Zhenguang Liu,Haiyu Ni,Lin Yu,Shuwen Xu,Ruonan Bo,Tianxin Qiu,Pengfei Gu,TianYu Zhu,Jin He,Adelijiang Wusiman,Shaowu Zhu,Jiaguo Liu,Yuanliang Hu,Deyun Wang

Recently, the cubosomes have been widely studied as drug carriers. It has been described that cubosomes could further stimulate the immune response after carrying the immune enhancer. Polygonatum sibiricum polysaccharide (PSP), one of the most important biologically active ingredients of Polygonatum sibiricum , has been reported as an immunostimulant to improve immune responses. This study was aimed to observe the immunomodulation effects of ovalbumin (OVA) absorbed cetyltrimethylammonium bromide-modified Polygonatum sibiricum polysaccharide cubosomes (CTAB-modified PSP-Cubs/OVA). Firstly, the antigen uptake of CTAB-modified PSP-Cubs/OVA by macrophages was determined in vitro. After that, mice were immunized with CTAB-modified PSP-Cubs/OVA. The activation of dendritic cells in lymph nodes, activation of lymphocyte, ratios of CD4 + to CD8 + , the concentrations of OVA-specific IgG in serum and the cytokines concentrations were analyzed. As the results showed, CTAB-modified PSP-Cubs/OVA could promote the production of OVA-specific IgG in serum. The ratio of CD4 + to CD8 + in CTAB-modified PSP-Cubs/OVA group was significantly increased compared with other groups. CTAB-modified PSP-Cubs/OVA could significantly activate dendritic cells and promote lymphocyte proliferation. The results indicated that CTAB-modified PSP-Cubs/OVA could promote the secretion of related cytokines and the proliferation of lymphocytes, stimulate the cellular immune response and increase the level of humoral immunity. Above all, CTAB-modified PSP-Cubs had good adjuvant activity.

Sustainable ultrasound-assisted extraction of Polygonatum sibiricum saponins using ionic strength-responsive natural deep eutectic solvents

Hongli Zhang,Xinpeng Li,Miao Kang,Zhanrong Li,Xiaowen Wang,Xu Jing,Jiajun Han

The sustainable extraction of saponins was investigated using natural deep eutectic solvents (NADESs) combined with ultrasound-assisted extraction. A novel NADES (butyric acid-urea) that was responsive to ionic strength was designed and used as the extractant. Ultrasound treatment and a catalyst ferric chloride with plant cell wall breaking function were applied to improve the extraction efficiency.   Since the solubility of the NADES varied significantly with ionic strength, 95% of NADES was readily separated from the water phase after the addition of sodium chloride, while saponins remained in the water phase for easy collection. The reuse capacity of NADES, the eco-friendliness of the extraction method, and the antioxidant activity of the extract were further evaluated.   NADES was continuously recovered and used to extract Polygonatum sibiricum powder: the yield of saponins did not decrease after five cycles of recovery and re-extraction. The penalty point on the “Eco-scale” suggested that the extraction method was “green” ( i.e . eco-friendly).   Compared with ethanol extracts, the NADES extracts showed a higher saponin concentration and antioxidant activity.   The study can contribute to the sustainable and green extraction of hydrophilic active substances in the food and pharmaceutical industries.

Optimized acid hydrolysis conditions for better characterization the structure of inulin-type fructan from Polygonatum sibiricum

Xinyan Zong,Zhe Wang,Shikang Chen,Si Li,Mingyong Xie,Shaoping Nie,Junyi Yin

Polygonatum sibiricum is an edible plant species in China known for its abundant polysaccharides . However, correlations between its analytical methods and fine structure have not been established. This is usually due to incomplete cleavage of the glycosidic linkages and instability of hydrolysis . In this study, a new optimal acid hydrolysis method for monosaccharide composition (2 M H 2 SO 4 for 1 h) and methylation analysis (2 mol TFA hydrolysis at 100 °C for 1 h) was developed for characterization of inulin-type fructans , resulting in significantly improved monosaccharide recovery and providing more reliable methylation data. The effectiveness of this method was demonstrated through its application to the study of polysaccharide from P. sibiricum (IPS-70S). The results showed that IPS-70S with a molecular weight of 3.6 kDa is an inulin-type fructans consisting of fructose and glucose in a molar ratio of 27:1. Methylation and NMR analysis indicated that IPS-70S contains →2)-Fru f -(6 → or →2)-Fru f -(1 → with branching →1,6)-Fru f -(2 → and terminates in Glc p -(1 → or Fru f -(2→. In conclusion, optimal acid hydrolysis applicable to the specific polysaccharides contribute to its structurally characterized. The newly optimized acid hydrolysis method for monosaccharide composition and methylation analysis offers a reliable and effective approach to the structural characterization of inulin-type fructans from P. sibiricum . Providing reliable basis for the overall work of NMR analysis and structural analysis, which have potential significance in the field of polysaccharides structural characterization.

