Multifunctional Prussian Blue Nanoparticles loading with Xuetongsu for efficient Rheumatoid Arthritis therapy through targeting inflammatory Macrophages and Osteoclasts

Yasi Deng,Bin Li,Hao Zheng,Ling Liang,Yupei Yang,Shiqi Liu,Mengyun Wang,Caiyun Peng,Bin Liu,Wei Wang,Huanghe Yu

Abnormal activation of macrophages and osteoclasts (OCs) contributes significantly to rheumatoid arthritis (RA) development by secretion of numerous inflammatory factors. Notably, these cells exhibit significant upregulation of folate receptor proteins on their surfaces. Unfortunately, there is a current lack of safe and effective therapeutic drugs for RA. Xuetongsu (XTS), a triterpenoid compound extracted from Kadsura heteroclita Roxb Craib, has demonstrated the ability to significantly inhibit the proliferation of RA fibroblast-like synoviocytes (RAFLS). However, its clinical application is hampered by poor targeting and short half-life. To address these drawbacks, we previously developed a nano-drug system named HRPS nanoparticles (NPs), which effectively targets RAFLS and inhibits synovial hyperplasia. However, this system overlooked the essential role of OCs in RA-related bone destruction. Therefore, we designed a novel folate-modified biomimetic Prussian blue (PB)-XTS NP (FMPX NP) for the selective delivery of XTS into inflammatory macrophages and OCs. The NP exhibits an excellent photothermal effect when assisted by laser irradiation, facilitating targeted release of XTS within inflammatory macrophages and OCs. The synergistic anti-inflammatory and reactive oxygen species scavenging effects of PB NPs and XTS are mediated by the inhibition of the NF-κB signaling pathway in inflammatory macrophages and RANK/RANKL/NFATc1 signaling pathway in OCs. In vivo experiments showed that FMPX NPs extended the half-life of XTS by 2.32 times, decreased hind foot swelling from 12.10 ± 0.49 to 8.24 ± 0.09 mm in the model group, and prevented bone damage. In conclusion, this study introduces a novel dual-targeted nano-based therapy for RA joints and highlights its potential for biochemical photothermal triple therapy for RA. FMPX NPs inhibit arthritis-related inflammation and bone destruction through a dual-target strategy, providing new insights for targeted drug therapies in clinical RA treatment.

Persulfate activation by low-rank coals for sulfamethazine degradation: Structural differences modulating activation pathways

Tianyuan Xu,Nanxi Zhu,Ruina Yuan,Yaowen Xing,Xiahui Gui

Carbon-driven persulfate (PS) activation is promising for antibiotic degradation, but conventional carbon catalysts often require thermal/acid-base modification or metal doping, suffering from low yields and high costs. Herein, we innovatively employed unmodified low-rank coals from Shengdong (LC SD ) and Sujiagou (LC SJG ) as cost-effective PS activators for sulfamethazine (SMZ) degradation. Characterizations including Raman, 13 C nuclear magnetic resonance, and electron paramagnetic resonance (EPR) revealed distinct structural-chemical features. LC SJG had more structural defects, –OH, and –COOH groups, while LC SD showed higher persistent free radical and C O group concentrations. LC SJG exhibited superior performance (99.3 % SMZ removal in 120 min) versus LC SD (84.2 %) and good tolerance to coexisting ions, indicating practical application potential. Possible degradation pathways of SMZ, as revealed by high-performance liquid chromatography-mass spectrometry, were proposed involving sulfonamide cleavage and pyrimidine ring opening. Mechanistic investigations via EPR, radical quenching, and electrochemical analysis revealed that 1 O 2 dominated SMZ degradation (59.3 %) in the LC SD /PS system via C O groups. Conversely, • OH and SO 4 •− contributed 62.7 % to degradation in the LC SJG /PS system through structural defects and –COOH groups. Overall, this study provides a novel strategy for developing low-cost, high-performance PS activators tailored for wastewater treatment.

Study on the slurry ability of low-rank coal blended with alkali-modified sludge

Xiaofang You,Wenqi Zhang,Xiaoteng Li,Chuandong Ma,Zhisen Li,Qingbiao Wang,Zhenhua Wang,Lin Li

