Ratiometric Electrochemical Determination of Ascorbic Acid Using a Copper Nanoparticle@Resin Nanosphere (CuNPs@RNS) Modified Glassy Carbon Electrode (GCE) by Differential Pulse Voltammetry (DPV)

Jie Zhang,Yulin Fu,Lin Li,Liqiang Yan,Xiongzhi Wu,Chenghong Lei

Ascorbic acid (AA) determination is of high importance in the diagnosis and treatment of diseases. Herein, we report a new ratiometric electrochemical sensor for ascorbic acid using a glassy carbon electrode (GCE) modified with a copper nanoparticle@resin nanosphere nanocomposite (CuNPs@RNS). The ratiometric strategy was established by immobilizing an internal reference (thionine) on the modified electrode using cyclic voltammetry (CV). The performance of the modified electrodes as well as the newly established ratiometric strategy was explored. The sensing platform had good electrocatalytic ability, reproducibility, and stability. In addition, the ratiometric strategy significantly improved the performance of electrochemical sensing with a wide linear range (0.0837 to 15.5 μM) and a low detection limit of 0.0279 μM under the optimal conditions. The original ratiometric electrochemical sensor was successfully applied to monitor ascorbic acid in tablets and urine. These results showed that the sensor provides a new strategy and broad prospects for biomolecular sensing with reliability and high sensitivity.

Establishment of a Customizable Fluorescent Probe Platform for the Organelle-Targeted Bioactive Species Detection

Dan Qiao,Landie Li,Tangliang Shen,Jiejie Yang,Hao Chang,Xiao Liang,Lu Zhang,Qianqian Wang,Ning Liu,Wei Zhao,Luqing Shang

A customizable fluorescent probe platform that can be used to detect various bioactive analytes offers significant potential for engineering a wide range of bioprobes with diverse sensing and imaging functions. Here, we show a facile and innovative strategy for introducing cis-amino-proline as a carrier scaffold, which is appended with three specific functional groups: a target group, a water-soluble group, and fluorophores with triggers. The potency of the designed strategy could be customized to generate variable multifunctional fluorescent probes for detecting bioactive species of interest, including reactive oxygen species (ROS), reactive nitrogen species (RNS), reactive sulfur species (RSS), ROS/RSS, and even enzymes. We designed and synthesized five representative water-soluble and organelle-targeted compounds, PMB, PMN, PMD, PRB, and PME, with emission wavelengths of these fluorescent probes varying from blue to red (465, 480, 535, 550, 565, and 640 nm). This strategy could be exemplified by its application to develop a mitochondria-/lysosome-targeting multifunctional fluorescent probe capable of imaging bioactive species of interest in live cells and nude mice.

Mitochondrial impairment and downregulation of Drp1 phosphorylation underlie the antiproliferative and proapoptotic effects of alantolactone on oral squamous cell carcinoma cells

Zhang Yafei,Yang Bingqian,Tu Chengwei,Ping Yifan,Chen Shuhong,Wu Tong,Zhao Zheyu,Mao Yixin,Yang Zhan,Cao Zelin,Li Jianmin,Huang Kate,Ding Xi,Wu Gang,Zou Peng,Deng Zhennan,Sun Xiaoyu

Background Oral squamous cell carcinoma (OSCC) is one of the most prevalent and fatal oral cancers. Mitochondria-targeting therapies represent promising strategies against various cancers, but their applications in treating OSCC are limited. Alantolactone (ALT) possesses anticancer properties and also regulates mitochondrial events. In this study, we explored the effects of ALT on OSCC and the related mechanisms. Methods The OSCC cells were treated with varying concentrations and duration of ALT and N -Acetyl- l -cysteine (NAC). The cell viability and colony formation were assessed. The apoptotic rate was evaluated by flow cytometry with Annexin V-FITC/PI double staining. We used DCFH-DA and flow cytometry to detect reactive oxygen species (ROS) production and DAF-FM DA to investigate reactive nitrogen species (RNS) level. Mitochondrial function was reflected by mitochondrial reactive oxygen species (ROS), mitochondrial membrane potential (MMP), and ATP levels. KEGG enrichment analyses determined the mitochondrial-related hub genes involved in OSCC progression. Dynamin-related protein 1 (Drp1) overexpression plasmids were further transfected into the cells to analyze the role of Drp1 in OSCC progression. Immunohistochemistry staining and western blot verified the expression of the protein. Results ALT exerted anti-proliferative and pro-apoptosis effects on OSCC cells. Mechanistically, ALT elicited cell injury by promoting ROS production, mitochondrial membrane depolarization, and ATP depletion, which were reversed by NAC. Bioinformatics analysis showed that Drp1 played a crucial role in OSCC progression. OSCC patients with low Drp1 expression had a higher survival rate. The OSCC cancer tissues presented higher phosphorylated-Drp1 and Drp1 levels than the normal tissues. The results further showed that ALT suppressed Drp1 phosphorylation in OSCC cells. Moreover, Drp1 overexpression abolished the reduced Drp1 phosphorylation by ALT and promoted the cell viability of ALT-treated cells. Drp1 overexpression also reversed the mitochondrial dysfunction induced by ALT, with decreased ROS production, and increased mitochondrial membrane potential and ATP level. Conclusions ALT inhibited proliferation and promoted apoptosis of oral squamous cell carcinoma cells via impairment of mitochondrial homeostasis and regulation of Drp1. The results provide a solid basis for ALT as a therapeutic candidate for treating OSCC, with Drp1 being a novel therapeutic target in treating OSCC.