Determination of polysaccharides composition in Polygonatum sibiricum and Polygonatum odoratum by HPLC-FLD with pre-column derivatization

Sheng Xu,Jianli Bi,Wenfang Jin,Baolei Fan,Chunqi Qian

A high-performance liquid chromatography-fluorescence detection (HPLC-FLD) method was established for the determination of seven monosaccharides in Polygonatum sibiricum and Polygonatum odoratum . The polysaccharides were de-esterified, extracted, hydrolyzed and derivatized with p-aminobenzoic acid (PABA) to obtain fluorescently labeled monosaccharide compounds, which were finally detected by HPLC-FLD. Inertsil ODS-3, C18 chromatographic column (250 mm × 4.6 mm, 5 μm) was used for chromatography. The excitation wavelength (E x ) was 313 nm, and the emission wavelength (E m ) was 358 nm. Ethyl acetate extraction reduced the peaks of chromatogram and improved the detection sensitivity than other agents. The established method had high sensitivity, strong specificity, good linear relationship and recovery efficiency. The results showed that the roots and fibrous roots of Polygonatum sibiricum and Polygonatum odoratum contained these seven monosaccharides, and the highest monosaccharide content was mannose. The method of PABA-HPLC-FLD for determination of monosaccharide content in Polygonatum sibiricum and Polygonatum odoratum was sensitive and accurate. The method established in this work provides a feasible analytical tool for the study of polysaccharides, and the findings on polysaccharides from Polygonatum sibiricum and Polygonatum odoratum can provide guidance for the natural medicine industry.

Study on the components changes of polysaccharides and saponins during nine steaming and drying of Polygonatum sibiricum

Qihong Zhang,Xinyu Lin,Weike Su

BACKGROUND In this study, the content and structure of Polygonatum sibiricum polysaccharides and saponins during different processing stages were determined. RESULTS After processing of Polygonatum , the content of polysaccharide and glucose decreased, and the content of galactose, glucuronic acid and sugar substitution gradually increased. The content of total saponins increased significantly. Only 18 compounds were found in raw Polygonatum and 17 new compounds were presented in processed Polygonatum . During the processing of Polygonatum , the polysaccharide was partially degraded into oligosaccharides, the molecular weight gradually decreased, and the neutral sugar was converted into uronic acid, resulting in a decrease in polysaccharide content. The saponins were partially degraded into sapogenins or modified. CONCLUSION This study clarifies the changes in the content and structure of polysaccharides and saponins in processed Polygonatum , which will pave the way for elucidating the processing mechanism. © 2024 Society of Chemical Industry.

Polygonatum sibiricum polysaccharide prevents depression-like behaviors by reducing oxidative stress, inflammation, and cellular and synaptic damage

Fengming Shen,Zhujin Song,Pan Xie,Lei Li,Bin Wang,Daiyin Peng,Guoqi Zhu

Ethnopharmacological relevance According to traditional Chinese medicine (TCM) theory ( Yi Xue Zheng Zhuan ), the main factors associated with the pathogenesis of depression are deficiencies relating to five zang organs, Qi, and blood. Polygonatum sibiricum F. Delaroche (PS), which may avert these pathological changes, has been used in a variety of formulas to treat depression. However, the effects and mechanism of action of PS, alone, and especially those of its main active component PS polysaccharide (PSP), on depression remain unexplored. Aim of the study To determine the effects of PSP on depression-like behaviors and to elucidate its mechanism of action. Methods PSP was isolated from dried PS rhizomes and qualified using transmission electron microscopy and Fourier transform infrared spectroscopy . Lipopolysaccharide (LPS) and chronic unpredictable mild stress (CUMS)-induced depression models were used to evaluate the antidepressive effects of PSP. Veinal blood and brain tissue were collected to determine the levels of hippocampal 5-HT, serum cortisol (CORT), brain and serum cytokines, and hippocampal oxidation-related indicators. The protein expression levels of phosphorylated extracellular signal-regulated kinase (p-ERK1/2), nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB), glial fibrillary acidic protein (GFAP), phosphorylated protein kinase B (p-Akt), phosphorylation of the mammalian target of rapamycin (mTOR), caspase-3, GluA1 and GluA2, and GluN2A and GluN2B were determined using western blotting and immunofluorescence. Nissl staining was performed to detect histopathological changes in brain tissues. Results Injection of LPS ( i.p. ) induced depression-like behaviors, reduced the level of hippocampal 5-HT, increased the serum CORT level and hippocampal oxidative stress (ROS), and prompted the activation of ERK1/2, NF-κB, and GFAP and an inflammatory response. Conversely, PSP administration reduced these changes and prevented depression-like behaviors. PSP administration also promoted hippocampal expression of p-Akt, p-mTOR, GluA1, and GluA2; reduced the expression of caspase-3, GluN2A, and GluN2B; and prohibited the loss of granular cells in the DG region. Conclusion These results indicate that PSP prevents depression-like behaviors, and synaptic and neuronal damage probably by reducing ROS/HPA axis hyperfunction and the inflammatory response.