In this study, alkali-modified municipal sludge (AS) was mixed with low-rank coal to prepare AS-coal water slurry (ASCWS). The influence of AS on the slurrying performance of coal water slurry (CWS) was investigated, and the slurrying mechanism of municipal sludge-CWS (SCWS) was elucidated. Research on the alkali modification of municipal sludge showed that the effectiveness of the modifiers was in the order Ca(OH) 2  > NaOH > KOH > K 2 CO 3 . Considering the apparent viscosity of the slurry as the evaluation criterion, the optimal alkali modification scheme was determined through the response surface optimization test, and 19.99 % Ca(OH) 2 was suitable to modify the sludge for 14.61 h Compared with the unmodified sludge, the content of oxygen-containing functional groups on the surface of AS was reduced, and the flocculent structure was disrupted, which helped improve the slurry formation performance of the SCWS. Blending the sludge with the CWS enhanced the stability of the slurry, and the water separation rates for the SCWS and ASCWS decreased from 18.62 % to 5.00 % and 5.26 %, respectively. The mechanism study showed that the zeta potential values of SCWS and ASCWS decreased from −10.67 mV to –32.23 mV and −39.22 mV, respectively. The increased electronegativity of the slurry owing to the addition of sludge facilitated particle dispersion and enhanced slurry stability. The adsorption patterns of sodium poly[(naphthaleneformaldehyde)sulfonate] (MF) on coal, sludge, and modified sludge were consistent with the Langmuir adsorption model. The pseudo-second-order kinetic model accurately described the adsorption process of MF on coal, sludge, and modified sludge.

Pyrolysis and catalytic upgrading of low-rank coal using a NiO/MgO–Al2O3 catalyst

Yi Li,Muhammad Nadeem Amin,Xingmei Lu,Chunshan Li,Fuqiang Ren,Suojiang Zhang

The catalyst NiO/MgO–Al 2 O 3 was prepared by loading magnesium and nickel onto the surface of γ-Al 2 O 3 beads, respectively, and was adapted to in situ upgrade the high temperature pyrolysis vapor derived from coal pyrolysis. The catalyst synthesis process, and structural variation were characterized by the XRD, SEM, and physical adsorption analyzer. In order to examine the catalytic effect on the different nickel loading ratio in the catalysts, the solid–liquid–gas product distribution, variation of gas and components of coal tar were studied by simulated distillation, GC/MS, elemental analysis and so on. The results shows both the tar yield and the content of the light fraction in the tar can be increased by catalytic up grade. The catalyst has a strong ability to convert the heavy component and polycyclic aromatic hydrocarbons in the tar to lower molecular weight components.

The antiresoptive effects of recombinant Lingzhi-8 protein against retinoic acid-induced osteopenia

Lihong Ruan,Ningning Jiang,Fengyang Guo,Hui Xu,Jingmin Zhang,Jingchun Sun

The antiresorptive agents still are the mainstay of osteoporosis treatment. This study aimed to investigate the efficacy of recombinant Lingzhi-8 (rLZ-8) on osteoclast in vitro and bone resorption in vivo . The rLZ-8 protein was derived from Ganoderma lucidum transformation and produced by a genetic system. Receptor activator of nuclear factor kappa-Β ligand induced RAW 264.7 cells to differentiate into osteoclastic cells in vitro . Cells were exposed to different doses of rLZ-8 for 7 days to measure differences of osteoclastic differentiation, apoptosis rate and gene expression. rLZ-8 was labeled with Alexa Fluor 568 to observe its intracellular distribution under super-resolution light microscopy. In addition, retinoic acid was administered to female rats for 14 days to develop osteopenia changes. Different doses of rLZ-8 were simultaneously administered to rats treated with retinoic acid to observe changes of bone mineral density, biochemical parameters and organ weight ratio. Results indicated that rLZ-8 regulated receptor activator of nuclear factor kappa-Β (RANK) - tumor necrosis factor receptor-associated factor 6 (TRAF6) - c-Jun N-terminal kinase (JNK) signaling pathway, by which rLZ-8 inhibited osteoclastic differentiation and promoted osteoclastic apoptosis. Through 3D-structured illumination microscopy, it was observed that rLZ-8 entered RAW264.7 cells and accumulated gradually into the cytoplasm but little into nucleus. Administration with rLZ-8 reversed loss of bone mass and improved ALP activity in osteoporotic rats. Low-to high-dose rLZ-8 treatments displayed little toxic effects on rat organs and did not seem to impact their overall health. All data suggested that rLZ-8 has possible action against osteoporosis.

Synergistic optimization of flotation reagent formulation for low-rank oxidized coal using Box-Behnken design

