humor, respectively, along with 3.30-fold and 2.34-fold prolongations in residence times compared to conventional eye drops. Ocular hypertension models showed TM@Am-P407-HA provided smoother pressure reduction curve with a prolonged duration of efficacy. Moreover, biosafety evaluations confirmed excellent biocompatibility of TM@Am-P407-HA through cytotoxicity, Draize irritation, and histopathological studies. This novel graft copolymer system represents a significant advancement in ocular drug delivery, offering enhanced bioavailability and reduced dosing frequency for glaucoma therapy

Development of Poloxamer Hydrogels Containing Antibacterial Guanidine-Based Polymers for Healing of Full-Thickness Skin Wound

Peipei Feng,Haofeng Qiu,Yang Luo,Jiaying Hu,Yuhao Cao,Qian Pang,Xianbo Mou,Ruixia Hou,Wenjia Hou,Yabin Zhu

A series of hydrogels containing guanidine-based polymers using a poloxamer as the matrix were prepared to provide novel wound dressings with antibacterial and repairing-promotion properties for skin wounds. Herein, we developed a series of antibacterial hydrogels, the cationic guanidine-based polymer polyhexamethylene guanidine hydrochloride (PHMG) with poloxamer aqueous solution (12%, w/w) simplified as PHMGP, chitosan (CS)-cross-linked PHMG (referred to as PHMC) with poloxamer aqueous solution simplified as PHMCP, and hyaluronic acid (HA)-modified PHMG (referred to as PHMH) with poloxamer aqueous solution simplified as PHMHP, for enhancing full-thickness skin wound healing. The characterizations, antimicrobial activity, cytotoxicity, and in vivo full-thickness wound-healing capability of these hydrogels were analyzed and evaluated. The results show that though PHMGP possesses great bactericide properties, its cytotoxicity is too strong to support skin regeneration. However, after modified with CS or HA, PHMCP and PHMHP showed good biocompatibility and antimicrobial properties against Gram-positive and Gram-negative bacteria that are commonly present in injured skin. Both PHMCP and PHMHP hydrogels exhibited upgraded wound-healing efficiency in full-thickness skin defects, characterized by a shorter wound closure time, faster re-regeneration, and the earlier formation of skin appendages, compared with those of control or pure poloxamer treatments. Their biological mechanism was detected. Both PHMCP and PHMHP can regulate the related biofactors during the skin repair process such as interleukin-1β (IL-1β), interleukin-6 (IL-6), transforming growth factor beta-1(TGF-β1), alpha-smooth muscle actin (α-SMA), and vascular endothelial growth factor, to promote wound healing with less serious scarring. In short, hydrogels with excellent capabilities to inhibit microorganism infection and promote wound healing were developed, which will shed light on designing and producing wound dressings with promising applications in future.

A Panax notoginseng saponins-loaded carboxymethyl chitosan/oxidized sodium alginate/poloxamer hydrogel for intrauterine adhesions therapy

Haoting Niu,Shuo Tang,Moyong Li,Lilu Guo,Lulu Qin,Liuyun Jiang,Anji Chen,Xiang Hu

A novel multifunctional double-network hydrogel composed of carboxymethyl chitosan-oxidized sodium alginate-poloxamer (PF127) loaded with Panax notoginseng saponins (OSA/CM/PF127@PNS) was developed for intrauterine adhesion therapy. FT-IR spectroscopy confirmed acetalation crosslinking between OSA and CMC, as evidenced by characteristic imine peak (1636 cm⁻ 1 ). SEM observations revealed the hydrogel possessed a porous microstructure, with robust self-healing capability validated (by rheological cyclic strain tests), a maximum swelling ratio of 1155%, and sustained PNS release reaching 52.2% at 24 h. In vitro degradation studies indicated the stability under physiological pH (7.4) was enhanced due to the dual-crosslinked network. Moreover, the hydrogel exhibited potent antibacterial activity (> 90% inhibition against E. coli and S. aureus ), antioxidant capacity, and excellent hemocompatibility (< 5% hemolysis). The hydrogel showed exceptional biocompatibility in cellular assays. Notably, PNS loading significantly suppressed pro-inflammatory cytokines (TNF- α , IL-1 β , IL-6) at the mRNA level and inhibited NF- κ B signaling at the protein level. Furthermore, it promoted angiogenesis in vitro, evidenced by upregulated angiogenesis-related proteins. Therapeutically, the hydrogel significantly accelerated wound healing in a murine model (> 90% closure by day 9) and effectively promoted endometrial regeneration in a rat IUA model. This work provides a novel and promising strategy based on natural polymer-based hydrogels, demonstrating its potential for the treatment of intrauterine adhesions and regenerative medicine.