A Groundbreaking Electric Field-Induced Cascade Gas Therapy Against Large Volume Solid Tumor Through Electro-Stress Storm

Gui Chen,Wenjia Zhang,Manchun Wang,Fengling Zhang,Mengliang Zhu,Yan Tang,Yixian Xie,Wen Ma,Peter Timashev,Massimo Bottini,Yingqiu Xie,Xing-Jie Liang,Meng Yu,Zhiqiang Yu

Gas therapy has been limited in its application as a robust standalone antitumor strategy due to the restricted gas production and cytotoxicity. To address this challenge, we employed electrotoxic PtRu composite metal nano-berries (PR) loaded with various therapeutic gas donors to construct a groundbreaking electric field-induced cascade gas therapy (EGT) platform, which generated a great electro-stress storm at tumor sites, exerting electrotoxicity and immunity functions against solid tumors, including those of large volume, through three pathways. Initially, electric field stimulation effectively boosted the release rate and yield of therapeutic gases from the EGT platform. Further, gas molecules reacted with reactive oxygen species (ROS) to either form oxidation coordination (CO and ROS) or generate more potent therapeutic components (RNS produced from ROS and NO), contributing to an electro-stress storm that augmented the cytotoxic potential of the gas components. Subsequently, this electro-stress storm further activated the tumor immune response, identifying and capturing escaped tumor cells, which held significant implications for treating tumors, including non-solid tumors with indistinct boundaries. In summary, the EGT platform leveraged an electro-stress storm to achieve ablation of large volume solid tumors and suppressed metastatic tumors, paving new pathways for gas-based therapeutic strategies.

2D MoSe2@PVP nanosheets with multi-enzyme activity alleviate the acute pancreatitis via scavenging the reactive oxygen and nitrogen species

Liying Zhang,Pei Xie,Hang Wu,Jiulong Zhao,Shige Wang

Acute pancreatitis (AP) is a disease with high mortality which could cause an inflammatory responses such as edema and necrosis of the pancreatic tissue. Studies have proved that the excessive produced reactive oxygen species (ROS) and reactive nitrogen species (RNS) play an important role in the pathogenesis of AP. Although progresses have been made for ROS and RNS elimination using artificial enzymes, biocompatible nanomaterials with advanced multi-enzymatic activities have been rarely studied. In this study, we constructed a simple and efficient method to fabricate a two-dimensional (2D) MoSe 2 @PVP nanosheets (NSs) as an artificial enzyme to treat oxidative stress-related diseases. Both in vitro and in vivo experiments proved that MoSe 2 @PVP NSs have good biocompatibility. Interestingly, the MoSe 2 @PVP NSs simultaneously showed the thermal durable peroxidase, catalase, superoxide dismutase, and glutathione peroxidase mimicking capacities, which endowed the material with good ROS and RNS scavenging abilities. The MoSe 2 @PVP NSs were engaged in treating the AP in an animal model, which led to a significant down-regulation of serum levels of amylase, IL-6, IL-1β, and TNF-α. Further H&E staining of pancreatic tissue validated the AP therapeutic effect of MoSe 2 @PVP NSs. We hope that our research will facilitate the development of multifunctional artificial enzymes for the treating of oxidative stress-related diseases.

Ultrasound-driven radical chain reaction and immunoregulation of piezoelectric-based hybrid coating for treating implant infection

Menglin Sun,Jiameng Wang,Xiaobo Huang,Ruiqiang Hang,Peide Han,Jiqiang Guo,Xiaohong Yao,Paul K. Chu,Xiangyu Zhang