Wenlong Meng,Hanbo Zhang,Yulong Cheng,Xiahui Gui,Yaowen Xing

The surface of low-rank oxidized coal is rich in oxygen-containing functional groups, making it highly hydrophilic and difficult to achieve efficient flotation using traditional oil-based collectors. Aiming to achieve optimal flotation reagent regulation, this study employed Box-Behnken design combined with response surface methodology (RSM) to establish a second-order regression model for combustible recovery in low-rank oxidized coal flotation, systematically investigating the synergistic effects between oxygenated polar collectors and frothers. Flotation experiments were conducted to examine the impacts of three critical parameters: polar/nonpolar component ratio in collector, collector dosage, and frother dosage on combustible recovery. Experimental results demonstrated that under optimal reagent conditions (40.60% polar component in collector, 1500.00 g/t collector dosage, and 602.44 g/t frother dosage), the combustible recovery reached 94.47%, closely matching the model prediction of 95.27%. Mechanistic analysis revealed that polar components in the collector selectively anchored onto oxygen-containing sites on coal surfaces through hydrogen bonding, while nonpolar components formed hydrophobic oil films, synergistically reducing coal-water interfacial energy. The interaction between frother and collector further stabilized bubbles and enhanced particle-bubble adhesion. This study provides theoretical guidance and technical support for the efficient and clean utilization of low-rank oxidized coal.

Comparison of sliding time of low-rank coal particles on air and oily bubble surfaces

Shiwei Wang,Xiuxiang Tao

In the present communication, the sliding time of low-rank coal particles on air and oily bubble surfaces was investigated. From the analysis of a high-speed camera, it was found that the sliding time of hydrophilic low-rank coal particles attached to air bubble was limited by the maximum angle of contact at about 61º whereas the maximum touching angle for particles attached to oily bubble was about 85º. Meanwhile, the sliding time of particles attached to the oily bubbles was always lower than that of particles attached to air bubbles during the slip process. Moreover, due to the Marangoni effect on the rising oily bubble surface, half of the sliding time of low-rank coal particles passing from about 3º to the bottom of the oily bubble was more than that of particles sliding from about 85º to the bottom of the oily bubble. Furthermore, based on the same hydrodynamic environment, the difference in the sliding time of low-rank coal particles attached to air and oily bubble surfaces was not the same as the difference in induction time between air or oily bubble and particle beds.

Influence of complex flotation reagents on flotation effect of low-rank coal and mechanism analysis

Weiwei Xie,Zidong Zhang,Lijuan Bie,Yaning Wang,Yifan Yu,Jiaxuan An,Lingmei Zhou

This study investigates the surface characteristics of Yongshengyuan (YSY) low-rank coal, a material known for its flotation challenges, employing a suite of analytical techniques such as Zeta potential analysis, atomic force microscopy (AFM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction phase analysis (XRD), and X-ray photoelectron spectroscopy (XPS). The flotation performance of low-rank coal was evaluated using three distinct surfactants (sodium petroleum sulfonate, hexadecyl dimethyl ammonium bromide, methyl oleate) in conjunction with 3# oil (base oil). The experimental results demonstrate that all surfactants enhance the flotation efficiency of low-rank coal, with the combination of sodium petroleum sulfonate and methyl oleate with 3# oil proving most effective. Comprehensive analyses of AFM, Zeta potential, contact angle and wetting heat of coal sample pre and post-application of the compound reagent, reveal that the surfactant preferentially interacts with the polar hydrophilic sites on the YSY low-rank coal surface. This interaction diminishes the presence of oxygen-containing functional groups on the coal surface, increases the ratio of hydrophobic groups, enhances the coal’s hydrophobicity, and consequently bolsters the flotation efficiency of the low-rank coal.

Strengthen mechanism of low-rank coal flotation: Analysis of interaction force and contact line behavior

Chunyun Zhu,Jun Chen,Yali Cheng,Fanfei Min,Guosheng Li,Yaowen Xing,Xiahui Gui

Interaction forces between oil and coal surfaces were analyzed by AFM with oil-probe. • The mechanism between polar/non-polar oil and coal surfaces was elucidated. • The forces and contact line between bubble and modified coal surface were analyzed. • A method based interaction force and coal property analysis can enhance flotation. The challenge in intensifying coal flotation is the precise regulation of interfacial hydrophobicity. In this study, coal models with controllable surface properties were employed to investigate the intensification mechanism of collectors, using atomic force microscopy (AFM) with oil droplet probes, adhesion force measurement, and flotation kinetics analysis. AFM results revealed that nonpolar oil exhibited “jump-in” adhesion exclusively on hydrophobic coal model surfaces, whereas polar oil could adhere to weakly hydrophobic surface. Not only the surface forces, but also the hydrodynamic forces influenced the collector droplet-particle interaction, increasing the repulsion to hinder the adhesion behavior. Adhesion tests demonstrated that the heterogeneous reagent adsorption induced localized pinning/recession of the contact line between bubbles and particles, thereby influencing adhesion strength. Flotation kinetics indicated that collectors with varying chain lengths achieved optimal performance at a nonpolar-to-polar ratio of 2:1. Notably, the optimal polar content (33.33%) in composite collectors exhibited a correlation with the coal surface’s polar composition (40.48%). Important theoretical insights for the accurate management of low-quality coal separation interfaces are provided by this study. pH-responsive HMDS terminated m-Cresol for inhibiting the spontaneous combustion in different-rank coals