Injectable thermosensitive hydrogel based on hyaluronic acid and poloxamer for sustained bupivacaine release and prolonged analgesia

Junhui Yang,Qien Zhao,Bo Lu,Yingying Lv,Wenting Jiang,Xiaowei Chen,Shengyu Zhang,Wei Zhao,Lei Jiang,Jiantao Zhang

Local anesthetics are commonly used for postoperative pain relief but have limited effectiveness due to their short half-life and low molecular weight. Various sustained-release systems have been explored but face challenges such as low encapsulation efficiency in liposomes, rapid degradation and limited release durations of hydrogel, and difficulties with injectability in combined microsphere/hydrogel. This study presents a double cross-linked hydrogel (Bup/PO) made from aminated poloxamer 407 (P407) and oxidized hyaluronic acid (OHA), cross-linked via oxime bonds and hydrophobic interactions. Rheological analysis showed that the hydrogel exhibited temperature sensitivity, with a storage modulus of up to 10,000 Pa at 37 °C, significantly higher than pure P407. In vitro, the hydrogel provided sustained bupivacaine release for up to 8 days. In vivo, a rat sciatic nerve block model demonstrated that the Bup/PO hydrogel significantly extended the mechanical pain threshold for 32 h and thermal pain threshold for 48 h. The hydrogel was fully biodegradable within 21 days, with mild reversible inflammation and no neurotoxicity. This study introduces a double-cross-linking mechanism that improves hydrogel stability, mechanical properties, and drug release, offering a promising solution for long-term postoperative pain management and regional anesthesia.

Alginate-poloxamer/silk fibroin hydrogels with covalently and physically cross-linked networks for cartilage tissue engineering

Jiaoyan Liu,Qing Fang,Hui Lin,Xiaofeng Yu,Hong Zheng,Ying Wan

Poloxamer was grafted onto alginate and the optimally synthesized alginate-poloxamer (ALG-POL) copolymer was combined with silk fibroin (SF) to produce thermosensitive ALG-POL/SF hydrogels with covalently and physically crosslinked networks. The formulated ALG-POL/SF gels were found to be injectable with sol-gel transitions near physiological temperature and pH. Rheological measurements showed that some ALG-POL/SF gels had their elastic modulus of around 5 kPa or higher with large ratio of elastic modulus to viscous modulus, indicative of their mechanically strong feature. The achieved ALG-POL/SF gels exhibited concurrent enhancement in strength and elasticity when compared to the gels built with either ALG-POL or SF alone. The microscopic insight into dry ALG-POL/SF gels validated that they were highly porous with well-interconnected pore characteristics. These ALG-POL/SF gels showed abilities to support the in-growth of seeded chondrocytes while effectively maintaining their chondrogenic phenotype. Results suggest promising attributes of ALG-POL/SF gels as alternative biomaterial for cartilage tissue engineering.

Weakening of the Transdermal Effect of p-Phenylenediamine Pigments by the Temperature-Sensitive Poloxamer Sodium Alginate Gel

Haiyan Gao,Yue Zhou,Jianguo Wang,Hujun Xu

A temperature-sensitive hair dye was prepared with a poloxamer-sodium alginate mixture. When the hair dye was applied to the hair (< 32 °C), it retained its flow dynamics. The hair dye was able to undergo a phase change and formed a solid as soon as it touched the scalp (32°C), which reduced the transdermal amount of p -phenylenediamine (PPD) and also adverse effects and toxicity. Compared to a conventional hair dye, the temperature-sensitive hair dye achieved a reduction of the transdermal amount of p -phenylenediamine by almost 65% at different concentrations. At the same time, the colouring effect was not significantly reduced. The poloxamer alginate hydrogel can be used as a novel colouring material to reduce the harm of the dye intermediate to the human body when colouring hair.

Study on an Injectable Chitosan–Lignin/Poloxamer Hydrogel Loaded with Platelet-Rich Plasma for Intrauterine Adhesion Treatment

Zhipeng Yu,Yang Min,Qi Ouyang,Yuting Fu,Ying Mao,Shuanglin Xiang,Xiang Hu,Liuyun Jiang

It is a great challenge to obtain an ideal hydrogel for the clinical treatment of intrauterine adhesion (IUA) disease. Here, a novel injectable chitosan–lignin/poloxamer hydrogel loaded with platelet-rich plasma (CL-PF127@PRP) was prepared by self-assembly at room temperature. Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), rheological analysis, and injectable writing were used to characterize the structure of the hydrogel. The results confirmed that the amino group of chitosan and the sulfonic group of sodium lignosulfonate were ionic-crosslinked by electrostatic attraction, which stabilized the three-dimensional structure of the PF127 hydrogel loaded with PRP, and PRP made the porous structure gradually become tight. Moreover, the CL-PF127@PRP hydrogel displayed good injectability and a solid state. The soaking experiment showed that the CL-PF127@PRP hydrogel had suitable degradation at pH = 7 and a good PRP release rate (PRP release 70% at 96 h). Cell experiments in vitro demonstrated that the CL-PF127@PRP hydrogel possessed good biocompatibility, an anti-inflammatory function, and pro-angiogenic activity. Furthermore, an animal experiment of skin wound and IUA confirmed that the skin wound closure rate of the CL-PF127@PRP hydrogel was over 50% on the seventh day. PRP improved the thickness of the endometrium and uterus receptivity, suggesting that the CL-PF127@PRP hydrogel offers great promise for the clinical treatment of IUA.