The poor efficiency of US-responsive coatings on implants restricts their practical application. Immunotherapy that stimulates immune cells to enhance their antibacterial activity is expected to synergize with sonodynamic therapy for treating implant infection effectively and safely. Herein, US-responsive hybrid coatings composed of the oxygen-deficient BaTiO 3 nanorod arrays and l -arginine (BaTiO 3-x /LA) are designed and prepared on titanium implants for sonocatalytic therapy-cooperated immunotherapy to treat Methicillin-resistant Staphylococcus aureus (MRSA) infection. BaTiO 3-x /LA can generate more oxidizing reactive oxygen species (ROS, hydroxyl radical (·OH)) and reactive nitrogen species (RNS, peroxynitrite anion (ONOO − )). The construction of nanorod arrays and oxygen defects balances the piezoelectric properties and sonocatalytic capability during US treatment. The generated piezoelectric electric field provides a sufficient driving force to separate electrons and holes, and the oxygen defects attenuate the electron-hole recombination efficiency, consequently increasing the yield of ROS during the US treatment. Moreover, nitric oxide (NO) released by l -arginine reacts with the superoxide radical (·O 2 − ) to produce ONOO − . Since, this radical chain reaction improves the oxidizing ability between bacteria and radicals, the cell membrane (argB, secA2) and DNA (dnaBGXN) are destroyed. The bacterial self-repair mechanism indirectly accelerates bacterial death based on the transcriptome analysis. In addition to participating in the radical chain reaction, NO positively affects macrophage M1 polarization to yield potent phagocytosis to MRSA. As a result, without introducing an extra sonosensitizer, BaTiO 3-x /LA exhibits excellent antibacterial activity against MRSA after the US treatment for 15 min. Furthermore, BaTiO 3-x /LA facilitates macrophage M2 polarization after implantation and improves osteogenic differentiation. The combined effects of sonodynamic therapy and immunoregulation lead to an effective and safe treatment method for implant-associated infections.

Constructing multifunctional Cu Single-Atom nanozyme for synergistic nanocatalytic Therapy-Mediated Multidrug-Resistant bacteria infected wound healing

Xiaochen Qiu,Liang Zhuang,Jian Yuan,Huizhen Wang,Xiaoyu Dong,Shan He,Shanyue Guan,Zhiyue Chang,Pengtao Bao

Developing an effective strategy to combat multi-drug-resistant (MDR) bacteria and promote wound healing without overuse of antibiotics remains an important and challenging goal. Herein, we established a synergistic reactive oxygen species (ROS) and reactive nitrogen species (RNS)-mediated nanocatalytic therapy, which was consisted of a multifunctional Cu single-atom nanozyme loaded with the l -arginine ( l -Arg@Cu-SAzymes) and a low level of hydrogen peroxide (H 2 O 2 ) as a trigger. l -Arg@Cu-SAzymes can possess excellent dual enzyme-like activities: catalase (CAT)-like activity that decompose H 2 O 2 into O 2 , and subsequent oxidase (OXD)-like activity that convert O 2 to cytotoxic superoxide anion radical (•O 2 − ). Meanwhile, l -Arg@Cu-SAzymes can also be triggered by H 2 O 2 to release nitric oxide (NO), which can continue to react with •O 2 – to generate more lethal peroxynitrite (ONOO − ). Collectively, the synergistic ROS and RNS mediated by l -Arg@Cu-SAzymes endow the treatment system with an outstanding antibacterial ability against MDR bacteria and reduce the inflammation at the wound site. Furthermore, l -Arg@Cu-SAzymes-mediated NO and O 2 release promote the cell proliferation , collagen synthesis, and the angiogenesis , as well as facilitate macrophage polarization to reparative M2 phenotype, thereby accelerating wound closure and tissue remodeling. Therefore, l -Arg@Cu-SAzymes-based synergistic nanocatalytic therapy can be regarded as a promising strategy for MDR bacterial infected wounds treatment, owing to their potent antibacterial efficacy and enhanced tissue remodeling effects.

Alendronate-Modified Nanoceria with Multiantioxidant Enzyme-Mimetic Activity for Reactive Oxygen Species/Reactive Nitrogen Species Scavenging from Cigarette Smoke

Xia Zhou,Weinan Zeng,Shu Rong,Heng Lv,Yonghong Chen,Yinghua Mao,Weilong Tan,Hong Li

Highly toxic radicals including reactive oxygen species (ROS) and reactive nitrogen species (RNS) in cigarette smoke play an important role in oxidative damage of the lungs, which cannot be efficiently scavenged by current filter techniques. Herein, a novel alendronate-coated nanoceria (CeAL) nanozyme is explored for cigarette filter modification for ROS/RNS scavenging. The CeAL nanozyme with an adjustable oxidation state and high thermal stability exhibits an excellent superoxide dismutase (SOD)-like activity, hydroxyl radical elimination capacity, catalase-mimicking activity, and nitric oxide radical scavenging ability. These synergistic antioxidant abilities make the CeAL nanozyme a lucrative additive for cigarette filters. The filter incorporated with the CeAL nanozyme can efficiently scavenge ROS/RNS in the hot smoke generated by burned commercial cigarettes, resulting in reduction of oxidative stress-induced pulmonary injury and acute inflammation of mice. The developed CeAL nanozyme opens up new opportunities for cigarette filter modification to decrease the toxicity of cigarette smoke and expands the application fields of nanoceria.