Efficient extraction of flavonoids from Polygonatum sibiricum using a deep eutectic solvent as a green extraction solvent

Hongli Zhang,Feilong Hao,Zhifang Yao,Junling Zhu,Xu Jing,Xiaowen Wang

Deep eutectic solvents (DESs) are becoming eco-friendly alternatives for extracting biologically-active substances from natural products. In this study, a DES is used as an eco-friendly solvent to extract flavonoids from Polygonatum sibiricum with the assistance of ultrasound. The antioxidant activity of the extracts was then investigated and the recyclability and reusability of the DES evaluated. Optimizing the DES extraction process produced the following optimum conditions: a betaine–acetic acid molar ratio of 1:4, water content of 30%, solid–liquid ratio of 1: 100 g mL −1 , extraction temperature of 50 °C, and extraction time of 30 min. The yield of extracted flavonoids reached 25.4 mg g −1 , which is about eight times that achieved using ethanol extraction. Furthermore, the antioxidant ability of the DES extract was more potent than that of the extract obtained using ethanol. Used DES can be efficiently recovered using ultrahigh capacitance porous activated carbon (the recovery yield of DES was around 92% of that of the original DES after ten reuses). In conclusion, DESs can be used to establish green ultrasound-assisted extraction methods for efficient extraction of plant flavonoids.

CO2-triggered switchable hydrophilicity solvent as a recyclable extractant for ultrasonic-assisted extraction of Polygonatum sibiricum polysaccharides

Jincui Chen,Yuan Xu,Zhijian Tan

In this study, CO 2 -triggered switchable hydrophilicity solvents (SHSs) based on different amines and water were employed in the ultrasonic-assisted extraction of Polygonatum sibiricum polysaccharides (PSPs). When CO 2 was pumped into the amine, the solution became hydrophilic and could be used as an extractant for PSPs extraction. When CO 2 was removed, the solution switched hydrophobicity, with the extractants being separated from the extract and further recycled and reused. The factors affecting the solid–liquid extraction of PSPs were studied. The maximum extraction yield of 399.2 mg/g was obtained at a solid–liquid ratio of 1:20, extraction time of 60 min, extraction temperature of 50 °C, and ultrasonic power of 500 W. SHSs could be recovered and reused with 88.4% recovery after the fifth cycle. The molecular weights and monosaccharide compositions of PSPs were also determined. This study provides a new strategy for sustainable extraction of plant polysaccharides and other bioactive ingredients.

Effects of different extraction methods on the structural characterization and bioactivities of polysaccharides extracted from Polygonatum sibiricum

Yang Mao-Hui,Zhou Xin,Yang Yan,Chen Hua-Guo

Polygonatum sibiricum is a traditional Chinese medicine with homology of medicine and food. Exploring different methods for extracting polysaccharides from Polygonatum sibiricum will provide broader insights into the study of its structural characteristics and biological activities. In this study, polysaccharides (PSPs) were extracted from by different methods with water (cold and hot), alcohol solutions (different concentrations) under ultrasound. The structure characterization of PSPs was mainly composed of Glc, Man and Gal with different molar ratio. HPLC data showed that the glycosidic bond of PSPs existed in α, β-D-configurations. Congo red experiment indicated that PSPs had similar triple helix structure. The PSPs extracted by different methods had different biological activities. PSP-5 had the best ability to inhibit AChE activity, and its mechanism was further explored. Results showed AChE inhibitory activity of PSP-5 was reversible, and the inhibitory mechanism was mixed type I competitive inhibition. Compared with the AChE inhibitors, PSP-5 had the potential to become an excellent AChE inhibitor.