Hydrochromic and piezochromic dual-responsive optical film derived from poloxamer and ethyl cellulose for visual fingerprints identification

Yuqian Guo,Xianhui An,Xueren Qian

Developing new materials that could identify fingerprint using the naked eye and observe the level 3 microscopic details is challenging. Here, we designed a novel hydrochromic and piezochromic dual-responsive optical film, which achieved the visual transparency transition. The performances of hydrochromic and piezochromic responses from high transparency to opaque whiteness were attributed to the introduction of poloxamer. The hygroscopic swelling of the disordered micelles led to light scattering, causing the hydrochromic response. The piezochromic response may be ascribed to the microcracks in the fragments of poloxamer crystals, which changed the refractive index of light. The fascinating combination of hydrochromic and piezochromic response was effectively applied in fingerprint identification. Hydrochromic response accurately recognized sweat pores, and piezochromic response could gradually reveal the ridges and valleys according to the different color of imprinted fingerprints. The film could identify fake fingerprints based on the differences in sweat pores between fake fingerprints and living fingers. More importantly, the film could easily detected not only the clear ridges but also the detailed sweat pores using the naked eye, indicating that the film has profound research significance in fingerprint analysis and liveness fingerprint detection.

Enhancement in bioavailability of ketorolac tromethamine via intranasal in situ hydrogel based on poloxamer 407 and carrageenan

Chenxi Li,Chunyan Li,Zheshuo Liu,Qiuhong Li,Xueying Yan,Yu Liu,Weiyue Lu

The objective of this study was to construct a new in situ gel system based on the combination of poloxamer 407 and carrageenan (carrageenan–poloxamer 407 hydrogel, CPH) for intranasal delivery of ketorolac tromethamine . CPH showed potassium ion concentration - dependent erosion characteristics which ensured slow erosion in aqueous environment containing potassium ion at the physiological level. Loading with ketorolac tromethamine influenced erosion, drug release and thermosensitive properties of CPH. CPH containing 15% ketorolac tromethamine showed suitable gelation temperature (near 35   °C) and in vitro sustained release profiles. Pharmacokinetic study of intranasal CPH containing 15% ketorolac tromethamine in rats demonstrated enhanced absolute bioavailability (68.8   ±   23.3%) and prolonged mean residence time (8.8   ±   3.5   h) in comparison with the intranasal solution group (24.8   ±   13.8%, 3.9   ±   0.6   h). Nasal ciliotoxicity evaluation on an in situ toad palate model preliminarily showed the safety of CPH for intranasal use. All results suggested the potential of CPH as a new sustained - release platform for the intranasal delivery of ketorolac tromethamine.

Enhancement of in vitro dissolution and in vivo performance/oral absorption of FEB-poloxamer-TPGS solid dispersion

Xiaoxia Sheng,Jingjing Tang,Jiayin Bao,Xiangjun Shi,Weike Su

In the present study, a febuxostat (FEB) solid dispersion was prepared via the solvent evaporation method, and the dispersion's solubility, dissolution rate and oral bioavailability were investigated. To select the appropriate carriers for the FEB solid dispersion, series of preliminary trials were performed on d-α-tocopherol polyethylene glycol 1000 succinate (TPGS), HPMC, PEG4000, PEG6000, Eudragital, PVA, PVP K30, poloxamer188 and poloxamer407. Based on the preliminary trials, TPGS and poloxamer188 were selected as the carriers for the FEB solid dispersion preparation. In addition, a mixture of FEB:poloxamer188:TPGS (1:5:1) exhibited outstanding characteristics; its saturation solubility was 7.31 times (in water), 17.67 times (in pH 1.2), 17.22 times (in pH 4.5), 3.60 times (in pH 6.0) and 1.99 times (in pH 6.8) higher than that of pure crystalline FEB. In an in vitro drug dissolution study, the mixture exhibited a drug release rate that was 1.94 times that of the FEB API. Meanwhile, oral bioavailability, C max and AUC, as evaluated in an in vivo pharmacokinetic study, also showed a notable enhancement.