Ao Gao,Wei Lu,Yuntao Liang,Zhenglong He,Jinliang Li,Yong Sun,Shuanglin Song,Xinfeng Cui,Xiang Gao

In this study, pH responsiveness of 1, 1, 1, 3, 3, 3-hexamethyldisilazane terminated m-Cresol (HTMC) in different-rank coals was investigated, and its inhibition performance regarding the spontaneous combustion of coal was explored. First, color changes in the deprotection reaction of HTMC under different acidic conditions were analyzed. Then, surface microstructures of these coals and pH values of their filtrates were tested. Subsequently, experiments were performed on different coal samples for grasping their low-temperature oxidation characteristics, weight loss rates, pore characteristics, and free radical variations. The experimental results show that HTMC can effectively protect phenolic hydroxyl groups in inhibitors, and the acidity of coal can promote the conversion rate of its deprotection reaction. For instance, in XAM coal (pH ≈ 6), sufficient H + stimulates HTMC to release massive m-Cresol which contains highly reactive phenolic hydroxyl, thereby effectively suppressing increases of the concentration and type of free radicals. The crossing point temperature (177.2 °C) and inhibition rate at 110 °C (75.9 %) of the HTMC-inhibited XAM sample are 33.1 °C and 17.9 % higher than those of the m-Cresol-inhibited one, respectively. This study confirms the applicability of pH-responsive HTMC to different-rank coals and may lay a theoretical foundation for the design of efficient reactive inhibitors.

Sophorolipid-enhanced methane production from low-rank coal: metagenomic insights into microbial and metabolic mechanisms

Wang Yaya,Di Zhiting,Akimbekov Nuraly S.,Liu Xiangrong,Tastambek Kuanysh,He Shihua,Zhou Anning,Bao Yuan

Anaerobic digestion (AD) represents a promising approach for enhancing coal resource utilization and advancing sustainable development within the coal industry. This study investigates the role of sophorolipid biosurfactants in enhancing methane production during coal AD. Sophorolipid supplementation significantly increased methane yield by reshaping the microbial community, enriching key bacteria ( Proteiniphilum , Petrimonas ) and methanogens ( Methanobacterium , Methanosarcina ). Metagenomic analysis revealed concurrent upregulation of glycolytic genes and critical pathways for pyruvate-to-acetate conversion ( aceE , PDH , ALDH ) and CO₂ reduction ( fdh , fdo ), driving methanogenesis through both acetoclastic and hydrogenotrophic pathways. Mechanistically, sophorolipid enhanced microbial membrane permeability and upregulated the TolC gene of the bacterial type I secretion system, facilitating extracellular enzyme release and coal depolymerization. These biological effects were complemented by physicochemical changes, including reduced liquid surface tension and increased ζ-potential, which improved microbe–enzyme–coal interactions. Fourier transform infrared spectroscopy and gas chromatography–mass spectrometry confirmed the degradation of aromatic and aliphatic hydrocarbons. Collectively, sophorolipid biosurfactants enhance coal biodegradation efficiency through synergistic microbial, genetic, and interfacial mechanisms, offering a promising strategy for cleaner coal conversion.

Low-temperature pre-carbonization modulation of the surface properties and floatability of low-rank coal

Yutong Xie,Zongxu Yao,Qun Wei,Yinshuang Guan,Zhiqiang Tang,Wei Jiang,Tianqi Xu,Zhijian Wang,Enhui Zhou,Chenlong Duan,Liang Dong

Low-rank coal (LRC) exhibits significant potential as a precursor for coal-based carbon materials due to its high chemical reactivity. However, achieving high-value utilization depends on the ultra-clean preparation of LRC. In this study, a combined process of low-temperature pre-carbonization and flotation is proposed to obtain ultra-clean coal. The evolution of surface chemical composition, morphology, pore structure, and wettability at different pre-carbonization temperatures was investigated, and the preparation mechanism was further explored. The results demonstrate that pre-carbonization effectively reduces oxygen-containing functional groups and enhances surface hydrophobicity. A critical transition occurs at 250 °C, where decarboxylation reduces the O = C-O content to 16.43% and increases the contact angle from 112.4º to 123.5º. Flotation tests on the 250 °C pre-carbonized sample yielded ultra-clean coal with an ash content of 0.81% and a yield of 40.36%. Calculations based on E-DLVO theory confirm that the enhanced flotation performance results from strengthened hydrophobic attraction and weakened electrostatic repulsion between coal particles and bubbles. This study elucidates the mechanism by which low-temperature pre-carbonization promotes flotation efficiency, offering a novel strategy for yielding ultra-clean coal with high application value.