Blood-Enriching Effects and Immune-Regulation Mechanism of Steam-Processed Polygonatum Sibiricum Polysaccharide in Blood Deficiency Syndrome Mice

Wang Juan,Wang Furong,Yuan Lixia,Ruan Hongsheng,Zhu Zhibiao,Fan Xiaoling,Zhu Lingyan,Peng Xin

Polygonatum sibiricum Red. has been used as a medicinal herb and nutritional food in traditional Chinese medicine for a long time. It must be processed prior to clinical use for safe and effective applications. However, the present studies mainly focused on crude Polygonatum sibiricum (PS). This study aimed to investigate the chemical properties, blood-enriching effects and mechanism of polysaccharide from the steam-processed Polygonatum sibiricum (SPS), which is a common form of PS in clinical applications. Instrumentation analyses and chemistry analyses revealed the structure of SPS polysaccharide (SPSP). A mice model of blood deficiency syndrome (BDS) was induced by acetylphenylhydrazine (APH) and cyclophosphamide (CTX). Blood routine test, spleen histopathological changes, serum cytokines, etc. were measured. The spleen transcriptome changes of BDS mice were detected by RNA sequencing (RNA-seq). The results showed that SPSP consists predominantly of Gal and GalA together with fewer amounts of Man, Glc, Ara, Rha and GlcN. It could significantly increase peripheral blood cells, restore the splenic trabecular structure, and reverse hematopoietic cytokines to normal levels. RNA-seq analysis showed that 122 differentially expressed genes (DEGs) were obtained after SPSP treatment. GO and KEGG analysis revealed that SPSP-regulated DEGs were mainly involved in hematopoiesis, immune regulation signaling pathways. The reliability of transcriptome profiling was validated by quantitative real-time PCR and Western blot, and the results indicated that the potential molecular mechanisms of the blood-enriching effects of SPSP might be associated with the regulating of JAK1-STAT1 pathway, and elevated the hematopoietic cytokines (EPO, G-CSF, TNF-α and IL-6). This work provides important information on the potential mechanisms of SPSP against BDS.

A New Method of Extracting Polygonatum sibiricum Polysaccharide with Antioxidant Function: Ultrasound-Assisted Extraction-Deep Eutectic Solvents Method

Chaoqun Sun,Guodong Wang,Jing Sun,Jiyong Yin,Jian Huang,Zizi Li,Di Mu,Menglu He,Tingting Liu,Jiali Cheng,Hanchen Du,Yan Chen,Wenjie Qu

Polygonatum sibiricumPolysaccharide (PsP) with antioxidant function is the main active component ofPolygonatum sibiricum(P.sibiricum). The currently poor extraction yield and extraction methods of PsP cannot meet the application of that in food industrial production. In this research, an ultrasound-assisted extraction-deep eutectic solvents (UAE-DESs) method, which has never been used in the PsP industry, was first used to extract PsP. The extraction conditions were optimized by the response surface method (RSM). Both the extraction yield and antioxidant function were simultaneously considered during the optimization process. The indicators of PsP’s level and antioxidant activity in vitro were used to present the extraction yield of the UAE-DESs method, the purity, and the antioxidant effect of PsP. Under the optimal conditions, which included a liquid–solid ratio of 26:1 (mL:g), extraction temperature of 80 °C, ultrasonic time of 51 min, and ultrasonic power of 82 W, the PsP extraction yield could reach (43.61 ± 0.09)%, which was obviously higher than single DESs (33.81%) and UAE (5.83%), respectively, and the PsP appeared favorably antioxidant function. This research proposed an efficient extraction method for PsP, filled the basic research gap, and further improved the development of PsP as a dietary supplement with antioxidant function in the food industry. Keywords:Polygonatum sibiricumPolysaccharide (PsP);ultrasound-assisted extraction-deep eutectic solvents (UAE-DESS);antioxidant function;response surface method (RSM)

Wenhao Xu,Jinli Yang,Xiangyang Gu,Wenjing Su,Faxiang Pu,Zhangfu Xie,Kongliang Jin,Weike Su,Lichan Mao

Ibuprofen (IBU) was a widely used NSAID (a type of nonsteroidal anti-inflammatory drug) worldwide, and many drug deliveries had been reported to enhance bioavailability. However, higher bioavailability would increase the danger of renal injury caused by oxidative stress. This study prepared IBU-Polygonatum sibiricum polysaccharide (IBU-PSP) drug delivery system via mechanochemical method. Due to drug delivery and renal protection effect of Polygonatum sibiricum polysaccharide (PSP), the solubility of IBU-PSP was increased 8.22 times, and the bioavailability was increased 2.52 times compared with IBU, carrageenin-induced rat paw edema test also increased. Meanwhile, short-term and long-term renal injuries induced by IBU were notable decreases. In conclusion, IBU-PSP was a multifunctional drug delivery system with superior anti-inflammatory and renal protection effects. It will benefit from developing high-efficiency NADIs preparations with safer clinical applications while providing an efficient and energy-saving technology for polysaccharide drug delivery.
